GLC를 이용 N-TFA n-butyl ester유도제화에 의한 아미노산 분석시 stainless steel packed column으로 효과적인 분리를 할 수 있는 고정상을 찾기 위하여 다음과 같은 2개의 column을 개발하여 표준 아미노산의 N-TFA n-butyl ester 유도체를 분석하였다. Column 1;3.36% OV-17+3.0% SE-30 on chromosorb W HP 100-120 mesh. Stainless steel column $(2m{\times}3mm\;I.D.)$ Column 2;1% NPGS+0.5% OV-17+0.5% SE-30 on chromosorb W HP 100-120 mesh. Stainless steel column $(2m{\times}3mm\;I.D.)$ Column 1에서 histidine을 제외한 19개의 아미노산 peak가 나타났으나 alanine과 valine peak는 분리되지 않았다. Colume 2에서는 15개의 아미노산이 좋은 분리를 이루었고 column 1에서 분리되지 않은 alanine과 valine.이 잘 분리되었으나 column 1에서 분리된 tryptophan, arginine 및 tyrosine이 나타나지 않았다. 분리된 모든 아미노산은 column 1, 2를 막론하고 아미노산 주입량 $1.25{\times}10^{-3}{\mu}mole-1.0{\times}10^{-2}{\mu}mole$의 범위에서 각 아미노산 peak의 높이와 내부표시제 peak 높이와의 비에 대한 주입량으로 나타낸 검정선이 선형을 나타내었고 모두 원점을 지났다. Glutamine, asparagine 및 cystine은 column 1,2에서 모두 glutamic acid, aspartic acid 및 cysteine peak와 일치하는 것으로 보아 N-TFA n-butyl ester 유도체를 만드는 과정에서 염산에 의해 각각 glutamic acid, aspartic acid 및 cysteine으로 전환되는 것으로 사료된다.
Gas chromatography (GC) separation technology and metal oxide semiconductor (MOS) gas sensors have been integrated for the effective analysis of volatile sulfur compounds (VSCs) such as H2S, CH3SH, (CH3)2S, and (CH3)2S2. The separation and detection characteristics of the GC/MOS system using diluted standard gases were investigated for the qualitative and quantitative analysis of VSCs. The typical concentrations of the standard gases were 0.1, 0.5, 1.0, 5.0, and 10.0 ppm. The GC/MOS system successfully separated H2S, CH3SH, (CH3)2S, and (CH3)2S2 using a celite-filled column. The reproducibility of the retention time measurements was at a 3% relative standard deviation level, and the correlation coefficient (R2) for the VSC concentration was greater than 0.99. In addition, the chromatograms of single and mixed gases were almost identical.
Triglyceride는 구성지방산의 종류에 따라 분자종이 다양하게 존재하여 식품의 가공, 저장중에 있어서 유지의 물성변화 및 산화특성을 효과적으로 규명하기 위해서는 구성 triglyceride 분자종의 상세한 해석이 요구된다고 하겠다. 율무 지질의 triglyceride를 reverse phase HPLC UV detector 210nm에서 분석한 결과 6개의 peak로 분획되었다. 각 획분을 GC로 acycl 탄소수별로 분획하고, 다시 DEGS column을 이용하여 fatty acid 조성을 분석하였다. 이 세가지 결과로부터 triglyceride 조성을 산정한 결과 11개의 TG 조성을 추산하였으며, 그 중 주요 triglyceride로는 C18 : 1 C18 : 2 C18 : 2(OLL)이 38.2%, C16 : 0 C18 : 2 C18 : 3(PLLn)이 15.7%, C18 : 1 C18 : 1 C18 : 2(OOL)이 12.6%였다. 한편 capillary GC와 GC-MS로 같은 시료를 분석하여 분자종을 확인하는 작업을 계속하고 있으며 위의 결과와 비교하여 다음에 보고하고자 한다.
A gas chromatographic (GC) method was developed to determine residues of captan, folpet, captafol, and chlorothalonil, known as broad-spectrum protective fungicides for the official purpose. All the fungicide residues were extracted with acetone containing 3% phosphoric acid from representative samples of five agricultural products which comprised rice, soybean, apple, pepper, and cabbage. The extract was diluted with saline, and dichloromethane partition was followed to recover the fungicides from the aqueous phase. Florisil column chromatography was additionally employed for final cleanup of the extracts. The analytes were then determined by gas chromatography using a DB-1 capillary column with electron capture detection. Reproducibility in quantitation was largely enhanced by minimization of adsorption or thermal degradation of analytes during GLC analysis. Mean recoveries generated from each crop sample fortified at two levels in triplicate ranged from 89.0~113.7%. Relative standard deviations (RSD) were all less than 10%, irrespective sample types and fortification levels. As no interference was found in any samples, limit of quantitation (LOQ) was estimated to be 0.008 mg/kg for the analytes except showing higher sensitivity of 0.002 mg/kg for chlorothalonil. GC/Mass spectrometric method using selected-ion monitoring technique was also provided to confirm the suspected residues. The proposed method was reproducible and sensitive enough to determine the residues of captan, folpet, captafol, and chlorothalonil in agricultural commodities for routine analysis.
분석 대상인 24종 농약을 극성이 다른 캐필러리 칼럼인 DB-17, SPB-608, 및 Ultra-2를 이용하여 전자적압력조절장치(Electronic pressure control, EPC)가 장착된 HP 5890 series II Plus 가스크로마토그래피에서 전자포획검출기로 동시분석법의 최적조건을 모색하였다. 본 실험에서 EPC 시스템의 압력조절기능을 도입하므로써 다음과 같은 분석향상을 얻었다. DB-17, SPB-608, 및 Ultra-2 캐필러리 칼럼에서 EPC 기능없이 동시 분석한 결과 일부 농약들이 분리되지 않았다. 그러나 EPC 시스템의 압력조절 기능을 적용한 결과 vinclozolin과 acetochlor를 제외한 22종 농약 모두 SPB-608 캐필러리 칼럼에서 동시 분석되었다. 또한 농약들의 분리 시간에도 영향을 주어 총 분석시간이 단축되었다. 즉 EPC 시스템 없이 분석한 시간이 $61{\sim}81$분인데 반해 EPC 시스템도입으로 45분으로 단축되었다. 각 농약들에 대한 검출한계는 $0.1{\sim}12.9$ ng/mL 검출되었다.
Essential oil components from tobacco powder were investigated as flavouring agent. The essential oil was isolated from tobacco powder by a simple distillation /extraction method The extracted essential oil was fractionated into basic, acidic and neutral groups. And the neutral group of essential oil was separated by column chromatography into 10 fractions. Above groups and fractions were tested for tobacco aroma and smoke aroma. The neutral group except most nonpolar fraction displays good flavouring properties which make them highly suitable for improving the flavour and aroma of tobacco and tobacco smoke. The most nonpolar fraction from neutral group was carefully investigated using preparative column, thin layer and gas chromatography ailed by GC/MS coupling. The major subfraction was identified as hydrocarbons on the basis of the IR spectrum. The 58 hydrocarbon components were identified by their mass spectra and was chromatographic retention times.
산${\cdot}$염기용매분배법과 규소산 칼럼 크로마토그라피법 및 용융 실리카모세관 칼럼크로마토그라피, 질량분석법을 사용하며 셀룰로오스와 리그닌의 열분해생성물중의 여러고리 방향족 화합물의 분리와 동정을 수행하였다. 16가지의 여러고리 방향족 화합물이 머무른 계수와 질량스펙트럼데이타에 의해서 확인되었다. 두가지 물질 모두에서 같은 종류의 여러고리 방향족 화합물이 생성되었으며, 그 함량에 있어서는 리그닌에서 훨씬 많은량이 생성되었다. 극히 발암성인 질소 및 유황을 함유하는 헤테로 고리 방향족 화합물은 거의 생성되지 않았다.
An analytical method was investigated for the meaiiurement of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) concentrations in air samples. Procedures required for column chromatographic clean up. silicagel (stage I) and gel permeation chromatography (stage II), were discussed. Identification and quantification of PCBs and OCPs were performed using a combination of gas chromatography/mass spectrometry/ selected ion monitoring. Recovery tests calculated from six samples are 68∼137% for PCBs and 58∼130% for OCPs except for endrin aldehyde. Instrumental detection limits determined for the PCBs and OCPs varied from 0.05 to 0.18 pg/m3 and from 0.71 to 16.82 pg/㎥, respectively. The method has been applied to the analysis of air samples collected at Ansung city, Kyonggi province, Korea. This method may serve as a screening protocol for the simultaneous determination of PCBs and OCPs in air.
An analytical method was developed to determine monocrotophos residues in apple, citrus, and soil using high-performance liquid chromatography (HPLC) with ultraviolet absorption detection. Monocrotophos was extracted with acetone from apple, citrus and moist soil samples. The extract was concentrated, added with saline water, and subjected to n-hexane washing to remove nonpolar co-extractives. Dichloromethane partition was then followed to recover monocrotophos from the aqueous phase. Silica gel column chromatography was employed to further purify the extract prior to HPLC determination. Reverse-phase HPLC using an oct-adecylsilyl column was successfully applied to separate and quantitate the monocrotophos residue in sample extracts at the wavelength of 230 nm. Overall recoveries of monocrotophos from fortified samples averaged $95.3{\pm}2.1%$ (n=6), $970{\pm}0.7%$ (n=6), and $92.8{\pm}4.3%$ (n=12) for apple, citrus, and soil, respectively. The proposed method was quite reproducible and sensitive enough to replace the troublesome gas-liquid chromatographic analysis for monocrotophos residues.
본 연구는 프탈산 에스텔류에 대하여 생물시료 중에 존재하는 가장 일반적인 9종의 분석을 기체크로마토그래피-질량분석기-선택이온검색법에 의해 수행하였다. 생물시료는 유기용매로 추출하고 Florisil 컬럼을 사용하여 정제하였다. 프탈산 에스텔류는 전처리 과정 중 가장 쉽게 오염되는 물질로 생물시료의 경우 전처리 단계가 수질이나 저질시료보다 복잡해지므로 각 전처리 단계의 오염정도를 비교 분석하였다. 표준물질이 첨가된 생물시료에 본 연구에서 개발된 분석법을 적용하여 구한 회수율은 79 - 117%정도로 나타났다. 또한 프탈산 에스텔류의 정량의 정확도, Florisil 컬럼 용리패턴, 검출한계 등을 조사하였다.
