Quality Changes in Fresh Wild Garlic Leaves Treated with Different Washing Solutions and Packaging Films during Storage (세척수 및 소포장 필름에 따른 산마늘의 저장 중 품질변화)

  • 산마늘은 마늘과 같이 독특한 향이 있어 입맛을 돋우며 웰빙식품으로 3월 중순에서 5월초에 채취하여 생채로 쌍을 먹거나 김치, 절임으로 생산되어 판매된다. 산마늘의 부가가치 향상을 위하여 살균소독 세척과 소포장용 필름포장방법을 구명하고자 본 실험을 수행하였다. 산마늘을 살균소독 세척수(수돗물, 0.2% 구연산 침지 후 50% 에탄올 분무처리)로 2분 세척하고 원심탈수 후, 포장필름은 $30{\mu}m$ oriented polypropylene (OPP), $30{\mu}m$ OPP 필름을 기본재료로 산소투과율 $5,000cc/m^2{\cdot}day{\cdot}atm$ (OPP_5)로 조정한 필름, $30{\mu}m$ micro perforated polypropylene (MPP) 필름에 밀봉하고, $30{\mu}m$ OPP필름에 개봉 포장하여 $4^{\circ}C$에서 42일 저장하였다. 산마늘은 구연산 침지 후 에탄올 분무세척 후 세척 전 보다 일반세균수가 1.5 log CFU/g 감소되었으며 수돗물 세척 후는 0.1 log CFU/g 정도 감소하여 수돗물 보다 구연산 침지 후 에탄올 분무에 의해 균수 감소효과가 보다 컸으며 저장기간이 경과한 후에도 그러한 경향은 유지되었다. 저장 35일 후 세척수에 관계없이 OPP 개봉과 MPP 밀봉 포장에서 의해 산마늘의 황화율과 부패율이 높게 나타났으며 OPP 밀봉에서 이취 발생 지수가 OPP_5 밀봉보다 높게 나타났다. 세척 및 포장처리에 의한 종합선도의 제한요인은 황화, 이취 및 부패취 발생 지수로 이를 종합하였을 때 산마늘의 상품성 향상을 위하여 소포장 하는 경우 구연산 침지 후 에탄올 분무 방법으로 세척한 다음 원심탈수하여 OPP_5필름으로 포장하는 것이 품질 향상에 적합하였으며 40일 이상 저장이 가능한 것으로 나타났다.

Identification of Novel Source of Resistance and Differential Response of Allium Genotypes to Purple Blotch Pathogen, Alternaria porri (Ellis) Ciferri

  • Purple blotch, caused by Alternaria porri (Ellis) Cifferi, is a serious disease incurring heavy yield losses in the bulb and seed crop of onion and garlic worldwide. There is an immediate need for identification of effective resistance sources for use in host resistance breeding. A total of 43 Allium genotypes were screened for purple blotch resistance under field conditions. Allium cepa accession 'CBT-Ac77' and cultivar 'Arka Kalyan' were observed to be highly resistant. In vitro inoculation of a selected set of genotypes with A. porri, revealed that 7 days after inoculation was suitable to observe the disease severity. In vitro screening of 43 genotypes for resistance to A. porri revealed two resistant lines. An additional 14 genotypes showed consistent moderate resistance in the field as well as in vitro evaluations. Among the related Allium species, A. schoenoprasum and A. roylei showed the least disease index and can be used for interspecific hybridization with cultivated onion. Differential reaction analysis of three A. porri isolates (Apo-Chiplima, Apn-Nasik, Apg-Guntur) in 43 genotypes revealed significant variation among the evaluated Allium species (P = 0.001). All together, the present study suggest that, the newly identified resistance sources can be used as potential donors for ongoing purple blotch resistance breeding program in India.

Ability of Lipophilic Extract Obtained from Plants to Inhibit Tyrosinase Activity in Reverse Micelles (역미셀계를 이용한 지용성 식물체 추출물의 tyrosinase 저해효과 분석)

  • The abilities of petroleum ether-extracts prepared from 75 plants to inhibit tyrosinase activity were evaluated in reverse micelles composed of isooctane/AOT(100 mM)/phosphate buffer(20 mM, pH 8.0) containing tyrosinase(105.3 units/mL) and 3,4-dihydroxyphenylalanine(0.18 mM). Compared with control which has no plant extracts, garlic could completely inhibit in vitro melanogenesis by tyrosinase, and Chinese quince, sweet potato, onion, radish bud and apple did more than 60%. Lipophilic extracts of medicinal plants and herbs such as rosemary, coriander, cinnamomi ramulus, crataegii fructus, ramulus biotae folium, mume fructus, menthae herba, eucommiae cortex and clove also inhibited tyrosinase activity more than 60%. When the extraction yield of lipophilic materials was considered together with their inhibition effect on tyrosinase, it was possible to select plants of which tyrosinase inhibitors could be produced in high quantity from unit weight. Using reverse micelles, the analysis of the capacity of lipophilic materials to inhibit tyrosinase activity which was difficult up to present could be possible.

Tobamovirus Coat Protein CPCg Induces an HR-like Response in Sensitive Tobacco Plants

  • When inoculated into sensitive tobacco Xanthi-nn plants, the crucifer and garlic-infecting Tobacco mosaic virus (TMV-Cg) induces local necrotic lesions that resemble those seen in the hypersensitive response (HR) of resistant tobacco plants. However, unlike these, tobacco Xanthi-nn plants do not become resistant to infection and the virus spreads systemically causing a severe disease characterized by necrotic lesions throughout the plant. To identify the viral protein that elicits this necrotic response, we used a set of hybrid viruses constructed by combination of TMV-Cg and the tobacco mosaic virus strain U1 (TMV-U1). In this study we present evidence that the coat protein of TMV-Cg (CPCg) is the elicitor of the necrotic response in tobacco Xanthi-nn plants. Local and systemic necrotic lesions induced by TMV-Cg and by the hybrid U1-CPCg -that carries CPCg in a TMV-U1 context- are characterized by cell death and by the presence of autoflorescent phenolic compounds and $H_2O_2$, just like the HR lesions. In addition, defense-related genes and detoxifying genes are induced in tobacco Xanthi-nn plants after TMV-Cg and U1-CPCg inoculation. We postulate that in our system, CPCg is recognized by sensitive tobacco plants that mount an incomplete defense response. We call this an HR-like since it is not enough to induce plant resistance.

Identification of Suitable Natural Inhibitor against Influenza A (H1N1) Neuraminidase Protein by Molecular Docking

  • The influenza A (H1N1) virus, also known as swine flu is a leading cause of morbidity and mortality since 2009. There is a need to explore novel anti-viral drugs for overcoming the epidemics. Traditionally, different plant extracts of garlic, ginger, kalmegh, ajwain, green tea, turmeric, menthe, tulsi, etc. have been used as hopeful source of prevention and treatment of human influenza. The H1N1 virus contains an important glycoprotein, known as neuraminidase (NA) that is mainly responsible for initiation of viral infection and is essential for the life cycle of H1N1. It is responsible for sialic acid cleavage from glycans of the infected cell. We employed amino acid sequence of H1N1 NA to predict the tertiary structure using Phyre2 server and validated using ProCheck, ProSA, ProQ, and ERRAT server. Further, the modelled structure was docked with thirteen natural compounds of plant origin using AutoDock4.2. Most of the natural compounds showed effective inhibitory activity against H1N1 NA in binding condition. This study also highlights interaction of these natural inhibitors with amino residues of NA protein. Furthermore, among 13 natural compounds, theaflavin, found in green tea, was observed to inhibit H1N1 NA proteins strongly supported by lowest docking energy. Hence, it may be of interest to consider theaflavin for further in vitro and in vivo evaluation.

Effect of Low Temperature Treatment of Seed Bulb and Planting Date on Plant Growth and Yield in Garlic (마늘의 파종기별(播種期別) 저온처리(低溫處理)의 차이(差異)가 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • In order to develop a cropping system that can produce garlic in the period of short supply from March to April, effects of low temperature treatment of seed bulbs and planting dates, starting date of low temperature treatment, days of low temperature treatment on plant growth, maturity and yield were studied in Southern strain, 'Namhae' and in Northern strain, 'Euiseong' of garlic (Allium sativum). The results obtained were as follows. In Sorthern strain, sprouting was significantly enhanced by low temperature treatment only in Sep. 14, and Sep. 29 plantings. Days to sprout were least in 30 days of low temperature treatment of Sep. 14 planting and in 45 days treatment of Sep. 29 planting. When considering on the beginning date of low temperature treatment, a marked difference was observed between treatments started before July 31 and after Aug. 15. Sprouting was most enhanced in 45 days low temperature treatment of Aug. 15 and Aug. 30 plantings. In Northern strain, sprouting was en hanced by low temperature treatment in planting from Sep. 29 to Nov. 13 and low temperature treatment for 60 days was most effective. Effect of low temperature treatment on early plant growth was observed in Sep. 14 and Sep. 29 plantings, but the effect on plant growth at intermediate stage or thereafter was observed in up to Oct. 29 plantings. Optimun days for low temperature treatment on growth enhancement was 45 and 60 days in Southern strain and 60 days in Northern strain in each planting dates. In Southern strain, the longer the low temperature treatment and the later the planting date the less the number of leaves developed. In Northern strain, normal leaves were not developed in plantings from Sep. 14 to Nov. 13. In Southern strain, clove differentiation and bulbing were earlist in 45 and 60 days treatment of Sep. 14, Sep. 29, and Oct. 14 planting initiated on July 31 and Aug. 15. In Northern strain, clove differentiation and bulbing were earlist in 60 days treatment of Oct. 14 planting initiated on Aug. 15 and Aug. 30. In treatment initiated later than above, longer the low temperature treatment the earlier the clove differentiation and bulbing in both Southern and Northern strains. The earlier the initiation date and the longer of low temperature treatment, the earlier bolting in southern strain. In Northern strain, bolting was most enhanced in 45 and 60 days of low temperature treatment initiated on Aug. 15 and Aug. 30. The longer the low temperature treatment in plantings thereafter, the earlier the bolting. The earlier the planting date garlic bulbs. Harvest date was earliest in 45 and 60 days low temperature treatment started from July 31 to Aug. 30 in Southern strain, and it was in 60 and 90 days low temperature treatment initiated from July 31 to Aug. 30 in Northern strain. Bulb weight was heaviest in 45 days low temperature treatment of Oct. 14 planting and next was in 45 days treatment of Sep. 29 planting in Southern strain. In Northern strain, bulb weight was heaviest in 60 days treatment of Oct. 14 planting and next was in 45 days treatment of Oct. 14 planting. When considered in the aspect of the beginning date of low temperature treatment, bulb weight was heaviest in 45 days treatment started on Aug. 30 in Southern strain and in 60 days treatment started on Aug. 15 in Northern strain. A high negative correlation between days to harvest and plant height on January 12, and a high positive correlation between days to harvest and days clove differentiation were observed. This indicates that enhanced plant growth and clove differentiation induced by low temperature treatment advanced the harvest date. A high negative correlation between bulb weight and days to clove differentiation, days to harvest suggests that the enhanced clove differentiation result and in heavier bulb weight. From the above results, it suggested that early crop of garlic can be harvested by planting at the period of Sep. 29 to Oct. 14 after 45 days of low temperature treatment of seed bulbs of Southern strain. Then harvest date can be shortened by 30 days compared to control and garlic can be harvested in early April.

Physiological Activity of Medicinal Plant Extracts (생약재 추출물의 생리활성 효과)

  • The physiological activity of 5 kinds of medicinal plant extracts were examined. Total polyphenol contents showed the highest value in 50 $\%$ ethanol extracts. Electron donating ability showed the highest value in 50 $\%$ ethanol extracts of Glycyrrhiza uralensis. SOD-like activity showed the highest activity in 50$\%$ ethanol extracts of Glycyrrhiza uralensis. Medicinal plant extracts showed the different nitrite scavenging abilities under the different pH conditions. Medicinal plant extracts showed the highest value in nitrite scavenging ability at pH 1.2. The maximum nitrite scavenging effect was found at pH 1.2 and decreased as pH increased. Angiotensin-converting enzyme inhibitory activity showed the highest value in 50 $\%$ ethanol extracts of dried garlic.

Effect of Plant Extracts with Superoxide Dismutase-like Activity on Survival of Fruit Flies under Oxidative Stress (Superoxide Dismutase유사활성을 지닌 식물체가 Oxidative Stress를 받고 있는 초파리의 수명에 미치는 영향)

  • Biological effect of aqueous extracts of 12 plants which showed superoxide dismutase (SOD)-like activity in vitro was evaluated using Drosophila melanogaster. Survival percentage of the flies was a criterion of effect when the flies were exported to paraquat, which generated superoxide anion radical in vivo. When flies were co-administered with paraquat and aqueous extracts of garlic, lettuce, kiwi, and nameko, they showed no defensive effect against of oxygen toxicity. If flies were exposed to 60 mM paraquat after adaptation to feed containing plant extracts with SOD-like activity for 10 days, however, survival percentage of flies fed with phytochemicals was $35{\sim}63%$ while that percentage of flies fed without phytochemical was only 11%. This result indicated that adaptation of flies to plant extracts with SOD-like activity could prevent the flies from oxidative injury. On the other hand, lettuce, kiwi, nameko, onion, persimmon, fern brake and cauliflower showed a reparative effect on an oxidative stress. Dropwort, shiitake, agaric mushroom and broccoli did not show such an effect.

Effect of Supplemental Medicinal Plants(Artemisia, Acanthopanax and Garlic) on Growth Performance and Serum Characteristics in Lactating Sows, Suckling and Weanling Pigs (사료내 약용식물(인진쑥, 오가피 및 마늘)의 첨가가 포유돈, 포유자돈 및 이유자돈의 성적 및 혈청특성에 미치는 효과)

  • In Exp. 1, a total of forty eight multiparous sows were used in a 21-d performance assay. All litters were standardized at eleven piglets within 24h of parturition. Sows were fed one of the four experimental diets 1) CON(basal diet; Control), 2) MP1(basal diet added 0.05% of medicinal plant mixtures), 3) MP2 (basal diet added 0.1% of medicinal plant mixtures) and 4) MP3(basal diet added 0.2% of medicinal plant mixtures). Backfat thickness difference from farrowing to weaning was tended to increase in CON treatment compared to those of medicinal plants mixture(MP) treatments. However, there were not significant differences among the treatments(P>0.05). Sow’s ADFI was increased in sows fed MP2 treatment compared to MP3 treatment(P<0.05). Piglet weight gain was tended to increase in MP1 treatment compared to CON treatment(P<0.05). The glucose concentration of MP3 treatment was higher that of CON treatment(P<0.05). Exp. 2, a total of one hundred twenty weaning pigs ($L{\time}Y{\time}D$, 4.70$\pm$0.63kg average initial body weight) were used in a 20-d performance assay. Weanling pigs were fed treatments diets included 1) CON(basal diet; Control), 2) MP0.05(basal diet added 0.05% of medicinal plant mixtures), 3) MP0.1(basal diet added 0.1% of medicinal plant mixtures) and 4) MP0.15(basal diet added 0.15% of medicinal plant mixtures). Through entire experimental period, as MP increased in the diets, there was an increase in ADG(linear, P<0.067), improvement gain/feed(linear, P<0.018) and a decrease in ADFI (linear, P<0.008). DM digestibility was significantly increased with addition of MP(linear, P<0.004; quadratic, P<0.030). In conclusion, sows fed MP2 had showed less body weight loss and increased weight gain for suckling piglet. In weaning pigs, as supplementation MP from 0.05 to 0.1% in diet had improved growth performance and DM digestibility.

Anti-Cancer Effect of the Combination of Thiacremonone and Docetaxel by Inactivation of NF-κB in Human Cancer Cells

  • Thiacremonone, the main component isolated from heated garlic (Allium sativum L.), is interested for using as a cancer preventive or therapeutic agent since garlic has been known to be useful plant in the treatment of cancers. Nuclear factor kappaB (NF-${\kappa}B$) is constitutively activated in the prostate cancer and activation of NF-${\kappa}B$ is implicated in drug resistance in cancer cells. Docetaxel, a semisynthetic analog of paclitaxel, is an antineoplastic drug widely used for advanced various cancer. In previous studies, we found that thiacremonone inhibited activation of NF-${\kappa}B$ in cancer cells and marcrophages. In the present study, we investigated whether thiacremonone could increase susceptibility of prostate cancer cells (PC-3 and DU145) to docetaxel via inactivation of NF-${\kappa}B$. We found that the combination treatment of thiacremonone (50 ${\mu}g$/ml) with docetaxel (5 nM) was more effective in the inhibition of prostate cancer cell growth and induction of apoptosis accompanied with the significant inhibition of NF-${\kappa}B$ activity than those by the treatment of thiacremonone or docetaxel alone. It was also found that NF-${\kappa}B$ target gene expression of Bax, caspase-3 and caspase-9 was much more significantly enhanced, but the expression of Bcl-2 was also much more significantly inhibited by the combination treatment. These results indicate that thiacremonone inhibits NF-${\kappa}B$, and enhances the susceptibility of prostate cancer cells to docetaxel. Thus, thiacremonone could be useful as an adjuvant anti-cancer agent.
