Control of Powdery Mildew by Garlic Oil in Cucumber and Tomato (마늘오일을 이용한 오이와 토마토 흰가루병 방제)

  • The effectiveness of four plant oils (garlic, ginger, cinnamon and lemongrass) against a range of plant pathogenic organisms was tested in vitro. Of the four oils, two oils (garlic and cinnamon) showed relatively good antimicrobial activities. Activity of garlic oil was shown against the plant pathogenic bacteria Agrobacterium tumefaciens, Ewinia carotovora, Ralstonia solanacearum, Xanthomonas campestris, and the fungi Phytophthora infestans, Fusarium oxysporum, Collectotricum sp., whereas Acidovorax avenae and Pythium sp. showed the resistance towards garlic oil. Results from the planta bioassays under greenhouse conditions indicated that garlic oil significantly reduced the cucumber powdery mildew (Sphaerotheca fusca) and tomato powdery mildew (Erysiphe cichoracearum) showing control value 70.0-74.6% and 71.2%, respectively.

Management of Tomato Root-knot Nematode Meloidogyne incognita by Plant Extracts and Essential Oils

  • The effect of plant extracts of eucalyptus (Eucalyptus chamadulonsis), garlic (Allium sativium), marigold (Tagetes erecta) and neem (Azadirachta indica) and essential oils were tested on the suppression of root-knot nematode Meloidogyne incognita under greenhouse and field conditions. In vitro study, all tested treatments had nematicidal effect on nematode juveniles after 24 and 48 hours from exposures. The highest percentage of nematode mortality was achieved by application of neem extract (65.4%), essential oils (64.4%) and marigold extract (60.5%), followed by garlic and eucalyptus extracts (38.7-39.5%). Under greenhouse and field conditions, neem extract and essential oils treatments were more effective in reducing population numbers of the M. incognita in soil and root gall index compared to other treatments. In field experiments, the maximum protection of tomato plant against root-knot nematode was obtained by application of neem and essential oil treatments, 44.2 and 32.6%, respectively.

  • Antimicrobial activity of garlic extracts of four local cultivars (Namdo, Goheung, Danyang and garlic imported from China) against several plant pathogenic bacteria and fungi was investigated. The garlic extracts exhibited marked in vitro antimicrobial activities against the Gram-positive and -negative bacteria as well as fungal pathogens. Of the four cultivars, two cultivars (Namdo and garlic imported from China) showed relatively good antimicrobial activities. Gram-positive bacteria, Clavibater michiganensis was most sensitive, whereas Burkholderia cepacia was the strong resistant towards garlic extracts. Results from the planta bioassays under greenhouse conditions indicated that garlic extracts significantly reduced the cucumber powdery mildew disease showing control value 59.7-61.9% compared with the nontreated controls.

Incidence of Viral Infection of Main Garlic Growing Areas of Gangwon Province in Korea (강원도 마늘 주산지 재배마늘의 바이러스 감염 실태)

  • This research was conducted to investigate the incidence of viral infection in Samcheok-shi and Jeongseon-gun, main garlic growing areas of Gangwon province in Korea. Four primers sets (GLV, LYSV, GCLV, OYDV) were used for RT-PCR to test of 100 samples collected from each location. Infection rates of GLV, LYSV, GCLV and OYDV of garlic samples from Samcheok-shi and Jeongseon-gun were 95, 95, 92 and 33%, respectively. All garlic samples tested in this research were infected at least one virus. Coinfection of 3 kinds of virus (GLV, LYSV and GCLV) and 4 kinds of virus (GLV, LYSV, GCLV and OYDV) were 60 and 25%, respectively. While infections of GLV, LYSV and GCLV were evenly found in both two regions, OYDV infection of garlic samples collected from Jeongseon-gun was higher than Samcheok-shi. Viral infections at garlic fields of whole country and Gangwon province are very serious now. Therefore, it is very necessary to develop technic for virus-free bulb and renew seed bulb of garlic.

Correlations between Soil Environment and Bioactive Constituents of Namdo Garlic Harvested in the Non-volcanic Ash Soil Distributed Western Jeju (제주 서부지역 비화산회토양에서 수확한 남도마늘의 기능성 성분 함량 및 토양환경과의 상관관계 분석)

  • In this study, we determined the ingredient analysis of harvested garlic bulb and soil analysis of four garlic-cultivated regions in Jeju, being one of the major areas of Namdo garlic production. Soil pH and electric conductivity were 7.02 and 1.03 dS/m, respectively. Soil organic matter was 4.31%. The mineral elements (potassium, calcium, magnesium, sodium, iron, manganese, copper and zinc) of Namdo garlic cultivated soil were analyzed by ICP, and calcium was the most highly contained mineral with $14.67cmol_+/kg$ and in the decreasing order of magnesium ($2.25cmol_+/kg$), potassium ($1.51cmol_+/kg$). Soluble solid and total acidity were 7.60 oBrix and 0.49%, respectively. The mineral contents of garlic bulb were in order of potassium (12,728 ppm) > sulfur (7,778 ppm) > phosphorus (4,916 ppm) > magnesium (691 ppm) > calcium (359 ppm). The content of total phenolic, total flavonoid and reducing sugar were 71.14 mg GAE/100 g, 17.64 mg QE/100 g and 26.53 mg GE/g, respectively. Alliin and allicin were 8.78 mg/g and 2.10 mg/g, respectively. The Pearson's correlation coefficients between mineral contents of soil and garlic bulb are analyzed. Macronutrients of soil is correlated with macronutrients of garlic (positive) and micronutrients of garlic (negative) contents.

  • We, Tong Yang Moolsan Co. Ltd. (TYM) set up the mass-production system for virus-free seed garlic via tissue culture technique. TYM's tissue culture technique is called as 'Multiple shoot propagation technique'. This technique can lead mass propagation of genetically homogeneous seed garlic in a short period because of its highly proliferation rate of in vitro shoots ($15^{10}$ /year). TYM researchers applied the technique to some selected garlic cultivars with superior characteristics and carried out field test of productivity in the inside and outside of the country for several years. According to the yearly results of field test with virus-free seed garlic, we ascertained that virus-free seed garlic can produce the highly yield increase (max. above $50\%$) and also can enhance the product quality. Consequently, we estimated that TYM's seed garlic will contribute to farmers with increase of income and can elevate the national position of garlic market in the world for its competitive power of technical and production cost.

The Chemical Basis of Green Pigment Formation ('Greening') in Crushed Garlic (Allium sativum L.) Cloves

  • The chemical processes involved in the formation of green pigment in crushed garlic cloves were investigated based on the principle of pink pigmentation in macerated onions. Intact greening and non-greening garlic cloves were either left untreated or heated at $90^{\circ}C$ for 3 min to inactivate enzyme activities. First, a colorless ether soluble compound referred to as color developer reacted with glycine (among all free amino acids) in garlic to form a second compound insoluble in ether. The latter compound then reacted with formaldehyde to yield the green colored pigment. Alliinase activity was necessary for the production of color developer and for the development of green pigment. In greening garlic that had been heat treated, green pigmentation did not proceed due to the heat-inactivation of alliinase, but the addition of alliinase solution into the garlic homogenates restored the pigmentation. However, this phenomenon was not observed in non-greening garlic with or without heat treatment. Finally, the mechanism of green pigment formation in crushed garlicis similar to that of pink pigment formation in macerated onions.

Biological Control of Garlic White Rot Accused by Sclereotium cepivorum and Sclereotium sp. Using Bacillus subtilis 122 and Trichoderma harzianum 23 (Bacillus subtilis 122와 Trichoderma harzianum 23에 의한 마늘 흑색썩음균핵병의 생물적 방제)

  • Trichoderma harzianum 23 WP and Bacillus subtilis 122 WP were formulated as antagonists of Sclerotium cepivorum and Sclerotium sp. of garlic white rot. In the field test, applications of Trichoderma harzianum WP and Bacillus subtilis WP reduced garlic white rot by Sclerotium cepivrum from 10.9% in the control to 4.1% and 6.2%, respectively at Taean. Also at Seosan, applications of Trichoderma harzianum 23 WP and Bacillus subtilis 122 WP reduced garlic white rot by Sclerotium sp. from 17.8% in the control to 1.2% and 2.6%, respectively. Treatment of Trichoderma harzianum 23 WP and Bacillus subtilis 122 WP increased garlic yield in two area. Therefore, Trichoderma harzianum 23 WP and Bacillus subtilis 122 WP have shown potential as biofungicides of garlic white rot in the two different pathogens.

Screening for Resistance of Garlic Cultivars to White Rot Caused by Sclerotium cepivorum (Sclerotirum cepivorum에 대한 마늘 재배종의 저항성 검정)

  • The optimal quantity of inoculum was determined to screen resistance of garlic cultivars against Sclerotium cepivorum and 30 cultivars was tested. The growth of the pathogen in detached roots, Disease incidence was increased when the inoculum density was raised form 10 to 100 sclerotia. The optimal inoculum density to differentiate resistance or susceptibility of garlic cultivars was seemed to be 50 sclerotia. The cultivars collected from England, Japan, Nepal and Turkey, and cultivars such as common red, PI1356104 and PI135693 were less than the other cultivars in their disease incidence. The growth of S. cepivorum in detached roots varied from 23 to 33 mm according to garlic cultivars. There was no relationship between the disease incidence and the growth in detached roots. The sclerotial germination was increased significantly when root extract was extract was added. The addition of only distilled water resulted in 13% germination, but the addition of 0.25 g of root extract in 100 ml distilled water resulted in more than 85% germination. There was no difference in the stimulation of sclerotial germination among cultivars which showed different resistance.

  • To determine the effect of garlic on acute liver damage caused by $CCl_4$, the values of GOT, GPT, LDH in blood were measured. GOT, GPT, LDH values from mouse treated 2,000mg/kg garlic substance powder were significantly lower than disitlled mouse(positive treatment) after distilled water ingestion only showed big death, but 2,000mg/kg garlic powder treated mouse rarely showed death of liver tissue.
