Identification of Gamma Irradiation of Imported Spice

  • Photostimulated Luminescence (PSL), Electron Spin Resonance (ESR) and Thermoluminescence (TL) analysis were conducted to detect irradiation treatment of imported whole and ground spices. The screening by PSL detected no irradiation treatment, except un the ground thyme and bay leaves which exhibited photon counts in the intermediate level. Irradiation of the two spices was detected after irradiating them at 1.0, 3.0, 5.0 and 10.0 kGy, and then subjecting them to PSL analysis, which resulted in the significantly low photons of non-irradiated spices compared to that at 1.0 kGy, indicating that the photon counts varied depending on the amount of inorganic mineral debris in the spices. To confirm a successful detection by using PSL, ESR and TL methods, some spices were selected, irradiated at 5.0 and 10.0 kGy, and subjected to the detection methods. PSL identified the irradiated spices except the cassia, which showed very weak PSL sensitivity, but was identified by ESR analysis. Also, the ESR and TL exhibited the typical signals induced by irradiation treatment and were able to successfully detect all of the irradiated spices. In addition, we found a positive correlation between the intensity of ESR and TL signals and irradiation doses.

감마선 조사한 총목피(Aralia elata Cortex) 추출물의 생리활성 (Biological Activities of Extracts from Gamma-irradiated Aralia elata Cortex)

  • 천연 약용식물에 있어서 감마선 조사 처리는 기능성 물질탐색에 적용 시 추출수율 증진, 색상 개선 등 산업적 응용을 용이하게 하는 방법으로 활용이 기대되고 있다. 본 연구에서는 감마선 조사가 총목피 추출물의 생리활성에 미치는 영향을 검토하였다. 감마선 조사 처리한 총목피의 감마선 조사여부를 확인하기 위하여 광자극 발광(PSL) 및 열 발광(TL) 방법을 활용하여 감마선 조사 판별 확인시험을 진행하였다. PSL법의 경우 비조사구 0 kGy에서 400.00 PCs로 음성(negative) 시료로 판단되었으며, 조사구 5, 10, 30 kGy에서 각각 90,100.00 PCs, 312,614.33 PCs, 321,661.67 PCs로 나타나 감마선을 조사한 양성(positive) 시료로 확인되었다. TL법 적용 시 비조사구 0 kGy에서 자연방사선에 의한 $300^{\circ}C$ 부근에서 조사구 5, 10, 30 kGy는 $150{\sim}250^{\circ}C$의 부근에서 특유의 glow curve를 나타내었다. TL ratio는 비조사구 0 kGy에서 0.011로 0.1 이하의 값을 나타냈으며, 조사구 5, 10, 30 kGy는 각각 1.105, 1.009, 2.206으로 0.1이상의 값을 나타내어 조사구로 확인되었다. 감마선 조사처리한 총목피의 total phenolics 함량을 측정한 결과 0, 10, 30 kGy 선량 중 10 kGy의 조사선량에서 총목피 열수 및 50% ethanol 추출물의 total phenolics 함량이 각각 $17.30{\pm}0.40mg/g$, $18.87{\pm}0.46mg/g$으로 가장 높은 phenolics 함량을 나타내었다. 감마선 조사 처리한 총목피의 angiotensin converting enzyme 저해효과와 xanthin oxidase 저해효과는 비조사구에 비해 조사구에서 열수 및 50% ethanol 추출물 모두 증가하는 것을 확인할 수 있었다. Pancreatin ${\alpha}$-amylase 저해효과와 ${\alpha}$-glucosidase 저해효과는 총목피 열수 및 50% ethanol 추출물 $200{\mu}g/mL$ phenolics 농도 처리 시 0~30 kGy의 모든 선량에서 60~100%에 가까운 높은 저해율을 나타내어 현재 당뇨병 치료제로 사용되고 있는 acarbose와 비교 시 월등히 우수한 항당뇨 효과를 확인할 수 있었고, 이를 통해 감마선 조사 처리한 총목피 추출물은 천연소재로써 항당뇨 관련 기능성 소재로 유용하게 사용될 수 있을 것으로 판단되었다. 이상의 결과 총목피 추출물은 다양한 생리활성을 가진 기능성 소재로 활용이 가능한 것으로 생각되었으며 감마선의 조사에 의해서 총목피의 생리활성이 증대되는 현상을 확인함으로써 감마선 조사가 식물체가 나타내는 생리활성을 증폭시킬 수 있는 수단으로 활용이 가능할 것이라 판단되었다.

Variation in Flowering Time of Soybean after Irradiation

  • The polygenic variation in flowering time of soybean irradiated with Co$^{60}$ gamma ray evaluated in the R$_2$ generation. The genetic variance in the irradiation treatment was about four times that of the control. The basic possibility of selection for the early and the late in flowering time of soybean after irradiated with Co$^{60}$ was suggested in this paper.

  • The environmental effects on radio-sensitivities of Lactobacillus and Saccharomyces cerevisiae were studied; Liquid suspensions of Lactobacillus and yeast were gamma-irradiated under various conditions; temperatures, hydrogen ion concentrations, amino acids and vitamins were treated seperately with variations of concentrations. (shown in figures) It is found that simultaneous heat treatment is effective to sterilize microorganisms than pre after treatment, and concentration of hydrogen ion does not affect the lethalty of yeast but or Lactobacilli was affect at the range of pH. 5.0 to 7.0. Ascorbic acid, thiamin and pyridoxine were protective dependently against lethal action of gamma-ray and its protective effects increase with the increasings of concentrations. Glutamic acid, aspartic acid, tyrosine and phenylalanine were proved to be protective for both strains at 0.1 between 1.0 percent. It can be suggested that industrial sterilizing doses of irradiation by gamma-ray for food should be applied more than those dose of saline or buffer suspension, because natural food stuffs are rich of vitamins and amino acids.

Selection of Herbicide Tolerant Cell lines from $\gamma$-ray-Irradiated Cell Cultures in Rice (Oryza sativa L. cv. Ilpumbyeo)

  • Herbicide tolerant rice (Oryza sativa L. cv. Ilpumbyeo) cell lines were selected from $\gamma$-ray-irradiated anther-derived cell cultures. The anther-derived cell clusters were small (300 to 400 ${\mu}{\textrm}{m}$ in diameter) and uniform ones that were screened by miracloth filtering. The cell suspensions were very efficient to plate one layer onto agar medium and to screen target cell lines. Herbicide tolerant cell lines were selected by 5 mg/L cyhalofop butyl (CHB) treatment by using the small cell suspensions on agar N6 medium containing 1 mg/L 2,4-D and 0.2 mg/L kinetin. Of the cell lines, one line (CHB-1) showed stable tolerance at 10 mg/L concentration after 6-month culture without herbicide suspension. Growth stability of CHB-1 was similar to that of control cell line on 10 mg/L CHB containing medium. In this experiment we established herbicide tolerant cell line selection system by using anther-derived uniform-cell suspensions with $\gamma$-ray-irradiation.

Effect of γ-Irradiation on the Physicochemical Properties of Zein Films

  • To elucidate the effect of gamma-irradiation on the physicochemical properties of zein films, the molecular and mechanical properties of the films were examined after irradiation at various irradiation doses. Gamma-irradiation of zein solutions caused the disruption of the ordered structure of the zein molecules, as well as degradation, cross-linking, and aggregation of the polypeptide chains based on an SDS-PAGE study. Gamma-irradiation increased the solubility of zein and decreased the viscosity due to cleavage of the polypeptide chains. Protein solubility of the zein films in urea/2-mercaptoethanol also increased with increasing irradiation doses. Alterations of the zein molecules by irradiation decreased water vapor per-meability by 12% and increased the elongation of zein films. However, mean tensile strength of the zein films was decreased by gamma-irradiation treatment. Measurement of Hunter color values indicated that irradiation caused a destructive effect on yellow pigments, resulting in a significant decrease in Hunter b values. The microstructure as observed by scanning electron microscopy showed that irradiated zein film had a smoother and glossier surface than the non-irradiated films.

감마선 조사가 열처리 겨우살이의 항당뇨 및 세포독성에 미치는 영향 (Effects of Gamma-irradiation on Anti-diabetic and Cytotoxic Activities of Heat-treated Mistletoe (Viscum album))

  • Mistletoe (Viscum album) has been widely used as a functional food material for various therapeutic purposes from ancient time. In this study, we examined anti-diabetic and cytotoxic activities of heated-treated mistletoe and the effects of gamma-irradiation on its activities. Heat-treated mistletoe extract was prepared by heating during different time (3, 6, 9 and 12 h) and gamma-irradiated with different doses of 0, 10, 30, 50, 70 and 100 kGy. Heat-treated mistletoe extracts showed a concentration-dependent cytotoxicity on rat insulinoma RINm5F cells and the effect was gradually decreased as heating time increased up to 12 h. 12 h heat-treated extract was no cytotoxic. Gamma-irradiation enhanced the reduction of heat-treated mistletoe-induced cytotoxicity and the decreasing effect was an irradiating dose-dependent. In particular, all of 70 kGy irradiated and heat-treated mistletoe extracts did not showed the cytotoxicity and the effect was comparable to 12 h heat-treated mistletoe extract. Among those extracts, 3 h heat-treated mistletoe extract gradually increased the insulin secreting activity by gamma-irradiation and the effect was the best at 70 kGy, whereas 12 heat-treated extract was no effect. On the test of ${\alpha}$-glucosidase inhibitory activity, 3 h heat-treated mistletoe extract showed the concentration dependent effects and gamma-irradiation induced more activity at 70 kGy, compared to non-irradiated 3 h and 12 h heated mistletoe extracts. These results suggest that the combination of heat treatment and gamma-irradiation might be more effective than only heat-treatment for improving the anti-diabetic activity of mistletoe extract and reducing its cytotoxicity.

Effect of gamma irradiation on the morphological and physiological variation from In vitro individual shoot of banana cv. Tanduk (Musa spp.)

  • Inducing genetic and morphological variation through conventional method is very difficult. Therefore, mutation induction through in vitro technology brings numerous advantages over the conventional breeding. Thus, the individual shoots (1 ~ 2 cm) were irradiated with gamma rays (10 ~ 70 Gy). The result revealed that the explants treated with higher doses (40, 50, 60, and 70 Gy) showed deleterious effects of ionizing radiation. The highest survival rate among ${\gamma}$ treated explants recorded was 71% in 10 Gy treatments while the lowest survivality was 15% in 70 Gy. Lethal dose 50% ($LD_{50}$) dose was found to be 33 Gy. In the in vitro condition, rooting reponse showed that increase in gamma irradiation dose resulted in the inhibition of root growth. Meanwhile, non-treated explants had the best rooting ability with the maximum number of root per explant (20) within a short period of time (6 days), with the highest root length of (15.1 cm). The longer period in rooting (12 days) and lowest number of root per explant (8) with shortest root length (10.1 cm) were recorded at 30 Gy treatment. The highest shoot length (13.6 cm) was observed at control treatment and the shortest shoot length (10.4 cm) was observed at 30 Gy. In the nursery, lowest leaf number (5) was observed at 30 Gy compared with other treatments. The highest chlorophyll content (49.8) was recorded at 10 Gy treated seedling. Irradiated explants with 10 Gy found to be superior over the control treatment and had positive effects in main growth parameters such as chlorophyll content.

저선량 감마선이 조사된 대두작물의 생육에 미치는 산성강우 효과 (Influence of Artificial Acid Rain on the Growth of Soybean Irradiated with Low Dose Gamma Radiation)

  • 저선량의 감마선이 조사된 대두종자로부터 자라난 작물체에 인공 산성강우가 미치는 영향을 알아보기 위하여 황금콩과 방사콩의 생육 양상을 조사한 결과 초장은 감마선조사 선량에 따라서만 생육 차이를 보였을 뿐 산성강우 무처리구(pH 6.00)와 처리구(pH 2.45와 2.20)간에는 큰 차이를 보이지 않았으며, 종자 저장기간에 따라서도 큰 차이를 보이지 않았다. 그러나 황금콩의 경우 생육 촉진을 위한 적정 조사선량이 산성강우 무처리구에 비해 처리구에서 낮아지는 경향을 보였다. 또한 피해 병징 양상은 산성강우 pH 2.20 처리구에서 심하게 나타났으며 방사콩에 비해 황금콩에서 산성강우 내성 효과가 컸으며, 종자 저장기간에 따라서도 차이를 보여 1995년산에 비해 1997년산 작물체에서 효과가 큰 것으로 나타났다. 그러나 방사콩의 생육 양상은 산성강우 처리 농도와 감마선 조사에 따라 큰 차이를 보이지 않았다.

감마선 조사에 의한 총목피(Aralia elata Cortex)의 추출수율 증대 및 항산화 효과 (The Effect on Anti-oxidative Activity and Increasing Extraction Yield of Aralia elata Cortex by Gamma Irradiation)

  • 본 연구에서는 감마선 조사가 총목피 추출물의 생리활성에 미치는 영향을 검토하였다. 감마선 조사한 총목피의 감마선 조사여부를 확인하기 위하여 전자스핀공명(ESR) 분석을 시행한 결과 대칭적인 unspecific central ESR 신호 좌우로 cellulose를 함유한 조사 식품에서 나타나는 한 쌍의 peak가 6.0 mT의 공간을 두고 나타나 감마선을 조사한 시료로 판별되었다. 감마선 조사된 총목피의 최적 추출조건은 10 kGy를 조사한 시료를 50% ethanol을 사용하여 15시간 추출이었다. 감마선을 조사한 총목피의 항산화 효과를 확인한 결과 DPPH radical 소거효과와 ABTS radical 저해는 총목피의 물 추출물과 50% ethanol 추출물의 경우, 비조사구와 조사구를 저 농도인 $50{\mu}g/m{\ell}$ 처리하였을 때 각각 80%와 98% 이상의 매우 높은 저해 효과를 나타내었다. 지용성 물질의 항산화 활성의 지표인 antioxidant Protention Factor (PF)는 2.18~2.78 PF로 매우 높은 PF값을 나타내었다. TBARs의 경우 물과 ethanol 추출물 모두 높은 TBARs값을 나타내었다. 물 추출물은 감마선의 조사에 의한 TBARs 저해능의 상승은 관찰되지 않았으나, ethanol 추출물은 10 kGy의 감마선조사에 의해 TBARs 저해능의 상승이 약하게 관찰되었다. 이상의 결과 총목피는 감마선의 조사에 의해서 총목피의 phenolic 성분들의 용출율과 항산화 효과가 증대되는 현상을 확인하였다. 또한 감마선 조사가 식물체로부터 생리활성물질의 용출 증대와 항산화 활성을 증폭시킬 수 있는 수단으로 활용이 가능할 것이라 판단되었다.
