흰쥐의 척수에서 Cyclic Nucleotides 및 Glipizide가 Baclofen의 심혈관반응에 미치는 영향 (Effects of Cyclic Nucleotides and Glipizide on the Cardiovascular Response of Baclofen in the Rats)

  • The purpose of present study is to investigate the influence of a spinal gamma-aminobutyric acid B($GABA_B$) receptor on a central regulation of blood pressure(BP) and heart rate(HR), and to define its mechanism in the spinal cord. In urethane-anesthetized, d-tubocurarine-paralyzed and artificially ventilated male Sprague-Dawley rats, intrathecal administration of drugs were carried out using injection cannula(33-gauge stainless steel) through the guide cannula(PE 10) which was inserted intrathecally at lower thoracic level through the puncture of a atlantooccipital membrane. Intrathecal injection of an $GABA_B$ receptor agonist, baclofen(30, 60, 100 nmol) decreased both BP and HR dose-dependently. Pretreatment with 8-bromo-cAMP(50 nmol), a cAMP analog, or glipizide(50 nmol), a ATP-sensitive $K^+$ channel blocker, attenuated the depressor and bradycardic effects of baclofen(100 nmol), but not with 8-bromo-cGMP(50 nmol), a cGMP analog. These results suggest that the $GABA_B$ receptor in the spinal cord plays an inhibitory role in central cardiovascular regulation and that this depressor and bradycardic actions are mediated by the decrease of cAMP via the inhibition of adenylate cyclase and the opening of $K^+$ channel.

GABAergic neuronal development in the embryonic mesencephalon of mice

  • This study presents neurogenesis and neuronal migration patterns of gamma-aminobutyric acid-ergic (GABAergic) neurons during mesencephalic development of mouse. After neurons from embryonic day (E) 10-16 were labelled by a single injection of 5-bromo-2'-deoxyuridine (BrdU), immunohistochemistry was performed. Neurogenesis were mainly generated in the mesencephalic region at E10 to E13. After E14, BrdU positive cells were observed only in the dorsal mesencephalon. GABAergic neurons were mainly originated in the ventrolateral region of the mesencephalon at the early embryonic stage, especially at E11 to E13. E10-labeled cells showed positive for GABAergic neuron in the basal plate of the mesencephalon at E13. At E15, GABAergic neurons were observed in the entire basal plate and some regions of the ventral and dorsal mesencephalon. They were present in the whole basal plate, the ventral and dorsal mesencephalon of E17, spreading more outward of the mesencephalon at P0. Our study demonstrates that major neurogenesis of GABAergic neurons occurs at E11 to E13. However, neuronal migration continues until neonatal period during mesencephalic development.

Intraocular Injection of Muscimol Induces Illusory Motion Reversal in Goldfish

  • Induced activation of the gamma-aminobutyric $acid_A$ ($GABA_A$) receptor in the retina of goldfish caused the fish to rotate in the opposite direction to that of the spinning pattern during an optomotor response (OMR) measurement. Muscimol, a $GABA_A$ receptor agonist, modified OMR in a concentration-dependent manner. The $GABA_B$ receptor agonist baclofen and $GABA_C$ receptor agonist CACA did not affect OMR. The observed modifications in OMR included decreased anterograde rotation $(0.01\sim0.03\;{\mu}M)$, coexistence of retrograde rotation and decreased anterograde rotation $(0.1\sim30\;{\mu}M)$ and only retrograde rotation $(100\;{\mu}M\sim1\;mM)$. In contrast, the $GABA_A$ receptor antagonist bicuculline blocked muscimol-induced retrograde rotation. Based on these results, we inferred that the coding inducing retrograde movement of the goldfish retina is essentially associated with the GABAA receptor-related visual pathway. Furthermore, from our novel approach using observations of goldfish behavior the induced discrete snapshot duration was approximately 573 ms when the fish were under the influence of muscimol.

  • In convulsion state by PTZ in rat, anticonvulsive effect and some of ${\gamma}-aminobutyric$ acid(GABA)-related mechanisms of Buthus extract in brain was experimented. It was inhibited GABA-T activity, lipid peroxide generation and xanthine oxidase activity as scheduled administration in vitro and vivo. And the content of brain glutathione was increased as scheduled administration in rat. In convulsion state by PTZ of previously managed rat by Buthus extract, onset time and duration were non-specific changes but recovery time and severity was remarkably reduced. In conclusion speculated that Buthus extract inhibits convulsion by control of GABA content In brain.

말초성 현기증의 감별진단 (Differential diagnosis of peripheral vertigo)

  • Dizziness can be classified mainly into 4 types: vertigo, disequilibrium, presyncope, and lightheadedness. Among these types, vertigo is a sensation of movement or motion due to various causes. The main causes of peripheral vertigo are benign paroxysmal positional vertigo (BPPV), acute vestibular neuritis (AVN), and Meniere's disease. BPPV is one of the most common causes of peripheral vertigo. It is characterized by brief episodes of mild to intense vertigo, which are triggered by specific changes in the position of the head. BPPV is diagnosed from the characteristic symptoms and by observing the nystagmus such as in the Dix-Hallpike test. BPPV is treated with several canalith repositioning procedures. AVN is the second most common cause of peripheral vertigo. Its key symptom is the acute onset of sustained rotatory vertigo without hearing loss. It is treated with symptomatic therapy with antihistamines, anticholinergic agents, anti-dopaminergic agents, and gamma-aminobutyric acid-enhancing agents that are used for symptoms of acute vertigo. Meniere's disease is characterized by episodic vertigo, fluctuating hearing loss, and tinnitus. It is traditionally relieved with life-style modification, a low-salt diet, and prescription of diuretics. However, diagnosis and treatment of the peripheral vertigo can be difficult without knowledge of BPPV, AVN, and Meniere's disease. This article provides information on the differential diagnosis of peripheral vertigo in BPPV, AVN, and Meniere's disease.

Cocaine- and Amphetamine-Regulated Transcript (CART) Peptide Plays Critical Role in Psychostimulant-Induced Depression

  • Cocaine- and amphetamine-regulated transcript (CART) peptide is a widely distributed neurotransmitter expressed in the central nervous systems. Previously, several reports demonstrated that nucleus accumbal-injected CART peptide positively modulated behavioral sensitization induced by psychostimulants and regulated the mesocorticolimbic dopaminergic pathway. It is confirmed that CART peptide exerted inhibitory effect on psychostimulant-enhanced dopamine receptors signaling, $Ca^{2+}$/calmodulin-dependent kinase signaling and crucial transcription factors expression. Besides modulation of dopamine receptors-related pathways, CART peptide also exhibited elaborated interactions with other neurotransmitter receptors, such as glutamate receptors and ${\gamma}$-aminobutyric acid receptors, which further account for attribution of CART peptide to inhibition of psychostimulant-potentiated locomotor activity. Recently, CART peptide has been shown to have anxiolytic functions on the aversive mood and uncontrolled drug-seeking behaviors following drug withdrawal. Moreover, microinjection of CART peptide has been shown to have an antidepressant effect, which suggests its potential utility in the mood regulation and avoidance of depression-like behaviors. In this review, we discuss CART pathways in neural circuits and their interactions with neurotransmitters associated with psychostimulant-induced depression.

간헐 분무와 음이온 방사를 가진 공기노출형 자동 현미발아시스템의 개발 (Development of an Automatic Brown Rice Germinating System of Air-Phase Type with Intermittent Water Spraying and Anion Radiation)

  • In this study an automatic germinating system for germinated brown rice (GBR) was developed for accelerating germination and sprouting of brown rice. With the developed germinating system, the effects of anion radiation treatment on germination ratio, sprout growth, color quality and ${\gamma}$-aminobutyric acid (GABA) content of brown rice were investigated. This system can fast produce GBR by controlling the environmental conditions such as ambient temperature, relative humidity, and proper anion radiation. Air-phase germination with intermittent water spraying improved both germination ratio and sprout growth by about 2.2 and 2.3 times, respectively, compared to the conventional water soaking method. Our results strongly suggest that this germinating system that provides air-phase germination with intermittent water spraying and anion radiation can produce germinated brown rice with good color quality, high GABA content, and fast germination ratio as well as sprout growth.

Functional Dissection of Glutamatergic and GABAergic Neurons in the Bed Nucleus of the Stria Terminalis

  • The bed nucleus of the stria terminalis (BNST)-a key part of the extended amygdala-has been implicated in the regulation of diverse behavioral states, ranging from anxiety and reward processing to feeding behavior. Among the host of distinct types of neurons within the BNST, recent investigations employing cell type- and projection-specific circuit dissection techniques (such as optogenetics, chemogenetics, deep-brain calcium imaging, and the genetic and viral methods for targeting specific types of cells) have highlighted the key roles of glutamatergic and GABAergic neurons and their axonal projections. As anticipated from their primary roles in excitatory and inhibitory neurotransmission, these studies established that the glutamatergic and GABAergic subpopulations of the BNST oppositely regulate diverse behavioral states. At the same time, these studies have also revealed unexpected functional specificity and heterogeneity within each subpopulation. In this Minireview, we introduce the body of studies that investigated the function of glutamatergic and GABAergic BNST neurons and their circuits. We also discuss unresolved questions and future directions for a more complete understanding of the cellular diversity and functional heterogeneity within the BNST.

Morus Nigra Extract Attenuates Cognition Impairment and GABAergic Interneuron Degeneration in Aged Rat Brain

  • Purpose: Aging process comes with cognitive impairment due to decreased neuronal cell number, activity, and neuronal circuit. Alteration of inhibitory neurons contributes to cognitive impairment in normal aging and is responsible for disrupting the excitation/inhibition balance by reducing the synthesis of gamma-aminobutyric acid (GABA). Morus nigra (Mulberry) is a natural physiologically active substance that has been proven to have anti-oxidant, anti-diabetic, and anti-inflammatory effects through many studies. This study aimed to evaluate the effects of the mulberry extract (ME) on cognitive function through anti-oxidant enzyme and GABAergic neuronal activity in aged rat brain. Methods: Sprague Dawley rats were randomly assigned as the young group (8 weeks, n= 8), aging group (67 weeks, n= 8), and aging+ mulberry extract group (67 weeks, n= 8). The aging+ mulberry extract group was orally administered 500 mg/kg/d mulberry extract for 6 weeks. Results: The aging+ mulberry extract group improved spatial and short-term memory. The antioxidant potential of ME increased the expression of superoxide dismutase-1 (SOD-1) and decreased inducible nitric oxide synthase (iNOS). Also, the aging+ mulberry extract group significantly increased the expression of GABAergic interneuron in hippocampus cornu ammonis1 (CA1) compared to the aging group. Conclusion: The number of GABAergic inhibitory interneurons was deceased and memory functions in the aging process, but those symptoms were improved and restored by mulberry extract administration.
