Occurrence of crown gall of chrysanthemum caused by Agrobacterium tumefaciens.

  • Incidence of crown gall on lower stem of chrysanthemum, Chrysanthemum morifolium Ramat., was first observed at Hwasung, Gyeonggi, Korea in 2001, Tumors on the stem were 1.5-2 cm in size and semi-round with rough surface texture of dark brown color. Four strains of bacteria isolated from the tumor tissues were characterized. Their colonies were convex, glistening, circular with an entire edge, and white to tannish-cream in color on PDA plus CaCO$_3$. They were gram negative, oxidase positive, and growing on DIM agar. The bacterial isolates inducing gall formation in chrysanthemum were identified as Agrobacterium tumefaciens based on biochemical and physiological characteristics, fatty acid profile using Sherlock Microbial Identification System, and substrate utilization patterns using Biolog Identification System. Young chrysanthemum plants inoculated with the bacteria developed typical galls within two to three weeks. Seedlings of tomato and slices of carrot roots also produced typical galls two to three weeks after inoculation. This is the first report on crown gall of chrysanthemum in Korea.

Gall structure and specificity in Bostrychia culture isolates (Rhodomelaceae, Rhodophyta)

  • The descriptions of galls, or tumors, in red algae have been sparse. K$\ddot{u}$tzing (1865) observed possible galls of Bostrychia but only presented a drawing. Intensive culture observations of hundreds of specimens of the genus Bostrychia over many years have revealed that galls appeared in only a small subset of our unialgal cultures of B. kelanensis, Bostrychia moritziana/radicans, B. radicosa, B. simpliciuscula, and B. tenella and continued to be produced intermittently or continuously over many years in some cultures but were never seen in field specimens. Galls appeared as unorganized tissue found primarily on males and bisexuals, but occasionally on females and tetrasporophytes. The gall cells usually were less pigmented than neighboring tissue, but contained cells with fluorescent plastids and nuclei. The galls were not transferable to other potential hosts. Galls could be produced from gall-free tissue of cultures that originally had galls even after transfer to new culture dishes. Electon microscopy of galls on one isolate (3895) showed that virus-like particles are observed in some gall cells. It is possible that a virus is the causative agent of these galls.

  • Recently, usual minor stress is getting more important in somatization disorder. This study is a clinical report of a patient with somatization disorder induced by stress treated with typical orintal medical therapy reinforcing the weakness of heart and gall blaader(Herb-med, acupunture, etc.) in combination with EFT. STAI, BDI were compared between before and after treatment. The results show the typical onntal medical therapy reinforcing the weakness of heart and gall blaader in combination with EFT is efficient in the treatment of Somatization disorder.

토끼 담낭절펼에 대한 Caerulein의 작용 (Effect of Caerulein on the Isolated Gall Bladder Strips of Rabbits)

  • The isolated rabbit gall bladder strips were prepared according to the technique described by Amer and Becvar (1969). The strips were placed in a bath containing 100 ml of Locke-Ringer solution maintained at $38^{\circ}C$. Oxygen was continuously bubbled through the solution. The tension of the muscle strip was initially adjusted to 0.7g. The contractile response was measured isometrically by a force-displacement transducer connected to a polygraph. The effect of a number of autonomic drugs were studied for their interaction with caerulein (Prof. V. Erspamer, F.I. 6934 Caerulein, Farmitalia, Italia), a gastrin or CCK.PZ like peptide, on isolated rabbit gall bladder strips. In this preparation caerulein produced contractions of CCK-PZ type, but the relative potency on a weight basis was 40 times that of CCK-PZ. The response of caerulein was not modified by either cholinergic or alpha or beta adrenergic blockade. However, the response of caerulein and of barium on the strips were prevented by papaverine or aminophylline. Isoproterenol, papaverine or aminophylline alone relaxed the preparation whereas caerulein, CCK-PZ, acetylcholine, serotonin, histamine or barium chloride contracted the preparation. In summary, it is concluded that caerulein on the gall bladder strip seems to act independently of the autonomic nervous system and mediated via mechanisms apparently similar to those involved in the action of barium chloride.

  • Conventionally, identification and classification methods of natural products include the morphological survey and assay of chemical disposition, sing these methods, however, is not satisfying for the precise identification of natural products because they are often valiable in the compositions and morphology To standardize the natural products identification and classification, genomic DNA analysis such as RAPD, RFLP and Amp-FLP can be adopted for this purpose. In this study, various ginsengs and bear gall bladder were tested for the development of genetic identification and classification method. Varieties of ginsengs such as, P. ginseng, P. quinquefolium, P. japonicus and P. notoginseng, were genetically analyzed by RAPD. Also, DNA isolated from Bear blood and gall bladder, Ursus thibetanus, Ursus americanus and Ursus arctos, were analyzed by the same method. The results demonstrated that the identification and classification of bear gall bladder and various ginsengs were possible by RAPD analysis. Therefore, this method was thought to be used as a additional method for the identification and classification of other natural products.

Extracting gall bladders from ultrasound images

  • Nowadays, the internal images of a human body can be easily provided by the ultrasound imaging, the X-ray CT, or the MRI device, among which the ultrasound imaging device has good resolution for soft tissues of a human body compared with the other devices. Furthermore, the use of ultrasound imaging devices will increase in future especially in the obstetrics, territory, since it does not give harm to the human body. Although several techniques have been investigated until now in order to extract organs from ultrasound images, very few of them have achieved satisfactory results because of low contrast and high noise nature of images. This paper proposes a technique for automatic extraction of the gall bladder area from ultrasound images. The proposed technique first extracts a small reliable area of a gall bladder from an ultrasound image employing smoothing, binarization, expanding and shrinking, and labeling, and then expands the area referring to the binarized version of the original image. The technique is examined its performance by real ultrasound images of a gall bladder and satisfactory results are obtained. Some problems to be solved are discussed finally.

인삼 Tumor Callus의 생장 특성 (Characteristics of the Growth of Ginseng Tumor Callus)

  • 인삼 crown gall tumor callus의 특성을 구명하기 위하여 Agrobacterium tumefaciens C58을 감염시켜서 얻어진 crown gall tumor를 phytohormone 무첨가 배지에서 배양하여 생육이 왕성한 tumor callus를 유기하였다. 이들 tumor callus는 정상조직에서 유기한 callus에 비해 외관상으로는 friable한 형태의 덩어리로 자라면서도 매우 단단한 양상을 보였으며, callus증식에 적합한 phytohormone 농도에서는 tumor callus의 생육이 오히려 억제되는 현상을 보였다. Tumor callus의 생육은 고체배양시 암배양이 광배양보다 생체중의 경우에는 2.4배, 건물중은 1.9배 가량이 더 많으며, 2,4-D가 함유된 최적조건에서 배양한 정상 callus의 생체중과는 비슷한 경향을 보였으나, 건물중은 tumor callus가 더 높았다. 그리고 현탁배양시에는 tumor callus가 정상 callus보다 생장속도가 2배 이상 빠른 경향이었다.

Observation of Root-knot Nematodes in the Root Gall Formed on Oriental Melon

  • Oriental melon, Cucumis melo L. cv. Geumssaragieuncheon, grafted on Shintozoa (Cucurbit maxima ${\times}$ Cu. moschata) was planted in a greenhouse infested with Meloidogyne arenaria and root galls were examined five months after planting. A gram of root gall was volumed at ca. 10 cm3 and contained in an average of 363 females (170 developing and 193 matured females), 2,120 secondstage juveniles (J2), and 13,074 eggs. In addition, there was 56 J2 per $cm^3$ soil around the infested plant. An oriental melon had an average of 134.6 g of root gall (70% of total root weight) per 0.72 $m^2$ area. In a conservative estimation, an oriental melon plant could accommodate ca. 1.2 ${\times}$ $10^7$ eggs and J2 per 0.72 $m^2$. The eggs contained in root tissues could be an important inoculum source to the next crop and the fate of these eggs are well worth further investigation.

한국산 양서류피부의 생물학적활성물질에 관한 검색 (Active Polypeptides in Korean Amphibian Skin Extracts)

  • The effect of skin extracts of Korean amphibian, poisonous snake and fresh-water fish were determined for their caerulein-like action on rabbit gall bladder strips. The isolated gall bladder strips were prepared according to the technique described by Amer and Becvar(1969). The strips were placed in a bath containing 100ml of Locke-Ringer solution maintained at $38^{\circ}C$. Oxygen was continuously bubbled through the solution. The tension of the muscle strip was initially adjusted to 0.7g. The contractile response was measured isometrically by a force-displacement transducer connected to a polygraph. In this rabbit gall bladder strip caerulein produced contraction of CCK-PZ type. The skin extract of Korean amphibian also elicited similar contraction as caerulein, which extracted from Australian amphibian, Hyla caerulea, by Erspamer et al. The calculated amount was approximately $2{\mu}g$ caerulein per gram of skin tissue in Korean amphibian and the potency was about 1/200 of that seen in Australian amphibian. The contraction of gall bladder strip by our amphibians occurs in decreasing order; Rana Nigromaculata coreana Okada, Rana nigromaculata Hallowell, Hyla arborea japonica Gunther and Bombina orientalis Boulenger. The skin extracts of poisonous snake and fresh-water fish produced no caerulein-like activity.

담낭염 환자에서의 $^{99m}Tc-HIDA$ Scan의 진단적 의의 (Clinical Application of $^{99m}Tc-HIDA$ Cholescintigraphy in the Cholecystitis)

  • Cholescintigraphic studies with $^{99m}Tc-HIDA$(dimethyl iminodiacetic acid) were performed in 22 cases of normal subjects, 21 of acute cholecystitis, 12 of chronic cholecystitis and 12 others, with the results of, 1) In normal control group, liver and intrahepatic biliary tree, CBD and gall bladder, and G-I tract appeared at 10, 20 and 30 minutes after intravenous injection of $^{99m}Tc-HIDA$ respectively, 2) In acute cholecystitis, 20 among 21 cases showed non-visualization of gall bladder with the diagnostic accuracy, sensitivity and specificity of 93.2%, 95.2% and 100% respectively. 3) In chronic cholecystitis, 5 among 12 cases showed non-visualization of gall bladder and remained 7 among 12 cases showed poor contraction of gall bladder (4), delayed visualization of gall bladder (1) and normal findings (2). 4) In the other disease group, the 12 cases which initially suspected as acute cholecystitis, revealed normal scan findings to exclude the cystic duct obstruction easily, With the above results, this scintigraphic procedure was found very rapid, accurate and easily available method for the determining of the cystic duct patency.
