Sensitivity of Colletotrichum spp. Isolated from Grapes in Korea to Carbendazim and the Mixture of Carbendazim Plus Diethofencarb

  • Thirty-six isolates of Colletotrichum spp. were obtained from infected grapes in two different locations of Korea; 18 isolates from Cheonahn, where carbendazim (MBC) and the mixture of MBC and diethofencarb (NPC) had been applied to control grape ripe rot, and 18 isolates from Cheongju, where no fungicides had been used. Sequences analysis of the internal transcribed spacer (ITS) and the $\beta$-tubulin gene identified 34 of the 36 isolates as Colletotrichum gloeosporioides. The remaining two isolates from Cheongju were identified as C. acutatum. Of the 18 isolates from Cheonahn, 12 were resistant to both MBC and the mixture (MBC+NPC), and six were sensitive to them. All C. gloeosporioides isolates from Cheongju, but not the two C. acutatum isolates, were sensitive to these fungicides. Sequence analysis of the $\beta$-tubulin gene in all isolates revealed that C. gloeosporioides resistant to MBC and MBC+NPC had a tyrosine instead of phenylalanine at the amino acid position 200. The appearance of resistance to MBC and the mixture in C. gloeosporioides correlated with the history of fungicide application in Korea.

살균제 Iprovalicarb 잔류물의 신속한 검출을 위한 바이오센서 (A Biosensor for the Rapid Detection of the Fungicide Iprovalicarb Residuess)

  • In this study, a biosensor was developed using an enzyme-linked immunosorbent assay (ELISA) to rapidly measure the fungicide iprovalicarb residues in agricultural products. The biosensor was designed to include micro-pumps and solenoid valves for fluid transport, a spectrophotometer cuvet as a reaction chamber, a photodiode with a light-emitting diode for optical density measurement, and a control microcomputer to implement assay. The rate of change in optical density of the cuvet was read as final signal output. Micro-pumps were evaluated to investigate their delivery capability, the highest values of the error and the coefficient of variation were 4.3% and 4.6% respectively. As the incubation period was reduced from 15 minutes to 11 minutes to shorten the total processing time, the sensor sensitivity was decreased as the antibody dilution ratio was reduced to a half. The maximum usable period of the coated cuvet was found to be two days with 1% error limit. To predict the concentration of the iprovalicarb residue in agricultural products, a linear calibration model was obtained with r-square values of 0.992 for potato and 0.985 for onion. In validation test for the samples of potatoes and onions against the high performance liquid chromatography, very high correlation values were obtained as 0.996 and 0.993 respectively. Using the cuvet immobilized with antigen, it took 21-minutes for the biosensor to complete the measuring process of the iprovalicarb residues.

Strobilurin계 살균제에 대한 잿빛곰팡이병균 Botrytis cinerea의 저항성 검정 (Monitoring for the Resistance of Strobilurin Fungicide Against Botrytis cinerea Causing Gray Mold Disease)

  • Strobilurin계 살균제인 azoxystrobin에 대한 잿빛곰팡이병균 Botrytis cinerea의 저항성 발현 여부를 조사하였다. B. cinerea 균주는 $1.0{\mu}g\;mL^{-1}$의 azoxystrobin에 대한 반응을 기준으로 실험 균주 중에서 46.5%가 감수성 균주로, 53.5%가 저항성 균주로 판명되었다. 병원균을 분리한 지역별로 저항성 균주의 출현 비율은 경기, 경남, 부산에서 28.6, 41.9, 36.8%이었으며, 충남에서 81.1%로 다른 지역보다 높게 나타났다. 기주별로 저항성 균주는 딸기, 오이, 토마토에서 분리한 균주에서는 각각 68.0, 40.0, 50.0%, 고추에서는 9.1%만이 나타났다. Azoxystrobin에 대해서 감수성이었던 B. cinerea 11-20은 kresoxim-methyl과 trifloxystrobin에 대해서도 감수성 반응을 보였으며, 저항성 균주인 11-74는 3종의 살균제 모두에 대해서 교차 저항성이 있음을 보여 주었다. 본 실험의 결과 실험에 사용한 B. cinerea 균주의 53.5%가 저항성 반응을 보일 정도로 높은 저항성 출현율을 보였으며, 동일 계열의 살균제에 대한 교차 저항성 반응을 보이고 있기 때문에, 포장에서 strobilurin계 살균제에 대한 철저한 관리가 요구되어진다.

국내 과실 탄저병을 일으키는 종 복합체와 종 다양성 및 살균제 감수성 (Colletotrichum Diversity within Different Species Complexes Associated with Fruit Anthracnose in South Korea and Their Fungicides In-Vitro Sensitivity)

  • 탄저병원균(Colletotrichum) 속의 많은 식물병원균은 한국을 비롯하여 전 세계 과수 생산에 큰 위협이 된다. 사과, 감, 자두, 복숭아, 대추, 포도 및 호두와 같은 과실에서 탄저병과 관련된 탄저병원균의 종이 동정되었다. 형태적, 다중 유전자의 계통발생적 및 병원성 시험이 다양한 접근으로 실시되었다. 동정된 탄저병원균의 종에 대한 감수성은 살균제에 대하여 평가하였다. 2종의 종복합체인 gloeosporioides와 acutatum에 속한 9종의 탄저병원균이 국내 과실에 탄저병의 주요 원인균으로 동정되었다. Colletotrichum gloeosporioides 종 복합체 안에는 6종인 C. aenigma, C. fructicola, C. gloeosporioides, C. horii, C. siamense 및 C. viniferum이 있는 반면, Colletotrichum acutatum 종 복합체 안에 종은 C. fioriniae, C. nymphaeae 및 C. orientalis이 동정되었다. 사과 탄저병원균은 C. fructicola, C. siamense, C. fioriniae 및 C. nymphaeae, 자두 탄저병원균은 C. siamense, C. fioriniae 및 C. nymphaeae, 복숭아 탄저병원균은 C. siamense, C. fructicola 및 C. fioriniae, 감 탄저병원균은 C. siamense, C. horii 및 C. nymphaeae, 오미자 탄저병원균은 C. fioriniae, 호두탄저병원균은 C. orientalis, 대추탄저병원균은 C. nymphaeae, 포도 탄저병원균인 C. aenigma, C. fructicola 및 C. siamense를 국내 과수류의 과실에 병을 일으키는 병원균의 종류로 처음 국제학술지에 보고되었다. 이들 병원균의 종에 대한 살균제의 감수성 시험에서 여러 살균제에 대한 탄저병원균의 종 간에 EC50값이 매우 다양하여 감수성의 차이가 크게 나타났다.

Potential of Cross-infection of Colletotrichum Species Causing Anthracnose in Persimmon and Pepper

  • Ninety isolates of Colletotrichum species from new persimmon tree twigs and 50 isolates from pepper plant fruits were isolated via single-spore isolation. Of the 140 isolates, 26 were examined for mycelial growth, carbendazim sensitivity, and ITS sequence. Four of the isolates from the persimmon trees, which were cultivated exclusively in an orchard, showed fast mycelial growth and sensitivity to carbendazim, while five of the pepper isolates showed slower mycelial growth and were resistant to the fungicide. However, 17 isolates from persimmon trees cultivated with pepper plants in the same orchard showed slow mycelial growth like the pepper isolates and they were sensitive to carbendazim like the persimmon isolates. ITS sequence analysis of these 27 isolates led to the identification of the 22 persimmon isolates as C. gloeosporioides and the five pepper isolates as C. acutatum. PCR with species-specific primers confirmed that the 90 isolates from persimmon were C. gloeosporioides whereas the 50 isolates from pepper were C. acutatum. The 90 persimmon isolates of C. gloeosporioides and 50 pepper isolates of C. acutatum were compared by a wound inoculation test to determine their capacity for host cross-infection. All of the C. acutatum isolates from pepper caused typical symptoms of anthracnose on the fruits of pepper plants and twigs of persimmon; they differed from the C. gloeosporioides isolates from persimmon, more than 90% of which were able to infect only persimmon. Amplified fragment length polymorphism analysis revealed the existence of two groups (C. gloeosporioides and C. acutatum isolates group). At 80% genetic similarity, the C. gloeosporioides group was defined within four clusters, while the C. acutatum group was within three clusters. However, these clusterings were unrelated with the virulence of Colletotrichum species against pepper fruits.

Identification and Characterization of Diplodia parva and Diplodia crataegicola Causing Black Rot of Chinese Quince

  • Fungal isolates from infected Chinese quince trees were found to cause black rot in Yeongcheon, Gyeongsangbuk Province, Korea. The quince leaves withered and turned reddish-brown and fruits underwent black mummification. To elucidate the cause of these symptoms, the pathogen was isolated from infected leaf and fruit tissues on potato dextrose agar and Levan media. Several fungal colonies forming a fluffy white or dark gray mycelium and two types of fungi forming an aerial white mycelium, growing widely at the edges, were isolated. Microscopic observations, investigation of fungal growth characteristics on various media, and molecular identification using an internal transcribed spacer, β-tubulin, and translation elongation factor 1-α genes were performed. The fungal pathogens were identified as Diplodia parva and Diplodia crataegicola. Pathogenicity tests revealed that the pathogen-inoculated fruits exhibited a layered pattern, turning brown rotting; leaves showed circular brown necrotic lesions. The developed symptoms were similar to those observed in the field. Fungal pathogens were reisolated to fulfill Koch's postulates. Apples were inoculated with fungal pathogens to investigate the host range. Strong pathogenicity was evident in the fruits, with browning and rotting symptoms 3 days after inoculation. To determine pathogen control, a fungicidal sensitivity test was conducted using four registered fungicides. Thiophanate-methyl, propineb, and tebuconazole inhibited the mycelial growth of pathogens. To the best of our knowledge, this is the first report on the isolation and identification of the fungal pathogens D. parva and D. crataegicola from infected fruits and leaves of Chinese quince, causing black rot disease in Korea.

Fitness is Recovered with the Decline of Dimethachlon Resistance in Laboratory-induced Mutants of Sclerotinia sclerotiorum after Long-term Cold Storage

  • After four years of cold storage, dimethachlon resistance of two laboratory-induced resistant Sclerotinia sclerotiorum isolates SCG7 and LA50 declined by 99.5% and 98.9%, respectively, and cross resistance to iprodione and procymidone also declined dramatically. Along with the decline of fungicide resistance, osmotic sensitivity to sodium chloride and glucose decreased tremendously; mycelial growth rate, sclerotia number and weight per potato dextrose agar (PDA) plate increased on average by 118.6%, 85. 5% and 64.5%, respectively; and virulence to detached leaves of oilseed rape increased by 72.7% on average. Significant negative correlations were detected between dimethachlon resistance levels and mycelial growth rate on PDA (r = -0.980, P = 0.021), and between resistance levels and lesion diameters on detached leaves of oilseed rape plants (r = -0.997, P = 0.002). These results have profound implications for assessing the potential risk for resistance development to dicarboximide fungicides in S. sclerotiorum.

돌연변이 식물 및 형질전환된 효모에서 phytochelatin synthase 발현이 살균제 tolclofos-methyl 분해에 미치는 영향 (Effect of Phytochelatin Synthase Expression on Degradation of Fungicide Tolclofos-methyl in Mutant Plant and Transformed yeast)

  • Phytochelatins (PCs) are small-sized peptides synthesized by PC synthase (PCS) using glutathione (GSH) as a substrate, and they play an important role in the detoxification of toxic heavy metals in plants, fission yeast, and other living organisms. Recently, it has been suggested that PCS is also involved in degradation of some xenobiotics including monobromobimane. PCS cleaves the Gly residue from GSH-xenobiotics conjugates resulting in ${\gamma}$-Glu-Cys-xenobiotics, and this is to degraded further. Therefore, our research is focus on whether PCS is also involved in degradation of tolclofos-methyl, an important pesticide which has been used in ginseng cultivated areas. Heterologous expression of Arabidopsis PCS confers tolerance to tolclofos-methyl in yeast. Furthermore, PCS-deficient Cad1-3 Arabidopsis mutant showed high sensitivity to tolclofos-methyl compared with wild-type plants. These results imply that PCS is involved in degradation of tolclofos-methyl as other xenobiotics.

Effects of Ionizing Radiation on Postharvest Fungal Pathogens

  • Postharvest diseases cause losses in a wide variety of crops around the world. Irradiation, a useful nonchemical approach, has been used as an alternative treatment for fungicide to control plant fungal pathogens. For a preliminary study, ionizing radiations (gamma, X-ray, or e-beam irradiation) were evaluated for their antifungal activity against Botrytis cinerea, Penicillium expansum, and Rhizopus stolonifer through mycelial growth, spore germination, and morphological analysis under various conditions. Different fungi exhibited different radiosensitivity. The inhibition of fungal growth showed in a dose-dependent manner. Three fungal pathogens have greater sensitivity to the e-beam treatment compared to gamma or X-ray irradiations. The inactivation of individual fungal-viability to different irradiations can be considered between 3-4 kGy for B. cinerea and 1-2 kGy for P. expansum and R. stolonifer based on the radiosensitive and radio-resistant species, respectively. These preliminary data will provide critical information to control postharvest diseases through radiation.

Distribution of Monilinia fructicola Isolates Resistant to Dicarboximide or to both Procymidone and Carbendazim in Korea

  • To evaluate the sensitivity of Monilinia fructicola to dicarboximides used in controlling brown rot of peach, the fungus was isolated from commercial peach orchards in Chochiwon (CH), Chongdo (CD), Gyeongsan (GY), and Youngduk (YO) in Korea. The population shift of dicarboximide-resistant isolates of M. fructicola was investigated for 3 years starting 1998. The frequency of procymidone-resistant isolates (PRI) was higher in CD and GY than in CH and YO. The frequency of PRI was higher in the mid season (July-August) than in the rest of the year. Cross-resistance rate of PRI to iprodione was over 87.8% during the investigation, and double-resistance to both procymidone and carbendazim was less than 10%. However, the rate of cross-resistant isolates to vinclozolin was low. In the orchards in GY and CH without any fungicide spray, the PRI population was persistent and did not vary for 3 years. The results suggest that dicarboximide resistance of M. fructicola could be a problem in controlling brown rot and blossom blight on peach trees because it may take a long time to recover the population with sensitive isolates even in the absence of these fungicides.
