Histone Acetylation in Fungal Pathogens of Plants

  • Acetylation of histone lysine residues occurs in different organisms ranging from yeast to plants and mammals for the regulation of diverse cellular processes. With the identification of enzymes that create or reverse this modification, our understanding on histone acetylation has expanded at an amazing pace during the last two decades. In fungal pathogens of plants, however, the importance of such modification has only just begun to be appreciated in the recent years and there is a dearth of information on how histone acetylation is implicated in fungal pathogenesis. This review covers the current status of research related to histone acetylation in plant pathogenic fungi and considers relevant findings in the interaction between fungal pathogens and host plants. We first describe the families of histone acetyltransferases and deacetylases. Then we provide the cases where histone acetylation was investigated in the context of fungal pathogenesis. Finally, future directions and perspectives in epigenetics of fungal pathogenesis are discussed.

Diversity and Bioactive Potential of Culturable Fungal Endophytes of Medicinal Shrub Berberis aristata DC.: A First Report

  • Bioactive natural compounds, isolated from fungal endophytes, play a promising role in the search for novel drugs. They are an inspiring source for researchers due to their enormous structural diversity and complexity. During the present study fungal endophytes were isolated from a well-known medicinal shrub, Berberis aristata DC. and were explored for their antagonistic and antioxidant potential. B. aristata, an important medicinal shrub with remarkable pharmacological properties, is native to Northern Himalayan region. A total of 131 endophytic fungal isolates belonging to eighteen species and nine genera were obtained from three hundred and thirty surface sterilized segments of different tissues of B. aristata. The isolated fungi were classified on the basis of morphological and molecular analysis. Diversity and species richness was found to be higher in leaf tissues as compared to root and stem. Antibacterial activity demonstrated that the crude ethyl acetate extract of 80% isolates exhibited significant results against one or more bacterial pathogens. Ethyl acetate extract of Alternaria macrospora was found to have potential antibacterial activity. Significant antioxidant activity was also found in crude ethyl acetate extracts of Alternaria alternata and Aspergillus flavus. Similarly, antagonistic activity of the fungal endophytes revealed that all antagonists possessed inhibition potential against more than one fungal pathogen. This study is an important step towards tapping endophytic fungal diversity for bioactive metabolites which could be a step forward towards development of novel therapeutic agents.

Monocerin and Ziganein: Phytotoxins from Pathogenic Fungus Exserohilum monoceras Inu-1

  • Two phytotoxic compounds were isolated from a culture of Exserohilum monoceras Inu-1, a fungal pathogen of Barnyard grass. The structure was determined by spectroscopic analyses including 2D NMR experiments. During the isolation procedure, the toxic components were monitored by the assay using Italian ryegrass (Lolium multiflorum Lam.), a host plant of the pathogen. The compounds inhibited the root growth of the host plant seedlings at a level of 100 ppm. While no substantial inhibition was observed even at 300 ppm in non-host plant seedlings such as lettuce and tomato.

Leveillula taurica(Lev.) Arnaud 의한 가지 흰가루병 발생 (Occurrence of Powdery Mildew on Eggplant Caused by Leveillula taurica (Lev) Arnaud in Korea)

  • Yellow spot or blotch symptoms were observed on the upper surface of eggplant (Solanum melongena. cv: Cheonryang) leaves in a commerical vinyl-house of Hapchon-gun, Kyongnam, Korea. We identified Leveillula taurica(L v.) Arnaud as a pathogen causing powdery mildew of eggplant which was observed newly in Korea. The fungal conidia from eggplant leaves were reinoculated to eggplant, tomato and pepper to confirm the same disease as the symptomatology and morphology of the pathogen.

Colletotricum gloeosporioides에 의한 소리쟁이 탄저병 (Anthracnose of Rumex crispus Caused by Colletotricum gloeosporioides)

  • An anthracnose of Rumex crispus was endemic in wet area around a fruit garden of Taejon in Korea. A fungal pathogen was repeatedly isolated from the leaf spot lesions of the weed plant and identified as Colletotrichum gloeosporioides. The plant was controlled completely by fungal inoculation with 5$\times$105 conidia/ml. The fungus has potential to be developed as a mycoherbicide for weed control.

Control of postharvest fungal spoilage of kiwifruit with TiO$_2$ photocatalytic ozonation.

  • TiO$_2$photocatalytic ozonation was attempted to disinfect fungal pathogens causing postharvest spoilage of kiwifruits and to decompose fungicide residuals on kiwifruits. TiO$_2$Photocatalytic ozonation process synergistically degraded organic compound and inhibited conidial germination of the fungal pathogen compared to single treatment of ozonation or photocatalysis. The efficient control of fungal spoilage and degradation of residual fungicide on kiwifruits indicate that TiO$_2$photocatalytic ozonation is a very attractive method for postharvest disease control of kiwifruits as an alternative to fungicides application.

Host-Induced gene silencing of fungal pathogenic genes confer resistance to fungal pathogen, Magnaporthe Oryzae in rice

  • Recently, host-induced gene silencing (HIGS) system has been successfully applied into development of resistant crops against insects, fungal and viral pathogens. To test HIGS-mediated resistance in rice against rice blast fungus, Magnaporthe oryzae, we first tested possibility of movement of small non-coding RNA from rice cells to rice blast fungus. The rice blast fungus expressing GFP transgene were inoculated to transgenic rice plants ectopically expressing dsRNAi construct targeting fungal GFP gene. Expression of dsRNAi construct for GFP gene in transgenic plants significantly suppressed GFP expression in infected fungal cells indicating that small RNAs generated in plant cells can move into infected fungal cells and efficiently suppress the expression of fungal GFP gene. Consistent with these results, expression of dsRNAi constructs against 3 fungal pathogenic genes of M. oryzae in transgenic rice specifically and efficiently suppressed not only the expression of fungal pathogenic genes, but also fungal infection. The conidia of M. oryzae applied on leaf sheath of transgenic rice expressing dsRNAs against 3 fungal pathogenic genes showed abnormal development of primary hyphae and malfunction of appressorium, which is consistent with the phenotypes of corresponding fungal knock-out mutants. Taken these results together, here, we suggest a novel strategy for development of antifungal crops by means of HIGS system.

A Putative Transcription Factor pcs1 Positively Regulates Both Conidiation and Sexual Reproduction in the Cereal Pathogen Fusarium graminearum

  • The plant pathogen Fusarium graminearum causes Fusarium head blight in cereal crops and produces mycotoxins that are harmful to animals and humans. For the initiation and spread of disease, asexual and sexual reproduction is required. Therefore, studies on fungal reproduction contribute to the development of new methods to control and maintain the fungal population. Screening a previously generated transcription factor mutant collection, we identified one putative $C_2H_2$ zincfinger transcription factor, pcs1, which is required for both sexual and asexual reproduction. Deleting pcs1 in F. graminearum resulted in a dramatic reduction in conidial production and a complete loss of sexual reproduction. The pathways and gene ontology of pcs1-dependent genes from microarray experiments showed that several G-protein related pathways, oxidase activity, ribosome biogenesis, and RNA binding and processing were highly enriched, suggesting that pcs1 is involved in several different biological processes. Further, overexpression of pcs1 increased conidial production and resulted in earlier maturation of ascospores compared to the wild-type strain. Additionally, the vegetative growth of the overexpression mutants was decreased in nutrient-rich conditions but was not different from the wild-type strain in nutrient-poor conditions. Overall, we discovered that the pcs1 transcription factor positively regulates both conidiation and sexual reproduction and confers nutrient condition-dependent vegetative growth.

Chitinase 유전자 도입 형질전환 감자식물체의 역병저항성 (Resistance to the Fungal Pathogen Phytophthora infestans of Transgenic Potato Plants Harboring of Chitinase Gene)

  • Chitinase 유전자가 삽입된 감자 Belchip 품종의 형질전환체와 대조구에 곰팡이 병원균을 접종하였다. 7개 계통의 형질전환식물체를 12cm정도 키운 후 병원균인 Phytophthora infestans의 zoospore를 접종하여 인공적으로 역병을 유발시켰다. 그 결과 발병율에 따라서 세 그룹으로 분리되었는데 대조구에 비하여 감염 정도가 심한 것 2개 계통. 비슷한 3개 계통, 발병 정도가 약한 2개 계통으로 구분되었다. 대조구에 비하여 저항성이 높았던 2개 계통과 발병이 심했던 1개 계통만을 대상으로 하여 2차실험을 실시한 결과, 1차실험과 비슷한 경향을 나타냈다. 포장에서 생육된 2개의 저항성 계통은 자연적으로 발생한 역병에 대해서도 역시 대조구에 비하여 역병저항성이 더 높았다.

매자기에 갈색무늬병(가칭)을 일으키는 Nimbya scirpicola (Nimbya scirpicola Causing Brown Spot of Bayonet-Gras (Scirpus maritimus))

  • A brown leaf and stem spot disease of bayonet-grass (Scirpus maritimus) was epidemic in reclaimed paddy fields of Chunbuk province, Korea. A fungal pathogen was repeatedly isolated from the necrotic lesions of the bayonet-grass and identified as Nimbya scirpicola. The pathogen induced disease symptoms only in bayonet-grass but not in 8 other plants tested; Brassica pathogen induced disease symptoms only in bayonet-grass but not in 8 other plants tested; Brassica compestris subsp. napus var. pekinensis, Cucumis sativus, Glycine max, Hordeum vulgare, Lycopersicon esculentum, Oryza sativa, Sesamum indicum and Triticum aestivum. The fungus has potential to be developed as a mycoherbicide.
