A Case Report on Fungal Contamination and Remediation in a Leakage-water Damaged Apartment

  • 이 사례연구는 누수가 있었던 고층 아파트의 공기중 진균 오염 실태를 조사하고, 이를 개선하기 위해 취해진 복원방법의 효과를 평가하기 위해 실시하였다. 공기중 진균은 주로 엔더슨 샘플러를 사용하여 측정하였다. 누수가 있었던 아파트의 복원 전 공기중 진균 농도는 매우 높았다(평균: $51,000\;cfu/m^{3}$, 기하평균: $40,163\;cfu/m^{3}$). 고농도의 오존처리와 오염된 벽체 철거 후 공기중 진균 농도는 상대적으로 매우 낮아졌으며(평균: $1,118\;cfu/m^{3}$, 기하평균: $899\;cfu/m^{3}$), 제거효율은 96.3%로 나타났다. 철거되지 않고 남아 있던 인근 벽체의 벽지를 제거하고 살진균제 처리를 실시한 결과, 공기중 진균 농도는 상대적으로 안전한 수준까지 낮아졌으며(평균: $95\;cfu/m^{3}$, 기하평균: $88\;cfu/m^{3}$), 제거효율은 99.7 %로, 나타났다. 결론적으로, 누수가 있었던 아파트의 공기중 진균 농도는 매우 높은 상태로 오염이 심한 상태이었으나, 본 연구에서 적용한 복원방법들은 공기중 진균 농도 저감에 있어서 매우 높은 효율을 나타냈다.

  • The present survey was undertaken to determine the mycoflora of broiler houses. Attempts were made to isolate and identify fungi in the dust, feed, litter and water from 21 broiler houses. A total of 166 isolates of fungi was identified as yeast spp. (44%), Aspergillus spp. (30.7%), Verticillium spp. (7.2%), Penicillium spp. (3.6%), Paecilomyces spp. (3.6%), Scopulariopsis spp. (3.0%), Cephalosporium spp. (3.0%), Chrysosporium spp. (2.4%), Cladosporium spp. (1.8%) and Absidia spp. (0.6%). Isolated of Aspergillus(A) spp. and Penicillium(P) spp. were identified as A. fumigatus, A. flavus, A. terreus, A. nidulans, A. niger, P. citrinum and P. palitan. Fungal contamination of the broiler houses predominated in the dust. The important point is that must of the isolates were pathogenic fungi.

Mycoflora of Chicken-Viscera with Aid of RAPD Technique as a Tool for Confirmation

  • A total of 100 samples of chicken-viscera were collected from different poultry-slaughtering houses in Austria;(20 samples of each of gizzard, heart, intestine, liver and spleen). Intestine and gizzard were heavy contaminated with moulds than other examined visceral organs($4.4{\times}10^5$ and $2.6{\times}10^4$ colonies/1g of the samples, respectively). Fungal contamination was not detected in all samples of heart and spleen. Eighty-five mould isolates were collected from the examined samples, the majority of isolates belonging to Aspergillus glaucus group(20.0%) and Trichoderma(14.1%). These isolates comprised 15 species belonging to 9 genera. Members of Aspergillus glaucus(telomorph: Eurotium) group and Trichoderma were further confirmed their identification using random amplified polymorphic DNA-polymerase chain reaction(RAPD-PCR) technique.

  • In this study, there has been investigated the simulation of irradiation dose using Monte Carlo methodology for the biological control of wooden cultural property. In the evaluation of fungal contamination on wooden cultural properties, Cladosporium tenuissimum, Aspergillus versicolor, Penicillium sp. were mainly identified from the Gimjeotgae. But these microorganisms were completely inactivated by 20 kGy gamma-rays. For dosimetry simulation of wooden cultural properties, Monte Carlo methodology with MCNP was used. The radiation absorbed dose distribution was predicted at 8.2~18.9 kGy. These results show that irradiation is effective for biologic control of wooden cultural properties and Monte Carlo methodology is useful for non-destructive conservation and preservation of wooden cultural properties.

DNA 기반 곰팡이 평가기법을 활용한 주택의 실내 곰팡이 노출수준 평가 (Evaluation of Indoor Mold Exposure Level in dwelling Using DNA-Based Mold Assessment Method)

  • Objective: Allergic diseases such as asthma due to fungal exposure in houses have increased, and proper management is urgent. Mold can grow in the air, floor, walls, and other areas according to environmental conditions, and there are many limitations to the conventional methodology for examining fungal exposure. For this reason, the degree of fungal contamination is being evaluated by ERMI (Environmental Relative Moldiness Index), a quantitative analysis method proposed by the EPA. In this study, we compared ERMI values between water-damaged dwellings and non-damaged ones to evaluate the effectiveness of Korean ERMI values. We also explored the association of ERMI values with the level of airborne mold and characteristics of dwellings. Methods: Floor dust was collected after installing a Dustream collector on the suction port of a vacuum cleaner. The collected samples were filtered to remove only 5 mg of dust, and DNA was extracted using the FastDNA SPIN KIT protocol. Results: The ERMI values were found to be 19.6 (-6.9-58.8) for flooded houses, 7.5 (-29.2-48.3) for leaks/condensation, and 0.8 (-29.2-37.9) for non-damaged dwellings. The airborne concentration of mold for flooded, leakage or condensed, and non-damaged houses were $684CFU/m^3$, $566CFU/m^3$, and $378CFU/m^3$, respectively. The correlation between ERMI values and the levels of airborne mold was low (R = 0.038), but a weakly significant association of the ERMI values with the concentration of particulate matter ($PM_{10}$) was observed as well(R=0.231,P<0.05). Conclusions: Our findings show that the reference value using ERMI can be used to distinguish water-damaged and non-damaged dwellings. It is believed that ERMI values could be a promising tool for assessing long-term fungal exposure in dwellings.

배양방법과 배지성분이 시루송편버섯 균사체 매트 형성에 미치는 영향 (Effect of culture method and medium components on Trametes orientalis mycelium mat formation (Yasuda) Imazeki)

  • 기존에 사용되는 1차 봉지배양 및 2차 매트생산 배양에 소요되는 기간을 단축시키기 위해 시루송편버섯 균사체에 Co-60 저준위 감마선 200 Gy 24시간 조사로 돌연변이 시킨 ToM 균사체를 활용하여 봉지배양이 생략된 박스 배양법을 시도하였다. 또한, 매트의 품질 및 물성을 강화시키기 위해 균액으로 ammonium nitrate가 3 g/L첨가된 mYMB6을 사용하였고, CaCO3를 1.5% 함유한 톱밥배지에서 균일한 품질의 균사체 매트 생산이 가능하다는 점을 확인하였으며, 기간 또한 기존의 생산방법과 비교하여 10일 이상 단축할 수 있다. 본 결과는 균사체 기반 친환경 소재 연구에 활용할 수 있을 것으로 생각된다.

미곡종합처리장 곡류시료 내 곰팡이 오염과 Fusarium 독소 발생 (Occurrence of Fungi and Fusarium Mycotoxins in the Rice Samples from Rice Processing Complexes)

  • 국내 미곡종합처리장(RPC)에서 벼와 이의 가공산물인 왕겨, 현미, 청치미, 쇄미, 색채미, 백미시료를 2010-2013년 동안 수집하고 미곡에 발생하는 곰팡이와 곰팡이독소 오염을 조사하였다. 전체 곰팡이와 Fusarium 속 곰팡이의 발생은 모든 시료 중 벼에서 가장 높았고, 왕겨가 다음이었으며 도정과정을 따라 감소하였다. 현미, 청치미, 색채미의 발생빈도는 서로 유사하였으며 백미가 가장 낮았다. Fusarium 속 곰팡이 중에서 붉은곰팡이 종복합체의 비율은 2012년에 87%로 가장 높았고, 2011년과 2013년에는 30%대에 머물렀다. Aspergillus 속과 Penicillium 속의 경우 발생빈도는 낮았으나 특정 RPC의 미곡시료에 높은 빈도로 발생하는 유형을 보였고 Alternaria, Nigrospora, Epicoccum 속 곰팡이의 발생유형은 전체 곰팡이의 것과 유사하였다. 미곡 시료의 곰팡이독소 오염여부를 조사하기 위해 2010-2012년 시료를 대상으로 데옥시니발레놀, 니발레놀, 제랄레논과 기타 트라이코쎄신을 분석한 결과, 가공산물 중 색채미의 오염빈도, 검출량, 독소 중복오염비율이 가장 높았고 다음은 청치미였다.

시판 건조 인삼류(백삼, 홍삼)의 곰팡이 발생 (Occurrence of Fungal Species in Dried Ginseng Products from Retail Market)

  • 시판 유통 중인 건조 인삼류(백삼 24점, 홍삼 26점) 포장 제품을 수집하여 곰팡이 발생 조사를 수행하였다. 백삼과 홍삼은 각각 50%와 46%의 시료에서 곰팡이가 검출되었고 검출 시료의 평균 곰팡이 오염도는 각각 0.5와 0.2 log10 CFU/g이었다. 백삼에서는 Penicillium polonicum, P. chrysogenum, Rhizopus microsporus가 각각 18.2%로 우점하였으나 홍삼은 Aspergillus spp.이 87.6%로 우점하였으며 이 중 A. chevalieri가 50.0%로 가장 높았다. 이 중 독성 종으로 알려진 P. polonicum, P. chrysogenum, P. melanoconidium, A. chevalieri 균주의 citrinin 독소 생성 가능성을 분석한 결과, 13 균주 모두 독소 유전자가 검출되지 않았다. 이 결과는 조사한 시료의 곰팡이독소 오염 위험은 매우 낮지만, 건조 인삼류에 곰팡이 오염이 가능함을 보여준다.

중점관리기준에 기초한 국내생산 당귀의 산지 수확 후 아플라톡신의 안전성 평가 연구 (Safety Evaluation from Aflatoxin risk of Korean Angelicae Gigantis Radix Based on Critical Control Points)

  • HACCP methodology was applied in the post-harvest processing and storage of domestic medicinal produces. Particularly in terms of mold and mycotoxin contamination, candidate critical control points (CCP) in the conventional practice in Korean farms were selected and monitored by comparing with on the standard guided processing and storage. When each processing of Angelicae Gigantis Radix were assessed for their safety, the drying steps such as the sun drying or the thermal drying depending on each farm made differences in mold contamination. Moreover, the storage conditions before or after the processing were another critical determinant in the fungal contamination. In other words, storage under $4{\circ}C$ rather than at room temperature was favorable for reducing mold growth in the harvested crops. Occurrence rate of Aflatoxin $B_1 \;(AFB_1)$ in Angelicae Gigantis Radix were 12.8%, but amount of $AFB_1$ in all the collected samples were below 10 ppb regulatory limit allowed in Korea. However, for a few samples of Angelicae Gigantis Radix, still relatively high levels of total amount of the major aflatoxins (aflatoxin $B_1\; +\; B_2\; +\; G_1\; +\; G_2$) were observed around 0.18~49.94 ppb, which is not regulated presently in Korea. It thus can be suggested that post-harvest processing and storage of Korean medicinal crops need further investigation and monitoring to establish the Good Agricultural Practice (GAP), particularly to minimize microbial risk including mold and mycotoxin contamination under the changing climate. Additionally, it is also warranted for new enacting of regulatory limits for total aflatoxins in the medicinal crops.

Microbial Population, Aflatoxin Contamination and Predominant Aspergillus Species in Korean Stored Rice

  • We evaluated microbial populations and aflatoxin production in unhulled and white rice from rice processing complexes of the National Agricultural Cooperative Federation in five regions in Korea and identified three predominant Aspergillus species. Fungal and bacterial populations in rice samples were significantly different between regions in 2007. Aflatoxins were also detected and varied at the levels of 2.45 - 3.43 ng per g unhulled rice grain and 1.29 - 2.09 ng per g white rice grain. Unhulled rice generally detected higher level of aflatoxins than white rice regardless of sampling regions; however, no significant differences were found in Anseong and Cheonan in 2005 and Cheonan and Gimpo in 2007. Aflatoxin production between sampling regions was not different regardless of rice type and sampling year. Although the fungal diversity was highly distinct from region to region, three Aspergillus isolates were predominant in the rice samples; thus, representative isolates AC317, AF57, and AF8 were selected and identified based on their morphological and molecular characteristics. Consequently, isolates AC317, AF57, and AF8 were identified as A. candidus, A. flavus, and A. fumigatus, respectively. These fungi can produce mycotoxins that are harmful for consumers and thus it is important to detect and reduce the population of storage fungi in rice.
