Antifungal Activity of Streptomyces griseofuscus 200401 against Pathogens causing Late Blight and Anthracnose on Pepper (역병균과 고추 탄저병에 대한 Streptomyces griseofuscus 200401의 항균활성)

  • To select microorganisms that exhibit antifungal activity against the fungal pathogens of pepper, Phytophthora capsici and Colletotrichum acutatum, soil samples from a forest and natural fields of Gajang-Dong, Sangju-city were tested in vitro and in vivo. Streptomyces griseofuscus 200401 was finally selected throughout antifungal activity test with dual culture, culture broth, and fruits. For the identification of the strain, nucleotide sequences of 16S rDNA and whole cell fatty acids were analyzed. It is like that the strain 200401 may be a novel biological control agent that can reduce application of chemical fungicides to control late blight and anthracnose on pepper.

  • Lactobacillus casei KC-324 was tested for its ability to inhibit aflatoxin production and mycelial growth of Aspergillus flavus ATCC 15517 in liquid culture. flatoxin $B_{1}$ biosynthesis and mycelial growth were inhibited in both simultaneous culture and individual antagonism assays, suggesting that the inhibitory activity was due to extracellular metabolites. produced in cell-free supernatant fluids of the cultured broth of L. casei KC-324. In cell-free supernatant fluids of all media tested, deMan, Rogosa and Sharpe broth, potato dextrose broth, and Czapek-Dox broth+1% yeast extract showed higher antiaflatoxigenic activity. In these case, fungal growths, however, was not affected as measured by mycelial dry weight. The antiaflatoxigenic metabolites from L. casei KC-324 were produced over wide range of temperatures between $25^{\circ}C$ and $37^{\circ}C$. However, these metabolites were not thermostable since the inhibitory activity of the supernatant was inactivated within 30 minutes at $100^{\circ}C$ and $121^{\circ}C$. The inhibitory activity was not influenced by changing pH of supernatant between 4 and 10. However, the antiaflatoxigenic activity was slightly reduced at pH 10.

Antifungical Activity of Autochthonous Bacillus subtilis Isolated from Prosopis juliflora against Phytopathogenic Fungi

  • The ability of Bacillus subtilis, strain ALICA to produce three mycolytic enzymes (chitinase, ${\beta}$-1,3-glucanase, and protease), was carried out by the chemical standard methods. Bacillus subtilis ALICA was screened based on their antifungal activity in dual plate assay and cell-free culture filtrate (25%) against five different phytopathogenic fungi Alternaria alternata, Macrophomina sp., Colletotrichum gloeosporioides, Botrytis cinerea, and Sclerotium rolfesii. The B. subtilis ALICA detected positive for chitinase, ${\beta}$-1,3-glucanase and protease enzymes. Fungal growth inhibition by both strain ALICA and its cell-free culture filtrate ranged from 51.36% to 86.3% and 38.43% to 68.6%, respectively. Moreover, hyphal morphological changes like damage, broken, swelling, distortions abnormal morphology were observed. Genes expression of protease, ${\beta}$-1,3-glucanase, and lipopeptides (subtilosin and subtilisin) were confirmed their presence in the supernatant of strain ALICA. Our findings indicated that strain ALICA provided a broad spectrum of antifungal activities against various phytopathogenic fungi and may be a potential effective alternative to chemical fungicides.

  • Arachidonic acid is a polyunsaturated fatty acid(PUFA) containing twenty carbon atoms with four double bonds. The family of w-6 PUFA, including arachidonic acid as well as r-linoleic acid, was served as intermediates in the formation of several key prostaglandin and leukotrienes. Several fungal strains of the genus Mortierella accumulate high amounts of arachidonic acid. In this study experiments were carried out to optimize the culture conditions for the mass production of fungus Mortierella alpina DSA -12 and lipid production with high proportion of polyunsaturated fatty acids, especially arachidonic acid. The batch culture was carried out in 500 L fermenter containing 50 g/L glucose, 18 g/L corn-steep powder and 100 mg/L MnS04 under $25^{\circ}C$, aeration rate of 0.5 vvm and agitation speed of 200 rpm without pH control. As a result, we could be obtained 22 g/L of cell mass with high contents of lipid 12.1 g/L) and arachidonic acid (5.1 g/L) The intermittent fed-batch culture was performed in the medium containing 20 g/L glucose and 10 g/L corn-steep powder. The final glucose concentration was 170 g/L and pH was maintained at 5.5 ${\sim}$ 6.0 by adding 14% ammonia solution. It was shown relatively high cell concentration (70.5 g/L) with high contents of lipid (45.8 g/L) and arachidonic acid 08.3 g/L). Therefore, when compared to batch cultures, the high concentration of arachidonic acid could be obtained by fed-batch culture using M. alpina DSA -12. These results imply that the fed-batch culture of M. alpina DSA -12 was feasible in industrial purpose and could be employed in the commercial production of arachidonic acid.

  • An antitumor substance was purified from the culture filtrate of phytopathogenic fungus Alternaria brassicicola SW-3 isolated from soil of a chinese cabbage patch, and its characteristics were investigated. Antitumor activity of A. brassicicola SW-3 was measured by MTT assay. The cytotoxic activity against human cancer cell line was detected in the culture filtrate of A. brassicicola SW-3, but no activity found in mycelium. Antitumor substance was isolated from the culture broth by ethyl acetate extraction and purified by silica gel column chromatography. Structure of the purified compound was analyzed by the instrumental analysis such as $^1$H-NMR, $^{13}$ C-NMR and IR spectroscopy. The purified fungal metabolite of an A. brassicicola SW-3, consists of 11 carbon chain with two hydroxyl groups and two epoxides which is identical to depudecin. The $IC_{50}$/ values of the active compound identified as depudecin were $69\mu$g/mL and $57\mu$g/mL against mouse melanoma B16BL6 cell line, and human hepatoma SK-HEP1 cell line, respectively.

Anti-Angiogenic Activities of Gliotoxin and 1ts Methylthio-Derivative, Fungal Metabolites

  • In the search for new naturally occurring angiogenic inhibitory we found that culture broths from two unidentified fungal strains exerted potent inhibitory activities on capillary-like tube formation of human umbilical vein endothelial cells (HUVEC) in vitro. Two active compounds were isolated by bioassay-guided separation and their structures were identified as gliotoxin (1) and its derivative methylthiogliotoxin (2) by spectroscopic analyses. These compounds significantly inhibited the migration of HUVEC assessed by in vitro wounding migration assay and exhibited at least 10 times more potent inhibition of proliferation of HUVECs as compared with that of cancer cell lines such as HeLa, MCF-7, and KB 3-1 cells. Especially, gliotoxin having disulfide group exerted more potent activities than methylthiogliotoxin, suggesting that gliotoxin could be a useful compound for further study as an anti-angiogenic agent.

  • Osmotically sensitive fungal protoplasts were liberated from the mycelium of several kinds of molds by mixed enzyme system of cellulase from Trichoderma viride $TO_4$ and chitinase from the culture filtrate of Streptomyces sp. 115-5. Relatively higher number of protoplast were released from young mycelium of Zygomycetes strains than Ascomycetes strains by using 10 mM phosphate buffer (pH 6.0) and 0.6 M NaCl as osmotic stabilizer. Protoplasts were released through ruptures in the wall, initially at the apices, but later also from old party of the hyphae.

Tobacco Growth Promotion by the Entomopathogenic Fungus, Isaria javanica pf185

  • Isaria javanica pf185 is an important entomopathogenic fungus with potential for use as an agricultural biocontrol agent. However, the effect of I. javanica pf185 on plant growth is unknown. Enhanced tobacco growth was observed when tobacco roots were exposed to spores, cultures, and fungal cell-free culture supernatants of this fungus. Tobacco seedlings were also exposed to the volatiles of I. javanica pf185 in vitro using I-plates in which the plant and fungus were growing in separate compartments connected only by air space. The length and weight of seedlings, content of leaf chlorophyll, and number of root branches were significantly increased by the fungal volatiles. Heptane, 3-hexanone, 2,4-dimethylhexane, and 2-nonanone were detected, by solid-phase micro-extraction and gas chromatography-mass spectrophotometry, as the key volatile compounds produced by I. javanica pf185. These findings illustrate that I. javanica pf185 can be used to promote plant growth, and also as a biocontrol agent of insect and plant diseases. Further studies are necessary to elucidate the mechanisms by which I. javanica pf185 promotes plant growth.

  • - The effects of sodium acetate on the production of $\gamma$-linolenic acid (GLA) and the secretion of the mycelial lipid into the culture medium with noninnic surfactants were studied with Mucor sp. KCTC 8405P. In the addition of 2.0%) sodium acetate to the basal medium, dry cell weight and total lipid content were increased from 7.8 g/l and 2.46 g/l to 16.0 g/1 and 4.77 g/l, but GLA content was decreased from 18.6% to 13.85%. The growth of Mucor sp. KCTC 8405P was greatly dependent on both the initial pH and the concentration of sodium acetate of culture medium, which was considered as the results of the formation of acetic acid because the fungal growth was completely inhibited at the concentration of acetic acid higher than 22 mM. With the decrease of the oxygen supply, the cell growth, total lipid, and GLA content were sharply decreased in the presence of 2.0% sodium acetate. For the secretion of mycelial lipid into the culture medium, the effects of the various nonionic surfactants were examined. In the addition of 0.5% Tween 80 or Span 80 to the basal medium, 194 mg/l or 263 mg/l of GLA was obtained in the culture medium.

  • The nematode-trapping process by Arthrobotrys conoides was investigated with the aid of scanning and transmission electron microscopy. 1. A. conoides captures nematode by means of three-dimensional network. 2. The wall of trap cell was thicker than that of vegetative hypha and the trap cell was more rich in cell organelles such as endoplasmic reticulum, mitochondria and electrondense granule. 3. The electron-dense granule, which could be found only in trap organs, gradually disappeared during its penetration into nematode cuticle. 4. The osmiophilic area was found at adhering site between the trap organ and nematode cuticle. 5. In some cases, any appressorium was not found at the site of penetration. 6. When the fungal-nematode culture was conserved for 2~3 weeks, numerous young nematodes were found to be adhered to spores, resulting in death.
