Functional Haplotypes and Evolutionary Analyses of SBE1 in Collected Rice Germplasm

  • The starch-branching enzymes (BEs) are responsible for synthesizing the amylopectin, which plays an important role in determining the structural and physical properties of starch granules. BE has two differently functioning isoforms (BEI and BEIIa/b) based on their difference in the chain-length pattern by the degree of polymerization (DP), which mainly contributes to the amylopectin chain length distribution in starch biosynthesis. In this study, we investigated functional haplotypes and evolutionary analyses of SBE1 in 374 rice accessions (320 Korean bred and 54 wild). The analyses were performed based on the classified subpopulations. Haplotype analysis generates a total of 8 haplotypes, of which only four haplotypes were functional carrying four functional SNPs in four different exons of SBE1 on chromosome 6. Nucleotide diversity analysis showed a highest pi-value in aromatic group (0.0029), while the lowest diversity value was in temperate japonica (0.0002), indicating the signal of this gene evolution origin. Different directional selections could be estimated by negative Tajima's D value of temperate japonica (-1.1285) and positive Tajima's D value of tropical japonica (0.9456), where the selective sweeps were undergone by both positive purifying and balancing selections. Phylogenetic analysis indicates a closer relationship of the wild with most of the cultivated subgroups indicating a common ancestor for SBE1 gene. FST-values indicate distant genetic relationships of temperate japonica from all other classified groups. PCA and population structure analysis show an admixed structure of wild and cultivated subpopulations in some proportions.

The Role of Functional and Playful Experiential Value on the Intention to Use ChatGPT (사용자가 인지하는 기능적, 유희적 경험가치가 챗GPT의 재사용 의도에 미치는 영향)

  • ChatGPT, a generative artificial intelligence(AI) technology that analyzes conversations to identify users' intentions and generates responses in consideration of the context of the conversation, is attracting attention from a user interface (UI) perspective that it can provide information through natural conversations with users. This study examined the effect of functional and playful values experienced by early users of ChatGPT on reuse intention and verified the structural relationship between technological efficacy, experiential values, and reuse intention. To verify the research model and hypotheses, a survey was conducted on college students who used ChatGPT for the first time. A total of 156 responses were received and 154 responses were used for analysis. As a result, both the functional experiential value and playful experiential value in the initial use process had significant effects on the intention to use ChatGPT. In addition, it was found that technological efficiency had a significant effect on functional and playful experiential values.

  • Taking 195 Korean businesses and sorting them out according to the unit types, this study looks into how flexibility strategy in labor market, which consists of numerical flexibility and functional flexibility, takes effect on the organization's outcome variable, which in turn consists of productivity, labor-management cooperation, and job security. In addition, the present study analyzes the role of labor union's moderating effect on the relationship among labor market flexibility, labor-management cooperation, and job security. It is found out as a result that numerical flexibility has no meaningful relation with productivity, labor-management cooperation, and job security, while functional flexibility has a considerably positive relation with them. This result confirms the importance of functional flexibility of business strategy in labor market. The West has continuously insisted the importance of functional flexibility, pointing out problems of numerical flexibility. On the other hand, considering that this study has confirmed functional flexibility's meaningful relation, while it has not found out numerical flexibility's negative relation, it is necessary to study further on the relationship between these two kinds of flexibility. The result of analysis on the role of union power's moderating effect confirms only the union power's interaction effect in the relation between numerical flexibility and labor-management cooperation. The possibility of union's opportunistic behavior can be detected in this result.

  • It is strongly expected that inkjet printing method will be play and important role on printable electronics such as 3D integration of embedded ceramic devices(capacitor, resistor, inductor and electrode or circuit), Si-TFT and organic TFT including display C/F, RFID, FPCB, and etc. A inkjet printing method had been center of attention to strengthen the competitiveness of flat panel display on market and to open the new world of manufacturing process of printable electronics. We will survey the industrial tendency of printable electronics and flat panel display including some examples of inkjet printing and present the considerable points of inkjet printing method and some role of materials for successful inkjet printing.

  • The success of new product is a key factor for getting competitive advantages. Marketing research has been investigating marketing capability, manufacturing technical capability, cross-functional integration, market knowledge competence, and competitive environment as the key success factors of new product advantage. Recently, the role of IT capability in enhancing new product advantage is assumed in the literature. However, the empirical studies on the role of IT capability are lacking. The purpose of this study is to identify the effects of IT capability on new product competitive advantages and the relationship between IT capability and the key success factors for new product advantage. Conducting an empirical field study, the IT capability is shown to have a direct effect on new product competitive advantages, and to be also the key success factor to make up for marketing capability and manufacturing technical capability. This study develops a conceptual model on the role of IT capability in enhancing new product competitive advantages by integrating marketing research and IS research.

  • Coffee is the most frequently consumed functional beverage world wide. The average daily coffee consumption is increasing. This crop, which plays an important role in the global economy is under great threat from climate change. To with stand the current climate change, farmers have to learn crop cultivation techniques, strategies to protect crops from diseases, and understand which type of seed varieties to use to avoid crop loss. The present review briefly discusses the coffee cultivation techniques, impact of climate changes on coffee production, processing techniques of coffee, and the importance of coffee in our society, including its chemical composition and prevention against, major diseases. Furthermore, the importance and role of advanced nanotechnology along with molecular approaches for coffee crop improvement and facing challenges are explained.

Study on Inhibition of Platelet Aggregation of Bioactive Constituents from Paeonia lactiflora (작약의 혈소판 응집억제작용에 관한 연구)

  • Methanol extracts from Paeonia lactiflora showed a strong inhibition against platelet aggregation on platelet activation test. Therefore, the bioactive constituents from Paeonia lactiflora were prepared using chromatography methods and were analyzed by NMR and reference data. Compound 1b was confirmed a same structure with henzoyloxypaeoniflorin, compound 2e was a same structure with paeoniflorin; main product of Paeonia lactiflora. Analytical data of compound 3a were not consistent with any known paeoniflorin soucture, but showed the souctural similarity with it. And also the aggregation inhibition activity of compound 3a showed a strong inhibition($\geq$ 90%) induced by collagen. Therefore it suggested that the structure of compound 3a may be the similar structure of benzoyloxypaeoflorin with a functional group in place of benzoyl group and/or a different functional group in stead of Rl. We suggested that benzoyl group of benzoyloxypaeoniflorin substitued instead of 5-carbon OH group on glycoside moiety paeoniflorin played role of the metabolite in case of a platelet aggregation inhibition activity. Paeoniflorin showed more strong inhibition by thrombin than collagen. Therefore, it may be destructed a calcium metabolite as a forming $Ca^2+$ chelate. Compound 3a may be that other functional group instead of OH group of 5-carbon on glycoside moiety of paeoniflorin and/or OH group of benzoyl moiety of paeoniflorin played role of the metabolite in a platelet aggregation inhibition.

Studies on the Functional Role of RNase G in the Regulation of Escherichia coli Enolase Expression Under Microaerobic Conditions (미세호기성 조건에서 Escherichia coli 에놀라아제의 발현에 있어서 RNase G의 역할에 대한 연구)

  • Enolase is one of the glycolytic enzymes, which are involved in a central energy metabolism present in nearly all organisms. In Escherichia coli, enolase constitutes RNA degradosome with RNase E, PNPase and RNA helicase, which are involved in most mRNA degradation and RNA processing. Recently, it has been reported that RNase G, an RNase E homolog, degrades eno mRNA. To examine a functional role of RNase G in enolase expression which is known to be up-regulated under microaerobic condition, we carried out experiments. Here, we report that expression levels of enolase and RNase G are not correlated under microaerobic condition. Based on this observation, we suggest the existence of an unknown factor(s) which regulate the activity of RNase G or enolase mRNA under microaerobic conditions.

Cyclin-Dependent Kinase Inhibitor 2A is a Key Regulator of Cell Cycle Arrest and Senescence in Endothelial Colony-Forming Cells in Moyamoya Disease

  • Objective : Endothelial colony-forming cells (ECFCs) have been reported to play an important role in the pathogenesis of moyamoya disease (MMD). We have previously observed stagnant growth in MMD ECFCs with functional impairment of tubule formation. We aimed to verify the key regulators and related signaling pathways involved in the functional defects of MMD ECFCs. Methods : ECFCs were cultured from peripheral blood mononuclear cells of healthy volunteers (normal) and MMD patients. Low-density lipoproteins uptake, flow cytometry, high content screening, senescence-associated β-galactosidase, immunofluorescence, cell cycle, tubule formation, microarray, real-time quantitative polymerase chain reaction, small interfering RNA transfection, and western blot analyses were performed. Results : The acquisition of cells that can be cultured for a long time with the characteristics of late ECFCs was significantly lower in the MMD patients than the normal. Importantly, the MMD ECFCs showed decreased cellular proliferation with G1 cell cycle arrest and cellular senescence compared to the normal ECFCs. A pathway enrichment analysis demonstrated that the cell cycle pathway was the major enriched pathway, which is consistent with the results of the functional analysis of ECFCs. Among the genes associated with the cell cycle, cyclin-dependent kinase inhibitor 2A (CDKN2A) showed the highest expression in MMD ECFCs. Knockdown of CDKN2A in MMD ECFCs enhanced proliferation by reducing G1 cell cycle arrest and inhibiting senescence through the regulation of CDK4 and phospho retinoblastoma protein. Conclusion : Our study suggests that CDKN2A plays an important role in the growth retardation of MMD ECFCs by inducing cell cycle arrest and senescence.

  • Natural product substances have historically served as the most significant also be prepared by source of new leads for pharmaceutical development. They can chemical synthesis(both semisynthesis and total synthesis) and have played a important role in the field of organic chemistry by providing synthetic targets. Rcently, they have also been extended for commercial purpose to refer to medicinal products, health functional foods, dietary supplements and cosmetics from natural sources. A large number of currently prescribed drugs have been either directly derived from or inspired by natural products. However, with the advent of robotics, bioinformatics, high throughput screening(HTS), molecular biology-biotechnology, combinatorial chemistry, in silico(molecular modeling) and other methodologies, the pharmaceutical industry has largely moved away from plant derived natural products as a source for leads and prospective drug candidates. The strategy for natural prduct industry is now changing from drug approaches to health foods by identifying effective natural products as preparations. In Korea, a lot of development of natural product based drugs have been done, but very few on health functional foods. The concept of natural product based health foods is not active components as lead compounds but standardized extracts or preparation mixed with other medicinal plants. The representative material has been recently known to be a standardized ginseng extract "Ginsana G 115" developed by Swiss Pharmaton company. The purpose of this presentation is to underline how natural products research continues to make significant contributions in the domain of discovery and development of new health functional foods. It is proposed to present the development of high value added health food or health functional foods through scientific investigation on efficacy and standardization of new materials form natural products.
