Antioxidative and Inhibitory Activities of Extract and Juice Powder from Hippophae rhamnoides L. against Nitric Oxide and Elastase Production (비타민나무 추출물 및 착즙 분말의 항산화 활성 및 Nitric Oxide 생성과 Elastase에 대한 저해활성)

  • Background: The flowering plant Hippophae rhamnoides L. has been used for many studies on fruit or leaf extracts. This study was conducted to investigate the development of a new cosmetic material from H. rhamnoides fruits and leaves that have by antioxidant, anti-inflammatory and wrinkle improvement activities. Methods and Results: The antioxidant abilities of H. rhamnoides extracts, including of a water-soluble fruit powder (FW), a fatsoluble fruit powder (FF), a supercritical extract of fruit by-product (BS), and a mixture of leaf and fruit (MIX), were investigated in vitro. A DPPH radical assay for antioxidant activity was performed for these fractions alongside assay to evaluate the total phenolic and flavonoid content (TPC and TFC). As expected, the MIX had the highest DPPH radical scavenging activity ($RC_{50}=10.27{\mu}g/m{\ell}$), and the TPC and TFC also were highest in MIX ($225.7mg{\cdot}GAE/g$, and $25.18mg{\cdot}QE/g$, respectively). Nitric oxide (NO) production in LPS-induced RAW264.7 cells was estimated and the results indicated an over 75% decrease of NO production in FF and MIX. In other assays, the highest elastase inhibitory activity was found in FW. Conclusions: These results revealed that H. rhamnoides extracts have a high potential for antioxidant, anti-inflammatory and antiwrinkle activities. H. rhamnoides products are suggested to be applied as the functional materials of cosmetic ingredients.

Development of UV-screening Polysilsesquioxane Spheres (자외선차단 기능을 갖는 폴리실세스퀴옥산 구형 입자의 개발)

  • A silsesquioxane precursor that contains p-methoxycinnamoyl group was synthesized to produce UV absorbing spherical polysilsesquioxane (PSQ) spheres with diameters of 10nm to a few ${\mu}m$ under different conditions. A dispersion of $10wt\%$ in Vaseline of these PSQ spheres with diameters of approximately $0.6{\mu}m$ showed SPF values of 5.7 and the identical dispersion did not turn white when applied to human hands. These UV absorbing PSQ spheres showed higher boosting effect of sun protection factor (SPF) than the well known UV blocking octyl methoxycinnamate (OMC). The results indicated that the PSQ spheres with p-methoxycinnamoyl group should be useful as the ingredients in UV screening functional cosmetics without causing allergies and whitening.

Effects of Hahella chejuensis-Derived Prodigiosin on UV-Induced ROS Production, Inflammation and Cytotoxicity in HaCaT Human Skin Keratinocytes

  • Prodigiosins, which are natural tripyrrole red pigments and synthetic derivatives, reportedly have multiple biological effects mainly on various types of cancer cells. However, the effects of bacterial prodigiosin on non-cancerous HaCaT human skin keratinocytes have not been reported. Therefore, the present study aimed to investigate the functional activities of prodigiosin derived from cultures of the bacterium Hahella chejuensis in HaCaT cells. Cell viability, the cell proliferation rate, and reactive oxygen species (ROS) production in vitro were assayed following treatment of HaCaT cells with prodigiosin. Prodigiosin did not cause cytotoxicity and notably increased proliferation of HaCaT cells. Furthermore, prodigiosin reduced ultraviolet (UV) irradiation-induced ROS production and the inflammatory response in HaCaT cells. More importantly, prodigiosin reduced matrix metalloproteinase-9 expression and increased collagen synthesis in UV-irradiated HaCaT cells, demonstrating that it elicits anti-aging effects. In conclusion, our results reveal that H. chejuensis-derived prodigiosin is a potential natural product to develop functional cosmetic ingredients.

  • Today, men's appearance is an important part of the global fashion industry, including Korea, with such fashion concepts as the Metrosexual and Ubersexual. Domestic and overseas beauty and cosmetics related organizations are using this phenomenon for marketing and researching to improve the function of cosmetics. The purpose of this study was to investigate the effect of Prunus persica L. Batsch leaf extract on the skin condition after clinical application to male subjects with oily skin to determine the possibility of functional cosmetics utilizing this extract. The cleansing bars were prepared by the MP method and the cleansing packs were prepared using natural ingredients. After 8 weeks of application, moisture, oil, erythema, melanin, pH, elasticity and tactile sensation were measured using MPA5. The results showed that there was a statistically significant difference in T-zone moisture and T-zone oil after using the natural cleansing bar, but there was no significant difference between the two groups for other items. However, the statistically significant changes were found in the individual skin condition change items of the subjects in each group, and it was judged that the two kinds of formulations could be used at the same time to have a higher skin improvement effect. Thus, Prunus persica L. Batsch leaf extract demonstrates potential to be used as a functional cosmetics item.

Carica papaya leaf water extract promotes innate immune response via MAPK signaling pathways

  • The emergence and rapid spread of the potentially fatal coronavirus disease 2019, caused due to infection by severe acute respiratory syndrome coronavirus-2, has led to worldwide interest in developing functional bioactive ingredients that act as immunomodulatory agents. In this study, we aimed to characterize Carica papaya extract and explore its potential as an immunomodulator by performing in vitro cell screening. Papaya leaf water extract (PLW) was found to significantly increase the levels of nitric oxide (NO) and prostaglandin E2 (PGE2) by upregulating inducible nitric oxide synthase and cyclo-oxygenase-2 activity, respectively. Additionally, PLW increased the production of tumor necrosis factor-α and interleukin 1β in RAW 264.7 cells. Furthermore, PLW activated the expression of c-Jun N-terminal kinase (JNK) and extracellular signal-regulated kinase (ERK) but not that of p38 mitogen-activated protein kinase. These results indicate that PLW increased the production of NO, PGE2, and pro-inflammatory cytokines by activating the JNK and ERK pathways in macrophages, thus demonstrating immunomodulatory properties. Finally, high-performance liquid chromatography fingerprint analysis indicated the presence of rutin, narirutin, and ρ-coumaric acid in PLW (6.30, 119.76, and 47.25 ppm, respectively). Treating cells with these compounds at non-toxic concentrations had no effect on NO production. Taken together, these results suggest that PLW may have potential as an immunity-enhancing supplement.

Skin-Mimicking Phantom for Measurment of Cosmetic Transdermal Absorption and Temperature Changes by Sonophoresis

  • Functional cosmetics containing various ingredients that improve skin health are currently being developed. In addition, technologies that help increase the absorption rate of such cosmetics have recently gained significant attention. Sonophoresis is a method to increase the transdermal absorption of cosmetics using ultrasound. A skin-mimicking phantom was fabricated using polydimethylsiloxane, Strat-MTM membrane, and thermochromic pigments. Gel-type cosmetics used in skin mask packs and epidermal-growth-factor-based nano-cosmetics were tested for their absorption rates at ultrasound frequencies of 1, 3, and 10 MHz in the single frequency mode, and 1/3 and 3/10 MHz in the dual frequency mode. The gel-type cosmetics and epidermal-grow-factor-based nano-cosmetics showed the highest absorption rate at 3/10MHz dual frequency. The size of the cosmetic particles decreased by 5-9 %. Furthermore, the temperature rise caused by ultrasound could be visually recognized by the thermochromic pigment in the phantom turning white. We presented a skin-mimicking phantom. The device can be customized according to the size of the ultrasound probe and has the advantage of quantitatively evaluating the transdermal permeability of cosmetics at a low cost. The development of the skin-mimicking phantom will be useful for determining the suitable conditions required to increase the absorption rate of cosmetics using ultrasound.

A Study on the Physiological Activity of Agastachis herba Extract and Fermented Products as Cosmetic Materials (화장품소재로서 곽향 추출물 및 발효물의 생리활성 연구)

  • Agastachis herba is so similar in use to herbs that it is called Korea Mint in the West. Western herbs are commonly used as ingredients in cosmetics. However, in Korea, Agastachis herba is primarily used as a medicinal herb. This study aims to compare the effectiveness of one cosmetic ingredient with another cosmetic ingredient. Antioxidant, whitening, polyphenol, flavonoid, and antibacterial experiments were conducted on lactobacillus, yeast ferments, essential oils, and floral water using extracts from Agastachis herba. Antioxidants exhibited significant effects in all samples, while essential oils displayed the highest efficacy in terms of whitening, polyphenol content, and antibacterial properties. Flavonoids exhibited the strongest effect in the 70% ethanol extract. The essential oil had the most significant functional impact. However, it was confirmed that the fermented products that underwent yeast fermentation with extracts were as effective as essential oils.

  • Cosmetic functional ingredients are products of emotional convergence technology. Unlike pharmaceuticals, it is very important that cosmetics ingredients contain emotional attractiveness because they are household products that have limitations in their effectiveness. Ginseng has long been used as a major prescription of oriental medicine for human health. Detailed records such as the origin and propagation of ginseng can lead to various cosmetical application of ginseng as an emotional convergence ingredients that utilizes the authenticity and excellence of ginseng as an asset of K-beauty. In the unfortunate reality that the first records of ginseng use depend on ancient Chinese literature, it is very meaningful to describe and leave the records of ginseng used in the field of beauty. This study examined the general historical records needed for the study of the cosmetics application of ginseng and the recent findings of ginsenoside in the area of skin function, and provided basic data that can be applied to new cosmetics development research.

Chemical Stability and Whitening Activity of Mixture of Functional Ingredients (미백 기능성 성분의 혼합물에 대한 미백 활성 및 안정성에 관한 연구)

  • Recently, arbutin, oil soluble licorice extact (GLY), ascorbyl glucoside (AA2G), and ethyl ascorbyl ether (EAE) have been widely used as functional whitening ingredients. To Investigate which combination between the above agents could be more effective for whitening effect, tyrosinase activity and MSH-induced melanin production in B-16 melanoma cells were investigated. Both GLY and arbutin dose-dependently inhibited purified tyrosinase activity. The inhibitory effects of GLY with AA2A or EAE on Drosinase activity were more potent than those of GLY alone, whereas that of arbutin with other ingreadients did not show those effects. In MSH-induced melanin production in B-16 melanoma cells, the mixture of Gly and EAE more significantly reduced melanin formation than Gly alone. Stability of mixture of GLY, arbutin, AA2A and EAE exposed at the temperature of $25^{\circ}C\;or\;45^{\circ}C$ for 30 days were also investigated. All of the combinations of whitening agents did not show any critical changes in their composition stability. These data suggest that the combination of GLY and vitamin C derivatives such as AA2G and EAE may be useful for the promotion of whitening effect.

Composition and Contents of Free Amino Acids and Phenolic Acid and Flavonoids of Imperata cylindrica Beauvois var. koenigii Root extracts (백모근 추출물의 유리아미노산 및 Phenolic acid와 Flavonoids 조성 및 함량)

  • This study was carried out to investigate the free amino acid and derivatives and phenolic acid and flavonoid compositions and contents of hot water and 95% ethanol extracts of Imperata cylindrica Beauvois var. koenigii root. The purpose of this study is to obtain basic data and to confirm their applicability as functional cosmetic and functional food ingredients on extracts. 15 kinds and 9 kinds of free amino acids were detected in both extracts, respectively. 5 kinds and 4 kinds of protein free-amino acid derivatives were detected in both extracts, respectively. Total phenol contents were 116.50 ± 0.06 and 140.10 ± 0.04 mg% in both extracts, respectively. The total flavonoid contents were 31.80 ± 0.03 and 43.90 ± 0.05 mg% in both extracts, respectively. 5 kinds and 6 kinds of phenolic acids were detected in both extracts, respectively. In the case of flavonoid analysis, no flavonoid compound was detected in the hot water extract while taxifolin was identified in the 95% ethanol extract. Based on the above results, the hot water and 95% ethanol extracts of I. cylindrica Beauvois var. koenigii root are rich in free amino acids, amino acid derivatives, phenolic acids and flavonoids which confirm their potential for applications in cosmeceuticals, nutricosmetics formulations and functional foods.
