Polysulfone/nanocomposites mixed matrix ultrafiltration membrane for the recovery of Maillard reaction products

  • Maillard reaction products like melanoidins present in industrial fermentation wastewaters are complex compounds with various functional properties. In this work, novel ultrafiltration (UF) mixed matrix membrane (MMM) composed of polysulfone (PSF) and nanocomposites was prepared through a phase inversion process for the recovery of melanoidins. Nanocomposites were prepared with acid functionalized multiwalled carbon nanotubes (MWCNTs) as the reinforcing filler for chitosan-thermoplastic starch blend. Higher nanocomposites content in the PSF matrix reduced the membrane permeability and melanoidins retention indicating tighter membrane with surface defects. The membrane surface defects could be sealed with dilute polyvinyl alcohol (PVA) solution. The best performing membrane (1% nanocomposites in 18% PSF membrane sealed with 0.25% PVA coating) resulted in uniform melanoidins retention of 98% and permeability of 3.6 L/m2 h bar over a period of 8h. This demonstrates a low fouling PSF membrane for high melanoidins recovery.

  • The king oyster mushroom (Pleurotus eryngii) is widely consumed because of its flavor, texture, and its functional properties such as antioxidant activity and prebiotic effects. However, long-term product storage and transportation (e.g., export) are difficult because of its limited durability. The shelf-life of king oyster mushroom is affected by environmental factors such as temperature, humidity, gas composition, and ventilation, which may affect sensory characteristics including respiration rate, texture, moisture, flavor, color, and pH. The major problems regarding storage of mushrooms are browning, flavor changes, and softening. To address these problems, novel preservation techniques were developed, and more durable variants were bred. Different drying methods, gamma irradiation, chitosan coating, modified atmosphere (MA) packaging, and controlled atmosphere (CA) storage were evaluated in order to extend the shelf-life of king oyster mushrooms. Freeze drying showed better results for the preservation of mushrooms than other drying methods. Irradiation with 1 kGy was more effective for extending mushroom shelf-life than higher doses. The preservative performance of chitosan-based films was improved by combining the compound with other hydrocolloids, such as oil, protocatechuic acid, and wax. The CA storage conditions recommended for king oyster mushrooms are 5kPa $O_2$ and 10 to 15kPa $CO_2$ at temperatures below $10^{\circ}C$. Active MA packaging with microperforated PP film was also effective for maintaining quality during storage.

Spray-Dried Powder Preparation and Physicochemical Properties of Green Coffee Bean Extracts (커피 생두 추출물을 이용한 분무건조 분말의 제조 및 물리화학적 품질특성)

  • In this study, the physicochemical properties of freeze dried (FD) and spray dried powders (SD-MD10, SD-MD20, SD-CD10, SD-CD20) from green coffee bean extract were investigated. Yields were 91.46% for FD powder and 71.63~87.98% for SD powders, respectively. The moisture content of SD powders (1.39~1.86%) was lower than that of FD powder (3.19%). L, a and b value of SD powders were higher than of FD powder. The particle size of the FD powder ($147.67{\mu}m$) was higher than that of the SD powders ($54.23{\sim}66.07{\mu}m$). The water absorption index of the SD powders (0.51-0.59) were lower than that of the FD powder (0.72). The water solubility index of the SD powders, 77.26-90.07%, was higher than that of the FD powder (70.07%). The total phenolic and flavonoid contents of SD-CD10 were 20.67 g/100 g and 8.92 g/100 g, respectively. The chlorogenic acid content of SD-CD10 (12.04 g/100 g) was higher than other SD powders. The DPPH and superoxide radical scavenging activities of SD-CD10 at $1,000{\mu}g/mL$ were 88.02% and 86.89%, respectively. The SD-CD10 was better than other SD powders on protective effects against oxidative stress in L-132 cells (87.33%). In conclusion, we provided experimental evidence that spray-dried powder of green coffee bean extracts have potential as functional food materials.

Processing of Functional Mackerel Fillet and Quality Changes during Storage (기능성 고등어 Fillet제조 및 저장 중 품질 변화)

  • In order to endow mackerel fillet with antioxidant effect, functionality and remove fishy smell, chitosan, oligosaccaride, extracts of green tea, dill weed and ginger was used as soaking solution additives. Quality characteristics of soaked mackerel fillet(SMF) was investigated during storage temperature and time after vacuum packing with polyvinyl chloride film. From the results of VBN measurement, initial spoilage was showed within 2 or 3 weeks in SMF soaked with brine including extracts of green tea, herb and ginger at 5$^{\circ}C$. In case of adding chitosan and oligosaccarides to the solution mentioned above, initial spoilage was found from 4 to 7 weeks, oxidation was reduced remarkably and SMF stored at -20$^{\circ}C$ maintained the freshness during 80 days. Viable cell counts were reached to 108 for 3 weeks storage at 5 and 0$^{\circ}C$. But no change of viable cell counts was founded at -20$^{\circ}C$. The shelf-lifes of SMF according to 5, 0 and -20$^{\circ}C$storage temperatures were 2 to 3, 4 to 7 weeks and about 3 months, respectively.

Functional Properties of Sulfated Polysaccharides in Ascidian(Halocynthia roretzs) Tunic (우렁쉥이 껍질 중 황산화다당의 기능적 특성)

  • Functional properties such as anti-blood coagulation, angiotensin converting enzyme (ACE) inhibitory activity, fat binding capacity, foaming properties, emulsifying properties and chemical components of sulfated polysaccharides isolated from ascidian tunics were investigated. The sulfated polysaccharide mainly consisted of sulfate, uronic acid, protein, and chondroitin sulfate, among which chondroitin sulfate showed higher concentration while sulfate and uronic acid did lower. Compositional menosaccharides were arabinose, xylose, glucose, galactose, glucuronic acid, N-acetylgalactosamine and N-acetyglucosamine. Especially, galactose content was dominant among them. And emulsifiability and foaminess of the sulfated polysaccharide was higher than the control group. Anti-blood coagulation of sulfated polysaccharide showed with respect to APTT (Activated partial thromboplastin time). ACE inhibitory activity showed about $16.7\%$.

  • The present study was conducted to elucidate functional properties of sulfated polysaccharides from ascidian tunics, In physical properties of the crude polysaccharides, emulsion ability and foaminess were more excellent compared with chitin and chitosan, particular dye binding capacity was prominent. Anti-blood coagulation of partially purified sulfnted polysaccharides showed with respect to APTT (Activated partial thromboplastin time). Especially, active fraction $(F_4)$ obtained by DEAE-cellulose ion exchange chromatography showed highest activity, which was approximately $20\%$ of the activity of heparin. ACE inhibitory activity also similar to anticoagulant activity. Active fraction $(F_4)$ obtained by DEAE-cellulose ion exchange chromatography showed about $34\%$, in ACE inhibitory activity.

Development of Disposable Immunosensors for Rapid Determination of Sildenafil and Vardenafil in Functional Foods

  • We introduced disposable amperometric immunosensors for the detection of Sildenafil and Vardenafil (SDF/VDF) based on screen printed carbon electrodes. The developed immunosensors were used as a non-competitive sandwich-type enzyme immunoassay with a horseradish peroxidase label. The sensors were constructed on screen printed carbon electrodes by the simple electrochemical deposition of a reduced graphene oxide and chitosan (ErGO-CS) composite. To evaluate the sensing chemistry and optimize the sensor characteristics, a series of electrochemical experiments were carried out including electrochemical impedance spectroscopy, cyclic voltammetry and amperometry. The sensors showed a linear response to SDF/VDF concentrations in a range from 100 pg/mL to 300 ng/mL. The lower detection limit was calculated to be 55 pg/mL, the sensitivity was calculated to be $1.02{\mu}Ang/mL/cm^2$, and the sensor performance exhibited good reproducibility with a relative standard deviation (RSD) of 7.1%. The proposed sensing chemistry strategy and the sensor format can be used as a simple, cost-effective, and feasible method for the in-field analysis of SDF/VDF in functional or health supplement food samples.

Effect of Pretreatment on Dyeability and Functionalities of Summer Rayon fabrics Finished by Gallnut Extract (전처리가 오배자 추출물에 의한 여름용 인견직물의 염색 및 기능성 향상에 미치는 영향)

  • Viscose rayon filament fabrics have been called 'artificial silk' and beloved as summer clothing materials for a long time in Korea. This is because the viscose rayon filament fabrics reveals glossy surface and cool touch feeling compared to other conventional fabrics composed of staple fibers. Therefore, we tried to prepare the higher value added viscose rayon filament fabrics for summer textile products. In this study, we applied gallnut extract to the viscose rayon filament fabric to develop summer fabrics with natural color and multi-functions such as antibacterial and antioxidant properties. This process also pursue eco-friendly and multi-functional fabric finishing from the natural material "gallnut". In addition, various pre-treatment with cationizer, chitosan, or chito-oligomer was applied to the finishing process to improve the finishing efficacy and durability. Consequently, it was found that the active component of gallnut extract was successfully incorporated to the viscose rayon filament fabric through a pad-dry-cure process. And, the treated viscose rayon filament fabrics showed excellent antibacterial and antioxidant properties. Therefore, it was expected that the rayon filament fabrics treated by gallnut extract could be used as effective summer fabrics preventing the growth of bacteria and skin ageing as well as providing cool touch feeling. However, the pre-treatments were not that meaningful on the functionalities but effective on coloring.

Polysaccharide-based superhydrophilic coatings with antibacterial and anti-inflammatory agent-delivering capabilities for ophthalmic applications

  • Medical silicone tubes are generally used as implants for the treatment of nasolacrimal duct stenosis. However, side effects such as allergic reactions and bacterial infections have been reported following the silicone tube insertion, which cause surgical failure. These drawbacks can be overcome by modifying the silicone tube surface using a functional coating. Here, we report a biocompatible and superhydrophilic surface coating based on a polysaccharide multilayer nanofilm, which can load and release antibacterial and anti-inflammatory agents. The nanofilm is composed of carboxymethylcellulose (CMC) and chitosan (CHI), and fabricated by layer-by-layer (LbL) assembly. The LbL-assembled CMC/CHI multilayer films exhibited superhydrophilic properties, owing to the rough and porous structure obtained by a crosslinking process. The surface coated with the superhydrophilic CMC/CHI multilayer film initially exhibited antibacterial activity by preventing the adhesion of bacteria, followed by further enhanced antibacterial effects upon releasing the loaded antibacterial agent. In addition, inflammatory cytokine assays demonstrated the ability of the coating to deliver anti-inflammatory agents. The versatile nanocoating endows the surface with anti-adhesion and drug-delivery capabilities, with potential applications in the biomedical field. Therefore, we attempted to coat the nanofilm on the surface of an ophthalmic silicone tube to produce a multifunctional tube suitable for patient-specific treatment.

Immune response to Salmonella Gallinarum experimentally infected layers fed with Guanosine 5'-monophosphate-chelated calcium and iron feed additives (Guanosine 5'-monophosphate 킬레이트 칼슘 및 철 사료 첨가제 급이 산란계의 Salmonella Gallinarum 인공감염에 대한 면역 반응)

  • The objective of this research was to evaluate the immune response to Salmonella Gallinarum experimentally infected layers fed with Guanosine 5'-monophosphate-chelated calcium and iron feed additives. Hy-Line brown, 34 week-olds layers were assigned to 3 groups; Group 1: basal diet feed, Group 2 (CaFe-GMP): basal diet feed mixed with chelated calcium and iron, and Group 3 (Fe-OCHT): basal diet feed mixed with chitosan for 4 weeks. There were challenged with 1.0×108 CFU/mL of the cultured Salmonella Gallinarum (SG) by oral administration on 28th feeding days. After SG challenge, Flow cytometric profiles showed that the CD4+/CD8+ T lymphocyte activation of Group 2 was much higher than Group 1 and Group 3 (P<0.05). In addition, the levels of interleukin-2 (13.37 mg/dl) and interferon-γ (2.35 mg/dl) in Group 2 were higher than Group 1 and Group 2. Populations of Lactic acid bacteria (3.5×1010 CFU/g) from cecum was highest observed in group 2. Re-isolation of SG from cecum in group 2 (8×105 CFU/g) was lower than group 1 (1.83×1010 CFU/g). The result of this study demonstrated that CaFe-GMP feed additive may be one of the potential candidates to control salmonellosis and functional feeds in layers.
