Survey on Fusarium Mycotoxin Contamination in Oat, Sorghum, Adlay, and Proso Millet during the Harvest Season in Korea (귀리, 수수, 율무, 기장의 수확기에 발생하는 Fusarium 곰팡이독소 오염도 조사)

  • A total of 244 cereal samples (oat, sorghum, adlay, and proso millet) were collected from fields to examine the contamination of Fusarium mycotoxins in cereals during harvest season in 2017 and 2018. The contamination levels of deoxynivalenol (DON), nivalenol (NIV), and zearalenone (ZEA) were analyzed individually by using the immunoaffinity column clean-up method with ultra performance liquid chromatography, and fumonisins (FUM) were analyzed by using the QuEChERS method with liquid chromatography-mass spectrometry. Highest level of NIV contamination (120.0-3277.0 mg/kg) was observed in oat samples among the analyzed cereals. In the adlay samples, DON contamination was the highest (maximum level 730.0 ㎍/kg). The proso millet samples had a high frequency of detection of NIV and ZEA (61.5% and 57.9%, respectively), but the levels were low (average detection level of NIV, 75.6 ㎍/kg, for ZEA, 21.5 ㎍/kg). Among the cereal samples, sorghum had the highest contamination frequency of DON, ZEA, and FUM, and the co-occurrence of Fusarium mycotoxin was 70.0%, which was higher than the average of 29.9%. In order to safely manage Fusarium mycotoxin levels in cereals, continuous research on the development of contamination prevention technologies together with monitoring of mycotoxin contamination is needed.

  • Objective: The objective was to characterize the fermentative and microbiological profile of Tifton 85 bermudagrass haylage with different layers of polyethylene film and storage time. Methods: The experimental design consisted of a randomized block design with four and six wrapping layers (100 and 150 microns in total. respectively) allocated in the main plots, through repeated measures analysis (30, 60, and 90 days of storage) with four replicates. Results: The storage time and number of wrapping layers did not show changes in the population of Clostridium and lactic acid bacteria. A decrease was observed in the enterobacteria population with an increase in the storage period in the two wrapping layers studied. Upon opening of the haylage at 30 days, the population of Bacillus was lower in haylages made with six layers of wrapping (3.63 log colony forming units/g). No growth of Listeria sp. or Salmonella sp. was observed during the experimental period. The fungal genera with a greater occurrence were Penicillium sp. and Fusarium sp. The following mycotoxins were not detected: ochratoxin A, fumonisins, and zearalenone. Relative to the organic butyric, propionic, and acetic acids, the haylages presented a low concentration of lactic acid; this may have prevented a drop in the pH, which was high when the silos were opened (5.4). The levels of ammoniacal nitrogen and soluble carbohydrates presented no variation among the number of wrapping layers, with an overall average of 35.55 and 38.04 g/kg. Conclusion: Tifton 85 bermudagrass haylage wrapped with four and six layers presented adequate fermentation and microbiological characteristics in the evaluated periods.

Differential Effects of Fumonisin $B_1$ on Cell Death in Cultured Cells: the Significance of the Elevated Sphinganine

  • Fumonisins are specific inhibitors of ceramide synthase in sphingolipid metabolism. An alteration in sphingolipid metabolism as a result of fumonisin exposure is related to cell death (Yoo et al., 1992). The objective of this study was to investigate whether elevated free sphinganine levels are related to the sensitivity of cultured cells to fumonisin exposure. Fumonisin $B_1$ elevated the intracellular free sphinganine concentraions in both LLC-$PK_1$ and Chinese hamster ovary (CHO) cells. However, CHO cells are resistant to fumonisin cytotoxicity at 50${u}m$, while LLC-$PK_1$ cells are sensitive at concentrations greater than 357M. The intracellular concentration of free sphinganine in LLC-$PK_1$ cells treated at 50${u}m$ fumonisin $B_1$ for 72 h was approximately 1450 pmol/mg protein relative to the 37 pmol observed in the control culture. Under the same conditions, the population of apoptotic cells in the 50${u}m$ fumonisin $B_1$-treated culture was approximately 37% of the total compared to 12% in the control. The caspase III-like activity after 72 h in the 50${\mu}$M fumonisin $B_1$-exposed culture Increased to approximately 50 $pmol/mg$ protein/hr compared to 6 $pmol/mg$ protein/hr in the control. L-cycloserine, a serine palmitoyltransferase inhibitory reduced the fumonisin $B_1$-stimulated caspase III-like activity down to the control level. Under the same culture conditions, the intracellular concentration of free sphinganine after-cycloserine plus fumonisin $B_1$ treatment was 140 pmol/mg protein compared to 1450 $pmol/mg$ protein in fumonisin $B_1$ alone. The intracellular concentration of free sphinganine in CHO cells treated with 50${u}m$ fumonisin $B_1$ for 72 h was al)proximately 460 pmol/mg protein, indicating that the mass amount of elevated free sphinganine in the CHO cells was about 32% of that in LLC-$PK_1$ cells. Adding exogenous sphinganine to the CHO cells along with 50${u}m$ fumonisin $B_1$ treatment for 72 h caused both necrosis and apoptosis. In conclusion, the elevated endogenous sphinganine acts as a contributing factor to the fumonisin-induced cell death.

  • Over recent years, it has become evident that food and agricultural products are easily contaminated by fungi of Aspergillus, Fusarium, and Penicillium due to rapid climate change, which is not only a global food quality concern but also a serious health concern. Owing to consumers' interest in health, resistance to preservatives such as propionic acid and sorbic acid (which have been used in the past) is increasing, so it is necessary to develop a substitute from natural materials. In this review, the role of lactic acid bacteria as a biological method for controlling the growth and toxin production of fungi was examined. According to recent studies, lactic acid bacteria effectively inhibit the growth of fungi through various metabolites such as organic acids with low molecular weight, reuterin, proteinaceous compounds, hydroxy fatty acids, and phenol compounds. Lactic acid bacteria effectively reduced mycotoxin production by fungi via adsorption of mycotoxin with lactic acid bacteria cell surface components, degradation of fungal mycotoxin, and inhibition of mycotoxin production. Lactic acid bacteria could be regarded as a potential anti-fungal and anti-mycotoxigenic material in the prevention of fungal contamination of food and agricultural products because lactic acid bacteria produce various kinds of potent metabolic compounds with anti-fungal activities.

Toxicity and Carcinogenicity of the Fusarium moniliforme MRC 826 Culture Material in Rats (랫드에서 Fusarium moniliforme MRC 826 배양물질의 독성 및 발암성에 관한 연구)

  • F. moniliforme MRC 826, a common fungal contaminant of com, has been known to produce a group of mycotoxins, the fumonisins. By thin layer chromatography, fumonisin $B_{1}$ was detected in the F. moniliforme MRC 826 com culture material(CM) extracts. This study was performed to compare the toxicity and carcinogenicity of F. moniliforme MRC 826 CM with those of aflatoxin $B_1(AFB_1)$ in rats. The toxicity was tested over a period of 7 days in ten female Sprague-Dawley (SD) rats. Treatment group were fed a 1 : 1 mixture(wt/wt) of ground CM and basal diet in powder form, while other negative control group were given basal diet alone. The principal pathological changes in rats treated with 50% CM were hepatocellular hydropic degeneration and renal tubular necrosis. The cancer-promoting activity of CM was evaluated in the rat liver diethylnitrosamine-two thirds partial hepatectomy(DEN-PH) model for carcinogenesis. 70 male SO rats(ca. 170 g) were randomized into 5 groups. Group I served as the positive controls and received the basal diet containing 2 ppm $AFB_{1}$ group 2 received 5% CM, group 3 received 2.5% CM, group 4 received 5% normal com and group 5 received 2.5% normal com. 5% treated group showed cancer promoting activity in rat liver using DEN as initiator and the induction of glutathione S-transferase placental form positive foci as an end point after 6 weeks of promotion.

A Study on the Safety of Mycotoxins in Grains and Commonly Consumed Foods (곡류 등 다소비 식품 중 곰팡이독소 안전성 조사 연구)

  • The purpose of this study was to investigate and evaluate the safety of the grains, nut products, beans and oilseeds being sold in Gyeonggi province by analyzing mycotoxins. A multi-mycotoxins analysis method based on LC-MS/MS was validated and applied for the determination of eight mycotoxins, including aflatoxins ($B_1$, $B_2$, $G_1$ and $G_2$), fumonisins ($B_1$, $B_2$), zearalenone and ochratoxcin A in 134 samples. The limit of detection (LOD) and limit of quantitation (LOQ) for the eight mycotoxins ranged from 0.14 to $8.25{\mu}g/kg$ and from 1.08 to $7.21{\mu}g/kg$, respectively. Recovery rates of mycotoxins were determined in the range of 61.1 to 97.5% with RSD of 1.0~14.5% (n=3). Fumonisin $B_1$, $B_2$, zearalenone, and ochratoxin A were detected in 22 samples, indicating that 27% of grains, 12.5% of beans and 11.8% of oilseeds were contaminated. Fumonisin and zearalenone were detected simultaneously in 2 adlays and 3 sorghums. Fumonisin $B_1$ and $B_2$ were detected simultaneously in most samples whereas fumonisin $B_1$ was detected in 1 adlay, 1 millet and 1 sesame sample. The average detected amount of fumonisin was $49.3{\mu}g/kg$ and $10.1{\mu}g/kg$ for grains and oilseeds, respectively. The average detected amount of zearalenone was $1.9{\mu}g/kg$ and $1.5{\mu}g/kg$ for grains and beans, respectively. In addition, the average amount of ochratoxin A was $0.08{\mu}g/kg$ for grains. The calculated exposure amounts of fumonisin, zeralenone and ochratoxin A for grains, beans and oilseeds were below the PMTDI/PTWI.
