Studies on the Organic Acids Composition in Shellfishes 2. Nonvolatile Organic Acids Composition of Oyster, Sea-mussel, Baby Clam, Hen Clam and Their Boiled-dried Products (패류의 유기산 조성에 관한 연구 2. 굴, 홍합, 바지락, 개량조개 및 그 자건품의 비휘발성유기산 조성)

  • In succession to the previous paper, the present study was directed to investigate the nonvolatile organic acids composition in raw and belied-dried products of oyster, sea-mussel, baby clam and hen clam by gas liquid chromatography. The results obtained are summarized as follows : In four kinds of the samples examined, eight kinds of organic acids were identified and determined in oyster, sea-mussel and baby clam, and nine kinds in hen clam. The major organic acids in oyster were pyroglutamic, succinic and malic acid which was $94.2\%$ of total quantity of organic acid, and those in sea-mussel, baby clam and hen clam were succinic and malic acid which were $90.8\%,\;89.7\%\;and\;86.4\%$ of total acids, respectively. The most abundant organic acid in sea-mussel, baby clam and hen clam was succinic acid that was $80.6\%,\;84.9\%\;and\;73.2\%$ in total acids, repectively. And that of oyster was pyroglutamic acid which marked $38.8\%$ in total acids, and the next one was succinc acid marked $34.4\%$. In the total quantity of organic acid, the highest was 913.0 mg/100g in oyster which showed 4.5 times as much as in hen clam, followed by 478.4 mg/100g in sea-mussel, 246.3mg/100g in baby clam, and the least was 201.2 mg/100g in hen clam. The decreasing rate of total quently of organic acids by boiled-dried procersing was the highest in oyster, $54.7\%$, followed by $46.5\%$ in sea-mussel, $37.1\%$ in hen clam and $29.4\%$ in baby clam. The decreasing rate of each organic acid shelved much difference according to the samples examined, in general, great in malic, fumaric and proglutamic acid ana less in succinic, lactic and oxalic acid.

Comparison of Nutritional Compositions between Amaranth Baby-Leaves Cultivated in Korea (국내 재배 아마란스 어린잎의 영양성분 비교)

  • In the present study, we compared and investigated the nutritional compositions of Amaranthus cruentus and Amaranthus hypochondriacus baby-leaves cultivated in Korea. Baby-leaves of two amaranthes consisted of more than 92% moisture, and A. cruentus contained a higher amount of moisture than A. hypochondriacus. Meanwhile, A. hypochondriacus contained higher levels of crude ash, crude protein, crude lipid, carbohydrates, and dietary fiber than A. cruentus. The major free sugars of the two amaranth baby-leaves were fructose and glucose. Fructose content of A. hypochondriacus was higher than that of A. cruentus, and glucose content of A. cruentus was higher than that of A. hypochondriacus. Acetic acid, malic acid, and fumaric acid were detected in two amaranth leaves, but succinic acid was not detected. Two amaranth leaves contained 17 amino acids except for methionine, proline, and tyrosine, and leaves contained the highest glutamic acid contents. In addition, A. cruentus and A. hypochondriacus leaves contained high contents of taurine and ${\gamma}$-aminobutyric acid and showed various biological activities. The major mineral and fatty acid of the two amaranth leaves were potassium and linolenic acid (C18:3), respectively. The ${\beta}$-carotene contents of A. cruentus and A. hypochondriacus leaves were $478.72{\mu}g/100g$ and $474.12{\mu}g/100g$, respectively. In vitamin B complex, $B_2$, $B_3$, and $B_5$ were detected in the two amaranth leaves whereas vitamins $B_1$, $B_6$, and $B_{12}$ were not detected. A. hypochondriacus contained higher amounts of vitamin C and E than those of A. cruentus. Overall, amaranth leaves contained high amounts of nutritional components. Therefore, amaranth leaves are expected to be useful for the development of a functional food. Moreover, these results will provide fundamental data for advancing sitological value, breeding new cultivars, and promoting leafy vegetable usage.

Effects of Dietary Supplementation of Organic Acid and Antibiotics Mixture on Growth Performances and Blood Metabolites in Growing Pigs (사료내 유기산제 및 항생제의 혼합첨가가 육성돈의 성장과 혈액성상에 미치는 영향)

  • This study was carried out to evaluate the effect of replacing antibiotics by organic acid mixture on growth performances and blood metabolites in growing pigs. Twenty-five crossbred pigs (Large White${\times}$Landrace) at the age of 79 days were fed five different diets by supplementing organic acid mixture and chlortetracycline. The experimental diets were consisted of diets without antibiotics supplementation (control), diets added 100mg/kg of chlortetracycline to control diet (T1), diets added 100mg/kg of chlortetracycline and 0.1% of $Acidomix^{(R)}$ (comprising formic acid 25%, sorbic acid 10%, fumaric acid 10%) to control diet (T2), diets added 0.1% of $Acidomix^{(R)}$ to control diet (T3), and diets added 0.3% of $Acidomix^{(R)}$ to control diet (T4). The changes in feed conversion ratio, average daily gain and blood metabolites were investigated. Twenty-five pigs were allotted to five treatments with five replications of each and the experiment was conducted on the basis of complete randomized design for 6 weeks. Average daily gain was significantly (p<0.05) different between T4 and control diets. All treatments including diets added $Acidomix^{(R)}$ and chlortetracycline were slightly higher than control diets. The feed intakes did not show a significant difference between the control and other treatments, and did not give change in feed intake by the addition of $Acidomix^{(R)}$. No differences on feed conversion ratio among treatments were observed but T4 and T3 treatments showed lower value than other treatments. The concentrations of blood urea nitrogen, total cholesterol and triglyceride were significantly (p<0.05) different among treatments, but their values were within the normal range representing that effects on blood profile by organic acid or antibiotics supplementation were not found. The results from this study indicated that adding chlortetracycline or organic acid mixtures to diets showed tendency to improve average daily gain and feed conversion ratio in growing pigs. These results showed that antibiotics could be replaced by organic acid mixture in growing pig diets, leading to stimulated growth and improved feed conversion ratio.

Changes in Quality of Welsh Onion (Allium fistulosum L.) during the Freezing Storage Period under Different Freezing Conditions (냉동조건에 따른 대파의 냉동저장 중 품질변화)

  • Purpose: This study was conducted to find the optimal freezing method and storage conditions for welsh onion. Methods: Cut welsh onions (0.3 cm) were packed in nylon/linear low density polyethylene (LLDPE) film bags, and frozen utilizing still-air freezing at -$20^{\circ}C$ (SAF20) and -$40^{\circ}C$ (SAF40), and immersed-liquid freezing at -$40^{\circ}C$ (ILF40); they were then stored at -$20^{\circ}C$ for 7 months. During storage, quality characteristics were measured monthly. Results: Drip loss was the lowest in the ILF40 packaging. Color difference in the stem (white part) did not differ significantly according to freezing conditions and storage time. Color difference in the leaf (green part) and stem was the lowest in SAF20. pH remained unchanged, while total aerobic bacterial count, pyruvic acid and moisture content decreased during storage. Pyruvic acid content of ILF40 was the highest among the freezing treatments. Fructose and glucose contents increased gradually during storage. Citric acid, malic acid, succinic acid and fumaric acid contents were unaffected, regardless of the freezing conditions. Conclusion: The optimal freezing method for welsh onions with the least quality changes was determined to be immersed liquid freezing, following by preservation up to 7 months by freeze-storing.

Development of succinate producing Cellulomonas flavigena mutants with deleted succinate dehydrogenase gene

  • This study was performed to produce succinic acid from biomass by developing mutants of Cellulomonas flavigena in which the succinate dehydrogenase gene (sdh) is deleted. For development of succinate producing mutants, the upstream and downstream regions of sdh gene from C. flavigena and antibiotic resistance gene (neo, bla) were inserted into pKC1139, and the recombinant plasmids were transformed into Escherichia coli ET12567/pUZ8002 which is a donor strain for conjugation. C. flavigena was conjugated with the transformed E. coli ET12567/pUZ8002 to induce the deletion of sdh in chromosome of this bacteria by double-crossover recombination. Two mutants (C. flavigena H-1 and H-2), in which sdh gene was deleted in the chromosome, were constructed and confirmed by PCR. To estimate the production of succinic acid by the two mutants when the culture broth was fermented with biomass such as CMC, xylan, locust gum, and rapeseed straw; the culture broth was analyzed by HPLC analysis. The succinic acid in the culture broth was not detected as a fermentation products of all biomass. One of the reasons for this may be the conversion of succinic acid to fumaric acid by sdh genes (Cfla_1014 - Cfla_1017 or Cfla_1916 - Cfla_1918) which remained in the chromosomal DNA of C. flavigena H-1 and H-2. The other reason could be the conversion of succinyl-CoA to other metabolites by enzymes related to the bypass pathway of TCA cycle.

Properties of Amino Acid and Volatile Flavor Compounds of Fermented Soybean Products by Soybean Cultivar (콩 품종에 따른 발효물의 아미노산과 향기성분 특성)

  • In this study, we analyzed the nutritional composition properties of soybeans and the organic acids, amino acids and volatile flavor compounds of fermented soybean products. We used five soybean cultivars including Pyeongwon, Jinpung, Saedanbaek, Saeolkong and Cheonga for this experiment. Physicochemical analysis of soybeans, showed that the cured protein and fat contents were 35.12~45.12 and 14.26~20.14%, respectively. The rank order of major organic acids was lactic acid > acetic acid > fumaric acid, with Saedanbaek being the highest. Total amino acid content of the samples was 358.12~657.28 mg/100 g, and glutamic acid, alanine, cysteine, valine, leucine, histidine and arginine were the major amino acids. We identified a total of 34 volatile aroma-compounds, including 7 alcohols, 7 acids, 7 ketones, 5 phenols, 2 esters, 1 furan, 4 pyrazines, and 1 miscellaneous compounds. As a result of this, could be applied to determine the suitability of cultivars and the quality for the process of the fermented soybean products.

Modification of Starch for Surface Sizing via Etherification and Esterification (에테르화 및 에스테르화를 통한 표면사이징용 변성 전분의 제조)

  • An oxidized starch was modified for surface sizing via etherification and esterification. Propylene oxide (PO), sodium monochloroacetate (SMCA), and acrylonitrile (AN) were used as etherification, and vinyl acetate monomer (VAM), maleic anhydride (MA), fumaric acid (FA), and itaconic acid (ITA) were used for esterification. Esterification and etherification of starch decreased both Brookfield viscosity and Brabender viscosity substantially even though the solids level was increased by 2% from 14 to 16%. Surface sizing performance of starches in tensile strength, stiffness and compressive strength was improved by esterification and etherification of the oxidized starch. Especially, SMCA etherification was found to be the most effective modification method.

Study on the Development of Horticultural Media using Recyled Used-mushroom-media (버섯폐배지를 재활용한 원예용 상토 개발에 관한 연구)

  • As for chemical characteristics of cultivated media waste, the total content of nitrogen, phosphoric acid, and potassium was 6.93%; organic matter content was 67.78%; pH and EC were 7.20 and 1.46 ds/m, respectively. Organic acids identified during stabilization included citric acid, oxalic acid, fumaric acid, and acetic acid, which was highest. During the stabilization of cultivated media waste, temperature decreased and pH increased on the 15th day, and the germination index was 88 after the period, indicating that at least 15 days of stabilization is needed when cultivated media waste is used for growth material of crop. As for germination characteristics, when the mixture rate of cultivated media waste was 20% and under, germination of cucumber and oriental melon was favorable without difference compared to the control plots. For lettuce and Chinese cabbage, no statistically significant difference was found when the rate was 10% and under compared to the control plots. As for seedling quality after 20 days of sowing, cucumber, oriental melon, and Chinese cabbage showed favorable growth only when the mixture rate of cultivated media waste was 10% and under; lettuce showed lower growth characteristics regardless of mixture rate when compared to the control plots. Cultivated media waste may be significantly useful for cucumber and oriental melon, but not for Chinese cabbage and lettuce.

Organic acidemias in Korea (한국의 유기산혈증)

  • Since we have started organic acid analysis on Jul. 1997, we have been collecting data about organic acidemias in Korea. The data presented here is our 3 years experience in organic acid analysis. We have collected 712 samples from major university hospitals all over the Korea, large enough for relatively accurate incidence of organic acid disorders. We are using solvent extraction method with ethylacetate, MSTFA for derivatization and quantitation of 83 organic acids simultaneously. Out of 712 patients sample, 498 patients sample (70%) showed no evidence of organic acid abnormalities. Out of 214 remaining samples we have found very diverse disorders such as methylmalonic aciduria(6), propionic aciduria (10), biotinidase deficiency (6), maple syrup urine disease (3), isovaleric aciduria (4), tyrosinemia type II (4), tyrosinemia type IV (1), glutaric aciduria type I (1), glutaric aciduria type II (22), 3-methylglutaconic aciduria type I (3), 3-methylglutaconic aciduria type III (7), HMG-CoA lyase deficiency (1), hyperglyceroluria (2), cytosolic 3-ketothiolase deficiency (55), mitochondrial 3-ketothiolase deficiency (3), 3-hydroxyisobutyric aciduria (2), L-2-hydroxyglutaric aciduria (2), fumaric aciduria (2), lactic aciduria with combined elevation of pyruvate (most likely PDHC deficiency) (28), lactic aciduria without combined elevation of pyruvate (most likely mitochondrial respiratory chain disorders) (35), SCAD deficiency (3), MCAD deficiency (1), 3-methylcrotonylglycineuria (1), orotic aciduria (most likely urea cycle disorders) (7) and 2-methylbranched chain acyl-CoA dehydrogenase deficiency (1). In conclusion, though the incidence of indivisual organic acidemia is low, the incidence of overall organic acidemia is relatively high in Korea. Most of the patients showed some signs of neurological dysfunction. In other words, organic acid analysis should be included in the diagnostic work up of all neurological dysfunctions.

Flavor Characteristics of Kakdugi by Radish Cultivars and Seasons (무 품종 및 계절에 따른 깍두기의 향미특성)

  • Chemical and sensory characteristics of kakdugi which was prepared with various radish cultivars and harvesting seasons were analyzed during 7-day storage. Average pH of small radish cultivar kakdugi was higher than that of large ones, and total acidity was lower in small ones. Reducing sugar content was the highest in kakdugi of autumn radish. Organic acids such as lactic, succinic and fumaric acid analyzed by GC increased until the third day of fermentation, whereas volatile isothiocyanates analyzed by GC/MS continued to decline. There was a significant difference in flavor characteristics of large radish kakdugi across seasons, in contrast to no significant difference in those of small radish kakdugi except sweet taste and reducing sugar content in which interaction existed between season and cultivar. Score of overall acceptability was higher in small radish than large ones with Dongja showing the highest score of overall acceptability. Overall acceptability of autumn Dongja kakdugi was positively correlated with radish kakdugi odor and sour odor, respectively, but negatively with total acidity, lactic acid content, sweet taste and pungency, respectively. By multiple regression analysis, overall acceptability in spring Dongja kakdugi is expressed as a function of overall acceptability = -0.1115 + 1.2519 savory taste + 1.5159 malic acid -0.0054 total isothiocyanate + ${\varepsilon}$.
