Isolation and Characterization of Exopolysaccharide-Producing Bacteria from Korean Fermented Vegetables (전통 침채류 유래 다당 생산균의 분리 특성)

  • Four bacteria producing viscous exopolysaccharides (EPSs) were isolated from Korean fermented vegetables (Cucumber kimchi, Young radish kimchi, Green onion kimchi) using a selection medium intended for isolating bacteria with tannin-degrading activity. They were identified phylogenetically by 16S rDNA sequence analysis and found to be very close to Enterobacter cowan ii, Escherichia senegalensis, Enterobacter asburiae, and Enterobacter ludwigii. Strain CK31, the most efficient EPS-producer, produced a heteropolysaccharide with an approximate molecular weight of 420 kDa. The neutral sugar fraction of the EPS was composed of rhamnose, fucose, arabinose, mannose, galactose, and glucose.

  • We have isolated an antithrombin active polysaccharide in red ginseng by procedures comprising three major steps involving alkaline extraction, anion exchange and gel permeation chromatography. Active polysaccharide behaved as a single band on cellulose acetate membrane electrophoresis. The average molecular mass was estimated to be about 177 kDa by gel filtration. This polysaccharide was found to be an acidic heteropolysaccharide that contains uronic acid moiety $(40.2\%)$, sulfate group $(9.2\%)$ and protein $(1.5\%)$ in addition to neutral sugar consisted of rhamnose, mannose, galactose, arabinose, glucose, fucose and xylose in a molar ratio of 1.00 : 0.88 : 0.86 : 0.78: 0.70 : 0.33 : 0.22. This polysaccharide inhibited blood coagulation via the intrinsic pathway like heparin in a dose-dependent manner. The clotting of fibrinogen by thrombin was also mitigated by the presence of this polysaccharide.

Glycoprotein in the Fruit Body of Sarcodon aspratus (능이자실체의 Glycoprotein)

  • This study was performed to investigate compositions of inorganic elements, amino acids and glycoprotein fractions as biological substances in fruit body of Sarcodon aspratus. The fruit body of Sarcodon aspratus contained Ca, Mg, Zn, Mn, Fe, Cu, and Pb, in particular high Ca and Na. Hot water extracts consisted of 54% of polysaccharide fraction and 32.6% of protein. In amino acids composition, fourteen free amino acids were detected, mainly glutamic acid, alanine and arginine. Fifteen kinds of total amino acids were contained with major components of glutamic acid, aspartic acid, serine and threonine. Concerned to glycoprotein extraction, 95% ethyl alcohol concentration gave the highest yields with 70.6% sugar fraction, 332% glycoprotein. Different ethyl alcohol concentration resulted in different protein precipitations, and lower concentration ethyl alcohol in the range of 30 to 70% gave more than 92% of higher sugar fraction. Crude glycoprotein (GP) was fractionated by P fraction of more than MW 300,000, P-1 fraction unadsorbed by DEAE-Sephadex, P-2 fractionated from P-1 by Sepharose 2B gel chromatography and P-3 fraction adsorbed by DEAE-Sephadex. Total sugars were increased and protein contents decreased during fractionation. GP and P-3 contained glucose, galactose, mannose and fucose. GP had high glucose with high contents of glutamic acid, serine, alanine and glycine. P-3 fraction contained high mannose with aspartic acid, glutamic acid, and glycine. P-2 fraction was 700,000 MW with high glucose and fucose, and low protein of 1.1%, high amounts of aspartic acid, glutamic acid and alanine, but no mannose and no cysteine.

Changes of Non-Cellulosic Neutral Sugars of Cell Wall in Soybean Sprouts (콩나물 생장중 세포벽 비섬유성 중성당의 변화)

  • This study was carried out to investigate the changes and composition of the non-cellulosic neutral sugars in cell wall of soybean sprouts during growth. The composition of non-cellulosic neutral sugars in cell of soybean sprouts was rhamnose, fucose, xylose, arabinose, mannose, galactose and glucose. The galactose content of cell wall was higher than other non-cellulosic neutral sugars, and was remarkably decreased during growth. The major non-cellulosic sugars of pectic substances were rhamnose, arabinose, and galactose. The arabinose content of pectic substance was increased in cotyledon and hypocotyl during growth. The contents of non-cellulosic neutral sugars were decreased in hypocotyl during growth. The galactose content of pectic substance was higher in cotyledon than those in hypocotyl, and was increased in cotyledon. The content of rhamnose was higher in ionically associated pectic substance than that in covalently bounded pectic substance. The major non-cellulosic neutral sugars of hemicellulose were glucose, rhamnose, arabinose and galactose. The galactose of hemicellulose was decreased remarkably during growth.

Human milk oligosaccharides: the novel modulator of intestinal microbiota

  • Human milk, which nourishes the early infants, is a source of bioactive components for the infant growth, development and commensal formulation as well. Human milk oligosaccharide is a group of complex and diverse glycans that is apparently not absorbed in human gastrointestinal tract. Although most mammalian milk contains oligosaccharides, oligosaccharides in human milk exhibit unique features in terms of their types, amounts, sizes, and functionalities. In addition to the prevention of infectious bacteria and the development of early immune system, human milk oligosaccharides are able to facilitate the healthy intestinal microbiota. Bifidobacterial intestinal microbiota appears to be established by the unilateral interaction between milk oligosaccharides, human intestinal activity and commensals. Digestibility, membrane transportation and catabolic activity by bacteria and intestinal epithelial cells, all of which are linked to the structural of human milk oligosaccharides, are crucial in determining intestinal microbiota.

  • 타액은 구강 환경을 조절하는 여러 가지 유기질과 무기질의 혼합물로 구성되어 있다. 구강 점막은 여러 타액 점액 단백질과 타액 항세균 단백질에 의해서 윤활이 되며 보호된다.타액의 다른 작용은 구강 점막을 축축하게 하고 음식을 부드럽게 한다. 구강건조증은 세균의 침착을 야기시키거나 점막면을 거칠게 하여 출혈이 되기 쉽게 하며 이로 인해 감염이 야기될 수 있다. 이러한 타액의 보호 작용은 mucin, fibronectins, proline-rich glycoproteins, histidine-rich proteins, $\alpha$-amylase, s-IgA 같은 특별한 타액 당단백질에 기인한다고 하는 것이 지난 30년 동안에 알려져 있다. 이러한 분자들의 구조, 구조와 기능사이의 관계, 타액 내 이러한 물질들의 농도에 관한 것들이 알려지고 있는 중이다. 이러한 타액 당단백질 특히 mucin, fibronectin, fucose-rich protein과 s-IgA의 구조와 기능에 대한 현재의 견해들을 이 논문에서 요약하고자 한다.

Characterization of Oilgosaccharide Moieties of Rat Intestinal Phytase

  • Phytase of rat intestine had a large amount of oilgosacchanrides ; The enzyme consisted of two different subunits with the molecular weights of 90 KDa and 70 KDa in its intack form, whereas the apparent molecular weights tumed to 72 KDa and 52 KDa, respectively, after deglycosylation. The treatment with glycopeptidase F reduced the molecular weights from 90 KDa and 70 KDa to 83 KDa and 52 KDa, respectively, While endoglycosidase H caused no change in their molecular weights. These results indicate that the 70KDa subunit contains only the N-linked oilgosaccharide chains, while the 90KDa subunit ocntains O-linked oilgosaccharides as well as N-linked ones. Enzyme-linked lectin assays suggeted that bisecting N-acetyl-D-glucosamine and galactose 1-4 N-acetyl-D-glucosamine structures were present and that fucose was included in these oilgosaccharide moieties. Sialic acid was not found in either subunit.

  • Carbohydrate analysis is important in studying structure and activity of complex polysaccharides. New analytical method was applied to get an information on the composition of polysaccharides showing antitumor activity. Monosaccharides were labeled with 7-amino-1, 3-naph-thalenedisulfonic acid (7-AGA) by reductive amination and separated by HPLC. Five kinds of polysaccharides from Basidiomycetes were hydrolyzed and analyzed in combination with electrophresis and HPLC. At the same time, alditol acetate derivatives were prepared and analyzed by gas chromatography. Two different techniques using different derivatization methods showed very similar results. The monosaccharides from Coriolus versicolor and Cordyceps militaris were glucose and galactose. Phellinus linteus composed of glucose, glactose, mannose, arabinose and fucose. The HPLC method with fluorescence detector was very sensitive compared to other methods.

  • The NCS(Non-Cariogenicity Sugar) from Bacillus sp. S-1013 was purified by cold acetone and methanol precipitation, and DEAE-cellulose ion-exchange and Sephadex G-100 column chromatographies, to yield an amorphous yellow syrup. The melting point and $[\alpha]_D^{20}$ were 155-$157^{\circ}C$ and +53, respectively. Instrumental analyses such as FT-IR, $^1H-NMR, and ^{13}C-NMR$ showed that the NCS contained an O-H group, C-H, C=O, $NH_2$, anomeric carbon, anomeric proton, N-acetylgalactose, fucose, and neuramic acid, thus, the NCS was determined to be a trisaccharide of Fuc($1\longrightarrow4$)GalNAc($2\longrightarrow6$) NeuAc.

Lectin-binding properties of chicken primordial germ cells during embryonic development

  • Lectins have great potential as to determine the alternation of the distribution of cell surface carbohydrates during cellular development and differentiation. Here, we investigated the presence and distribution of cell surface carbohydrates on chicken primordial germ cells (PGCs) during the migration and gonadal stages using a variety of lectins. A total of six FITC-labelled lectins from several specificity classes were used: ConA (glucose/mannose), WGA (N-acetylglucosamine), STA (N-acetylglucosamine), DBA (N-acetylgalactosamine/galactose), UEA-I (fucose) and PHA-E (oilgosaccharide). As a results, PGC-specific binding was observed in STA. PGCs of migration stage (2.5- and 5.5-day embyos) were STA-positive whereas PGCs of 10-day embryonic gonad were not. The results suggest that N-acetylglucosamine residuse are present specifically in migrating chicken PGCs and changes during development.
