감귤 내 생리활성성분이 간암세포의 생존에 미치는 영향 (Effect of active ingredients in the Citrus fruits on the proliferation of hepatocellular carcinoma cells)

  • Previous studies have suggested that Citrus fruits might suppress the proliferation of various cancer cells. However, little is known about any specific ingredients in the extract of Citrus fruits to exert its anti-proliferative activity in cancer cells. The present study aimed to identify the active ingredients in Citrus fruits to suppress the proliferation of rat hepatocellular carcinoma cells. Among tested compounds, two polymethoxylated flavones (nobiletin and tangeritin) showed significant anti-proliferative activity whereas other compounds (synephrine, rutin, hesperidin) did not. Interestingly, nobiletin as well as tangeritin also decreased the protein amount of gluconeogenic enzymes, PEPCK and G6Pase. The possible involvement of gluconeogenic activity in the proliferation of hepatocellulacarcinoma cells are further to be investigated.

  • As consumption of high-quality fruits increases and sales and packaging units become smaller, the demand for automatic fruit grading systems is increasing. Compared to other crops, the quality of fruit is determined by visual characteristics such as shape, color, and scratches, rather than just physical size and weight. Accordingly, this study presents a CNN model that can effectively extract and classify the visual features of fruits and a perceptron that classifies fruits using physical features, and proposes a stacking ensemble model that can effectively combine the classification results of these two neural networks. The experiments with AI Hub public data show that the stacking ensemble model is effective for grading fruits. However, the ensemble model does not always improve the performance of classifying all the fruit grading. So, it is necessary to adapt the model according to the kind of fruit.

  • To investigate the effect of several bactericides and fungicides against fruit & vegetable decay grapefruit seed extract(GFSE) was tested in a dilution solution. GFSE was shown to be effective against decay and rind breakdown and to extend the shelf-life of fruits and vegetables during storage and tiansport. Antimicrobial efficiency of GFSE on a wide spectra of gram + & - bacteria, moulds and yeasts was demonstrated by the measurement of minimal inhibitory concentrations performed (Bacteria 10-500ppm, Fungi:250-1,000ppm, Yeasts:100-250ppm). GFSE was separated and extracted into water-soluble fraction, water-insoluble and non-dialyzed fraction to isolate the antimicrobial substances. The water-soluble fraction showed the most active antimicrobial effect. The antimicrobial substances were isolated by gas chromatography. As the result of the isolation using GC, Peak-D was found to be the antimicrobial compound in GFSE. The identification of the most antimicrobial substance was carried out by using GC-MS.

Antimicrobial Activity of Various Parts of Tomato Plants Varied with Different Solvent Extracts

  • The antimicrobial activity of acetone, hexane, dichloromethane, and methanol extracts from leaves, stems, immature green fruits, and red fruits of tomato plants was examined against six phytopathogens. The minimum inhibitory concentration (MIC) of the acetonic extracts from these four plant parts was lower than that of the other solvents. Among the acetonic extracts, tomato leaves had a lower MIC than the other tomato parts. The acetonic extract from tomato leaves was therefore selected as a source of antimicrobial substances. The acetonic extract from tomato leaves inhibited mycelial growth of Fusarium oxysporum f. sp. lycopersici, Glomerella cingulata, and Rhizoctonia solani. Mycelial growth of R. solani treated with acetone extract from leaves showed more susceptibility than the other phytopathogens. Using 0.31 mg/ml of the acetonic extract from leaves, mycelial growth of R. solani on days 1, 2, and 3 decreased by 50.0, 52.1, and 64.0%, respectively, compared with acetone solvent treatment. The antimicrobial compounds effective against R. solani were identified as linolenic acid and caffeic acid by bioautography and GC-MS. These two compounds were used to treat six phytopathogens to confirm their antimicrobial activities. Linolenic acid inhibited mycelial growth of R. solani, while caffeic acid showed only slight antimicrobial activity. Results indicated that we propose extracts from tomato leaves which included antimicrobial compounds may provide a new lead in the pursuit of new biological sources of agrochemical candidates.

오매의 간디스토마 살충성 물질에 관한 연구 (The Anthelmintic Principle of "O-Mae", the Roasted Fruits of Prunus mume, Against Clonorchis sinensis)

  • The anthelmintic substance of the roasted fruits of Prunas mume against Clonorchis sinensis was isolated and its structure was identified by chemical and physical methods. The results obtained from the experiments are as follows: 1) The methanol extract of the roasted fruits of P. mume was fractionated into four parts: petroleum ether, ethyl ether, ethyl acetate and water soluble part. Among these, etherial fraction was found to have strong wormicidal effect on liberated metacercaria of Clonorchis sinensis. 2) From the etherial fraction, the wormicidal substance was isolated by silica gel, polyamide and sephadex column chromatography and identified to be 5-hydroxymethylfurfural (5-HMF) by chemical and spectral data. 3) 5-HMF was synthesized and administered to the rabbits infected with C. sinensis. On the 2nd day after administration, the EPG (eggs per gram in feces) reached to the maximal value. Among the adult worms isolated from the bile duct of the treated animal, 84% of worms were damaged morphologically. 4) The content of 5-HMF in the fruits of P. mume which were roasted in an oven at $90-110^{\circ}C$ for 52 hours and that in the fresh fruits was evaluated by HPLC. The content of 5-HMF was 0.8% in the roasted fruits and 0.02% in the fresh ones.

담배나방(Heliothis assulta)유충의 먹이 유인성 (Food Attractancy of the Oriental Tobacco Budworm, Heliothis assulta, Larvae)

  • 한국에서 담배나방(Heliothis assulta Guen e) 유충은 주로 고추과실과 담배의 어린잎을 가해한다. 따라서 본 실험에서는 이 두가지 외에 피망과 관상용 고추에 대한 1영기유충의 유인효과를 비교하였다. 다른 어느 종류보다도 고추과실에 더 많은 수의 유충이 유인되었다. 고추식물에서도 꽃이나 잎에는 거의 유인되지 않고 비교적 숙성된 고추과실에 가장 많이 몰려들었다. 이와같은 경향은 생과즙이나 10%에탄올 추출물 경우에도 비슷하며 2~4영기유충에서도 같은 경향으로 나타나 담배나방유충을 유인하는 화합식이 고추과실에 존재함을 시사하였다.

Grapefruit 종자추출물을 이용한 밀감의 저장효과 (Efficacy of Grapefruit Seed Extract in the Preservation of Satsuma mandarin)

  • 수확한 밀감(Satsuma mandarin)을 grapefruit 종자 추출물(GFSE)로 처리하여 $15{\sim}20^{\circ}C$, 상대습도 60% 조건하에서 저장하면서 밀감과피의 파손 및 연화정도를 대조구(GFSE 무처리구)와 비교하고, 오염곰팡이를 분리하여 (GFSE 처리가 곰팡이 생육에 미치는 항균효과 실험을 실시함과 동시에 투과전자현미경을 이용하여 곰팡이 포자의 형태변화를 중심으로 항균효과를 관찰하였다. 저장 8주후, 대조구의 경우, 밀감시료중 80% 정도는 과피가 심하게 곰팡이로 오염되어 파손되었으며 내부과육조직도 연화되어 폐기현상을 보인 반면, 100ppm의 GFSE 처리시험구의 경우, 밀감시료의 27% 정도 곰팡이가 오염되었고, 그 중 반수정도만 과피가 분해 파손 되었으며 250ppm GFSE 처리시험구는 단지, 13%의 밀감과피에서만 곰팡이가 오염되었을 뿐아니라, 박피해낸 과육의 조직감도 우수하였고 과즙액의 용출현상도 보이지 않아 기호도면에서 저장초기의 밀감시료에 뒤떨어지지 않는 것으로 나타났다. 한편, 변패밀감과피에 오염된 Penicillium속 곰팡이를 분리하여 GFSE 처리에의한 곰팡이 생육저해율을 조사한 결과, 100ppm 농도에서 71.8%, 250ppm 농도에서 91.2%, 500 ppm 이상의 농도처리로 100% 저해율을 나타냄으로써, 곰팡이에 의한 밀감변패현상을 방지하기 위한 GFSE의 최소저해농도는 $250{\sim}500ppm$ 정도로 판명되었다. 또한, GFSE 처리 전후의 곰팡이 포자 conidia의 세포구조를 비교 검토한 결과, GFSE 처리에 의하여 포자벽주위의 세포구조가 붕괴되고 포자내용물의 유실현상이 심화되어. 곰팡이의 증식기능이 크게 상실되어 곰팡이를 사멸케 하는 결과를 초래하는 것으로 나타났다.

과실유래 단백질 조효소액과 과육의 근원섬유 분해 효과에 관한 연구 (Proteolytic Effect of Fruit Flesh and Crude Enzyme Extract from Fruits on Myofibrilar Protein)

  • 국내산 무화과, 키위, 배, 파인애플은 단백분해효과가 알려져 조리 중에 쓰여지고 있으나 체계적인 연구가 부족한 상태이다. 이에 이들 과실로부터 조효소액을 추출하여 그 특성을 연구하고 과실을 직접 소고기에 처리하여 연육효과를 살펴보고자 하였다. 과실에서 조효소액을 추출한 후 카제인과 근원섬유에 대한 단백분해 능력을 측정한 결과 파인애플 > 키위 > 무화과 > 배의 순서로 나타났다. 이 조효소액을 근원섬유 단백질에 처리하여 전기영동을 실시한 결과 배를 제외하고 조효소액의 농도를 증가시켜 처리할수록 myosin heavy chain이 170,000D 이하로 붕괴됨을 볼 수 있었다. 연육작용 측정에 가장 적합하다고 알려진 소편화율을 관찰한 결과, 파인애플 > 키위 > 무화과 > 배로서 근원섬유단백질 분해 활성의 순서와 같았다. 소고기에 갈은 과육을 5% 및 10%로 처리하였을 때 침지액의 가용성 질소량을 측정한 결과, 과실 처리구 모두에서 대조구보다 높은 가용성 질소량을 나타내었다. 과실을 갈아 소고기에 처리한 후 가열하여 전단력을 측정한 결과, 파인애플이 연육에 가장 효과적인 것으로 나타났으며, 배는 효과가 적은 것으로 나타났다. 소고기에 갈은 과실을 넣고 숙성하여 가열한 후 텍스쳐를 측정한 결과, springiness와 gumminess의 경우 파인애플이 가장 효과적인 것으로 나타났으며 키위도 상당히 효과적인 것으로 나타났다. 과육의 연육효과를 평가한 결과, 5% 첨가시 연도의 경우는 파인애플과 키위가 가장 연한 것으로 나타났고 기호도의 경우 모든 과실에서 10% 첨가보다 5%가 더 높은 기호도를 나타내었다.
