Acute Oral Toxicity of Extract Derived from Fruiting Body of Phellinus gilvus in Rats

  • This study was carried out to investigate the acute oral toxicity of a crude extract derived from fruiting body of Phellinus gilvus (PGE) using male and female SD rats. Groups consisted of five male and female rats were treated with a single dose of the test substance intragastrically at 0, 500, 1,000, 2,000, and 5,000 mg/kgaj, respectively. Clinical signs, body weight change, and food and water consumption change were observed for 14 days after administration. No mortality or abnormal clinical signs in animals were shown during the observation period at the dose used in this study. Also there was no difference in net body weight gain, water and food consumption or gross pathological findings at terminal sacrifice among the groups of rat treated with different doses of the test substance. The results suggested that acute oral toxicity of PGE in rats is very low at the conditions employed in this study and $LD_{50}$ of PGE was estimated to be over 5,000 mg/$\textrm{m}{\ell}$ in both sexes of rats.

Production of Vegetable Wasp and Plant Worm by Injection of Paecilomyces japonica to Pupae in the Chinese Oak Silkworm, Antheraea pernyi (종균의 주사에 의한 작잠번데기 동충하초 생산)

  • Injection method of Paecilomyces japonica to Antheraea pernyi pupal body was investigated on feasibility for production of the corresponding vegetable wasp and plant worm. Infection rate of P. japonica to the pupae was 100%, and also fruiting body formation rate was 97%. The initial pupal body weight of 9.38 g befor injection of P. japonica to the pupae was finally reduced to 7.87g after two weeks in vivo culture. From these results, the injection method to pupal body will be useful for the production of Antheraea vegetable wasp and plant worm in future.

Immuno-stimulating and Antitumor Effects on Mouse Sarcoma 180 by Crude Polysaccharides Extracted from Fruiting Body of Hericium erinaceus (노루궁뎅이(Hericium erinaceus) 자실체 추출 조다당류의 생쥐 Sarcoma 180에 대한 면역증강 및 항암 효과)

  • Hericium erinaceus, an edible and medicinal mushroom belonging to the Basidiomycota family, has been used for curing gastric ulcers and stomach cancers in human beings and is also known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma in mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to as Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from the fruiting body of the mushroom. In in vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cell lines such as Sarcoma 180, HepG2, HT-29 and NIH3T3 at concentrations of $10{\sim}2,000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited a life prolongation effect of 29.1~54.1% in mice previously inoculated with Sarcoma 180. Fr. Na increased the numbers of spleen cells by 2.9 fold at a concentration of $50\;{\mu}g/ml$ compared with the control. Fr. Na improved the immuno-potentiating activity of B lymphocytes by increasing alkaline phosphatase activity by 5.5 fold compared with the control at a concentration of $200\;{\mu}g/ml$. Fr. NaCl increased the numbers of peritoneal exudate cells and circulating leukocytes by 4 and 2.3 folds at a concentration of 50 mg/kg, respectively. Therefore, the crude polysaccharides extracted from the fruiting body of H. erinaceus could improve antitumor activities in mice.

Antitumor and Immuno-potentiating Activity against Mouse Sarcoma 180 by Crude Polysaccharides Extracted from Fruiting Body of Tricholoma matsutake (송이(Tricholoma matsutake)의 자실체에서 추출한 조다당류가 생쥐의 Sarcoma 180에 미치는 항암 및 면역증강 작용)

  • Tricholoma matsutake, one of edible and medicinal mushroom belonging to Tricholomaceae of Agaricales, has been known to contain some curing effect on gastric cancer and ulcer, and inhibitory effect on sarcoma 180 and Ehrlich sarcoma. Neutral salt soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HepG2, HT29 and NIH3T3 at the concentration of 2.0 mg/ml. Intraperitoneal injection with crude polysaccharides showed life prolongation effect of 23.4$\sim$37.2% in mice previously inoculated with Sarcoma 180. Fr. MeOH and Fr. HW exhibited the immuno-potentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by 2.2$\sim$11.9 folds compared with control at the concentration of 0.2$\sim$0.5 mg/ml. In case of Fr. NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 6.0 and 1.5 folds at the concentration of 50 mg/kg, respectively. Therefore, it is concluded that crude polysaccharides extracted from fruiting body of Tricholoma matsutake showed antitumor and immuno-potentiating activity against Sarcoma 180 of mouse.

Effects of mixed LED light sources on the fruiting body growth of oak mushroom (Lentinula edodes) 'Nongjingo' (LED 혼합광이 표고 '농진고' 자실체의 생육에 미치는 영향)

  • In this study, we investigated the morphological characteristics and antioxidant ability of mushroom cultivar Lentinula edodes 'Nongjingo' fruiting bodies after exposure to various light conditions. Color differences between mushrooms treated with mixtures of LED light revealed that mushrooms displayed lighter color shades when compared to the control group (fluorescent light treated mushrooms). Redness increased and yellowness decreased after exposure to all treatments other than the fluorescent control. Measurement of growth characteristics of 'Nongjingo' fruiting bodies showed increases after exposure to all mixed LED treatments. In addition, the uniformity of fruiting bodies was higher when using LED light compared to fluorescent light. The measurement of stem diameters did not show a significant difference between the treatments, however, diameters were slightly larger with exposure to white-green LED. Moreover, stem length was longer in the mixed LED treatments when compared to those exposed to fluorescent light. Examination of the ratio of stem diameter to stem length revealed that the diameter of the stem was greater than the length. The antioxidant activity of water extracts made from Nongjingo fruiting bodies grown under mixed LED conditions was compared to those from mushrooms grown under fluorescence light conditions. The highest antioxidant activity was observed from mushrooms treated with white LED; however, no significant difference was found between mushrooms exposed to white-green LED compared to white-blue LED. The treatment showed higher antioxidant ability than vitamin C. Our results confirm that treatment of white LED and white-blue LED affects the growth and antioxidant ability of Nongjingo mushroom fruiting bodies.

Analysis of antioxidant activities, β-glucan, and amino acid contents of Phallus indusiatus (망태버섯 추출물의 항산화, 베타글루칸 및 아미노산 함량 분석)

  • The antioxidant activities and β-glucan contents of hot-water extracts of the eggs and fruiting bodies of Phallus indusiatus were investigated using different drying methods, and the amino acid contents as nutritional components of the eggs and fruiting bodies of P. indusiatus were analyzed. DPPH radical scavenging and nitrite scavenging activities of hot-water extracts of the eggs of P. indusiatus obtained after hot-air drying were 59.4% and 15.6%, respectively, at 1 mg/ml concentrations, which showed higher activities than those of freeze dried samples. Total polyphenol and β-glucan contents in hot-air-dried hot-water extracts of the eggs of P. indusiatus were 8.25 mg GAE/g and 45.9%, respectively, which were the highest among all samples. Additionally, 17 amino acids were detected from the hot-water extracts of the eggs and fruiting bodies of P. indusiatus, and the amino acid contents were higher in the eggs than in the fruiting bodies. Cysteine, phenylalanine, and glutamic acid were the most abundant essential and non-essential amino acids in the analyzed extracts. The results of this study showed that the physiological activities of the antioxidants from P. indusiatus, well known as wild edible mushroom, were greater when extracted from the dried samples. Further, the amino acid contents were higher in the egg extracts than in the extracts from the fruiting bodies of P. indusiatus.

Characteristics of Lentinula edodes Fruiting Bodies Formed by Mono-mono Hybridization Using Mating Type Analysis (교배형 분석을 활용한 표고 교배균주 육성 및 자실체 발생 특성)

  • In this study, dikaryotic strains of Lentinula edodes were generated by mono-mono hybridization using mating type analysis and their fruiting body characteristics were investigated. Approximately 100 monokaryotic strains were isolated from the basidiospores of Sanbaekhyhang, and homokaryotic strains were isolated from Chungheung 1ho using protoplast isolation and regeneration. Their mating types were evaluated and a total of 60 dikaryotic strains were hybridized. Using these strains, fruiting bodies were produced and their characteristics were examined after cultivation on sawdust media. The results indicated that the rate of hybridization was 100% and that 55 of 60 strains formed fruiting bodies. These showed normal pileus and gill structures; however,10 strains also produced fruiting bodies with abnormal pileus and gill structures. The weight and size of the fruiting bodies differed depending on the strains. Overall, further studies are needed for predicting the characteristics of hybridized strains based on their parental strains.

Biological Activities and Cultural Characteristics of an Entomogenous fungus, Paecilomyces tenuipes (Peck) Samson (눈꽃동충하초의 배양적 특성 및 생리활성)

  • To develop technique for the production of P. tenuipes stromata on a large scale, the infection of P. tenuipes and the growth of stroma were investigated by silkworm (Bombyx mori) variety. Also, studied about biological activites of fruiting body formed on silkworm. Infection rate of the 5th instar larvae of the silkworm with P. tenuipes was the highest in Yangwonjam, followed by Hachojam, Baegokjam and Chilbojam in that order. Also, as the inoculation times was increased, infection rate tended to be raised. The rate of fruiting body formation of the silkworm pupae infected with P. tenuipes was the highest in Baegokjam, followed by Yangwonjam and Chilbojam in the order. But, actually the fruiting body formation of the 5th instar larvae of the silkworm tested was good in Chilbojam, followed by Yangwonjam and Baegokjam in that order in 3 times spraying inoculation. The fruiting bodies of Yangwonjam and Chilbojam infected with P. tenuipes had high amount of Mannitol, but Baegokjam and Hachojam had high concentration of Glucose on a dry weight basis. The mean content of total amino acid in the fruiting bodies of P. tenuipes was 1.03 ${\mu}mole/g$. The distribution rate of amino acid components decreased in the order of Arginine (12.2%)>Glycine (10.5%)> Proline (9.6)>Tyrosine (8.9%)>Serine>Leucine>Threonine. The most abundant amino acid in the fruiting bodies of the Baegokjam, Chilbojam and Hachojam infected with P. tenuipes was arginine, while Yangwonjam was Glycine. The most abundant fatty acid in P.tenuipes was Oleic acid on a dry weight basis. The unsaturated fatty acids such as Oleic acid, Linoleic acid and Linolenic acid accounted for more than 78% of the total fatty acids.

Morphological Characteristics of Artificially Cultivated Dictyophora echinovolvata (인공재배한 흰돌기망태버섯(Dictyophora echinovolvata)의 형태적 특징)

  • This study was conducted to investigate morphological characteristics of veiled lady mushroom (Dictyophora sp.). Morphological observation of mycelium showed the presence of chlamydospore-like swelling cells, and typical mycelial champ connections. Fruiting body of the veiled lady mushroom consists of cap, velum, stipe, and vulva. Oval-shaped eggs were surrounded by exodermis, gelatin layers and endodermis, and the fruiting bodies remained inside volva as liquid gelatin layer. Dark green spore layer was formed at the top of cap. The surface eggs in the cultivated veiled lady mushroom was covered with white needle-shaped mycelial projection that was oval in shape. The velum of fruiting body stretched down to the top of volva. The net was regularly shaped, and smelled like fragrance with chestnut flower. The strain KACC 50650 was identified as Dictyophora echinovolvata based on the morphology of eggs and fruiting bodies, and named as Huin-Dol-Gi-Mang-Tae mushroom in Korean.

Stain improvement in the white button mushroom 'Seolgang' and its varietal characteristics in Agaricus bisporus

  • The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, 'Seolgang' was developed by crossing two monokaryons 'CM020913-27' and 'SSU423-31'. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of 'Seolgang' on CDA was better at $20^{\circ}C$ and $25^{\circ}C$ when it was compared with that of '505 Ho'. The mature cap shape of new strain 'Seolgang' is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain '505 Ho', the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of '505 Ho'. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of '505 Ho'. Genetic analysis of the new strain 'Seolgang' showed different profiles compared to '505 Ho', CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.
