Feasibility of Determining the Ripeness of Strawberry Fruit Flesh by Fourier Transform Infrared Spectroscopy (Fourier 변환 적외선 분광분석법에 의한 딸기 과육의 성숙도 측정 가능성)

  • Fourier transform - infrared spectroscopy (FT-IR) provides biochemical profiles containing overlapping signals from a majority of the compounds that are present when whole cell extracts are analyzed. We attempted to determine the ripeness of strawberry fruit flesh by FT-IR. Fruit ripeness was divided into four developmental stages based on fruit skin color: 'yellow-green', 'pink-green', 'pink', and 'red' stages. Principal component analysis of FT-IR data of inside fruit flesh extracts clustered samples of four different developmental stages into three discrete groups: (1) 'yellow-green' group, (2) 'pink-green' group, and (3) 'pink' and 'red' group. The most remarkable difference between four different developmental stages was found in the carbohydrate fingerprint region $(1,000-1,100cm^{-1})$ of the FT-IR spectrum, indicating that differences in carbohydrate compounds represented the ripeness of strawberry fruit. Overall results indicate that FT-IR in combination with PCA enables discrimination of the ripeness of strawberry fruit flesh.

Effects of Urea and K2SO4 treatment on the mineral nutrient concentration and fruit skin color of 'Mibaekdo' peach fruits at harvest (Urea와 K2SO4 처리에 의한 복숭아 '미백도'에서 수확 시 과실의 무기성분 농도 및 과피색 변화)

  • This study has been conducted to investigate the effect of Urea and K2SO4 treatment at stone hardening stage and 20 days before harvest on soil chemical properties, mineral nutrient concentration and quality of 'Mibaekdo' fruit peach. K concentration after Urea and K2SO4 treatment in soil was increased significantly by Urea 162g+K2SO4 188g/tree(standard amount) treatment at stone hardening stage, K2SO4 1.0% tree-spray, Urea 81g+K2SO4 94g/tree(half amount), Urea 162g+K2SO4 188g/tree and Urea 324g+K2SO4 376g/tree(double amount) soil treatment before harvest 20 days compared to control. T-N, K and Ca concentration in leaf was increased significantly by all treatment. but Na concentration in leaf was increased by Urea 0.5% and K2SO4 1.0% tree-spray treatment before harvest 20 days. T-N concentration in fruit skin was increased significantly by standard amount soil treatment, which decreased by K2SO4 1.0% tree-spray and half amount soil treatment. T-N, K and Ca concentration in fruit flesh(1~10mm depth flesh from peel) were increased markedly by all treatment excepted Urea 0.5% tree-spray. The leaf weight at harvest was increased markedly by Urea 0.5% tree-spray, standard amount and double amount treatment before harvest 20 days. Fruit weight was increased significantly by standard amount compared to all treatment. Red fruit skin(Hunter a value) progress was effective by K2SO4 tree-spray, half amount and double amount treatment before harvest 20 days. Fruit SSC was increased significantly by Urea 0.5% and K2SO4 tree-spray before harvest 20 days, standard amount treatment at stone hardening stage compared to control.

Efficiency of virus elimination in apple calli (cv. Hongro) derived from meristem culture of dormant buds (사과 품종 홍로의 휴면아 분열조직 배양을 통해 형성된 캘러스에서의 바이러스 제거효율)

  • Various sizes (0.2 ~ 1.2 mm) and developmental stages (referred to as Stage 1 ~ 3) of apical and lateral meristems were excised, together or separately, directly from dormant buds of apple 'Hongro'. They were mixed infected by Apple scar skin viroid (ASSVd), Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV) and Apple stem grooving virus (ASGV), which are major viruses attacking apples. A total of 31 callus lines (> 10 mm in diameter) were obtained by culturing the explants on Murashige and Skoog (MS) medium supplemented with 3% sucrose, 3.0 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-butyric acid (IBA), and they were subjected to RT-PCR analysis for virus detection. A high rate of virus elimination (expressed as the percentage of calli that did not amplify during RT-PCR, i.e., RT-PCR negative calli per total number of calli obtained) was achieved for ACLSV (100%), ASSVd (93.7%), and ASPV (93.7%), whereas it was only 25.8% for ASGV. ASPV was detected in the presence of 2 ~ 3 bracts. Simultaneous virus elimination of ASSVd, ASPV, ACLSV, and ASGV occurred during the meristem culture, in which the early stages of the dormant buds (Stage 1) were used, because ASGV was mostly eliminated during that stage. The results of the present study will be valuable for the production of virus-free apple trees.

Production System of Virus-free Apple Plants Using Heat Treatment and Shoot Tip Culture (열처리와 경정배양을 이용한 바이러스 무병 사과 생산 시스템)

  • In worldwide, viral diseases of apple plants has caused the serious problems like reduced production and malformation of fruits. Also, the damages of apple plants by virus and/or viroid infection (Apple chlorotic leaf spot virus, Apple stem grooving virus, Apple mosaic virus, and Apple scar skin viroid) were reported in Korea. However there is few report about the protection approach against the infection by apple viruses. Therefore, this paper introduced the experimental protocol for the development of virus-free apple cultivars (Danhong, Hongan, Saenara, Summerdream). Apple plants were treated at $37^{\circ}C$ for 4 weeks and shoot tips were cultured in vitro. After heat treatment, the detection of apple viruses was performed by RT-PCR using virusspecific detection primers in new apple cultivars. With the heat treatments followed by in vitro shoot tip culture, the proportion of virus-free stocks of 'Danhong', 'Hongan', 'Saenara', and 'Summerdream' was 28%, 16%, 12%, and 12%, respectively. Taken together, this approach can be a good tool for production of virus-free apple stocks.

Responses of Young 'Fuyu' Persimmon Trees to Summer Fertilization Rate and Leaf-fruit Ratio

  • Small-sized persimmons produced by high crop load are better accepted in the export markets. However, maintaining high crop load frequently results in weakness of tree vigor, deterioration of fruit quality, and increase of the risks for alternate bearing. This experiment was conducted to determine the combined effects of fertilization rate and leaf-fruit (L/F) ratio on container-grown 3-year-old 'Fuyu' persimmon trees. Application of 3.6-g N, 2.1-g $P_2O_5$, 2.7-g $K_2O$, 2.7-g CaO, and 0.6-g MgO was for the control fertilization rate (CF) and that of a 3-fold CF was for the high fertilization rate (HF). Commercial fertilizers were surface-applied to a container on July 6, July 17, and August 10 in three equal aliquots. Single tree for each fertilization rate was assigned for 12 L/F ratios (5, 6.3, 7.7, 9, 10.4, 13, 15.5, 18, 21, 24, 27, and 33) mostly by fruit thinning or rarely by defoliation on July 1. HF did not affect the yield, weight and soluble solids of the fruits but decreased skin color. As L/F ratio increased, yield decreased but average weight, skin color, and soluble solids of fruits increased. With HF, N and K concentrations in leaves, fruits, and shoots increased to some extent but soluble sugars in dormant shoots decreased. Many shoots were cold-injured with low L/F ratio especially at the HF. HF did not increase number of flower buds the next spring either on a shoot or on a tree basis but increased shoot length, compared with the CF. Increasing L/F ratio markedly increased number of flower buds and shoot growth the following year at both fertilization rates. Therefore, an appropriate combination of fertilization rate and L/F ratio should be necessary to maintain stable fruit production and tree vigor at high crop load.

Damage of Sweet Persimmon Fruit by the Inoculation Date and Number of Stink Bugs, Riptortus clavatus, Halyomorpha halys and Plautia stali (노린재류 접종시기와 밀도에 따른 단감 피해과율)

  • This study was conducted to investigate the fruit damage according to the number of released stink bugs and date of adult introduced on non-astringent persimmon. The fruit damage differed significantly by the number, date, and the source of feed for Riptortus clavatus. Damages on the fruits appeared when five bugs were released to mother branches bearing fruits. Damages to the fruits was heavy in the case that bugs were introduced in September and October, compared with in June, July and August. Persimmon fruit damage was influenced significantly by the available sources to R. clavatus, persimmon fruit alone or additional supply of kidney bean (Phaseolus vulgaris) and water. Characteristic damage symptoms by R. clavatus showed only scars on the fruit skin, without fruit dropping. Halyomorpha halys (=mista) showed significantly higher fruit damage when they were released in July, August, and September compared with other periods. Number of H. halys to the fruit also showed significant differences in fruit damages. Five H. halys in July showed 100% damaged fruits. The fruit damage by Plautia stali was not significantly different according to dates of introduction but the number of insects gave significant differences in the fruit damage ratio at the level of 1, 3, 5 bugs introduced to the mother branche bearing friuts. The damage symptoms by P. stali was fruit dropping in July and August but from September damaged fruits were found in the branches.

Study on skin anti-inflammatory activity of fig (Ficus carica L.) fruit extract fractions (무화과(Ficus carica L.) 열매 추출 분획의 피부 항염증 활성 연구)

  • Figs has known to have antioxidant, whitening, anti-inflammatory, and antibacterial effects in their leaves, roots, stems, latex, and fruits. In order to develop cosmetic materials based on natural products, we have studied on the skin activity of the ficin in latex as well as the whitening function of the fruit extract with 70% ethanol, and used it as a raw material for released cosmetic product. However, there is little research on the demand for the development of new eutectic solvent extraction methods and its ability to control skin inflammation and psoriasis regulation. Thus, in this study, we evaluated the effectiveness of fig fruit extracts and fractions using eutectic solvent extraction for skin inflammation control and psoriasis. First, fig fruits were extracted under optimal eutectic solvent conditions and fractionated with n-hexane, dichloromethane, ethyl acetate, and butanol. First, the antioxidant activity and inhibition of nitric oxide (NO) production were confirmed in mouse macrophage RAW264.7 cells. In addition, as a result of observing the mRNA expression through RT-PCR, pro-inflammatory cytokines such as TNF-α, IL1α, and IL-1β were suppressed significantly in the hexane, dichloromethane, and ethyl acetate fractions. In addition, it was confirmed in TNF-α stimulated HaCaT keratinocyte model. Finally, chemokine CC motif ligand 20 (CCL20), marker gene of human psoriasis skin disease, was significantly suppressed in the hexane, dichloromethane, and ethyl acetate fractions. These results suggested its anti-inflammatory and skin soothing effect and the possibility of development as an excellent skin soothing natural cosmetic material in the future through future clinical trials.

Influence of Air Temperature during Midsummer on Fruit Sunburn Occurrence in 'Fuji'/M.9 Apple Tree (한여름 기온이 '후지'/M.9 사과나무의 과실 일소증상 발생에 미치는 영향)

  • This study was carried out to investigate the effects of air temperature during midsummer on sunburn occurrence that had some problems recently on 'Fuji'/M.9 planting system in Korea. There was a positive relationship between sunburn incidence and accumulated days with high air temperature over $31^{\circ}C$, or accumulated total quantum per day. 'Fuji'/M.9 apple trees with relatively open canopies, compared to 'Fuji'/M.26, were more susceptible to sunburn injury because the fruit was more exposed to sunlight. In 'Fuji'/M.9 apple trees, those that were poor in vigor or with short bourse shoot length were more susceptible to sunburn injury. Sunburn incidence was affected by neither regions nor row orientations. Sunburn was observed most often on the fruits on the south and west sides of the trees. The reason was due to high skin temperature of the fruits reaching $40{\sim}45^{\circ}C$ under full sunlight.

Relationships of Food Habits & Life Style and Skin Health of Young Females (젊은 여성의 식습관 및 건강관련 생활습관과 피부건강과의 관련성 연구)

  • This study was performed to investigate the effects of food habits and life style on the skin health of young Korean females in their late teens and twenties. The study focused on facial skin and used a self-administered questionnaire survey, an examination of skin type, and pH tests. More than half of the females had combination skin type or pH level higher than 5.8 which is regarded as normal. The variables that turned out to significantly (p<0.05) effect their facial skin types were body mass index (BMI), the type of residence, the amount of water intake, whether or not they intake drinks, the frequency and amount of alcohol drinking, favorite foods as side dishes at meals, intake frequency of fruit, the dairy, seaweed, and fast food, the frequency of washing their faces per day, the method of how they used water during face washing, whether or not they had professional skin care, and whether or not they wore color make up. The variables of the females' age, BMI, the amount of alcohol drinking, the frequency and duration of exercise, the controlling methods of stress, their favorite foods as side dishes at meals, and the intake frequency of soy products or animal foods significantly affected their facial pH levels (p<0.05). The results of this study indicate that some food habits and life styles are significantly related with facial skin health. More importantly BMI and favorite foods as side dishes during daily food intake could have remarkable effects on facial skin health.

Effect of Low Pressure Fog and External Watering on the Fruit Quality of Korean Melon Grown in Sumer (여름철 참외 재배시 저압포그 및 외부살수가 과실의 품질에 미치는 영향)

  • The objective of this study was to examine the changes in temperature drop and fruit production due to low pressure fog system in plastic greenhouses during summer cultivation of Korean melon. The indoor temperature of plastic house was dropped by $7.6^{\circ}C$ compared to control on July 26th, 2015 from 10:00 to 18:00. Fruit weight was smaller and lighter by 96g compared to control. The sugar content and color parameter were also enhanced due to application of low pressure fog system. The fraction of malformed fruits was decreased by 15.3% in plots where low pressure fog system was applied. The fraction of marketable fruit and yield were increased by 15.3% and 26% compared to control, respectively. As a result, high quality fruit production within plastic house of summer was increased by applying low pressure fog system and it is positively affected the drop of indoor temperature.
