Application of electric pulsed power on fruit body production of edible and medicinal mushrooms

  • Effect of pulsed power was investigated on fruit body formation of 10 edible mushrooms, Lentinula edodes, Glifola frondosa, Pholiota nameko, Flammulina velutipes, Hypsizygus marmoreus, Pleurotus ostreatus, Pleurotus eryngii, Pleurotus abalonus, Agrocybe cylindracea and Sparassis crispa. Pulsed power of 100-170 kV was directly charged to the substrate just before fructification. The effect of the pulsed power resulted to promote for 10 edible mushrooms fructification. The treatment especially stimulated the fructification on Pleurotus species.

  • In order to elucidate the human impact on the distribution of cellular slime molds, samples were collected from 3 types of forest ; natural forests(Mt. Deogyu and Mt. Tsukuba), semi-natural forests(Seoul Great Zoo and Tama Zoo), artificial forests(Seoul National University and Tokyo Gakugei University) .The distribution of cellular slime molds in mountains was different from that of zoo and universities. In mountains, endemic species was occurred and species diversity was higher than in zoo and universities. In zoo and universities disturbed by human, Dictyostelium sphaerocephalum was occurred with higher importance value than in mountains. 6 species were selected to investigate the effect of temperature on froctification; Polysphondylium canlidum, D. delicatum. D. firmibasis, D. sphaerocephalum P. violaceum, D. purpureum. P. violaceum and D. purpureum had an optimum temperature for fructification around 25~3O˚C but the others around 22~23˚C. The degree of sensitivity to temperature was as follows; P. candidum >D. lelicatum > D. firmibasis > D. sphaerocephalum > P. violaceum > D. purpureum. 일본잎갈나무 천연갱신지의 종자 결실 및 벌채 유형별 종자 비산 특성 (Seed Fructification and Seed Dispersal Characteristics by Harvest Types in Natural Regeneration Sites of Larix kaempferi)

  • 본 연구는 일본잎갈나무 인공림에서 천연하종갱신을 이용한 후계림 조성 방법을 제안하고자 수행되었다. 이를 위해 2016년 경기도 가평에 모수작업과 개벌작업을 실시하여 시험지를 조성하고, 2014년에 조성된 경상북도 봉화의 시험지와 함께 천연갱신지의 종자결실 및 종자낙하 특성을 분석하였다. 그 결과, 결실이 좋은 해에 V영급 일본잎갈나무의 개체 목당 종자 수는 690,340립이며, 모수가 ha당 25본인 일본잎갈나무 천연갱신지에서는 ha당 1,380만립의 종자가 결실되는 것으로 추정되었다. 종자의 비산은 9월부터 시작되었으며, 11월까지 낙하된 종자의 발아율은 종자품질 기준인 40%를 넘는 것으로 나타났다. 갱신유형별 종자 낙하량은 보잔목작업, 모수작업, 개벌작업 순이었고, 보잔목작업이 종자의 균등한 비산에 유리한 것으로 나타났다. 따라서 일본잎갈나무의 천연갱신을 위한 벌채와 갱신상 조성 시기는 종자가 비산하는 9월 이전이 효과적이고, 보잔목작업이 균일한 종자공급에 유리한 방법인 것으로 판단된다.

Relationship between some Phenological Parameters and Somatic Embryogenesis in Theobroma cacao L.

  • The relationship between three phenological parameters and somatic embryogenesis was investigated during a two-year period. Staminodes and petals from six hybrids and two clones as controls were sowed on three distinct primary callus growth media. Flowering level, fructification level, and leaf thrusts rhythm as phenological parameters were measured simultaneously during the weekly harvest of flower buds. Mean and coefficient of variation of the measured parameters highlighted stable phenological phases. The relationship between phenological parameters and somatic embryogenesis was investigated first by comparing the variation of somatic embryogenesis and that of the phenological parameters, and second by using Pearson's linear correlation. Except for the fructification level in both control clones the first year, the other parameters recorded stable phenological phases, regardless of the genotype and year. Favorable and unfavorable phases for the somatic embryogenesis were identified. In hybrids, favorable phases included February, August, September, and October. In both control clones, time interval propitious to embryogenesis stretched from February to December. The significance of the coefficient of correlation seemed to establish a relationship between somatic embryogenesis and phenology. However, a causal link could not be established. Leaf thrusts rhythm was revealed to be the phenological parameter most linked to somatic embryogenesis. Attempts to optimize embryogenesis during unfavorable phases, showed that a correction of 2.4 D/TDZ concentration is not the solution.

대추나무 '황실'의 수령에 따른 결실 및 과실특성과 수확량 (Fructification, Fruit Characteristics, and Yield According to Tree Age of Jujube Tree (Zizyphus jujuba var. inermis) 'Hwangsil')

  • 대추나무 '황실'의 고품질 생대추 생산기술 개발을 위한 기초자료를 확보하기 위해 수령(3~7년생)에 따른 생육특성, 과실품질, 수확량 등을 분석하였다. 대추나무 '황실'의 평균 수고와, 수관면적, 근원경, 지하고, 주지수, 주지간거리는 각각 230.8 cm(224.4~247.2 cm), 3.0 m2(2.1~3.8 m2), 4.8 cm(2.4~6.2 cm), 69.1 cm(46.6~78.0 cm), 12.9개(8.6~19.6 개), 8.1 cm(7.4~9.0 cm)인 것으로 나타났다. 각 항목 간 상관분석 결과, 수령과 수관폭, 근원경, 지하고는 정의 상관을 보인 반면, 주지수는 음의 상관을 나타냈다. 결과모지 및 본당 평균 과실수는 20.0개(14.3~26.3개)와 302.8개(257.3~373. 5개)인 것으로 각각 나타났다. 특히 수령과 결실특성(주지당 결과모지수, 결과모지당 결과지수, 결과지당 과실수) 간에는 유의한 차이가 없는 것으로 추정된다. 대추나무의 수형은 생육면적과 하우스 형태에 따라 일정하게 관리하기 때문이다. 평균 과중은 24.2 g(22.4~26.8 g)이었으며 과실의 경도와 당도는 각각 28.4 N(27.3~30.0 N)과 19.0%(17.1~19.8%)인 것으로 조사되었고, 수령 간에 유의한 차이는 각각 없는 것으로 분석되었다. 본당 수확량은 7.4 kg/본(5.7~9.1 kg/본)으로, 6년생과 7년생에서 통계적으로 가장 많은 수확량을 나타냈다.

Screening of-Lyophyllum Decastes-Highly Productive Cultivable Strains

  • In order to explore mycelial growth and fruiting body formation of Lyophyllum decates on different media, ten cultivation media by using cottonseed hull, sawdust, corn cob etc. as main components were designed for seven strains. The results showed that the mycelial colour of all strains are mainly snow-white, and the formula of media using corn cob as main materials was better than that using cottonseed hull and sawdust for mycelial growth, but no fruiting body was formed. The cottonseed hull medium with a small amount of sawdust, plant leaves, humus or fermented material and wheat was beneficial for fruiting formation. The incubation period for fruiting formation of strain 3001 was 108 days and the highest yield was-214.80 g/bag. Fructification of the strains tasted occurs successively in order of 3001, 1035, 1004 and 1013. It was concluded that different medium composition had significant effect on the mycelial growth and fruiting body formation.

Morphological and Molecular Characterization of Pseudocercospora chionanthi-retusi Causing Leaf Spot on Chionanthus retusus in Korea

  • Leaves of Chionanthus retusus were found to be damaged by leaf spot disease associated with a fungus in Iksan, Korea. Leaf spots were angular to irregular, vein-limited, scattered, 1-8 mm diameter, brownish-gray to dark brown when dry, with heavy fructification. The pathogen causes premature defoliation of C. retusus plant and was identified as Pseudocercospora chionanthi-retusi based on morphological and molecular-phylogenetic analyses. The phylogenetic tree was constructed using multi-locus DNA sequence data of partial actin (actA), partial translation elongation factor 1-alfa (tef1), partial DNA-directed RNA polymerase II second largest subunit (rpb2) genes, and internal transcribed spacer regions. Current study provides detail morphological description of P. chionanthi-retusi on C. retusus in Korea, with supports of phylogenetic analysis and pathogenicity test.

The Uneven Distribution of Mating Type Genes in Natural and Cultivated Truffle Orchards Contributes to the Fructification of Tuber indicum

  • The aim of this study was to investigate the pattern of distribution of mating type (MAT) genes of Tuber indicum in ectomycorhizosphere soils from natural T. indicum-producing areas and cultivated truffle orchards and ascocarp samples from different regions. Quantitative real-time PCR and multiplex PCR were used to weight the copy numbers of MAT1-1-1 and MAT1-2-1 in natural truffle soils and cultivated orchard soils. The effect of limestone on the pattern of truffle MAT genes and the correlation between soil properties and the proportion of MAT genes were also assessed. These results indicated that an uneven and nonrandom distribution of MAT genes was common in truffle-producing areas, cultivated truffle orchards, and ascocarps gleba. The competition between the two mating type genes and the expansion of unbalanced distribution was found to be closely related to truffle fructification. Limestone treatments failed to alter the proportion of the two mating type genes in the soil. The content of available phosphorus in soil was significantly correlated with the value of MAT1-1-1/MAT1-2-1 in cultivated and natural ectomycorhizosphere soils. The application of real-time quantitative PCR can provide reference for monitoring the dynamic changes of mating type genes in soil. This study investigates the distributional pattern of T. indicum MAT genes in the ectomycorhizosphere soil and ascocarp gleba from different regions, which may provide a foundation for the cultivation of T. indicum.

Identification and Characterization of Cercospora malayensis Causing Leaf Spot on Kenaf

  • In September 2013 and 2014, a significant number of kenaf plants showing symptoms of leaf spots with approximately 50% incidence were found in experimental plots in Iksan and Namwon, Korea. Leaf spots were circular to irregular, more or less vein-limited, reaching to 10 mm in diameter. The spots were initially uniformly brown to reddish brown, turning pale brown with a purplish margin and showing grayish patches on the lesion due to heavy fructification. The causative agent of the leaf spot disease was identified as Cercospora malayensis. The pathogenicity test was conducted with similar results, which fulfilled Koch's postulates. This is the first report of C. malayensis infection of kenaf in Korea.

Effect electric pulse application on the fruit body production of Tricholoma matsutake-In situ condition

  • Effect of electric pulse stimulation was tested on the fruit body formation of valuable mushroom Tricholoma matsutake in the field of natural habitat of this mushroom. After applying the electric stimulator to the specific area of pine forest we found that the treatment especially stimulated the fructification of T. matsutake. And the most valuable findings of our study was that only our treatment plots showed fruit body formation whereas the control plots and the whole natural habitats of our study area showed zero production of this mushroom during the same time. From the point of view of mushroom production, our experiment shows that fruit body production can be upgraded by using pulsed power as an electrical stimulation in the field of the natural habitat of this mushroom. These findings from our experiment confirm the effectiveness of the significance of pulsed power technology for the improvement of T. matsutake fruit body production in the natural habit of this mushroom.
