Physiological Activities of Extract from Edible Mushrooms (주요 식용버섯 추출물의 생리활성 효과)

  • This study was conducted to investigate the physiological activity of extracts of fresh mushrooms. The components were extracted by hot water; subsequently, the hot-water extract was subjected to 60% ethanol precipitation to yield high-molecular-weight (HMW) and low-molecular-weight (LMW) fractions. Total polyphenol contents, $\beta$-glucan contents, electron-donating ability (EDA), superoxide dismutase (SOD)-like activity, nitrite-scavenging activity, fibrinolytic activity, nitric oxide (NO) production, and inhibition of NO production of the mushroom extracts were measured using lipopolysaccharide (LPS)-stimulated murine macrophages, RAW 264.7 cells. The extracts of Lentinus edodes (Berk.) Singer and Pleurotus ostreatus (Fr.) Kummer contained the highest levels of $\beta$-glucan (33.5% and 25.57%, respectively). Further, the LMW fractions of the Phellinus linteus contained the highest levels of polyphenols (233.23 mg/g). The EDA of LMW fractions (10 mg/mL) of the Phellinus linteus and Agaricus bisporus were 80.74% and 51.35%, respectively. Further, SOD-like activities of the LMW fractions were high as compared to those of the HMW fractions. Nitrite-scavenging activities of the LMW fractions (pH 1.2; concentration, 10 mg/mL) of the Phellinus linteus and Pleurotus ostreatus (Fr.) Kummer were 75.95% and 41.05%, respectively. The fibrinolytic activity of the LMW fractions of all mushrooms showed no enzyme activity by fibrin plate assay. The fibrinolytic activity of the extracts of Tricholoma matsutake was the greatest inhibitory activity at 60.4%. Further study revealed that the mushroom extracts exhibited anti-inflammatory effects on RAW 264.7 cells. The LMW fraction ($500\;{\mu}g/mL$) of the Phellinus linteus considerably inhibited NO production (100%).

Effects of Oak Wood Vinegars on Mycelial Growth, Fruiting Body Production, and Mushroom Quality of Lentinula edodes (참나무류 목초액이 표고의 균사생장, 버섯 생산량 및 품질에 미치는 영향)

  • Mycelial growth of Lentinula edodes on solid or liquid culture media supplemented with differentconcentrations of oak wood vinegar varied depending on the types of wood vinegar or mushroom varieties used.Oak wood vinegar obtained from traditionally carbonizing kiln (TWV) inhibited mycelial growth of L. edodes atthe dilution level of less than $5{\times}10^{-2}$, but stimulated at $10^{-3}$ to $2{\times}10^{-3}$. Wood vinegar from mechanicallycarbonizing kiln (MWV) inhibited at $10^{-3}$, but stimulated at $2{\times}10^{-3}$. In liquid culture media, both wood vinegarinhibited at $5{\times}10^{-2}$, but stimulated at $2{\times}10^{-3}$. Sanjo-302-ho grown in liquid culture media at $2{\times}10^{-3}$, and Sanrim 2and 3-ho grown at $4{\times}10^{-3}$ showed relatively high degree of wood decay (DWD) and growing ability within wood(GAWW) when these isolates were inoculated onto oak wood logs. TWV completely inhibited mycelial growth ofgreen mold fungi, Trichoderma species, tested at $5{\times}10^{-1}$ dilution level, while MWV inhibited at $5{\times}10^{-1}$ to $5{\times}10^{-2}$dilution level. For Diatrype stigma, TWV inhibited mycelial growth at the dilution level of less than $5{\times}10^{-2}$, whileMWV did 80% of mycelial growth at $10^{-2}$, and 100% at $5{\times}10^{-1}$ dilution level. Fresh and dry weight of fruitingbodies harvested after soaking of wood logs into wood vinegar solutions with different concentrations werecompared, and were the highest at $2.5{\times}10^{-2}$ dilution level. Storage test of fruiting bodies at $10^{\circ}C$ for 10 daysshowed that fruiting bodies harvested after soaking in the solution with $2.5{\times}10^{-2}$ dilution level showed the bestfreshness by general test and color changes. In addition, shear force value of L. edodes fruiting bodies measuredby using texture analyzer showed that $2.5{\times}10^{-2}$ dilution level was the best concentration for keeping flesh texture.

Study on the Controlling Mechaniques of the Environmental Factors in the Mushroom Growing House in Chonnam Province (전남 지방에 있어서의 양송이 재배에 최적한 환경조건 조절법 분석에 관한 연구)

  • The important results which have been obtained in the investigation can be recapitulated as follows. 1. As demonstrated by the experimental results and analyses concerning their effects in the on-ground type mushroom house, the constructions in relation to the side wall and ceiling of the experimental house showed a sufficient heat insulation on effect to protect insides of the house from outside climatic conditions. 2. As the effect on the solar type experimental mushroom house which was constructed in a half basement has been shown by the experimental results and analyses, it has been proved to be effective for making use of solar heat. However there were found two problems to be improved for putting solar house to practical use in the farm mushroom growing: (1) the construction of the roof and ceiling should be the same as for the on ground type house, and (2) the solar heat generating system should be reconstructed properly. 3. Among several ventilation systems which have been studied in the experiments, the underground earthen pipe and ceiling ventilation, and vertical side wall and ceiling ventilation systems have been proved to be most effective for natural ventilation. 4. The experimental results have shown that ventilation systems such as the vertical side wall and underground ventilation systems are suitable to put to practical use as natural ventilation systems for farm mushroom house. These ventilation systems can remarkably improve the temperature of fresh air which is introduced into the house by heat transfers within the ventilation passages, so as to approach to the desired temperature of the house without any cooling or heating operation. For example, if it is assuming that X is the outside temperature and Y is the amount of temperature adjustment made by the influence of the ventilation system, the relationships that exist between X and Y can be expressed by the following regression lines. Underground iron pipe ventilation system. Y=0.9X-12.8 Underground earthen pipe ventilation system. Y=0.96X-15.11 Vertical side wall ventilation system. Y=0.94X-17.57 5. The experimental results have 8hown that the relationships existing between the admitted and expelled air and the $CO_2$ concentration can be described with experimental regression lines or an exponent equation as follows: 5.1 If it is assumed that X is an air speed cm/sec. and Y is an expelled air speed in cm/sec. in a natural ventilation system, since the Y is a function of the X, the relationships that exist between X and Y can be expressed by the regression lines shown below: 5.2 If it IS assumed that X is an admitted volume of air in $m^3$/hr. and Y is an expelled volume of air in $m^3$/hr. in a natural ventilation system, since the Y is a function of the X, the relationships that exist between X and Y can be expressed by the regression lines shown below. 5.3 If it is assumed that expelled air speed in emisec. and replacement air speed in cm/sec. at the bed surface in a natural ventilation system are shown as X and Y. respectively, since the Y is a function of the X. the relationships that exist between X and Y can be expressed by the following regression line: GE(100%)-CV (50%) ventilation system. Y=$127.18{\times}1.0093^{-x}$ 5.6 The experimental results have shown that the ratios of the cross sectional area of the GE and CV vent to the total cubic capacity of the house, required for providing an adequate amount of air in a natural ventilation system, can be estimated as follows: GE(admitting vent of the underground ventilation) 0.3-0.5% (controllable) CV(expelling vent of the ceiling ventilation) 0.8-1.0% (controllable) 6. Among several heating devices which were studied in the experiments, the hot-water boilor which wasmodified to be fitted both as hot-water boiler and as a pressureless steam-water was found most suitable for farm mushroom growing.

Effect of Selenium Supplementation on Beef Color Stability (셀레늄 급여가 쇠고기 육색 안정성에 미치는 영향)

  • The objective of this study was to investigate the supplementation effect of selenium on beef color stability. A total of 15 Hanwoo steers were divided into 3 groups and 2 groups were administered with 0.9 ppm of one of two organic-selenium products, Organic-Se and Se-SMC (Se-spent mushroom compost) for 4 mon. The third group was the control group, which was not with fed selenium during the same period. The result of this study showed that there was no significant difference in meat color between the control and treatments when Hunter $L^*$, $a^*$, $b^*$, chroma, hue and total color difference (${\Delta}E$) were measured after 30 min of blooming. When the oxymyoglobin (OxyMb) contents were measured after beef samples were ground and stored for 48 h at $20^{\circ}C$ in an incubator, they were 26.04%, 28.52% and 33.78% for the control, Organic-Se and Se-SMC after 14 d of storage and 12.65, 18.98 and 18.72 after 21 d of storage at $4^{\circ}C$, respectively (p<0.05). The control had a significantly higher metmyoglobin (MetMb) content than Organic-Se and Se-SMC (p<0.05). This result indicated that selenium supplementation was effective in preventing the oxidation of myoglobin(Mb) and production of MetMb and thus was able to maintain the purplish fresh red color of the meat.

Physical and Sensory Evaluation of Tenebrio molitor Larvae Cooked by Various Cooking Methods (조리방법별 갈색거저리 유충의 물리적 및 관능적 특성)

  • Recently, the Tenebrio molitor larva was recognized as a novel food ingredient by the Ministry of Food and Drug Safety in Korea. Accordingly, we investigated its physical and sensory characteristics to establish the cooking conditions that may increase the demand of T. molitor larvae as a food. In this study, T. molitor larvae were cooked by various methods such as hot air dry, oven-broil, roast, pan fry, deep fry, boil, steam, and by microwave. In the physical evaluation of texture, the hardness and fracturability values were highest when larvae were cooked in the microwave. The adhesiveness, springiness, and chewiness values were highest when larvae were boiled. Boiled and steamed larvae had the highest lightness (L value), while oven-broiled larvae had the highest redness (a value) and yellowness (b value) values. Sensory evaluations assessed the appearance, aroma, flavor, and texture of cooked T. molitor larvae. Steamed and boiled larvae sizes were significantly large and the form was well preserved similar to fresh larvae. The moisture heat cooked (steamed and boiled) T. molitor larvae had the aroma and flavor of steamed corn, canned pupa, and boiled mushroom. In case of oven-broiled T. molitor larvae, the aroma and flavor of mealworm oil, seafood, sweet and roasted sesame were higher than in those cooked by other methods. In texture among sensory evaluation, the hardness and crispiness were the highest in the hot air dried and oven-broiled larvae, whereas juiciness was significantly higher in the boiled and steamed. Accordingly, we suggest that oven-broiled T. molitor larva will be prefered by consumer, due to its the rich aroma and flavor.

Effects of Various LEDs on the Seed Germination, Growth and Physiological Activities of Rape (Brassica napus) Sprout Vegetable (몇 가지 LED가 유채의 종자발아, 초기 생장 및 생리활성에 미치는 영향)

  • All the seeds of rape (Brassica napus) sprout vegetables were germinated within three days after seeding irrelevant to the light colors. The total fresh weight of rape sprout vegetables at 6 days after seeding have been increased when grown under green and white color lights by 0.339g/10plants and 0.339g/10plants compared with the control. The total flavonoid contents in rape sprouts were increased under red and blue lights by 72.5 and 70.9mg${\cdot}L^{-1}$. Those DPPH radical scavenging activities at 2,000mg${\cdot}L^{-1}$ were increased by 90.0 and 90.3% which were sprouted and grown under blue and white lights. Nitrite radical scavenging activity of sprouts were most decreased compared with the control when grown under the red light by 57.4mg${\cdot}L^{-1}$. And mushroom tyrosinase inhibition activity of rape sprouts was extremely increased under the green light by 22.5mg${\cdot}L^{-1}$.

Aroma Characteristics of Applemint (Mentha rotundifolia(L.) Huds) with Different Extraction Methods (추출방법에 따른 애플민트의 향기특성)

  • Aroma was extracted from Applemint(Mentha rotundifolia(L.) Huds) with SDE(simultaneous distillation and extraction), SFE(supercritical fluid extraction) and headspace method and the compounds of aroma were tentatively identified with GC-MS. The functionality of aroma compounds were determined with GC-olfactometry. Total 67 compounds were identified. Among them, 39 compounds were determined from SDE, 42 from SFE and 16 from headspace extract. Many terpene compounds were extracted with SDE and headspace methods but hydrocarbones with SFE. The major constituents of aroma obtained from SDE and SFE, were piperitenone oxide, germacrene-D and trans sabinene hydrate, but those from headspace method were 3-octanol, 1,8-cineol, camphene and benzeneacetaldehyde. Results of sniffing test, determining characteristics and strength of aroma showed that the major constituents of SDE extract were refreshing sweet and apple-like(ethyl-2-methyl butanoate), sweet and fruity-like$({\alpha}-thujene)$, fresh mushroom-like(1-octen-3-ol, 3-octanol), and bitter herb-like$({\delta}-cadidene)$. Major constituents of aroma extracts obtained from headspace method were alcoholic, refreshing sweet and apple-like(ethyl 2-methyl butanoate), unpleasant chemical, and bitter herb and grassy-like(camphene).

Effects of Chlorine Dioxide Treatment on Quality and Microbial Change of Agaricus bisporus Sing during Storage (이산화염소 처리가 저장 중 양송이버섯(Agaricus bisporus Sing)의 품질 및 미생물학적 변화에 미치는 영향)

  • Effects of chlorine dioxide ($ClO_2$) on weight loss, polyphenol oxidase activity, and microbial change of fresh mushrooms (Agricus bisporus Sing) were investigated. Mushrooms were treated with 5, 10, and 50 ppm of $ClO_2$ solution. Mushrooms were stored at room temperature and $4^{\circ}C$. Weight loss for $ClO_2$ treated mushrooms was smaller than that of the control group. Polyphenol oxidase activity of the $ClO_2-treated$ samples stored at room temperature increased until day 2 of storage and then decreased, and the activity of the samples stored bacteria, yeast and mold counts than the control during storage. Aerobic bacterial counts for 50 ppm of $ClO_2$ treatment at $4^{\circ}C$ increased from $1.62{\times}10^2\;CFU/g\;to\;1.66{\times}10^7\;CFU/g$ at day 10 of storage, while control groups were from $1.00{\times}10^3\;CFU/g\;to\;3.72{\times}10^8\;CFU/g$. These results indicate that $ClO_2$ treatment could be useful in improving the quality and microbial safety of mushrooms.

Comparative Study on the Sawdust Cultivation and the Antioxidants of Hericium spp. (노루궁뎅이버섯류의 톱밥재배와 항산화물질 비교 연구)

  • As a preliminary study in order to develop new varieties of Hericium species, this study was carried out to investigate the optimal temperature for mycelial growth, to figure out the applicability to sawdust cultivation on Quercus mongolica substrate, and to analyze the antioxidant capacity of ergothioneine and polyphenols in Hericium strains preserved in Korea Forest Research Institute (KFRI). In the results of optimal temperature for mycelial growth of eight Hericium erinaceus, it was $20^{\circ}C$ in a strain (KFRI 842), $25^{\circ}C$ in five strains (KFRI 507, 508, 509, 843, 845), and $30^{\circ}C$ in two strains (KFRI 582, 844). Optimal temperature for mycelial growth of H. coralloides (KFRI 713) was $25^{\circ}C$. Four strains (KFRI 508, 843, 844, 713) out of the total nine Hericium strains showed full mycelium growth within 20 days at the optimal temperature on PDA medium in petri-dish (85 mm in diameter). The other strains have need of more time for full mycelium growth. Mushroom production of H. erinaceus ranged from 215 to 384 g of fresh weight and its dry weight was 7 to 9% of it, whereas that of H. coralloides was 299 g of fresh weight and its dry weight was 10% of it. The contents of ergothioneine and polyphenols of H. erinaceus strains were different by strains and those were in the range of $1.6{\sim}3.7$ mg/g dw. and $5.9{\sim}7.8$ mg/g dw., respectively. On the other hand, those of H. coraloides were in the range of 1.7 mg/g dw. and 3.9 mg/g dw., respectively. From the results of correlation ($R^2$ = 0.1) between ergothioneine and polyphenols in the strains, it was found that the total contents of them differ by strains but the ratio of the two compounds was not very different in the strains.
