연포장재 필름으로 개별 포장된 수삼의 저장 중 품질 변화 (Changes in Quality of Fresh Ginseng Packed Individually with a Soft Film during Stnrage)

  • 수삼을 물로 세척하고 기능성 연포장재 필름주머니(ONY/LDPE/L-LDPE; 20$\times$30 cm, 93${\mu}m$)로 개별 포장하여 $25^{\circ}C$에서 20일간 저장하면서 수삼의 외관품질 변화 및 관능적 특성을 조사하였다. 수삼의 외관품질은 저장 후 10일째부터 열화되기 시작하였고 15일이 경과되면서 열화빈도가 급격히 증가하여 20일째에는 전체 수삼의 75%가 열화되었다. 저장 중 수삼의 외관품질 열화는 주로 경도 저하에 의하여 일어났으며 곰팡이 발생 빈도는 낮았다. 20일이 경과된 수삼 중 정상 수삼은 조직이 단판하고 인삼냄새와 쓴맛이 강했던 반면, 경도가 저하된 수삼은 조직이 물렁거리고 끈적이는 정도가 심하였으며 자극적인 냄새, 신냄새, 단맛 및 신맛이 강한 경향을 나타냈다.

마우스 수컷 생식세포에서 비스페놀 A에 대한 인삼 에탄올 추출물의 보호 효과 (Protective Effect of Panax ginseng Ethanol Extracts Against Bisphenol A (BPA) in Mouse Male Germ Cells)

  • This study was carried out to evaluate the preventive effect of three forms of Korean ginseng roots (fresh, white and red) against bisphenol A (BPA) toxicity in mouse male germ cells (GC-2spd, TM3, TM4). ROS (reactive oxygen species) generation were measured by DCF-DA (2',7'-dichlorohydrofluorescein diacetate) assay. Also, semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) was performed to quantify the mRNA expression levels of apoptosis-related genes, Bax (pro-apoptotic gene) and Bcl2 (anti-apoptotic gene). ROS generation was increased by $50{\mu}M$ BPA, but definitely decreased by treatment with Korean ginseng extracts (fresh, white and red) in mouse male germ cells. In especial, Korean fresh ginseng extract reduced significantly ROS production to normal control. In addition, Korean fresh and white ginseng extracts suppressed the apoptosis of mouse male germ cells by fine-tuning mRNA levels of apoptotic genes changed by BPA. In general, Korean fresh ginseng extract was more effective than white ginseng extract for reducing BPA-induced oxidative stress and apoptosis in mouse male germ cells. Therefore, Korean fresh and white ginseng may help to alleviate biphenol A toxicity in mouse male germ cells.

Thermal Conversion Pathways of Ginsenosides in Red Ginseng Processing

  • According to the results of my study on the chromatographic analysis of fresh ginseng (Panax ginseng C. A. Meyer) roots, most of the contents of protopanxadiol ginsenosides $Rb_1$, Rc, $Rb_2$, and Rd are derived from the corresponding malonyl ginsenosides in fresh ginseng by a heat process. Also, I confirmed that acetyl ginsenosides are naturally occurring constituents in fresh ginseng, not decarboxylates from malonyl ginsenosides. Seven neutral ginsenosides $Rg_1$, Re, Rf, Rc, $Rb_1$, $Rb_2$, and Rd were transformed to specific conversions in red ginseng preparation conditions. The conversion paths progress by three rules concluded from my study. These conversion rules are I: the ether bond is stable at positions 3 and 6 in the dammarane skeleton, II: the ether bond between sugars is stable in glycosides, and III: the ether bond to glycosides is unstable at position 20 in the dammarane skeleton.

상황 균사체 배양에 의한 파삼의 항암 활성 증진 (Enhancement of Anticancer Activities of Low Quality Ginseng by Phelinus linteus Fermentation)

  • Low quality fresh ginseng was fermented by Pheliuus linteus mycelium at $22^{\circ}C$ for 30 days, then extracted by water solvent at $100^{\circ}C$ for 180 min. On human normal cell lines (HEK293), cytotoxicity was about 10% lower in adding extracts of the fermentation ginseng than that from low quality ginseng. The fermented extracts also inhibited the growth of several human cancer cells. Among them, respectively, digestive organs related cancer cells, such as human stomach adenocarcnioma and human epithelial adenocarcinoma were most effectively inhibited up to 85% and 90%, respectively. Then, selectivities were in the ranges of 3 to 5, compared to 2 to 3 from low quality fresh ginseng. Generally, fermented ginseng extract showed higher anticancer activities as well as higher DPPH radical sacavening activity, possibly due to high contents of total phenolic components as 6.96 mg/g. It was very interesting that the fermented ginseng contained very higher contents of ginsenoside-Rc+$Rb_2$, compared to others in low quality fresh ginseng because of partition digestion of mycelium growth. The results can tell that low quality fresh ginseng can be utilized by the fermentation with Pheliuus linteus mycelium.

세척처리에 따른 수삼 표면의 미생물 제어효과 (Reduction of Microbial Populations on the Surface of Fresh Ginseng by Various Washing Treatments)

  • 현재 대부분의 수삼은 흙이 묻은 상태로 유통되고 있는데 소비자의 편이성을 고려하여 수삼을 세척한 후 포장한 상태로 유통시키기 위한 연구의 일환으로 수삼의 세척처리 방법에 따른 표면세척효과 및 미생물 감균 효과를 조사하였다. 수확 직후 수삼의 부위별 생균수는 5.85~6.63 log CFU/ea범위로 뇌두 부위가 가장 많았고 다음으로는 지근과 주근 순이었다. 부위별 곰팡이 수는 3.67~5.12 log CFU/ea범위로 뇌두 부위가 많았으며 주근과 지근은 거의 유사한 수준이었다. 수삼표면을 상압살수, 고압살수 및 솔질 처리를 하였던 바 고압살수처리에 의해 세척도가 증가하고, 표면 미생물을 감소시킬 수 있었으나 그 효과는 솔질처리의 경우보다는 낮았다. 세척한 수삼의 표면 미생물 수를 더 낮추기 위해 부가적으로 에탄올용액, 차아염소산나트륨 용액, 과산화수소용액, 전해산화수, 오존수 및 중온수를 각각 처리하였던 바 전해산화수 처리 시 생균수와 곰팡이 수가 세척한 수삼에 비해 각각 0.79 CFU/ea, 0.51 log CFU/ea 정도 감소하여 다른 처리구에 비하여서는 우수하였으나 처리구간의 통계적인 유의성은 인정되지 않았다.

Purification and characterization of polyphenol oxidase from fresh ginseng

  • Polyphenol oxidase (PPO) was purified from fresh ginseng roots using acetone precipitation, carboxymethyl (CM)-Sepharose chromatography, and phenyl-Sepharose chromatography. Two isoenzymes (PPO 1 and PPO 2) were separated using an ion-exchange column with CM-Sepharose. PPO 1 was purified up to 13.2-fold with a 22.6% yield. PPO 2 bound to CM-Sepharose, eluted with NaCl, and was purified up to 22.5-fold with a 17.4% yield. PPO 2 was further chromatographed on phenyl-Sepharose. The molecular weight of the purified PPO 2 from fresh ginseng was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and was about 40 kDa. The optimum temperature and pH were $20^{\circ}C$ and 7.0, respectively, using catechol as a substrate. Pyrogallol showed the highest substrate specificity. The effect of a PPO inhibitor showed that its activity increased slightly in the presence of a low concentration of citric acid. High concentrations of acidic compounds and sulfite agents significantly inhibited purified ginseng PPO 2.

A549 human lung cancer cell과 Sarcoma-180 복강암에 대한 수삼(水蔘) 증류약침(蒸溜藥鍼)의 항암효과(抗癌效果) (The Study on Anti-cancer Effects of Distilling Fresh-ginseng Herbal acupuncture against implanted Sarcoma-180 in vivo and A549 human epithelial lung cancer cells in vitro)

  • Objectives ; This study was to investigate the anti-cancer effects of herbal acupuncture with distilled fresh ginseng. The herbal acupuncture was injected to Chung-wan($C.V_{12}$) and Wisu($BL_{21}$) of mice that were subjected to Sarcoma-180 adbominal cancer cell and A549 human epithelial lung cancer cells in vitro. Methods : Anti-cancer effects of distilled fresh ginseng herbal acupuncture were tested by measruing Cox, Bcl-2, and Bax by using RT-PCR in A549 human epithelial lung cancer cells in vitro. And four weeks old Balb/c line male mice weighing around $20\;{\pm}\;3g$ were used to measure survival rate and anti-cancer effect to outputs of interleukin-2 and interleukin-4 using flow cytometry, possibility of mRNA menifestation using RT-PCR, and Cox mRNA. The results are as follows. Results : 1. In measuring mRNA menifestation in Cox, Bcl-2, and Bax by using RT-PCR in A549 human epithelial lung cancer cells in vitro, the result showed that fresh ginseng decreased Cox-2 which is directly involved in Inflammation process. 2. Survival rate was measured in an anti-cancer effect experiment against Sarcoma-180 abdorminal cancer. Median survival time of controlled group was 27 days, of experiment group I was 21 days, and of experiment group II was 27 days. Therefore, experiment group I showed -22.2% increase in survival rate and experiment group II showed no difference compare to controlled group. 3. There was no difference between condition group and controlled and experiment group in measuring outputs of interleukin-2 and interleukin 4 by using flow cytometry 4. In measuring outputs of interleukin-2 by using ELISA, there was no significant difference between condition group and controlled group and there was decrease in experiment group II compared to conditioned and controlled group. 5. In measuring cytokine mRNA menifestation by using RT-PCR, experiment group I showed increase of mRNA menifestation in interleukin-2,4 and $interferon-{\gamma}$ and experiment group II showed no significant difference in $interferon-{\gamma}$. Conclusion : According to the results, fresh ginseng herbal-acupuncture took a little effects in cancer. In using distilled fresh ginseng herbal acupuncture has effect on Cox-2 decrease. However, the difference in concentration of fresh ginseng showed no effect on killing cancer cell. It is assumed that inaccurate concentration of herbal acupuncture and fresh ginseng component could be the reason for this result. Therefore, future consideration will be studies on herbal acupuncture concentration.

생마늘과 무취마늘의 휘발성 향기 성분의 비교 (Comparison of Volatile Flavor Components between Fresh and Odorless Garlic)

  • An attempt was made in this study to analyze volatile flavor components of fresh and odorless garlic (Allium sativum L.), Essential oils in fresh garlic and odorless garlic were isolated by a simultaneous steam distillation and extraction(SDE) method using n-pentane/diethy ether as solvent. A total of 21 and 22 components were identified by GC/MS from the essential oils of fresh garlic and odorless garlic, respectively. Diallyl trisulfide, diallyl disulfide and methyl allyl disulfide were found to be major volatile flavor components in fresh garlic and odorless garlic. By the preparation of ordorless garlic, 77.5% of diallyl trisulfide, 15.0% of diallyl disulfde, 72% of methyl allyl disulfide, and 78.4% of allyl thiol components in fresh garlic were lost. Eleven compounds including 2-vinyl-4H-1,3-dithiin, eugenol and 2-methoxy-4-vinyl-phenol were identified in odorless garlic, but not m fresh garlic.

연포장재 필름으로 개별포장한 수삼의 저장조건에 따른 외관품질 변화 및 개체무게 감소 (Quality Change and Weight Loss of Fresh Ginseng Individually Packaged in a Soft Film According to its Storage Condition)

  • 수삼을 물로 세척한 후 한 뿌리씩 연포장재 필름 주머니(ONY/LDPE/L-LDPE, 200$\times$300mm, 90$\mu\textrm{m}$)에 넣고 밀봉하여 $25^{\circ}C$, 1$0^{\circ}C$ 또는 4$^{\circ}C$에서 저장하면서 외관품질과 개체무게를 조사한 결과 수삼의 외관품질은 저장온도에 관계없이 주로 연화에 의하여 불량해졌으며 외관품질 변화 속도상수와 저장온도의 역수뿐만 아니라 개체무게 변화속도상수와 저장온도의 역수간에도 각각 5% 유의수준에서 양호한 직선관계를 나타내었다. 따라서 $25^{\circ}C$에서의 온도학대시험을 통하여 저온에서 저장중인 개별포장 수삼의 저장수명 및 개체무게 변화율을 단기간에 예측할 수 있을 것으로 판단된다.

반응표면분석법을 이용한 인삼 Root 액체배양조건의 최적화 (Optimization of Submerged Culture Conditions for the Production of Ginseng Root Using Response Surface Method)

  • 식물조직배양기술을 이용하여 인삼 root를 생산하고자, 식물생장조절물질로 유도된 인삼 root를 사용하여 최적액체배양조건을 RSM을 이용하여 조사하였다. 최적액체배양조건을 배지의 pH, sucrose 농도, nitrogen 농도, phosphate 농도의 3 level-4factor의 fractional factorial design에 의하여 조사한 결과, 인삼 root의 생장율은 최저 1.00g에서 최고 2.33g까지 나타났다. 다중회기분석으로 구한 model식을 가지고 등고분석과 3차원분석을 수행한 후 독립변수의 최저 또는 최고수준에서 조속변수가 최대치를 나타내지 않는 배지의 pH와 sucrose농도의 변수에 대하여 model식을 편미분한 결과 인삼 root의 최적액체배양조건은 pH5.6, sucrose 3.8%, nitrogen 50mg/L, phosphate 80.7mg/L로 예측되었다. 이렇게 결정된최적조건값들을 model식에 대입하여 얻은 예상치는 2.36g 이었다.
