To examine the developmental capacity of manipulated embryos after ultrarapid refreezing and thawing, mouse embryos were biopsied at 4-cell stage, frozen twice at 4-cell and morula stages, respectively, and then transferred to rec-ipients. Single blastomeres were biopsied from 4-cell embryos by a modified aspiration method. Biopsied 4-cell embryos were equilibrated into freezing medium at room temperature for 2.5 min, loaded into 40 $\mu$I of freezing medium in 0.25 ml plastic straw and then directly immersed into liqiud nitrogen. Freezing medium for 4-cell embryos consisted of 4.0 M ethylene glycol and O.25 M sucrose in dPBS supplemented with 6 mg/lm BSA. Morulae were frozen into freezing medium containing 5.0 M glycerol instead of ethylene glycol. Thawing was conducted by agitating each straw in 3TC water for 20 sec. The c content of each straw was expelled into 0.5 ml of dilution medium, which consisted of 0.25 M sucrose and 3 mg/ml BSA in dPBS. The thawed embryos were rehydrated in dilution medium for 10 min, washed 3 times with dPBS and then cultured in M16 medium at 37$^{\circ}C$, 5% CO$_2$ in air. Blastocysts that developed from frozen or refrozne biopsied embryos were transferred to recipients on Day 3 of pseudopregnancy, respectively. In vitro and in vivo developmental rates of the biopsied and intact 4~cell embryos after freezing and thawing were 78 (10l/130) and 25% (10/40), and 91 (114/125) and 30% (12/40), respectively. The stone Buddha and Shrine of Unjusa temple (Korean Treasure No. 797) at Hwasun formed in Koryo Dynasty is unique style which faces each other the back parts of south and north within the stone Shrine. The stone Buddha and Shrine is highly evaluated in historical, artistic and academic respects. But, the stone properties have been exposed in the open system various aspects of degradations weathered for a long time without specific protective facilities. To inquire into relative deterioration and environmental factor, air temperature and relative humidity of the stone Buddha and Shrine were monitored for a year of the indoor and outdoor, respectively. As a result, the temperature shows increase and decrease according to the seasons in the tendency to clear. While the relative humidity is high to keep all four seasons. Highly relative humidity environment induces dew condensation on the interior of stone Buddha and Shrine. The dew condensation is recorded at the spring, summer and winter season. The summer season is double the total of spring and winter season. In the case of summer, dew condensation is long time continued due to high temperature and relative humidity that is kept by more than nearly 100%. There is progress towards chemical weathering throughout dissolve rock properties and alteration on the rock surface. In the case of winter, dew condensation is not kept for a long time as summer. In the winter, which showing a below zero may add physical weathering throughout moisture that happen by dew condensation to repeat freezing and thawing. Therefore, the reduction plan of the relative humidity effect on dew condensation should be prepared.
In this study, a commercial production stepwise method for restorative rice with high quality and microbial safety was developed. The stepwise treatment method included steaming, refrigerated aging, and low temperature drying. The soaking rice was steamed twice at $90-100^{\circ}C$, and then, the rice was aged at $0-10^{\circ}C$, frozen at $-20^{\circ}C$, and dried at low temperatures with 5 m/s wind speed at $1-20^{\circ}C$ and 85% relative humidity. Applying the three steps improved sensory qualities compared with the conventional hot air drying and made storage at room temperature for 3 months possible. Specifically, the moisture content of the restorative rice was increased to 30%, which was 4.3 times higher than the 7% of the conventional air dried rice, and the rice grain shape was well maintained. The texture and appearance of the three-step rice were significantly improved (p<0.05) in a sensory evaluation.
To get well matured farm yard manure from ricestraw as quickly as possible during winter season, straw piles wrapped with polyethylene film and/or straw thatch were stored in the vinyl house or open air. Their maturities and changes of temperature in heap were investigated from the beginning of December 1983 to March of next year. Heat increment in vinyl house was high $2-5^{\circ}C$ than at open air at the lowest temperature but it didn't rise over the freezing point. However, the highest temperature was arisen over than $20^{\circ}C$ averagely at the vinylhouse compared to those of open air during three months. Temperature in piles of straw manure was reached to about $70^{\circ}C$ in maximum and rose again very rapidly after repiling in the vinyl house, whereas increment of temperature after repiling was delayed and took long times to reach the maximum temperature at open air. Wrapping with P.E. film also affected the insulation of decomposing heat of straw pile and promoted the repeat of piling even at open air. By these results, ricestraw would be decomposed rapidly by insulation with P.E. film in the vinyl house and could be reached to matured compost for application to field on next spring season. P.E. film covered for vinyl house was endured until May of next year without tear by weathering.

A new type of passive samplers were designed and produced by authors. After evaluating the quality of activated carbon by measuring recovery rate of organic vapors and steadiness of sampling rate, activated carbon with 30 - 35 mesh produced by Company S in Korea was selected. In each passive sampler, an amount of 400 mg of the activated carbon was filled in 25-mm cassette and covered by fixed screen (or wire screen with 100 mesh). In addition to the fixed screen, a wind screen (or wire screen with 300 mesh) was also attached at outer face. The sampling rate of the new Korean passive samplers was estimated Conclusions obtained in the study are as follows. 1. Sampling rates of the newly developed Korean passive samplers were affected by sampling time. For n-hexane, sampling rates of 15- and 60-minute samples were 70.92 and 37.45 ml/min, respectively. Sampling rate of both 200- and 450-minute samples was 25.96 ml/min. It is concluded that, when passive samplers are used for measuring organic vapors, samples be collected longer than 60 minutes. 2. Sampling rate of the passive samplers was also affected by airborne concentration of organic vapors. Lower sampling rates were determined at level of 1/2 threshold limit values (TLVs) recommended by the American Conference of Governmental Industrial Hygienists (ACGIH). It is recommended that sampling rate of the passive samplers be obtained at site by measuring concentrations using both the NIOSH Method and passive samplers simultaneously. 3. When the passive samplers, which collected organic vapors, were exposed to clean air for five hours, there was no significant loss of organic vapors due to reverse diffusion. 4. When samples were stored at room temperature ($21.8{\pm}0.7^{\circ}C$) and refrigerator ($3.8{\pm}0.7^{\circ}C$), there was no significant difference in the accuracy of results. For trichloroethylene and n-hexane, accuracies were within 25 % at both temperatures until seven days. However, poor accuracy exceeding 25 % was indicated in toluene from the first day. It is recommended that samples be stored at freezing temperature below $0^{\circ}C$. 5. Sampling efficiency was significantly affected by direction of the passive samplers. Results of samplers facing wind and down, respectively, were compared. Lower amount of organic vapors were collected when the sampler was oriented down. It is recommended that, when air velocity is low in plants, the passive samplers be oriented to the wind. However, when air velocity is high, the passive samplers be oriented down.
To extend shelf life of mulberry fruits, usually expensive freeze drying of freezing process is required. In this study an economic to multipurpose agricultural products dryers with mulberry dryer and technique was developed to reduce postharvest processing cost. Fuel and electricity consumption for drying were lowest when drying temperature was $60^{\circ}C$. And various drying conditions (fuel 3 hrs, heat 1 hr, room temperature air velocity 51 hrs) of the developed dryer were analyzed to improve drying performance. Dried mulberry fruits had dark brown color and marketable high quality explain the semi-dried characteristics.
BACKGROUND: The freezing injury by pruning can be reduced by suspending pruning work when severe cold weather (-23 to -49℃) is forecast. Minimum air temperature of the study area, Gunwi region at February 3, 2012 was -21.9℃, and the subzero temperature continued until April 8, 2012. This study was conducted in two years to investigate the effect of delaying winter pruning until full bloom on shoot growth and fruit quality of 'Fuji'/M.9 apple trees. METHODS AND RESULTS: The time of pruning were March 26 for dormant, April 3 for bud break, and May 2 for full bloom. The winter pruning at full bloom significantly reduced fruit weight for two years compared with the control (winter pruning at dormant), and shoot growth was reduced only in the following year. There was no significant effect of delaying winter pruning at bud break on soluble solid content, fruit red color, return bloom, and pruning weight for two years compared with the control. CONCLUSION(S): These results indicated the delaying winter pruning at bud break of 'Fuji'/M.9 apple tree did not offer any disadvantage over comparable dormant pruning, since the fruit quality was not affected. The delayed pruning at full bloom resulted in decreased fruit weight, though shoot growth, fruit quality, and return bloom were not affected by the delayed pruning. So, the delayed pruning should be considered carefully only for the fruit tree orchards in diseases.
Dryers becoming commercially available for experimental and industrial use are classified to general drying oven, hot-air dryer, vacuum dryer, freezing dryer, etc. and kinds of them are various from the function, size and volume, etc. But the moisture measurement is not applied although it is important factor for the quality control and the performance improvement of products, and then now is very passive because the weight is weighed arbitrarily after dry-end. Generally the method for measuring moisture is divided by a direct measurement method and a indirect measurement method, and the former such as the change of weight or volume on the front and rear of separation of moisture, etc. is mainly used. Relatively a indirect measurement is very limited to apply due to utilize measurement apparatuses using temperature conductivity and micro-wave etc. In this research, we easily designed the moisture measurement system using the open-source based Arduino, and monitored moisture fluctuations and weight profiles in the real-time without the effect of external environment. Concretely the temperature-humidity and load cell sensors were packaged into a drying oven and the various change values were measured, and their sensors capable to operate 60℃ and 80℃ were selected to suitable for the moisture sensitive materials and the food dry. And also the performance safety using the organic samples of banana, pear, sawdust could be secured because the changes of evaporation rate as the dry time and temperature, and the measurement values of load cell appeared stable response characteristics through repeated experiments. Hereafter we judge that the reliability can be improved increasingly through the expansion of temperature-humidity range and the comparative analysis with CFD(Computational Fluid Dynamics) program.
This study suggests a method of alleviating global warming by the increase of the Earth surface albedo through Artificial Sea ice Increasing (ASI) over the Available Freezing Areas (AFA). The method is developed based on the fact that the large sea surface area in or near the Arctic and the Antarctic has no ice even though both water and air temperatures are below zero and the artificial sea ice generation is thus available. The mean energy of $0.85Wm^{-2}$, which was suspected of adding to the earth by the global warming effect was calculated to offset at once when the sea ice area about $4.09{\times}10^6km^2$ was additionally increased. In addition, three techniques for producing ice plates on the sea surface (using ships, installation apparatus, and floating matter such as Green Cell Foam) for ASI were proposed. According to the result of simple analysis using the energy balance model, when ASI was maximally operated only for 3 months (September, October, and November) over AFA, it is expected that the annual mean temperature of earth surface would be decreased about $0.11^{\circ}C$ in the following year. On the other hand, in case of generating the artificial sea ice in all four seasons, a risk of triggering snowball earth was detected.

This study was carried out to establish a spatial decision support system for evaluating climatic aspects of a given geographic location in complex terrains with respect to the quality apple production. Monthly climate data from S6 synoptic stations across South Korea were collected for 1971-2000. A digital elevation model (DEM) with a 10-m cell spacing was used to spatially interpolate daily maximum and minimum temperatures based on relevant topoclimatological models applied to Jangsoo county in Korea. For daily minimum temperature, a spatial interpolation scheme accommodating the potential influences of cold air accumulation and the temperature inversion was used. For daily maximum temperature estimation, a spatial interpolation model loaded with the overheating index was used. Freezing risk in January was estimated under the recurrence intervals of 30 years. Frost risk at bud-burst and blossom was also estimated. Fruit quality was evaluated for soluble solids, anthocyanin content, Hunter L and A values, and LID ratio, which were expressed as empirical functions of temperature based on long-term field observations. AU themes were prepared as ArcGlS Grids with a 10-m cell spacing. Analysis showed that 11 percent of the whole land area of Jangsoo county might be suitable for quality 'Fuji' apple production. A computer program (MAPLE) was written to help utilize the results in decision-making for site-selection of new orchards in this region.
