세포내 인산농도에 의한 일일초 모상근생장 및 Catharanthine 생산의 조절 (Regulation of Growth and Catharanthine Production by the Intracellular Phosphate Level in Hairy Root Cultures of Catharanthus roseus)

  • 일일초 모상근배양에서 세포내 인산농도와 Catharanthine 생산과의 관계를 규명하였다. 모상근의 생장은 생장배지의 인산농도에 비례하여 증가하였다. Catharanthine 함량은 인산을 첨가하지 않은 배지에서 모상근을 배양하였을 때 최대값을 나타내었고 배지내의 인산농도를 증가함에 따라 비례적으로 감소하였다. 그리고 세포내의 인산농도에 비례하여 모상근의 생장은 증가하나 catharanthine의 함량은 감소하였다. Catharanthine 함량은 g 건조세포당 인산농도가 100mole 이하일 때에 현저하게 증가하였다. 이상의 결과로 세포내 인산농도는 모상근의 생장과 이차대사산물의 조절에 중요하게 관여함을 시사한다.

감귤원 토양에서 분리한 인산염 가용화 미생물 Bacillus sphaericus PSB-13의 특성 (Characteristics of Bacillus sphaericus PSB-13 as Phosphate Solublizing Bacterium Isolated from Citrus Orchard Soil)

  • 본 연구는 감귤원 토양유형별로 고정되어 있는 난용성 인산염함량을 파악하고 이를 생물학적으로 이용하고자 인산가용화 우수미생물을 선발하여 특성을 조사하였다. 토양중 난용성 인산염의 형태별 분포는 Al-P>Ca-P>Fe-P 순으로 화산회토 토양이 비화산회토 보다 많았다. PDA-P배지를 이용하여 인산염 가용화능이 우수한 Bacillus sphaericus를 분리하였으며 pH 4~5와 $30^{\circ}C$에서 생육이 제일 좋았다. Bacillus sphaericus은 $AlPO_4$에서 324.5 ppm, $Ca_{10}(PO_4)_6(OH)_2$에서 334.8 ppm, $Ca_3(PO_4)_2$에서 207.0 ppm의 유리인산을 생성하였으며 인산효소활성은 $35^{\circ}C$에서 배양할 때 높았다. Bacillus sphaericus를 Bentonite 등 담체에 고정 후 온도를 달리하여 7개월 보존기간 동안 밀도변화를 조사한 결과 $10^5c.f.u.\;g^{-1}$ 수준과 인산가용화능을 유지하여 추후 생물비료로 이용이 가능할 것으로 기대된다.

척추동물 뇌조직의 Energy 대사에 관한 계통학적 비교연구 (A Comparative Study on the Energy Metabolism of Brains of Several Vertebtrates with Respect to Their Phyogeny)

  • The present paper deals with the comparative study on phylogenic difference in the patterns of energy metabolism of brain slices of several vertebrate species by measuring oxygen consumptionwith glucose-6-phosphate, glucose-1-phosphate, glyceraldehyde-3-phosphate or glutamate as respiratory substrate employing Warburg's manometric method, by determination of the utilization rate of glucose using glucose-1-C14 by analyzing patterns of free amino acid distribution , and by histochemical determination using glucose-1-C14 by analyzing patterns of free amino acid distribution acid distribution , and by histochemical determination of glycogen contents. 1. Glucose enhances the oxygen consumption of brain slices of animals belinging to reptile, aves and mammalia while it shows a tendency to decrease that of animals belonging to pisces and amphibia. 2. Glucose-6--phosphate increase oxygen consumption more than glucose in every species examined, while glucose-1-phosphate and glyceraldehyde-3-phosphate increase that of Rana nigromaculata only . In general m, it appears that phosphosugars are more effective as a respiratory substrate to those species which have less endogenous respiration than to those having larger endogenous respiration. 3. Similar patterns of free amino acid distribution and the relative amount are found among the species and in every species examined glutamic acid is detected in the larges amount . ${\gamma}$-Amino butyric acid, glycine, alanine and aspartic acid are found in every species. 4. Ophicephalus showed less oxygen consumption than endogenous respiration when glutamate was added to the medium. When sodium fluoride was added, the oxygen consumption was some what increased . Such phenomenon wasnot found in the frog. 5. The result of histochemcial analysis of the brain showed that glycogen was abundantly present in the fish , amphibia , and especially in the reptile and that no distinctive grains of glycogen were found in the bird and mammal . From these facts, it may be supposed that anaerobic glycolysis as energy source dominates in fish and amphibia and aerobic respiration through the oxidation of glucose dominates in bird and mamal , the reptile occupying transitional position between these two categories. The way of obtaining energy for brain activity by the oxidation of glucose supplied from the circulating blood is seemed to be first acquired by reptile and the function is completed both in aves and mammal.

Application of Immobilization Technology in Solubilization of Rock Phosphate

  • Phosphates solubilizing bacterial strains belong to Pantoea, Burkholderia and Enterobacter were isolated and employed in assessing their solubilization ability of Ca phosphate and ER phosphate (Eppawala Rock Phosphate). Among the bacterial strains used, PSB-13 (Pantoea rodasii) showed higher Ca-phosphate solubilization ($1100{\mu}g\;ml^{-1}$) as well as rock phosphate solubilization ($168{\mu}g\;ml^{-1}$). The strain was then immobilized in agar to further assess its phosphate solubilization ability. According to the results, agar encapsulated strain solubilized 0.3%, 7.31%, 20.24%, and 20.62% more Ca-phosphate and 11.53%, 15.29%, 28.48%, 36.55% (respectively in 4 cycles) more ER-phosphate than free cells. The reuse efficiency of agar entrapped bacterial cells for Ca-phosphate and ER-phosphate solubilization was greater than that by freely suspended bacterial cells. In conclusion, immobilization could enhance the phosphate solubilization capacity of the strains and thus could be used effectively in enhancing solubilization of ER phosphate.

GALK Hyperactivity로 인한 갈락토스혈증의 임상적 특성에 관한 연구 (Clinical and Laboratory Characteristics of Galactokinase Hyperactivity)

  • Purpose: Galactose is metabolized to galactose-1-phosphate by galactokinase (GALK), galactose-1-phosphate uridyltransferase (GALT) and UDP-galactose-4-epimerase (GALE), and galactosemia occurs when each enzyme is deficient. In Korea, unlike foreign countries, classic galactosemia is rare and transient galactosemia due to GALK hyperactivity is reported, but studies on frequency, clinical significance, and genetic variation are lacking. In this study, we analyzed the clinical characteristics of patients with galactosemia due to GALK hyperactivity. Methods: We investigated 85 patients who had an elevated galactose level in the neonatal screening test without deficiency of enzymes at Department of Pediatrics, Seoul & Bucheon Soonchunhyang University Hospital from January 2008 to June 2016. We investigated the level of galactose, galactose-1-phosphate, GALK and duration of galactose normalization, and analyzed the correlation between GALK elevation and galactose, galactose-1-phosphate and duration of galactose normalization. And the levels of galactose, galactose-1-phosphate, and duration of galactose normalization were compared between the galactose-free formula feeding group and non-feeding group. Results: Mean age of visit was $26.7{\pm}16.1days$. Duration of galactose normalization was $35.3{\pm}20.5days$. Mean galactose level was $18.5{\pm}7.3mg/dL$ in the neonatal screening and follow-up galactose level in serum was $2.3{\pm}5.4mg/dL$. The mean value of galactose-1-phosphate was $6.0{\pm}4.7mg/dL$ and the mean GALK level was $3.84{\pm}1.28{\mu}mol/Hr/gHb$. There was no significant correlation between GALK levels and galactose levels in the neonatal screening test (P=0.351), and we analyzed the correlation between GALK levels and follow-up galactose levels in serum, there was no significant correlation (P=0.101). There was a significant correlation between GALK levels and galactose-1-phosphate (P=0.015), and the correlation between GALK levels and duration of galactose normalization was not statistically significant (P=0.176). 49% of the patients were fed galactose-free formula, and 45% were not. Galactose and galactose-1-phosphate levels in the neonatal screening test were statistically significantly higher (P=0.004, 0.034) in using galactose-free formula group. Duration of galactose normalization was not related to the use of galactose-free formula (P=0.266, 0.249). Conclusion: Galactosemia due to GALK hyperactivity seems to be a temporary phenomenon and may not require galactose restriction. More research is needed on the role of the nuclear protein, racial traits and genetic variations in Korean patients.

Ascorbic Acid와 Pyrophosphate로부터 Ascorbic Acid-2-Phosphate의 효소적 생산

  • Microorganisms capable of producing ascorbic acid-2-phosphate (AsA2P) from ascorbic acid (AsA) and pyrophosphate (PPi) were screened from the culture collection of this laboratory. Among them, Cellulomonas sp. AP-7 showed the highest productivity of AsA2P. The optimal conditions for the production of AsA2P from AsA and PPi with cell-free extract as an enzyme source were investigated. The reaction mixture for the maximal production of AsA2P consisted of 21 g protein of cell-free extract per liter as the enzyme source, 250 mM AsA, 200 mM sodium pyrophosphate, 150 mM sodium acetate buffer (pH 4.5). By using this reaction mixture, 31.9 mM of AsA2P, which corresponded to a 12.76% yield based on AsA, was produced after incubation of 48 hr at 33$\circ$C.

Chlorella 세포의 핵산 인단백질 및 인지질의 생합성에 미치는 마그네슘의 결핍 효과 (Effect of magnesium-deficiency on the biosynthesis of nucleic acid, phosphoprotein, and phospholipid in Chlorella cells)

  • Chlorella ellipsoidea were grown in a Mg-free medium. Aliquots of the algal cell were taken out at the beginning and predetermined time intervals during the culture and were analyzed the contents of phosphate in various fractions of the cell constituents. The results obtained were compared with those of the control. When Chlorella cells were grown in a Mg-free medium, the contents of phosphate in the DNA protein, RNA-olyphosphate complex, nucleotidic-lbileP, and PCA-soluble, fractions decreased compared with those of the control, while the content of acid insoluble polyphosphate increased significantly. On the otherhand, RNA-P and lipid-P showed the tendency of decrease at the early stage of the culture, but they were increased more than those in the control as culture proceeds. It is showed that phosphate turnover from acid-insoluble polyphosphate into DNA, protein, and RNA-polyphosphate complex was inhibited by magnesium-deficiency of the cells.

인산가용화 사상균 Penicillium sp. PS-113의 고체배양 (Solid Culture of Phosphate-solubilizing Fungus, Penicillium sp. PS-113)

  • A fungus, Penicillium sp. PS-113, isolated from soil showed the high phosphate-solubilizing activity in patato dextrose broth-rock phosphate to produce free phosphates to the culture broth with the concentrations of 585 ppm against rock phosphate. In this medium, the optimum temperature and initial pH to solubilize rock phosphate were 30$^{\circ}C$ and pH 7.5, respectively. In order to make the mass production of the conidia from this fungus, we cultured in on various solid-based media like barley, corn, wheat, rice, rice bran, and compost. As a result, the fungus highly produced conidia ranging from 2.1 to $5.1{\times}10_9$ conidia/g${\cdot}$media on these solid media except compost-based medium, which was 0 times less than others. Effects of inoculation of the phosphate solubilizing fungus as a biofertilizer were studied in perlite-based pot cropped with Zea mays Suwon 19. Inoculation of Penicillium sp. PS-113 increased in plant height (1.4 times), plant weight (5.2~8.1 times) and root length (1.1~1.2 times) at 60-day cultivation, compared to Hogland solution either without $NH_4H_2PO_4$ or displace $NH_4H_2PO_4$ to powdered rock phosphate, a phosphorus source for plant growth.

여러 가지 형태에 따른 Alcohol-Oxidase의 특성 비교 (Comparison of the Characteristics of Alcohol-Oxidase by the Various Forms)

  • The properties of the alcohol-oxidase from yeasts assimilating of methanol(Hansenula polymorpha CBS 4732, Pichia pastoris CBS 2612 and Candida boidinii CBS 8106) as free(cellules, purified enzymes) and immobilized forms(immobilized cellules, immobilized enzymes) were investigated. Immobilization enhanced the activity and stability of alcohol-oxidase to a certain degree. The optimum temperature of the immobilized alcohol-oxidase was lower than those of the free forms. The pH / activi쇼 profiles of alcohol-oxidase did not change by immobilization, but changed by the microorganisms. When the immobilized cellules were stocked at 4$^{\circ}C$ in 10mM potassium phosphate buffer(pH 7.5 or 8.0), alcohol-oxidase was more stable than those were stocked in potassium phosphate buffer containing 0.65M sucrose. The immobilization modifies the conditions of oxidation on the various substrates. alcohol-oxidase in immobilized forms showed some with higher Km value for methanol than that in free ones.
