Effects of Reactive Oxygen Species on DNA Stability in Humnn Spermatozoa

  • This study was designed to investigate the effects of reactive oxygen species (ROS) on DNA stability in human spermatozoa. To verify human spermatozoa were incubated with xanthine-xanthine oxidase (X 100$\mu$M-XO 50 mlU ~ 400 mIU), $H_2O_2$ (125 $\mu$M ~ 1 mM), sodium nitroprusside (SNP 0.1 $\mu$M ~ 100 $\mu$M) or lymphocyte. Otherwise, spermatozoa were incubated under low $O_2$ (5%) condition. Damage of sperm DNA was analyzed by single cell electrophoresis (Comet assay) and flow cytometry after acridine orange staining. In the presence of ROS, there was increase in DNA damage. The rate of DNA single strand breakage (9.0$\pm$1.0% ~ 46.0$\pm$4.6%) and DNA fragmentation (7.51$\pm$1.0% ~ 29.5$\pm$4.6%) were similar regardless of the kinds of ROS and exposure time. DNA damage in the lower $O_2$ condition (5%) was lower than ambient $O_2$ condition (20%). Taken together, it suggested that sperm DNA might be damaged by ROS. In the presence of ROS, increase in DNA damage and chromatin instability was obvious in spite of short exposure. Although present study reconfirmed that sperm incubation in the low concentration of ROS have the benefit m the induction of capacitation and Ah, the increase in DNA damage by ROS and possible genetic problem should be considered before the human trials.

3-Hydrogenkwadaphnin Induces Monocytic Differentiation and Enhances Retinoic Acid-mediated Granulocytic Differentiation in NB4 Cell Line

  • Recently, we have reported that 3-hydrogenkwadaphnin (3-HK), a diterpene ester isolated from Dendrostellera lessertii (Thymealeaceae), is very effective against leukemia cell lines without any detectable effects on normal cells (Moosavi et al., 2005b). In this study, we report that 3-HK induces $G_1$ cell-cycle arrest, differentiation and apoptosis in APL NB4 cell line. Indeed, the drug between 24 to 96 h induced 7-65% growth inhibition of NB4 cells. Cell viability was also decreased by 2-55% between 24 to 96 h treatments with the drug, respectively. These effects of the drug were also dose-dependent. According to flow cytomtry results, 3-HK (15 nM) induced a significant G1-arrest up to 24 h which was consequently followed with appearance of sub-$G_1$ peak at 72 to 96 h. Hoechst 33258 staining and DNA fragmentation assays confirmed the occurrence of apoptosis among the treated cells. On the other hand, NBT reducing assay, Wright-Giemsa staining, phagocytic activity and expression of cell surface markers (CD11b and CD14) confirmed that the inhibition of proliferation is associated with differentiation especially toward macrophage-like morphology. Interestingly, 3-HK at 5 and 10 nM enhanced the effects of all-trans retinoic acid (ATRA) in NB4 cells. Based on these results, 3-HK might become an ideal candidate for treatment of APL patients pending full exploration of its biological functions.

  • The purpose of this study is to review the features of eroticism expressed in men's fashion, to explain psychological and social contexts which engender eroticism and to analyze in what way eroticism is portrayed in men s fashion in this context. Eroticism as the metonymy of forbidden sexual desire, has been embodied mostly in visual forms such as picture and photo. From a psychoanalytic view, the context in which eroticism is formed can be explained by primary narcissism and fetishism. Primary narcissism is the feeling of satisfaction with the self in which the subject who is admiring and the object of admiration are one and the same. Accordingly, in order to give rise to eroticism, both subject and object have to exist. Fetishism, the metonymy of castrated penis, is also one of the factors to produce eroticism. Metaphorically seen as a woman who has a penis, a man who dresses in the same manner as a woman dresses can be a source of eroticism to gays. From a sociological view, the context in which eroticism is fostered depends on the dynamic relations among social powers. In these relations, who is a subject or who is an object has been continuously changing. In the Post-modern culture appearing in the late 20th century, power begins to take various forms, and gays and women who had never been subjects begin to make man an object of eroticism. The other point is that social morality, ignoring desire itself, objectifies sexual desire and seeks to remove It by exchanging it for objectified symbols. The design elements provoking eroticism in men s fashion are exposure and decoration. In particular, models in exposure and decorative fashion have been objectified through the methods of fragmentation rather than showing the whole figure or removing a person s individuality or will,.

Protective Effects of Gyungokgo on Oxidative Stress-Induced Apoptosis of H9c2 Cardiomyoblast Cells (산화적 손상으로 유발된 심근세포 고사에 대한 경옥고의 방어효과)

  • Backgrounds & Objectives : The water extract of Gyungokgo (GOG) has traditionally been used for treatment of general weakness and hemoptysis in oriental medicine. However, little is known about the mechanism by which the water extract of GOG rescues cells from these damages. This study was designed to investigate the protective mechanisms of GOG on H2O2­induced cell death in H9c2 cardiomyoblasts. Methods : In this study, we used H9c2 cells. Cells were treated with oxidative stress in the absence and presence of 1000㎍/ml GOG for 12hrs. Cells were treated with various concentrations of GOG for 12hrs. Cell viability was measured by MTT assay. Oxidative stress, which markedly decreased the viability of H9c2 cells, was characterized by apparent apoptotic features such as chromatin condensation as well as fragmentation of genomic DNA and nuclei. Results : GOG significantly reduced H₂O₂-induced cell death and apoptotic characteristics. The cotreatment of GOG and H₂O₂ in H9c2 cells also induced the phosphorylation of ERKs in a time-dependent manner. Moreover, PD098059, a MEK1 (upstream activator of ERK) inhibitor attenuated the protective effect of GOG on H₂O₂-induced cytotoxicity in H9c2 cardiomyoblast cells. Conclusions : These results suggest that MEK/ERK pathways play important roles in the protective effects of GOG in H9c2 cells. Taken together, they suggest that the protective effects of the water extracts of GOG against oxidative damages may be mediated by the regulation of HO-1, Fas/FasL and Bcl-XS proteins.

Antitumor Effect of Gagamjengac-tang and $As_2O_3$ in Human Lung Cancer Cell Line, H-157 (폐암세포주 H-157에서 가감증액탕과 $As_2O_3$의 병용처리에 의한 항종양효과)

  • Objectives : This study was designed to elucidate the synergistic cytotoxic mechanisms of the co-treatment of Gagamjengac-tang (GGJAT) and As₂O₃ in human lung cancer cell line, H-157. Methods : The combination of GGJAT and As₂O₃ synergistically augmented the cytotoxicity of GGJAT and As₂O₃ in H­157 cells. The cytotoxicity by the combination of these two drugs was revealed as apoptosis which was characterized by chromatin condensation and fragmentation in DAPI staining. Results : Antioxidant NAC completely blocked the apoptotic death of H-157 cells by GGJAT and As₂O₃. The apoptotic cytotoxicity of GGJAT and As₂O₃ was accompanied by the induction of DR4 and DR5 in RT-PCR. In addition, antioxidant enzymes such as SOD1, GSH synthetase and GSH reductase were also increased in H-157 cells treated with GGJAT and As₂O₃. However, of note, p53, Fas, FasL and TRAIL were not detected in H-157 cells treated with GGJAT and As₂O₃ by RT-PCR. Conclusions : These results suggest that the synergistic cytotoxicity of the co-treatment of H-457 cells treated with GGJAT and As₂O₃ may cause induction of death receptors DR4 and DR5 as well as reactive oxygen species.

  • Objective : In this study, the ability of Oriental medicine Samultanggamibang(SMTG) to induce apoptosis was investigated in B16F10 melanoma cells. Method : Tetrazolium-based colorimetric assay was performed for cytotoxicity test. Several new assays for the basis of biochemical events associated with apoptosis such as DNA fragmentation by a flow cytometry, caspase-3 activation and PARP cleavage by Western blotting should be carried out potentially useful for the basis of biochemical events associated with apoptosis such as a flow cytometry and caspase-3 activation. Results : (1) The number of B16F10 melanoma cells was less than 30 % after exposure to 1 mg/ml SMTG for 48 h. SMTG increased cytotoxicity of B16F10 melanoma cells in a dose- and time-dependent manner. (2) The percentage of apoptotic cells by flow cytometric analysis of the DNA-stained cells increased to 21 % at 24 h and 25 % at 48 h after treatment with 1 mg/ml SMTG. (3) SMTG-induced apoptosis was accompained by the activation of caspase-3 and the specific proteolytic cleavage of poly-ADP-ribose polymerase. (4) SMTG induces the activation of caspase-3 and the specific proteolytic cleavage of poly-ADP-ribose polymearse and eventually leads to apoptosis through c-Jun NH2-terminal protein kinase (JNK)-dependent manner in B16F10 melanoma cells. Conclusion : SMTG had a strong cytotoxic effect of B16F10 melanoma cells.

  • This paper proposes three methods for improving the accuracy of auto-music transcription considering with time-varying tempo from monophonic sound. The first one that uses TV(Total Variation) filter for smoothing the pitch data reduces the fragmentation in the pitch segmentation result. Also, the measure finding method that combines three different ways based on pitch and energy of sound data, respectively as well as based on rules produces more stable result. In addition the temporal result of note-length encoding is corrected in optimal way that the resulted encoding minimizes the sum of quantization error in a measure while the sum of note-lengths is equal to the number of beats. In the experiment with 16 children songs, we obtained the improved result in which measure finding was complete, the accuracy of encoding for note-length and pitch was about 91.3 and 86.7, respectively.

Fusarium mangiferae as New Cell Factories for Producing Silver Nanoparticles

  • Finding a safe and broad-spectrum medication is a goal of scientists, pharmacists, and physicians, but developing and fabricating the right medicine can be challenging. The current study describes the formation of silver nanoparticles (AgNPs) by Fusarium mangiferae. It involves the antibiofilm activity of the nanoparticles against Staphylococcus aureus. It also involves cytotoxic effect against mammalian cell lines. Well-dispersed nanoparticles are formed by F. mangiferae. The sizes of the nanoparticles were found to range from 25 to 52 nm, and UV-Vis scan showed absorption around 416-420 nm. SEM, TEM, and AFM results displayed spherical and oval shapes. Furthermore, the FTIR histogram detected amide I and amide II compounds responsible for the stability of AgNPs in an aqueous solution. AgNPs were observed to decrease the formation of biofilm at 75% (v/v). DNA reducing, smearing, and perhaps fragmentation were noticed after treating the bacterial cells with 50% (v/v). Additionally, cell lysis was detected releasing proteins in the supernatant. It was also observed that the AgNPs have the ability to cause 59% cervical cancer cell line (HeLa) deaths at 25% (v/v), however, they showed about 31% toxicity against rat embryo fibroblast transformed cell lines (REF). The results of this study prove the efficiency of AgNPs as an antibiofilm against S. aureus, suggesting that AgNPs could be an alternative to antibiotics. It must also be emphasized that AgNPs displayed cytotoxic behavior against mammalian cell lines. Further studies are needed for assessing risk in relation to the possible benefit of prescribing AgNPs.

New HDAC inhibitor, IN2001 induces apoptosis/cell cycle arrest in human breast cancer cells

  • The acetylation of histone is one of the mechanisms involved in the regulation of gene expression and is tightly controlled by two core enzymes, histone acetyltransferase (HAT) and deacetylase (HDAC). There are several reports that imbalance of HAT and HDAC activity is associated with abnormal behavior of the cells in morphology, cell cycle, differentiation, and carcinogenesis. Recently, an increasing number of structurally diverse HDAC inhibitors have been identified that inhibit proliferation and induce differentiation and/or apoptosis of tumor cells in vivo and in vitro. In this study, we have investigated the effects of novel HDAC inhibitors, IN2001 on ER positive and ER negative human breast cancer cell lines. The growth inhibition, cell cycle arrest and apoptosis of cells by HDAC inhibitors were determined using SRB assay, DNA fragmentation, and flow cytometry. We found that IN 2001 as well as Trichostatin A inhibited cell growth dose-dependently in both ER Positive and ER negative human breast cancer cell lines. The growth inhibition with HDAC inhibitors was associated with profound morphological change. The result of cell cycle analysis after 24 h exposure of IN2001 showed G2-M cell cycle arrest in MCF-7 cell and apoptosis in T47B and MDA-MB-231 cell. In summary, IN2001 has antiproliferative effect on human breast cancer cells regardless of the expression of estrogen receptor. These findings heights the possibility of developing HDAC inhibitors as potential anticancer therapeutic agents for the treatment of breast cancer.

Structural characterization and degradation efficiency of degradation products of iopromide by electron beam irradiation (전자선 처리 후 생성된 Iopromide의 분해산물 구조 규명 및 분해 효율)

  • Iopromide is an X-ray contrast agent that has been detected frequently with high concentration level in surface waters. Structural characterization of degradation products and measurement of degradation efficiency of iopromide by an electron beam irradiation were performed. For the fortified sample with iopromide, electron beam irradiation (UELV-10-10S, klysotrn, 10 MeV, 1 mA and 10 kW) was performed. The chemical structures of I_D_665 and I_D_663, which are degradation products of iopromide, were proposed by interpretation of mass spectra and chromatograms by LC/ESI-MS/MS. The mass fragmentation pathways of mass spectra in tandem mass spectrometry were also proposed. Iopromide was degraded 30.5~98.4% at dose of 0.3~5 kGy, and 97.8~30% in the concentration range $0.5{\sim}100{\mu}g/kg$ at electron beam dose of 0.3 kGy, respectively. Thus, increased degradation efficiency of iopromide by electron beam irradiation was observed with a higher dose of electron beam and lower concentration.
