Stable Isotope Chemistry of Bone Collagen and Carbonate Assessed by Bone Density Fractionation

  • This paper presents a stable isotope chemistry of bone collagen and carbonate. Bone carbonate has the potential to provide additional isotopic information. However, it remains controversial as to whether archaeological bone carbonate retains its original biogenic signature. I used a novel application of bone density fractionation and checked the integrity of ${\delta}^{13}C_{apa}$ values using radiocarbon dating. Diagenesis in archaeological bone carbonate still remains to be resolved in extracting biogenic information. The combined use of bone density fractionation and differential dissolution method shows a large shift in the ${\delta}^{13}C_{apa}$ values. Although ${\delta}^{13}C_{apa}$ values are improved in lighter density fractions, a large percentage of contamination in bone carbonate was reported via $^{14}C$ dating compared to that noted with bone collagen.

Temperature Controllable HPLC Column for Preparative Fractionation of Polymers

  • An HPLC column with a self-contained temperature control device was constructed for preparative temperature programmed interaction chromatography. Two Peltier plates were attached to a large bore column ($120{\times}22\;mm$ i.d.) and the column temperature was controlled by PID mode feed back control. At a flow rate of 1.5 mL/min, the column temperature could be increased and decreased at a rate as high as $50^{\circ}C/min$ and $10^{\circ}C/min$, respectively, which is much faster than using a column jacket and bath/circulator. The rapid heating and cooling rates allows a high repetition rate of chromatographic fractionation. The performance of the temperature controllable column was demonstrated successfully by the fractionation of homo-polymer precursors from diblock copolymers.

Increase in δ15N of Nitrate through Kinetic Isotope Fractionation Associated with Denitrification in Soil

  • To observe the changes in isotopic composition (${\delta}^{15}N$) of $NO_3{^-}$ during denitrification, an incubation experiment using soil treated with nitrification inhibitor (2-chloro-6-trichloromethyl-pyridine) under water-saturated condition was conducted for 153 h. The $NO_3-N$ concentration decreased from 73.3 to $20.6mg\;kg^{-1}$ during the incubation period, with denitrification rate constant of $0.00905h^{-1}$, and ${\delta}^{15}N$ values of $NO_3-N$ increased from +0.9 to +25.5‰ with decreasing the $NO_3-N$ concentration. The increase in the ${\delta}^{15}N$ values of $NO_3-N$ is due to kinetic isotope fractionation, which always results in $^{15}N$ enrichment of the substrate. The isotopic fractionation factor calculated in this study was 1.0196, an indication that 1.96% more $^{14}NO_3{^-}$ reacted at a given time interval than a comparable number of $^{15}NO_3{^-}$. The ${\delta}^{15}N$ values measured through the incubation study showed a good agreement with the results calculated from the Fochts isotope fractionation model. Our results suggest that when the ${\delta}^{15}N$ of $NO_3{^-}$ is used for tracing the fate of N, the kinetic isotope fractionation associated with denitrification must be taken into consideration.

Reduction of Saturated Fatty Acid Methyl Esters of Biodiesel Produced from Beef Tallow by Acetone Fractionation (우지로부터 합성된 바이오 디젤의 포화도 감소를 위한 용매 분별 연구)

  • It is known that the content of saturated fatty acids methyl ester (SFAME) affect the pour point of biodiesel at low temperature. In this study, biodiesel (BD) was produced from beef tallow (TAL) by alkali catalyst. To reduce the saturation in BD, acetone fractionation was applied. Besides, TAL was also solvent-fractionated to reduce the saturated fatty acid (SFA) content for further producing BD. With acetone, TAL or TAL methyl ester (5:1 v/w) were fractionated at 10, 0, -10, and $-15^{\circ}C$, respectively. At $-10^{\circ}C$, 17.35% of SFA was observed in fractionated TAL (liquid part, -10TAL) when 5:1 solvent ratio was used for 24 hr. Under the same condition, fractionated BD (liquid part, -10BD) showed SFA (33.14%) with 78wt % yield. Also, fractionation of BD with different concentration of crystallizer 209 (0.1, 0.5, and 1%) along with different time (2, 6, 12, and 24 hr.) was observed. The best condition for reducing the SFA was 0.5% of crystallizer 209 addition for 12 hr of fractionation time at $-10^{\circ}C$, in which 30.14% of SFA content was observed in BD (liquid part). Among different crystallizer, ps 66 showed the least content of SFA content (23.28%) in BD after fractionation ($-10^{\circ}C$ and 24 hr) with 0.5wt% addition.

Effect of Sample-loading on Fractionation Efficiency (FE) in a Large Scale Splitter-less Gravitational SPLITT Fractionation (GSF)

  • Gravitational SPLITT fractionation (GSF) provides separation of colloidal particles into two subpopulations in a preparative scale. Conventionally, GSF is carried out in a thin rectangular channel having two inlets and two outlets at the top and bottom of the channel, respectively. And the channel is equipped with two flow-splitters, one between the top and bottom inlets and another between the top and bottom outlets. A large scale splitter-less GSF system had been developed, which was designed to operate in the full feed depletion (FFD) mode. In the FFD mode, there is only one inlet through which the sample is fed, thus preventing the sample dilution. In this study, the effect of the sample-loading (in the unit of g/hr) on the fractionation efficiency (FE, number% of particles in a GSF fraction that have the sizes expected by theory) of the new large scale splitter-less FFD-GSF system was investigated. The system was tested in the sample-loading range of 3.0-12.0 g/hr with polyurethane latex beads (PU) and sea-sediment. It was found that there is an optimum range in the sample-loading for a FFD-GSF separation. It was also found that there is a general tendency of FE decreasing as the concentration of the sample suspension increases.

Optimization of Fractionation Conditions for Natural Organic Matter in Water by DAX-8 Resin and its Application to Environmental Samples (DAX-8 레진의 수중 자연유기물의 분획조건 최적화 및 환경시료에의 적용)

  • Natural organic matter (NOM) is a heterogeneous mixture of organic matter with various polarities and molecular weights in an aquatic environment. This study investigated the effects of separation conditions (resin volume, organic matter, etc.) and the repeated use of the resin for the fractionation of organic components in the DAX resin fractionation method. The distribution characteristics of the organic components ((hydrophilic [Hi], hydrophobic acid [HoA], and hydrophobic neutral [HoN]) under the derived fractionation conditions were also analyzed. Constant fractionation results (i.e. HoA/Hi ratio) were obtained in the column capacity factor (i.e. the packed resin volume) in the range of 50 to 100. The resin-packed column maintained constant separation efficiency for up to two repeated uses. The above conditions were applied to wastewater and stream water samples (before and after rainfall). The results showed that the concentration of organic matter in the wastewater effluent was 2-15 times lower with an increased ratio of hydrophilicity to hydrophobicity (i.e. Ho/Hi) compared to the influent depending on the industrial wastewater classification. Particularly, HoN was found to have a high content distribution, 10.2-50.4% of the total dissolved organic matter (DOM), in the effluents. For the stream water, the content of Hi or HoN increased significantly after rainfall, suggesting a correlation with the distribution characteristics of pollutants from the stream watershed. The results provide useful data to enhance the reliability of the DAX resin fractionation and its application to environmental samples.
