Studies on the Productive Structure and the Productivity of × Populus albaglandulosa Plantation (은수원사시나무의 조림지(造林地)의 생산구조(生産構造)와 생산성(生産性))

  • The productive structure and the productivity of ${\times}$Populus albaglandulosa plantations, where are middle parts of the Korean peninsula, were studied by means of stratifying clip technique and of allometry. The densities of stands in the sample areas were 693 trees/ha in 6 year-old plantation and 527 or 625 trees/ha in 9 year-old one. The photosynthetic part of the productive structure was not shown normal conical form but layering. So this was efficient to transmit solar radiation into the stand floor. The standing crop of the terrestrial parts of 6 year-old plantation was 18.11 ton/ha and that of 9 year-old one 38.8 to 47.3 ton/ha. The wood volume to trunk to 6 year-old was $31.3m^3/ha$ and that of 9 year-old was 68.8 to $83.9m^3/ha$. The annual net production of 6 year-old plantation was 4.8 ton/ha/year and that of 9 year-old one was 10.0 to 11.7 ton/ha/year and its wood volume of trunks was 17.9 to $21.1m^3/ha/year$. In the 9 year-old plantation the standing crop or the annual net production was different between two sample areas. This seemed that the cause was not due to chemical character but to physical character of soil.

Use of Zymography for Identification of the Same Clone in a Clone Bank of Pinus densiflora Sieb. et Zucc (Isoperoxidase 변이형(變異型)에 의(依)한 소나무 Clone 감별(鑑別))

  • Using of zymography for identification of same clone of Pinus densiflora, the two year old needle leaves of 48 ramtes including 8 clones (6 ramets per clone) were collected in the clone bank which has been established on the near campus of Institute of Forest Genetics, Suwon, in 1962. All 8 bands are named from cathod to anode, G, H, K, M, N, Q, S and Y. Only CB-1 clone shows all bands, while KW-3 clone reveals only 5 bands. Other 6 clones were found 7 bands but the occurred frequencies of those bands are variable among those clones. Though the grafting stock are used various individuals grown seed propagation of P. densiflora, moreover, three stocks have been different species and that one has been P. rigida and two individuals have been P. koraiensis, the zymograms of the ramets belonging to the same clone reveals the identified patterns. The results show that the stocks for grafting have not been affected on the isoperoxidase patterns of their scions in P. densiflora. Among 48 ramets of 8 clones, 4 ramets are found the different isoperoxidase patterns from that of the remained rametes within same clone. Thus, it is conluded that zymography is useful for testing genuineness of the grafted clones of P. densiflora.

Isolation and Culture of Protoplasts from Embryonic Cotyledons of Pinus densiflora S. et Z. (소나무 자엽(子葉)을 이용(利用)한 원형질체(原形質體) 나출(裸出) 및 배양(培養))

  • Protoplasts were isolated from cotyledons of germinating Pinus densiflora S. et Z. seeds. The seeds were germinated for nine days, and excised embryonic cotyledons about 3-4mm long were incubated with Cellulase Onozuka R-10 and Macerozyme. After 8 hours of incubation, large number of viable protoplasts were isolated. Isolated protoplasts were cultured in a medium containing basal salts of $B_5$ medium, vitamines, amino acids, organic acid, sugars, and growth hormones. The first evidence of protoplast budding was observed after twelve hours in culture, and it suggested that high potential of the embryonic cotyledons for rapid cell division affected the early budding, rather than effect of culture medium was shown in twelve hours. The three- to four-cell stage was reached after three to four days of culture. Most cell divisions were achieved by additional buddings rather than equal binary cell division. No further cell division was observed beyond the four-cell stage. Protoplasts isolated from fully expanded cotyledons (germinated for 17 to 24 days) seldom initiated or failed to initiate cell division.

Photosynthesis, Chlorophyll Contents and Leaf Characteristics of Ilex rotunda under Different Shading Treatments (비음처리에 따른 먼나무의 광합성, 엽록소 함량 및 엽 특성)

  • Two-year-old seedlings of Ilex rotunda were grown under control (full sunlight) and three different shading condition. Those conditions were full sunlight (PPFD 1600${\mu}mol\;m^{-2}\;s^{-1}$), 30% (PPFD 400${\mu}mol\;m^{-2}\;s^{-1}$), 50% (PPFD 250${\mu}mol\;m^{-2}\;s^{-1}$) and 70% treatment (PPFD 100${\mu}mol\;m^{-2}\;s^{-1}$). Total chlorophyll contents were inverse proportion to light intensity. Seedlings under full sunlight showed the highest photosynthetic activity such as photosynthetic rate, intercellular $CO_2$ concentration and water use efficiency. Photosynthetic activity trend was increased at the higher light intensity than the lower treatment over PPFD 500${\mu}mol\;m^{-2}\;s^{-1}$. Especially, seedlings under 70% treatment showed the worst photosynthetic activity at high light intensity. That result was regular for adapted plant in low intensity environment. Leaf area was also inverse proportion to light intensity, while dry weight per leaf area was shown the opposite trend.

  • Somatic embryo induction, plant regeneration, and field establishment were investigated from tissue cultured winter buds of a 10-year-old tree Aralia elata. Embryogenic calli were obtained from cultures of winter buds on MS medium supplemented with 2,4-D. A number of somatic embryos were regenerated from the calli on an embryo induction medium supplemented with 2,4-D and BA. Although abnormal somatic embryos were frequently observed, most of the embryos formed were morphologically normal. All somatic embryos at the later stage of maturity germinated successfully, but only 14% of them could be developed into plantlets on MS basal medium. The plants regenerated from the somatic embryos survived well in the field (survival rates : more than 95%) and have grown normally for three years after transplanting.

Berberine Production by Cell Suspension Cultures of Cork Tree (Phellodendron amurense Rupr)

  • Various culture conditions for cell growth and berberine production in cork tree (Phellodendron amurense Rupr.) were investigated. Callus was induced from cambium tissue of cork tree, and cultured on LS liquid medium supplemented with 0.5 mg/1 2,4-D, 0.1mg/1 BA, and 3% sucrose. Several factors enhancing berberine production and cell growth in cork tree cell cultures were found. Some of them enhanced both cell growth and berberine production, but others resulted in a decoupling of cell growth and berberine production with significant in the specific levels. High level of nitrate (80mM), high level of phosphate (8.98mM), and sucrose (7%), 1.0mg/l IAA were effective in berberine production, whereas low level of nitrate (40mM), and phosphate (2.25mM), and high level of sucrose (7%) in the medium were effective in cell growth. Two stage culture(first stage for cell growth, and second stage for berberine production) increased berberine production almost twice (5.06mg/g dry weight) as much as single stage cultures in berberine production.

  • Multivariate analysis methods were used to examine the relationships between top and bottom growth variables of hybrid poplars after coppicing and to discriminate between clones in coppice growth potential. Strong and linear relationship was exhibited between top and bottom growth variables. Clone 5328 was different from the other clones and the best among the clones in coppice growth potential.

First Report of Gymnosporangium globosum Causing American Hawthorn Rust in Korea

  • Field surveys and specimen collections of the rust fungal pathogen Gymnosporangium were carried out for 15 years from 1985 through 1999 in various locations of Korea. Macroscopic and microscopic examinations of morphological characteristics of aecia from the collected specimens revealed that Gymnosporangium globosum is the causal agent of American hawthorn rust disease on Crataegus pinnatifida and C. pinnatifida varmajor. The host plants are new for this rust fungus. G. globosum was found only in Gyeonggi and Chungbuk provinces, indicating that its distribution in Korea is limited. This is a first full description on morphological characters of aecia of G. globosum in Korea.

  • The hybrid poplar, populus alba ${\times}$ P.glandulosa $F_1$ shows a strong ability to produce root suckers. The relationship between crown density and root sucker was studied and the other factors that may be related to production of root sucker were also discussed. The possibility was also discussed to use its root suckers for fuelwood forests and vegetation in the area that erosion control is needed.
