Since its introduction in the forensic field, quantitative PCR (qPCR) has played an essential role in DNA analysis. Quality of DNA should be evaluated before short tandem repeat (STR) profiling to obtain reliable results and reduce unnecessary costs. To this end, various human DNA quantification kits have been developed. Among these kits, the PowerQunat® System was designed not only to determine the total amount of human DNA and human male DNA from a forensic evidence item, but also to offer data about degradation of DNA samples. However, a crucial limitation of the PowerQunat® System is its high cost. Therefore, to minimize the cost of DNA quantification, we evaluated kit performance using a reduced volume of reagents (1/2-volume) using DNA samples of varying types and concentrations. Our results demonstrated that the low-volume method has almost comparable performance to the manufacturer's method for human DNA quantification, human male DNA quantification, and DNA degradation index. Furthermore, using a reduced volume of regents, it is possible to run 2 times more reactions per kit. We expect the proposed low-volume method to cut costs in half for laboratories dealing with large numbers of DNA samples.

Purpose: This study examined the patterns of drugs, poisons, and chemicals detected in autopsy samples performed in the Seoul Institute and other regional forensic offices of the National Forensic Service (NFS) between 2014 and 2016. Methods: The investigation carried out using the laboratory information management system. Forensic toxicological identification and quantitation were performed in autopsy samples, including heart blood, peripheral blood, liver, kidney, vitreous humor and etc. Gas chromatography/mass spectrometry (GC-MS) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) were used to analyze the drugs and poisons. Results: Forensic autopsies were performed on 9,674 cases in this period. Based on the autopsy reports, 699 cases (7.2%) were considered as unnatural deaths caused by fatal intoxication. The number of male deaths was higher than that of female deaths, with the age of 50-59 being the most common age group. Conclusion: Drugs comprised the largest number of deaths due to poison, followed by alcohol, agrochemicals, drug with alcohol, carbon monoxide, and cyanide, in that order. Zolpidem was the most frequently used drug in all drug-related intoxication cases.

A variety of Y-STR analysis kits have been developed and used in the forensic field. Prior to the forensic application of a new kit, laboratory validation and sensitivity tests are essential processes in selecting suitable alternatives and for assuring that standard operating procedures are followed. In this paper, we have performed a sensitivity study of a new commercial kit, the Yfiler® PLUS PCR Amplification Kit (Yfiler plus kit, released in 2014) by comparing it with the AmpF/STR® YfilerTM PCR Amplification Kit (Yfiler kit, released in 2004). The Yfiler plus kit includes the 17 Y-STR loci of the Yfiler kit and has been supplemented with 10 new Y-STR loci. First, we analyzed the sensitivity difference between the two kits using commercial control DNA 2800M and 007. In addition, we compared the detection rate between the two kits from the 16 selected forensic casework samples of less than 0.5 ng concentrations. The results show that the sensitivity and detection rate of the Yfiler plus kit are higher than the corresponding rates of the Yfiler kit. In addition, we were able to obtain more Y-STR profiles with the use of the new kit. Thus, we suggest that Yfiler plus kit is a more effective forensic tool to detect Y-STR profiles from forensic casework samples of low concentrations.

Ethyl glucuronide (EtG) and ethyl sulfate (EtS), which are ethanol metabolites, are direct indicators of ethanol intake; they have been studied in a variety of biological samples in forensic science. It is necessary to analyze ethanol metabolites to determine whether the ethanol detected in autopsy cases was due to alcohol consumption before death or due to the ethanol produced from post-mortem decay. In general, EtG and EtS are detected in the blood together with ethanol; however, it may be difficult to secure blood depending on the extent of decay. Therefore, the aforementioned method should be replaced by detecting the ethanol metabolites using tissue biological samples. In this study, we determined the optimal experimental conditions for detecting EtG and EtS from spleen samples using Liquid Chromatography - Tandem Mass Spectrometry (LC-MS/MS). Herein, the test method was validated, and an analysis method was applied to the actual autopsy cases.

Analysis techniques using DNA profiling are widely used in various fields including forensic science and new technologies such as the Direct PCR amplification method are being developed continuously in order to acquire the DNA profiles efficiently. However, it has a limits such as non-specific amplification according to the quality of crime scene evidence samples. Especially, split peaks caused by excessive DNA samples are one of the important factors that could cause the debate to allow researchers to interpret the DNA profile results. In this study, we confirmed the occurrence rate of split peaks in each STR (short tandem repeats) locus of the $GlobalFiler^{TM}$ kit and investigated the possibility of improving the split peaks using several PCR additives such as DMSO (dimethylsulfoxide), $MgCl_2$, Betaine and Tween-20. As a result, we could make three groups according to the occurrence rate of split peaks in Direct PCR and it was confirmed that the ratio of split peaks could be reduced by DMSO (87.4 %), $MgCl_2$ (84.5 %) and Betaine (86.1 %), respectively. These results indicate that PCR additives such as DMSO, $MgCl_2$ and Betaine can be improve the split peaks in Direct PCR and thereby facilitate subsequently a successful DNA profile results.

Blood is a commonly found body fluid at crime scenes, and plays an important role in identifying suspects and in the reconstruction of crime scenes. Although serological detection of blood has been widely used in the field of forensic science, research on the detection of old bloodstains is scarce. This work aimed to compare various methods for the detection of old bloodstains and validate the reliability of their results. Four presumptive tests-Tetramethylbenzidine, $Bluestar^{(R)}$, Leucomalachite Green, Kastle-Meyer tests-and two confirmatory tests-Fecal Occult Blood (FOB) and Rapid Stain $Identification^{(TM)}-Blood$ ($RSID^{TM}-Blood$) tests-were compared. Bloodstain samples from post-mortem cases were collected on gauzes and then stored at room temperature for periods from 7 to 30 years. All the presumptive tests were positive, even for the 30-year-old sample. However, FOB and $RSID^{TM}-Blood$ provided false negative results for some samples stored for 17 years or more (1988 to 2001). The results indicate that FOB and $RSID^{TM}-Blood$ are not reliable for the detection of old bloodstains. These findings can be useful in the selection of an appropriate detection method for serological testing of old bloodstains. In addition, the information will be useful background knowledge when applied in the field of forensic practice.

Fetal DNA (fDNA) detection in maternal serum is a challenge due to low copy number and the smaller size of fDNA fragments compared to DNA fragments derived from the mother. Massively parallel sequencing (MPS) is a useful technique for fetal genetic analysis that is able to detect and quantify small amounts of DNA. In this study, seven clinical samples of maternal serum potentially containing fDNA were analyzed with a commercial single nucleotide polymorphism (SNP) panel, the HID-Ion $AmpliSeq^{TM}$ Identity Panel, and the results were compared to those from previous studies. Reference profiles for mothers and fetuses were not available, but multiple Y chromosomal SNPs were detected in two samples, indicating that fDNA was present in the serum and thereby validating observations of autosomal SNPs. This suggests that SNP-based MPS can be valuable for fDNA detection, thereby offering an insight into fetal genetic status. This technology could also be used to detect small amounts of DNA in mixed DNA samples for forensic applications.
Paint, fiber and dye which play a critcal role in proving the relationship between a suspect and a victim or a crime scene, are one of the most frequently encountered trace evidences at a forensic laboratory, however, in usual, because of infinitesimally small sizes of forensic samples, investigation of the spectroscopic characteristics of such samples is becomming more and more prevalent in forensic science as a non-destructive method. In this study, transmittance/reflectance profiles at ultraviolet-visible region (240-780nm), were investigated by UV/visible microspectro- photometer and used to analyze the spectral characteristics of different types of 14 microfibers, 12 inks of four colors and 44 automotive paints of two colors. Good results for discrimination were given from spectra of these samples due to the characteristic bands in uv/vis region, respectively.
This paper includes a review of 555 drug-facilitated sexual assault (DFSA) cases analyzed at the National Forensic Service (NFS), South Korea, between 2006 and 2012. The results of toxicological analyses of blood and urine samples were also reported, and furthermore the results were interpreted with respect to the number of drugs detected. The number of DFSA cases was highest during warmer summer months and the mean age of the victims was 25 years, with 48% being between 20 and 29 years. Accommodations or entertainment places were the most frequent place of the sexual assault (57%); and the assailant was a stranger in 72% of the DFSA cases. Drugs were identified in the blood or urine samples in 145 cases (26%) and sedative-hypnotics, such as benzodiazepines and zolpidem, were the most commonly detected, along with sedative antihistamines such as doxylamine and diphenhydramine. The frequent presence of sedative drugs in biological samples tends to implicate their use in chemical submission. However, interpreting the analytical results in terms of voluntary vs. surreptitious administration of drugs requires further detailed investigation and knowledge of the victim's health status and medication used at the time of event.

The successful identification of hit-andrun a and traffic accidental vehicles from evidential paint fiagments is greatly facilitated with a comprehensive laboratory collection of reference paint samples and the technique for direct analysis without sample preparation. The Pyrolysis-Gas Chromatography(PGC) is a precise and reliable method for performing both quantitative and qualitative analysis of polymeric materials and forensic samples. Our Forensic Laboratory is conducting the examination and identification of 73 reference paint samples; 4 colors of each domestic automotive make that is popular in Korea, by Curie Point Pyrolyzer(JHP-3) and GC with capillary column(ultra alloy-5). This method can be used not only to compare paint traces with their suspected sources, but also to identify the type, make and model of the automotive car.
