Anti-inflammatory Activity of Ethanol Extract of Undaria pinnatifida Root in RAW 264.7 Cells (RAW 264.7 세포에서의 미역(Undaria pinnatifida) 뿌리 에탄올 추출물의 항염증활성)

  • The anti-inflammatory effects of the ethanol extract of Undaria pinnatifida root (UPREE) were investigated using the lipopolysaccharide (LPS)-induced inflammatory response in RAW 264.7 cells by measuring the production of nitric oxide (NO), interleukin (IL)-6, tumor necrosis factor (TNF)-${\alpha}$, and IL-$1{\beta}$, and cell proliferation. We found that NO levels were reduced by 34% at $100{\mu}g/mL$. Moreover, the production of IL-6 and TNF-${\alpha}$ was suppressed by the UPREE treatment. In particular, the IL-6 production was inhibited by more than 30% at $100{\mu}g/mL$ UPREE. The proliferation of RAW 264.7 cells was measured by MTT assay, and we found no cytotoxicity in those cells treated with UPREE compared to the control. Our results suggest that UPREE shows promise as a therapeutic anti-inflammatory treatment.

Succinyl Daidzin and Succinyl Genistin are New Isoflavone Derivatives Found in Cheonggukjang

  • Two new fermentation products were separated from cheonggukjang by high performance liquid chromatography (HPLC) and tentatively identified as succinyl daidzin and succinyl genistin by HPLC-mass spectrometry (MS). Both succinyl daidzin and succinyl genistin were detected in all 4 commercially available and one laboratory prepared cheonggukjang and 1 commercially available natto. However, these compounds were not detected in commercially available 4 home-made and 4 factory-produced doenjang. Peak areas of succinyl genistin were about 1.95-2.45 times higher than those of succinyl daidzin in cheonggukjang, which may be due to the higher concentration of genistin derivatives than daidzin derivatives in soybeans. This is the first report on the succinyl derivatives as isoflavone metabolites from cheonggukjang and these 2 isoflavone derivatives could be characteristic indicators for cheonggukjang.

  • Hot water extracts of Angelica keiskei aerial parts were solvent-fractionated with ethyl acetate (EtOAc) and buffers (5% $NaHCO_3$, pH 8.0; 1.0 N HCl, pH 3.0) to obtain EtOAc-soluble acidic and neutral fractions. EtOAc-soluble neutral fraction was purified by Sephadex LH-20 column chromatography and reverse phase HPLC. Assay for purification of antioxidative compounds was performed by spraying DPPH solution on thin layer chromatography. Two isolated substances were identified as pseudoisopsoralen and 8-methoxypsoralen(xanthotoxin) by FAB-MS and NMR analyses.

Anti-Atopic Activities of Sargassum horneri Hot Water Extracts in 2,4-Dinitrochlorobezene-Induced Mouse Models

  • Atopic dermatitis (AD) is a chronic inflammation associated with skin hypersensitivity caused by environmental factors. The objent of this study was to assess the hot water extracts of Sargassum horneri (SHHWE) on AD. AD was induced by spreading 2,4-dinitrochlorobenzene (DNCB) on the BALB/c mice. The efficacy of SHHWE was tested by observing the immunoglobulin E (IgE), cytokine, skin clinical severity score and cytokine secretions in concanavalin A (Con A)-stimulated splenocytes. The levels of interleukine (IL)-4, IL-5 and IgE, the pro-inflammatory cytokines that are closely related, were notably suppressed in a does-dependent manner by SHHWE, whereas the level of interferon γ (IFN-γ), the atopy-related Th1 cytokine inhibiting the production of Th2 cytokines, was increased. Therefore, these results show that SHHWE has a potent anti- inhibitory effect on AD and is highly valuable for cosmetic development.
