The pathogenic Vibrio genus contains halophilic bacteria that are distributed in marine and freshwater environments. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are potent human pathogens and leading causes of septicemia, wound infection, and seafood-borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrio species in seawater off the west coast of Korea. Sixty-four seawater samples were obtained from different sites in Gomso Bay and Byeonsan from April 2018 to November 2018. Pathogenic Vibrio species were detected using a combination of most probable number (MPN)-polymerase chain reaction methods. V. cholerae, V. vulnificus, and V. parahaemolyticus were found in 0.0%, 20.3%, and 65.6% of seawater samples, respectively. Quantitative results revealed 3.6-23 MPN/100 mL of V. vulnificus, and 3.6-930 MPN/100 mL of V. parahaemolyticus in the samples. Overall, these results provide new insight into the necessity for seawater sanitation in Gomso Bay and Byunsan; they also provide evidence that will help reduce outbreaks of seafood-borne illness caused by pathogenic Vibrio species.
The pathogenic Vibrio genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. V. cholerae, V. vulnificus, and V. parahaemolyticus are the main species that can be potent human pathogens and the leading cause of septicemia, wound infections, and seafood borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrios in seawater. We obtained a total of 80 seawater samples from the Geum River estuary area in the west coast of Korea from April to December 2021. Pathogenic Vibrios was determined using a combination of the most probable number-polymerase chain reaction (MPN-PCR) methods. The detection levels of V. cholerae, V. parahaemolyticus, and V. vulnificus in the seawater samples were 7.5%, 68.8%, and 30.0%, respectively. The quantitative results were as follows: 3.6-3.6 MPN/100 mL in V. cholerae, 3.6-3,400 MPN/100 mL in V. parahaemolyticus, and 3.6-4,300 MPN/100 mL in V. vulnificus. Overall, these results provide novel insight into the necessity for seawater sanitation in the Geum River estuary area, and could help reduce the risk of seafood-borne outbreaks caused by pathogenic Vibrios.
The current study was conducted to evaluate the antibacterial and antioxidant activities of the essential oil fraction from the roots of Angelica tenuissima Nakai and its main components. We extracted the essential oil fraction from the roots of A. tenuissima using steam distillation and isolated its main components. Their antibacterial activities were determined by broth dilution test against food-borne pathogenic bacteria. Antioxidant activities were evaluated by DPPH-scavenging assay and reducing-power test. Also tested was their ability to inhibit the growth of two gastrointestinal cancer cell lines, Caco-2 and MKN-45. The A. tenuissima oil fraction and its main components, ligustilide and butylidene phthalide exhibited marked inhibitory effects against most of the tested antibiotic-susceptible and antibiotic-resistant bacterial strains with minimum inhibiting concentrations (MICs) from $0.21{\pm}0.08$ to $3.60{\pm}0.89mg/ml$. They also showed growth-inhibiting activity against Caco-2 and MKN-45 cells. The oil fraction showed significant antioxidant activities in DPPH radical scavenging assay and reducing-power test. Taken together, A. tenuissima essential oil could be used as a safe additive for preventing food contamination by pathogenic bacteria. Additionally, its antioxidative activity and the ability to inhibit gastrointestinal carcinoma cell lines could increase its value for functional foods and prevention of cancer.
식용이 간편한 즉석섭취용(ready-to-eat)샐러드 제품의 수요는 나날이 증가하고 있으나 국내 유통 샐러드의 미생물 오염수준은 기준 및 규격이 설정되어있지 않고 그 수준 또한 알려져 있지 않다. 따라서 소비가 집중되어 있는 대도시 지역을 중심으로 백화점, 대형할인점 또는 패밀리레스토랑 등에서 판매되는 즉석섭취용 샐러드 제품 120건을 대상으로 하여 coliform bacteria, E. coli, E. coli O157:H7, Staphylococcus aureus, Salmonella spp. Listeria monowtogenes 및 Bacillus cereus의 분리시험을 실시하고 일부 제품을 대상으로 일반세균수 및 세척효과를 조사하였다. 제품 120건 중 총 73건에서 coliform bacteria 등이 검출되어 60.8%의 검출율을 보였다. 5. coli가 17건, Staphylococcu aureu가 3건, Salmonella spp.가 1건에서 각각 검출되었고 기타 coliform bacteria는 59건에서 검출되었다. 샐러드는 양식방법에 따라 유기농샐러드와 비유기농샐러드로 나누었고, 종류는 야채로 구성된 야채샐러드와 튀긴 닭 등이 첨가된 혼합샐러드로 나누었으며 판매형태에 따라 완포장제품과 샐러드바 제품으로 나누었다. 양식방법 및 포장방법과 미생물오염과의 통계적 연관성은 없는 것으로 나타났다. 그러나 분리된 균주 수에 있어서는 야채샐러드와 혼합샐러드 간에 통계적으로 유의한 차를 보여 야채샐러드의 경우 좀 더 다양한 균종이 분리되는 것으로 나타났다 (p<0.005). E. coli는 일반 재배 야채샐러드의 경우 90건 중 10건에서 분리되었고 유기농재배 야채샐러드 의 경우 30건 중 7건에서 분리되었다. 식중독 원인균은 비유기농 재배 야채샐러드 4건에서 Staphylococcus aureu가, 1건에서 Salmonella arixonia가 검출되었으나 유기농재배 야채에서는 검출되지 않았다. 널리 판매되는 일부 제품 4종을 5회에 걸쳐 검사한 결과 총 호기성 세균수는 평균 4.8$\pm$0.19 log$_{10}$ cfu/g으로 조사되었다. 같은 제품을 생리식염수와 야채용세척제를 이용해 3분간 침지 후 세척효과를 본 결과 생리식염수는 5회, 야채용세척제는 1회 세척 시 95.5%의 세척효과를 내는 것으로 나타났다.다.
본 연구에서는 높은 분리능을 가지고 있을 뿐만 아니라 실험의 재현성과 경제성의 측면에서도 많은 장점을 갖고 있는 ERIC DNA sequence를 응용한 ERIC-PCR을 이용하여 Salmonella, E. coli, Shigella, Vibrio 등 4속의 주요 그람 음성 식중독유발 세균들의 분리 동정 방법을 확립하고자 하였다. ERIC-PCR 결과, E. coli의 경우 0.3kb, 0.42kb 및 1.2kb의 band가 모든 균주에서 공통적으로 확인되었고, Salmonella속으로부터는 0.22kb, 0.4kb 및 0.7kb의 band가 증폭되었다. Shigella속은 모든 표준균주와 분리균주로부터 0.33kb와 1.25kb의 band가 증폭되었으며, S. sonnei의 경우 위의 주요 2개 band 이외에도 대부분의 균주에서 0.44kb, 2.0kb 및 3.05kb의 band가 증폭되어 다른 종의 Shigella와 구별되는 fingerprinting pattern을 나타내었다. 그리고 V. parahaemolyticus의 경우 표준균주와 분리균주 모두 0.51kb와 1.5kb의 band가 증폭되어 V. cholerae, V. mimicus 등과 같은 다른 종의 Vibrio와 구별되는 fingerprinting pattern을 나타내었다. 이와 같이 4속의 모든 식중독 균주마다 ERIC-PCR후 생성되는 fingerprinting pattern에서 3-5개의 공통적인 band가 증폭되는 것이 확인되어 이를 이용한 속 수준의 분리 동정과 이러한 주요 band들 이외의 부수적인 band들을 고려하여 종 수준까지의 분리도 가능함을 확인하였다. 따라서 본 연구의 결과는 ERIC 반복적 DNA 염기서열을 이용한 ERIC-PCR이 식중독균의 분리 동정 방법으로 사용될 수 있음을 확인하였으며, 나아가 더 많은 속(genus)의 식중독세균을 대상으로 한 새로운 분리 동정 방법을 확립하는데도 응용이 될 수 있을 것이다.
연구는 대기압 유전체장벽방전 플라즈마 처리에 따른 식품유해 미생물 사멸효과를 조사하기 위해 수행되었다. 플라즈마 처리 시, 활성종 생성 및 농도에 영향을 미치는 노출시간, 노출거리, 산소비율, 전력 변화에 따른 E. coli의 사멸효과를 조사한 결과, E. coli의 사멸율은 플라즈마 처리를 위한 노출시간, 산소비율, 전력의 증가에 따라 증가한 반면, 노출거리의 증가에 따라서는 사멸율이 감소하였다. 이 결과는 미생물 시료가 플라즈마에 노출되는 시간이 증가됨으로서 시료 내 NO 농도가 증가되고, E. coli의 사멸율 역시 증가되는 결과로 뒷받침할 수 있고, 미생물 사멸효과를 높이기 위해서는 활성종의 농도가 증가되어야 함을 의미한다. E. coli와 함께 B. cereus, B. subtilis, B. thuringiensis, B. atrophaeus를 대상으로 대기압 유전체 장벽방전 플라즈마에 의한 살균효과를 조사한 결과, 72.3~91.3%의 높은 사멸율을 나타내었다. 이러한 결과로 미루어, 대기압 유전체장벽방전 플라즈마기술은 다양한 미생물에 적용될 수 있는 유용한 살균기술임을 확인하였다.
Antimicrobial coating on chicken carcasses may reduce the effects of cross-contamination and improve product shelf-life and safety. Thyme oil was mixed at 0.5%(v/v) with a pre-gelatinized pea starch coating solution. The coating solution was spread on chicken breast meat after inoculation with selected spoilage and pathogenic bacteria. After inoculation, the chicken meats were packaged in plastic bags and stored at $4^{\circ}C$. During 12 day storage, total aerobic bacteria, lactic acid bacteria, and inoculated organisms were counted at 4 day intervals. Thyme oil treatments reduced the viability of Salmonella as well as the growth of Listeria and Pseudomonas by 2 log CFU/g, and appeared to eliminate inoculated Campylobacter during storage. The addition of thyme oil increased the viscosity of the pre-gelatinized pea starch solution. The results suggested that thyme oil inclusion in an edible starch coating may be a satisfactory delivery system to enhance the safety of processed fresh meat.

Titanium dioxide ($TiO_2$) shows antibacterial effects when exposed to near ultra violet (UV) light. In this study, $TiO_2$ photocatalytic continuous reactor was designed and applied to food-borne pathogens such as Vibrio parahaemolyticus ATCC 17802, Salmonella choleraesuis ATCC 14028, and Listeria monocytogenes ATCC 15313. $TiO_2$ films were prepared by conventional sol-gel dip-coating method using titanium tetra iso-propoxide (TTIP). The antibacterial activity of photocatalytic reactor with various flow rates and UV-A illumination time showed effective bactericidal activity. As the UV-A illumination time increased, survival rates of those bacteria decreased. After 60 min of UV-A illumination, the survival rates of V. parahaemolyticus and S. choleraesuis were less than 0.1%. However, that of L. monocytogenes was about 5% at that time point. These results present an effective way to exclude pathogenic bacteria from aqueous foods.
The aim of this study was to determine the production of folate, short chain fatty acids (SCFAs), and antimicrobial activity exhibited by Lactobacillus plantarum G72 for potential dietary application in pregnant women. L. plantarum G72 has been reported to possess characteristic activities and functionality including β-galactosidase activity and antioxidant activities. L. plantarum G72 showed antibacterial activity against pathogenic bacteria (Listeria monocytogenes ATCC 15313, Salmonella typhimurium P99, Escherichia coli ATCC 25922, and Staphylococcus aureus KCCM 11335) using a modified method, and formation of the largest inhibition zone was observed against S. aureus KCCM 11335 (12.0-17.0 mm). The adherence of four food-borne pathogenic bacteria to HT-29 cells was inhibited by L. plantarum G72 (0.13 to 0.92 log CFU/ml). The most considerable inhibition of adherence to HT-29 cells was observed by using L. plantarum G72 against S. typhimurim P99. Additionally, folate production by L. plantarum G72 was 50.1 ng/ml, and L. plantarum G72 produced relatively more lactic acid (11,176.73 mg/kg) than acetic, propionic, or butyric acids. Therefore, the results of this study suggest that L. plantarum G72 may serve as a multifunctional food additive in the health industry.
BACKGROUND: Contaminated water was a major source of food-borne pathogens in various recent fresh produce-related outbreaks. This study was conducted to investigate the microbial contamination level and correlations between the level of sanitary indicator bacteria and the detection ratio of pathogens in agricultural water by logistic regression analysis. METHODS AND RESULTS: Agricultural water was collected from 457 sites including surface water (n=300 sites) and groundwater (n=157 sites) in South Korea from 2018 to 2020. Sanitary indicator bacteria (total coliform, fecal coliform, and Escherichia coli) and food-borne pathogens (pathogenic E. coli, E. coli O157:H7, Salmonella spp., and Listeria monocytogenes) were analyzed. In surface water, the coliform, fecal coliform, and E. coli were 3.27±0.89 log CFU/100 mL, 1.90±1.19 log CFU/100 mL, and 1.39±1.26 log CFU/100 mL, respectively. For groundwater, three kinds of sanitary indicators ranged in the level from 0.09 - 0.57 log CFU/100 mL. Pathogenic E. coli, Salmonella and Listeria monocytogenes were detected from 3%-site, 1.5%- site, and 0.6%-site water samples, respectively. According to the results of correlations between the level of sanitary indicator bacteria and the detection ratio of pathogens by logistic regression analysis, the probability of pathogen detection increased individually by 1.45 and 1.34 times as each total coliform and E. coli concentration increased by 1 log CFU/100mL. The accuracy of the model was 70.4%, and sensitivity and specificity were 81.5% and 51.7%, respectively. CONCLUSION(S): The results indicate the need to manage the microbial risk of agricultural water to enhance the safety of fresh produce. In addition, logistic regression analysis is useful to analyze the correlation between the level of sanitary indicator bacteria and the detection ratio of pathogens in agricultural water.
