Identification of protease-resistant proteins from allergenic nuts using two-dimensional gel electrophoresis and mass spectrometry

  • Nuts are one of the most common sources of allergies in individuals of all ages. In order for a particular protein to render an allergic reaction, it must resist proteolytic digestion by intestinal enzymes. In this study, three well-known allergenic nuts, almonds, cashew nuts, and peanuts, were used as samples, and enzyme digestion with Bacillus protease and porcine pepsin was tested. A proteomic approach using two-dimensional gel electrophoresis and an MS/MS analysis was applied to visualize and identify the proteins that were resistant to enzyme digestion. Among the 150 protein spots tested, 42 proteins were assigned functions. Due to the lack of genomic databases, 41% of the identified proteins were grouped as hypothetical. However, 12% of them were well-known allergens, including AraH. The remainder were grouped as storage, enzymes, and binding proteins.

Research on the Allergic Potential of Insecticidal CrylAc Proteins of Genetically Modified Rice

  • In Korea, different kinds of genetically modified (GM) crops are under development, including GM-rice expressing insecticidal crystal (Cry) proteins of Bacillus thuringiensis (Bt) modified to change a single amino acid. In this study, amino acid (aa) sequences of modified Cry proteins were compared to that of known allergens, and Cry proteins expressed in GM-rice were identified by using Cry protein specific polyclonal antibody. The antigen-antibody reactions were compared between GM and commercial rice to assess the allergic risk of Cry proteins. This analysis showed no known allergen to have more than 35% aa sequence homology with modified Cry proteins in Bt rice over an 80 aa window or to have more than 8 consecutive identical aa. Sera from allergic patients showed some IgE reactivity via immunoblotting and enzyme-linked immunosorbent assay (ELISA), although no differences were seen between GM and commercial rice. Based on these results we conclude that GM rice with modified Cry proteins has no differences in its protein composition or allergenicity relative to commercial rice.

청소년과 성인의 식품 알레르기 예방을 위한 식품 알레르기 발생 실태 및 개선안 연구 (Study on Incidence and Improvement of Food Allergies for Prevention of Damage in Adolescents and Adults)

  • 본 연구는 식품 알레르기의 발생 실태를 조사하여 식품 알레르기로 인한 소비자의 피해를 예방하고, 국민의 식생활을 안정시키기 위한 기초 자료를 구축하기 위해서 10대 이상의 청소년과 성인 남녀를 대상으로 식품 알레르기 발생 실태와 식품 알레르기로 인한 소비자 피해를 예방하기 위한 개선안에 대해 알아보았다. 첫째, 조사 대상자의 일반적 특성으로 성별은 남자가 32.9%, 여자는 67.1%인 것으로 나타났고, 연령은 40~50세 미만이 31.0%로 가장 높게 나타났다. 학력을 보면 대학교 재학 졸업이 56.4%로 가장 높았고, 직업은 사무관리직과 주부가 21.6%로 가장 높게 나타났다. 월 평균 소득에서는 100~200만 원 미만과 300~400만 원 미만이 18.8%로 가장 높게 나타났고, 월 평균 식비에서는 30~50만 원 미만이 23.5%로 가장 높게 나타났다. 둘째, 식품 알레르기 발생 실태에 대해서 조사한 결과는 다음과 같다. 전체의 41.1%가 식품으로 인한 알레르기를 한번이라도 경험한 것으로 조사되었다. 식품 알레르기 발생 시 증상 부위로는 피부가 80.3%로 가장 높았는데, 연령에 따라서는 피부에서 13세 이상 20세 미만이 88.9%로 가장 높게 나타났고, 호흡기는 40~50세가 15.9%로 가장 높게 나타났으며, 소화기(20.0%)와 전신(6.7%)은 50세 이상에서 가장 높게 나타났다(p<0.05). 식품 알레르기 발생 횟수로는 연 2~3회가 42.7%로 가장 높게 나타났다. 식품 알레르기 발생 시 대처 방법으로 병원치료를 한다는 응답이 전체의 53.4%로 가장 높게 나타났고, 다음으로 약물치료 20.6%, 별다른 대처하지 않음 16.0%, 기타 6.1%, 알레르기 원인 식품 제한 3.8% 순으로 나타났다. 50세 이상을 제외하고 모든 연령에서 식품 알레르기 발생 시 병원치료를 한다고 응답한 비율이 가장 높게 나타나서, 연령에 따라서 식품 알레르기 발생 시 대처하는 방법이 다른 것으로 나타났다(p<0.05). 식품 알레르기의 유발 식품으로는 총 44종의 식품 중에 인스턴트 식품이 가장 많은 것으로 나타났으며, 패스트푸드, 계란, 생선, 우유, 돼지고기, 새우, 갑각류, 견과류 등에 의한 알레르기 발생 비율이 높고, 과일에 의한 알레르기는 복숭아를 제외하고는 적은 것으로 나타났다. 식품 알레르기로 인한 소비자의 피해를 예방하기 위해서 가장 중요하게 생각되는 항목에 대해서는 알레르기 식품 표시제의 개선이 가장 높게 나타났고, 다음으로 소비자의 관심과 노력, 식품 제조업자의 교육과 관리, 알레르기 식품 표시제의 홍보, 정부의 관리 감독 강화, 전문가에 의한 진단과 관리, 소비자 대상 예방교육 순으로 나타났다. 이상의 연구 결과, 식품 알레르기를 경험한 사람이 비교적 많은 것에 비해서 식품 알레르기로 인한 증상이 발생되었을 때에 적절한 진단과 체계적인 치료를 받지 못하는 것으로 조사되었고, 식품 첨가물의 섭취 증가로 인해 인스턴트 식품과 패스트푸드에 의한 알레르기 발생 비율도 높은 것으로 나타났다. 또한 식품 알레르기로 인한 소비자 피해를 예방하기 위해서는 알레르기 식품 표시제의 개선이 필요한 것으로 나타났다. 유럽연합이나 미국 등지에서는 알레르기 식품 표시 대상 품목을 포괄적인 통칭명으로 규정하고 있으므로, 이들 국가와 FTA로 식품의 수입과 수출이 증대되는 상황에서 국제적인 교역증대를 위해서 뿐만이 아니라, 국내 수입식품의 관리를 위해서라도 13개 품목으로 한정되어 있는 알레르기 식품 표시를 포괄적인 통칭명으로 규정하도록 하고, 가공식품에 대해서만 표시를 의무화하던 기존 정책을 비포장식품에 대해서도 표시를 의무화하도록 알레르기 식품 표시제를 개선할 필요가 있다. 서구화된 식생활과 가공식품의 섭취 증가로 식품 알레르기가 점차 증가하는 추세이므로 알레르기 식품의 표시제의 개선과 적극적인 홍보, 그리고 대체식품 안내를 위한 소비자 교육 등도 반드시 필요할 것으로 사료된다. 본 연구는 전문가의 진료와 진단을 통하지 않고 소비자들의 판단으로만 설문 조사를 수행한 한계점을 가지고 있지만, 향후 국가적인 차원에서 식품 알레르기에 대해 정책과 관리가 동행된다면 식품 알레르기로 인한 위해를 미연에 방지하여 국민 보건을 증진할 수 있을 것이라고 생각한다. 최근에는 해외로 수출된 국내 식품들이 해당 국가에서 표시위반으로 리콜되는 사례도 빈번한 만큼, 식품 알레르기 표시제가 국제적인 기준에 적합하도록 개선이 요구된다. 본 연구 결과에서 보듯이 식품 알레르기 표시제의 전반적인 개선을 통해 식품 알레르기로 인한 소비자의 피해를 사전에 예방하고, 국민의 식생활 안정을 도모하는데 기여할 수 있는 종합적인 식품 안전 관리 시스템을 구축할 것으로 기대해 본다.

  • Allergy skin prick test and intradermal test represent one of the major tools in the diagnosis of IgE-mediated diseases like as atopic asthma, allergic rhinitis, atopic dermatitis, food and drug allergy, and insect bite when properly performed. Skin tests are of particular importance in fields such as allergen standardization, pharmacology, and epidemiology. Even if skin tests seem easy to perform, adequate and proper interpretation requires well-trained physicians who can recognize the numerous factors that may modify the results of skin tests.

Perception and practice regarding allergen labeling: focus on food-related employees

  • BACKGROUND/OBJECTIVES: Most consumers are able to recognize allergenic foods. However, the frequency of checking such foods is reportedly low, resulting in higher prevalence of food-related allergic reactions in Korea compared to other countries. Thus, this study was performed to investigate the overall perception of allergenic food labeling and its practice level in food manufacturing company employees. SUBJECTS/METHODS: The survey was administered to food safety employees and food development teams at food companies located in metropolitan areas. A total of 399 (93.8%) valid samples were used in the final analysis. Statistical analyses, including Frequency Analysis, t-test, Anova, PCA (Principal Component Analysis), and Pearson Correlation Analysis using SPSS ver. 21.0, were performed. RESULTS: The correct answer rate in the analysis of allergy-related knowledge level ranged from 15.0% to 89.7%. Analysis of differences in allergy-related perception by knowledge level showed significant differences in introduction of a food recall system, strengthening of relevant laws and regulations, content labeling, description of substitutional food, and differentiated package by age. CONCLUSIONS: It can be concluded that labeling of allergenic foods should be made easier and more convenient for checking by employees, developers, and consumers, and it is necessary to provide contents through the development of publicity, guidelines, or APP along with labeling.

우리나라 초등학생의 식품알레르기 현황과 영양(교)사의 식품알레르기 인식 변화에 대한 고찰, 1995~2015 (Changes of prevalence of food allergy in elementary school student and perception of it in school nutritionist in Korea, 1995~2015)

  • Purpose: The aim of this study is to summarize and report on the change of food allergy in elementary school students and perception and practices in school nutritionists in Korea from 1995 to 2015. Methods: The search strategy was "(food allergy AND elementary school AND Korea) AND (nutritionist OR perception OR practice)". The search was conducted via KISS, DBPIA, RISS, NDSL, PubMed, Scopus, and Google scholar and full text and abstracts on the topic of food allergy evaluating prevalence, allergen, symptom, perception and practices were included in this review. Results: Out of 1379 records found in the sources, 13 related studies were included in the final analysis. The results showed that the number of students who had experienced food allergy was increasing. The two frequent allergenic foods were eggs and milk. The perception and practices of food allergy in school nutritionists was gradually increased. Conclusion: Further objective evaluations are required to confirm the food allergy status and its management in school.

Regulation of IgE-Mediated Food Allergy by IL-9 Producing Mucosal Mast Cells and Type 2 Innate Lymphoid Cells

  • Due to the increasing prevalence and number of life-threatening cases, food allergy has emerged as a major health concern. The classic immune response seen during food allergy is allergen-specific IgE sensitization and hypersensitivity reactions to foods occur in the effector phase with often severe and deleterious outcomes. Recent research has advanced understanding of the immunological mechanisms occurring during the effector phase of allergic reactions to ingested food. Therefore, this review will not only cover the mucosal immune system of the gastrointestinal tract and the immunological mechanisms underlying IgE-mediated food allergy, but will also introduce cells recently identified to have a role in the hypersensitivity reaction to food allergens. These include IL-9 producing mucosal mast cells (MMC9s) and type 2 innate lymphoid cells (ILC2s). The involvement of these cell types in potentiating the type 2 immune response and developing the anaphylactic response to food allergens will be discussed. In addition, it has become apparent that there is a collaboration between these cells that contributes to an individual's susceptibility to IgE-mediated food allergy.

Genomic DNA Sequence of Mackerel Parvalbumin and a PCR Test for Rapid Detection of Allergenic Mackerel Ingredients in Food

  • Mackerel (Scomber japonicus) often causes severe allergic reactions in sensitive people. Food containing undeclared mackerel may pose a risk to such people. The major allergenic protein in fish such as mackerel, codfish, and Alaska pollack has been found to be parvalbumin. In this study, we developed a polymerase chain reaction (PCR) method to detect mackerel DNA using primers corresponding to the parvalbumin gene. We cloned and sequenced 1.5 kb of parvalbumin gene by PCR using mackerel genomic DNA as a template. Nucleotide sequence analysis of genomic parvalbumin gene, composed of 4 exons and 3 introns, allowed the selection of two pairs of oligonucleotide primers specific for mackerel. These primers successfully enabled PCR amplification of specific regions of genomic parvalbumin DNA from mackerel, but no amplification from 8 other fish samples, surimi, and 6 boiled fish pastes. The sensitivity of this method was sufficient to detect 5 ng of purified mackerel DNA mixed with 50 ng of surimi DNA. This rapid and specific method for the detection of allergenic mackerel would be beneficial in reducing food allergy caused by the ingestion of hidden allergen in processed food.

Influence of age at complementary food introduction on the development of asthma and atopic dermatitis in Korean children aged 1-3 years

  • Background: Complementary food in infancy is necessary for human growth, neurodevelopment, and health. However, the role of allergen consumption in early infancy and its effects on the development of food allergy or tolerance remain unclear. Purpose: To investigate the influence of age at the time of complementary food introduction on the development of asthma and atopic dermatitis in Korean children aged 1-3 years. Methods: We combined data from the Korea National Health and Nutrition Examination Survey collected from 2010 to 2014 and analyzed 1619 children aged 1-3 years who were included in the survey. Multivariate regression analysis was used to identify associations among type of feeding, age at the time of complementary food introduction, and doctor-diagnosed atopic dermatitis and asthma. Results: Age at the time of complementary food introduction was not significantly associated with doctor-diagnosed atopic dermatitis and asthma in children aged 1-3 years. In the univariate analysis, children with asthma showed higher water and sodium intake levels than nonasthmatic children. However, this relationship was not significant in the multivariate regression analysis. Conclusion: The present study revealed no statistically significant relationship between age at the time of complementary food introduction and the risk of atopic dermatitis and asthma in young Korean children. A national prospective study is needed to clarify the influence of age at the time of complementary food introduction on the development of allergic diseases.

A Duplex PCR Assay for Differentiating Native Common Buckwheat and Tartarian Buckwheat, and Its Application for the Rapid Detection of Buckwheat Ingredients in Food

  • One of the major allergenic proteins in common buckwheat (Fagopyrum elculentum) was found to be a BW10KD. In this work, allergenic BW10KD genomic DNAs from the native common buckwheat 'Pyeongchang' and Tartarian buckwheat 'Clfa47' were cloned by polymerase chain reaction (PCR), and their nucleotide sequences were determined. In addition, a novel PCR assay targeting the allergenic BW10KD gene was developed to detect and differentiate both buckwheat species in food. The nucleotide sequences of the BW10KD genomic DNA from 'Pyeongchang' and 'Clfa47' were 94% identical. Base differences in the nucleotide sequences of the BW10KD genes are probably useful as a molecular marker for species-specific identification. The 'Pyeongchang'-specific primer set 154PF/400PR and the 'Clfa47'-specific primer set 154DF/253DR generated 247 and 100 bp fragments in singleplex PCR, respectively. A duplex PCR assay with 2 species-specific primer sets simultaneously differentiated the 'Pyeongchang' and 'Clfa47' in a single reaction. The PCR assay also successfully allowed for the rapid detection of buckwheat ingredients in foods.
