A study on the behavior of CF, CF2 radicals in an inductively coupled plasma using Laser Induced Fluorescence (레이저 유도 형광법을 이용한 유도 결합 플라즈마내의 CF, CF2 라디칼의 거동에 관한 연구)

  • CF & $CF_2$ radicals in a $C_4F_8$ inductively coupled plasma were observed with laser induced fluorescence. 251.9nm UV laser was used for the $CF_2$ excitation and 265.3nm UV emitted light for the detection which has the maximum intensity among many induced fluorescence lights. In the case of CF radical detection, 232.9nm UV laser was used for the excitation and 247.6nm for the detection. $CF_2$ radical density increased toward substrate, while CF radical had its maximum at about 10nm away from the substrate. The atomic fluorine density which was studied by the actinometry increased as the position moves away from the substrate. This phenomena was thought to have a close relation with the polymer growth on the wafer. When the bias voltage increased, $CF_2$ , CF radicals decreased while the atomic fluorine increased tio some extent and then decreased, which was thought to be due to the change in the ionization and dissociation.

Autofluorescence Loss in Photobleaching for Human Dentin ex vivo

  • Two-photon fluorescence microscopy was performed on the enamel-dentin junction area of a human tooth using a femtosecond pulsed laser. We obtained a clear image contrast between the bright dentin and dark tubules with the autofluorescence generated from the endogenous fluorophores in dentin. The autofluorescence shows a broad spectrum due to complex cross links between dentinal collagens, which extend from blue to orange wavelengths (470-590 nm), but a gradual autofluorescence loss in photobleaching was observed for a long-term exposure under strong excitation. For increasing excitation power, we found that two-step decay becomes significant in the spectrally integrated autofluorescence.

Prediction of Trihalomethanes Formation Potential of Dissolved Organic Matter with Various Sources Using Differential Fluorescence 3D-Excitation-Emission Matrix (EEM) (차등 3차원 형광 여기-방출 매트릭스를 이용한 다양한 기원의 용존 유기물질 트리할로메탄 생성능 예측)

  • This study aimed to maximize the potential of fluorescence 3D excitation-emission matrix (EEM) for predicting the trihalomethane formation potential (THMFP) of DOM with various sources. Fluorescence spectroscopy is a useful tool for characterizing dissolved organic matter (DOM). In this study, differential spectroscopy was applied to EEM for the prediction of THMFP, in which the difference between the EEM before and after chlorination was taken into account to obtain the differential EEM (DEEM). For characterization of the original EEM or the DEEM, the maximum intensities of several different fluorescence regions in EEM, fluorescence EEM regional integration (FRI), and humification index (HIX) were calculated and used for the surrogates for THMFP prediction. After chlorination, the fluorescence intensity decreased by 77% to 93%. In leaf-derived and effluent DOM, there was a significant decrease in the protein-like peak, while a more pronounced decrease was observed in the humic-like peak of river DOM. In general, leaf-derived and effluent DOM exhibited a relatively lower THMFP than the river DOM. Our results were consistent with the high correlations between humic-like fluorescence and THMFP previously reported. In this study, HIX (r= 0.815, p<0.001), FRI region V (r=0.727, p<0.001), humic-like peak (r= 0.827, p<0.001) from DEEM presented very high correlations with THMF P. When the humic-like peak intensity was converted to a logarithmic scale, a higher correlation was obtained (r= 0.928, p<0.001). This finding suggests that the humic-like peak in DEEM can serve as a universal predictor for THM formation of DOM with various origins.

  • A polymeric photobase generator containing oxime-urethane groups is applied to a fluorescence micropatterning material. Polymer bearing oxime-urethane groups was prepared by copolymerization of methyl methacrylate with methacryloyloxyethyl benzophenoneoxime urethane (MBU). The reaction of amino groups in the irradiated copolymer film with dansyl chloride (Dns-Cl) was monitored by using UV absorption, IR absorption and fluorescence spectroscopy. The fluorescence spectrum of the Dns-Cl-treated irradiated copolymer film shows a strong fluorescence with a fluorescence maximum wavelength at 510 nm. A blue fluorescent micropattern with a line width of $2{\sim}3{\mu}m$ was obtained. Treatment of the irradiated copolymer film with Dns-Cl and rhodamine B mixture led to the formation of green, red, and orange-colored fluorescence micropatterns. Thus, various colored micropatterns on a single polymer film can be obtained by selective excitation of each dye molecules.

  • Primary fluorescence characteristics of ten standard solutions of organophosphates, sea water, and water from agricultural land were investigated by fluorescence contour. All the standard solutions of organophosphates has shown characteristics countours. Their emission maxima were shown between 296 nm and 437 nm. According to their numbers of emission maxima on the fluorescence contours, the organophosphates can be categorized in two different groups. Ateric and Diazinon are the first group with two emission maxima. DDVP with other seven standard organophosphates belong to the second group. The second group has two subgroups. One is characterized by the similar emission and excitation maxima, which are 310 nm and 280 nm, respectively. Those are DDVP, Hinosan, Kitazin, Locsion, Meta. The other sub-group shows quite different emission and excitation maxima from the first sub-group. They are Monopho, Thaconyl and Gropho and their emisson maxima were in far longer (437 nm) or shorter wavelength (296 nm). From agricutural samples, one of the investigated organophosphates was detetected by its characteristic retention time $(t_r=12min)$. HPIC-fluorescence data gave an additional parameter for differentiation between two organophosphates which has similar excitation and emission maxima.

  • Fluorescence excitation spectrum of the trans.trans-1,3,5,7-octatetraene(OT)-Xe van der Waals clusters formed in supersonic jet expansions has been obtained. The transition lines corresponding to the van der Waals cluters of OT with Xe are observed in the lower frequency side of the OT band origin. Based on the spectral shifts, fluorescence lifetimes, and concentration dependence of the peak intensities, most of the transition lines are assigned to the $OT-Xe_n$ (n = 1, 2, 3, 4) clusters. Long progressions of a van der Waals vibrational mode are observed for n = 1, 2, 3 and 4 clusters and assigned to rocking of the OT moiety with respect to the Xe atom with the help of ab initio quantum mechanical calculation.

  • It has been reported that at low temperature region, poloxamers existed as a monomer. Upon warming, an equilibrium between unimers and micelles was established, and finally micelle aggregates were formed at higher temperature. In this study, the fluorescence spectroscopy was used to study the micelle formation of the poloxamer 407 in aqueous solution. The excitation and emission spectra of pyrene, a fluorescence probe, were measured as a function of the concentration of poloxamer 407 and temperature. A blue shift in the emission spectrum and a red shift in the excitation spectrum were observed as pyrene transferred from an aqueous to a hydrophobic micellar environment. From the $I_1/I_3 and I_{339}/I_{333}$ results, critical micelle concentration (cmc) and critical micelle temperature (cmt) were determined. Also, from the fluorescence spectra of the probe molecules such as 8-anilino-1-naphthalene sulfonic acid and 1-pyrenecarboxaldehyde, the blue shift of the $\lambda_{max}$ was observed. These results suggest a decrease in the polarity of the microenvironment around probe because of micelle formation. The poloxamer 407 above cmc strongly complexed with hydrophobic fluorescent probes and the binding constant of complex increased with increasing the hydrophobicity of the probe.

Haematococcus pluvialis Cell-Mass Sensing Using Ultraviolet Fluorescence Spectroscopy

  • A simple whole-cell-based sensing system is proposed for determining the cell mass of H. pluvialis using ultraviolet fluorescence spectroscopy. An emission signal at 368 nm was used to detect the various kinds of green, green-brown, brown-red, and red H. pluvialis cells. The fluorescence emission intensities of the cells were highest at 368 nm with an excitation wavelength of 227 nm. An excitation wavelength of 227 nm was then selected for cell-mass sensing, as the emission fluorescence intensities of the cell suspensions were highest at this wavelength after subtracting the background interference. The emission fluorescence intensities of HPLC-grade water, filtered water, and HPLC-grade water containing a modified Bold's basal medium (MBBM) were measured and the difference was less than 1.6 for the selected wavelengths. Moreover, there was no difference in the emission intensity at 368 nm among suspensions of the various morphological states of the cells. A calibration curve of the fluorescence emission intensities. and cell mass was obtained with a high correlation ($R^2=0.9938$) for the various morphological forms of H. pluvialis. Accordingly, the proposed method showed no significant dependency on the various morphological cell forms, making it applicable for cell-mass measurement. A high correlation was found between the fluorescence emission intensities and the dry cell weight with a mixture of green, green-brown, brown-red, and red cells. In conclusion, the proposed model can be directly used for cell-mass sensing without any pretreatment and has potential use as a noninvasive method for the online determination of algal biomass.

  • 2-Dimensional fluorescence sensor has a wide range of excitation and emission wavelengths, that some biogenic fluorphors in a biological process can be monitored simultaneously. The production processes of 5-aminolevulinic aicd (ALA) by recombinant E. coli BL21 (DE3) pLysS harboring plasmid pFLS45 were on-line monitored by a 2-dimensional fluorescence sensor The characteristics of fluorescence spectrum was dependent upon physical and biological factors of a bioprocess such as culture pH, cell mass etc. Some off-line data were correlated to the fluorescence intensity well, which was monitored at some combination of excitation and emission wavelengths by the 2-dimensional fluorescence sensor.

  • The fluorescence excitation (FE) spectrum of the S1-S0 (1B2-1A1) transition of dimethyldiazirine cooled in supersonic jet expansions has been obtained. Dispersed fluorescence (DF) spectra have also been taken for some prominent features of the FE spectrum. Vibrational analyses of the FE and DF spectra with the help of an ab initio molecular orbital calculation lead to some new vibrational assignments and refined fundamental frequencies.
