Nutritive Value and Functional Properties of Protein Concentrate Fractionated from Chrysanthemum Flowers

  • Some functional properties and nutritive value were determined for the protein concentrated fractionated from chrysanthemum flower in orer to renew interest in the flowers as food. Proximate components of chrysanthemum flower protein concentration (FPC) showed 61.2% protein, 2.0% fat and 35.2% carbhydrate on a dry basis. In amino acid composition of FPC, glutamic acid was the highest in the content, follwoed by aspartic acid, leucine and lysine. The ratio of essential/ total amino acids(E/T) was 0.42, showing a higher level of essential amino acids compared to the FAO reference protein. Digestibility of chrysanthemum FPC by pepsin and trypsin was lwoer than that of casein and was negatively correlative to both water and fat absorptions. Similar characteristics were determined between chrysanthemum FPC and milk casein in their emulsifying activity and emulsion stability. This results indicate that flowers or petals of chrysanthemum might be developed as a good source of protein.

Functional Analysis of ESTs from the Flower Bud of Korean Ginseng

  • In order to study gene expression in a reproductive organ, we constructed a cDNA library of immature flower buds in Korean ginseng and generated expressed sequence tags (ESTs) of 3,360 clones randomly selected. The ESTs could be clustered into 1,844 non-redundant groups. Similarity search of the non-redundant ESTs against public non-redundant databases of both protein and DNA indicated that 1,254 groups show similarity to genes of known function. These ESTs clones were divided into sixteen categories depending upon gene function. The most abundant transcripts were unknown protein (72), chlorophyll a/b-binding protein (48), and stylar glycoprotein. There are no useful informations of gene expression during the development of flower bud in Korean ginseng. These results could help to understand the development of flower bud in Korean ginseng.

대청 추출물의 항산화 효소 활성 및 항균 효과 (Antioxidant Enzyme Activity and Antimicrobial Activity of Isatis tinctoria Extract)

  • 대청의 이용성 향상을 위한 자료 확보 측면에서 식물체 부위 및 추출 용매별에 따른 항산화 효소 활성 및 항균효과를 조사하였다. APX(Ascorbate Peroxidase) 활성은 줄기의 에탄올 추출물, 잎의 메탄올 추출물, 잎의 증류수의 추출물 순으로 높아 각각 1601.7, 1133.7 및 524.3(Unit/mg protein)을 나타냈다. CAT(Catalase) 활성은 꽃의 에탄올 추출물, 잎의 메탄올 추출물, 꽃의 증류수 추출물 순으로 높았는데, 각각 177.1, 120.8 및 55.4(Unit/mg protein)를 나타냈다. POD(Ascorbate Peroxidase) 활성은 꽃의 에탄올 추출물, 꽃의 메탄올 추출물, 줄기의 증류수 추출물 순으로 높았으며, 각각 27.1, 14.6 및 10.4(Unit/mg protein)를 나타냈다. SOD(superoxide dismutase) 활성은 뿌리의 증류수 추출물, 꽃의 메탄올 추출물, 뿌리의 에탄올 추출물 순으로 높았으며, 각각 90.8, 80.1, 75.5%를 나타냈다. 대청의 꽃 추출물은 Vibrio parahaemolyticus에 대해서만, 뿌리는 Staphy lococcus aureus에 대해서만, 줄기 추출물은 Bacillus subtilis, Escherichia coli, Staphy lococcus aureus에 대해서만 용매에 관계없이 항균활성을 나타냈다. 특히 대청 잎의 증류수 추출물은 Bacillus subtilis, Escherichia coli에 대해 높은 항균활성을 나타내어 저해환 직경이 각각 30.0 및 24.0 mm이었다. 이와 같은 결과는 약용식물로서 대청의 가치가 높음을 시사해 주었다.

홍화(Carthamus tinctorius L.)의 부위별 이화학적 특성 (Properties of the Chemical Composition of Safflower (Carthamus tinctorius L.))

  • In order to develop new materials for the functional feed, the components of safflower (Carthamus tinctorius L.) was studied. Chemical composition, minerals, amino acids, and fatty acids of the seed and the flower of safflower were analyzed. The chemical composition of safflower seed was 3.48% (w/w) moisture, 17.10% crude protein, 30.10% crude fat, 6.11% crude ash and these of safflower flower was 7.47% moisture, 26.30% crude protein, 11.50% crude fat, 5.73% crude ash. Mineral contents of the seed were K 170.70ppm, P 14.82ppm, Ca 13.17ppm, Mg 7.83 ppm whereas these of the flower were K 64.99 ppm, P 49.90 ppm, Ca 10.43ppm. Other mineral contents were less than 7.00ppm in all parts. The composition of the amino acid were approximately as follow, the major amino acid in all parts were aspartic acid, leucine, glycine, and arginine, the contents of these were 12.17mg/g, 11.52mg/g, 8.27mg/g, 6.99mg/g, 4.86mg/g in the seed, 19.35mg/g, 31.67mg/g, 10.30mg/g, 9.06mg/g, 12.51mg/g in the flower, respectively. The mahor fatty acids in the all parts were linoleic acid (C아래첨자). The linoleic acid and the palmitic acid (C아래첨자) in the seed and the flower parts were 77.75% (w/w), 19.32% and 8.37%, 25.62% respectively. On the basis of chemical analysis, the safflower showed to have relatively high contents of crude protein and crude fat, minerals, polyunsaturated fatty acid as linoleic acid. These results suggested that safflower was found to be a useful material of natural health food for the functional food development.

  • A cDNA encoding desiccation-related protein was isolated from a flower bud cDNA library of Chinese cabbage (C-DH) and its nucleotide sequence was characterized. It contains 679 bp nucleotides with 501 bp open reading frame. The amino acid sequence of the putative protein showed the highest amino acid sequence homology (79 % identity) to dehydrin protein in Gossypium hirsutum. Also, the C-DH shares 48-52% amino acid sequence identity with the other typical dehydrin proteins in plant cells. When the amino acid sequence of their proteins were aligned, several peptide motifs were well conserved, of which function has to be solved. Particularly the C-DH contains 15 additional amino acids at its N-terminus. Genomic Southern blot analysis using the coding region of C-DH showed that the C-DH consists of a single copy gene in Chinese cabbage genome. The C-DH mRNA, whose transcript size is 0.7 kb, was expressed with a tissue-specific manner. It was highly expressed in seed, flower buds and low expression as detected in root, stem or leaf tissues of Chinese cabbage. And the transcript level of C-DH was significantly induced by the treatment of plant hormone, abscisic acid and water-deficit conditions.

초음파 처리 인삼화뢰 추출물의 콜라겐 합성작용 및 MMP-1 발현저해 (Inhibition of MMP-1 Expression and Collagen Synthesis Activity of Ultrasonication Processed Ginseng Flower Buds Extract)

  • This study was to evaluate the effect of isopropyl alcohol fraction of ultrasonication processed ginseng flower buds(GFB-IF) on the collagen synthesis activity and inhibition of MMP-1 suppression in UV-irradiated human dermal fibroblasts. The higher contents of ginsenoside Rg2(8.234%), Rh1(5.749%), F4(3.881%) in isopropyl alcohol fraction of ginseng flower buds obtained by ultrasonication process at 600W(100℃) for 16 hours. GFB-IF had collagen synthesis effect. GFB-IF induced a significant dose-dependent decrease in the expression for MMP-1 protein. These results suggest that GFB-IF is a potential candidate for the prevention and treatment of wrinkle improving.

Molecular Cloning and Characterization of a Flower-specific Thionin in Chinese Cabbage

  • Thionins are a family of low molecular weight cysteine-rich antimicrobial peptides. We isolated a cDNA encoding thionin gene from a flower bud cDNA library of Chinese cabbage (CFT). The gene contains 611 by nucleotides with 60 bp, and 150 by untranslated regions at its N- and C-terminal, respectively. The deduced amino acid sequence encoded 133 amino acids containing precursor polypeptide. The protein reveals that the precursor has a tripartite structure: a putative signal sequence at the N-terminus, followed by a mature thionin peptide, and a C-terminal acidic domain, which facilitates transport of the mature thionin through membrane. Genomic Southern blot analysis suggests that the CFT gene may be present as a single or two copy gene in the Chinese cabbage genome. Northern blot analysis shows that the gene is specifically expressed in flowers, but not in leaves, stems, or roots. When we analyzed the antifungal activity of the recombinant CFT protein, which was expressed in E. coli using the truncated cDNA region corresponding to the mature protein part, it was not active on fungal growth inhibition.

Screening and isolation of antibacterial proteinaceous compounds from flower tissues: Alternatives for treatment of healthcare-associated infections

  • Healthcare-associated infection represents a frequent cause of mortality that increases hospital costs. Due to increasing microbial resistance to antibiotics, it is necessary to search for alternative therapies. Consequently, novel alternatives for the control of resistant microorganisms have been studied. Among them, plant antimicrobial protein presents enormous potential, with flowers being a new source of antimicrobial molecules. In this work, the antimicrobial activity of protein-rich fractions from flower tissues from 18 different species was evaluated against several human pathogenic bacteria. The results showed that protein-rich fractions of 12 species were able to control bacterial development. Due its broad inhibition spectrum and high antibacterial activity, the protein-rich fraction of Hibiscus rosa-sinensis was subjected to DEAE-Sepharose chromatography, yielding a retained fraction and a non-retained fraction. The retained fraction inhibits 29.5% of Klebsiella pneumoniae growth, and the non-retained fraction showed 31.5% of growth inhibition against the same bacteria. The protein profile of the chromatography fractions was analyzed by using SDS-PAGE, revealing the presence of two major protein bands in the retained fraction, of 20 and 15 kDa. The results indicate that medicinal plants have the biotechnological potential to increase knowledge about antimicrobial protein structure and action mechanisms, assisting in the rational design of antimicrobial compounds for the development of new antibiotic drugs.

Molecular Cloning of Peroxidase cDNA from Flower Bud of Ginseng(Panax ginseng)

  • A full-length cDNA (PPrx1) encoding peroxidase has been isolated and its nucleotide sequence determined from flower bud in ginseng plant (Panax ginseng). A PPrx1 cDNA is 1192 nucleotides long and has an open reading frame of 1062 bp with a deduced amino acid sequence of 354 residues (pI 7.53). The deduced amino acid sequence of PPrx1 matched to the previously reported peroxidase protein genes. The PPrx1 showed a high similarity with the $64\%$ identity with peroxidase of N. tabacum (AAK52084). In the phylogenetic analysis based on the amino acid residues, the PPrx1 was closer with peroxidase of G. max (AAD37376).

Immunostimulatory effects of the Pueraria lobata flower extract via MAPK signaling in RAW264.7 cells

  • Objectives: In this study, we investigated the effects of Pueraria lobata Ohwi flower extracts (PLFE) on macrophages and their underlying mechanism(s) of action. PLFE increased the production of NO and cytokines (IL-6 and TNF-𝛼) in a dose-dependent manner, indicating its immunostimulatory property. Furthermore, PLFE upregulated iNOS, COX-2, and mitogen-activated protein kinase (MAPK) signaling in RAW264.7 cells. Additionally, PLFE enhanced the phosphorylation of I𝜅B𝛼 and subsequent I𝜅B𝛼 degradation, thereby enabling the nuclear translocation of NF-𝜅B. Taken together, these findings demonstrate that the immunostimulatory effects of PLFE are mediated by the nuclear translocation of the p65 subunit of NF-𝜅B and subsequent secretion of cytokines (IL-6 and TNF-𝛼), upregulation of iNOS and COX-2, and stimulation of MAPK signaling (JNK, ERK, and p38). Thus, PLFE may be a potential immunostimulatory therapeutic.
