The success of embryo cloning depends on numerous factors; interaction between recipient ooplasm and donor nucleus, nuclear reprogramming, oocyte activation, and donor cell cycle and type. In this study, the cell cycle and apoptosis of bovine fetal fibroblast as a donor cell for embryo cloning were evaluated following different activation treatments. (omitted)
Objectives : This study is aimed to investigate the effects of bee venom and melittin on cell death in synovial cell line. Methods : It was evaluated by using MTT assay, morphologic method, DNA fragmenation, NO generation, flow cytometry, immunocytochemistry analysis, RT-PCR, Western blot. Results : The obtained results are summarized as follows: 1. The MTT assay demonstrated that synovial cell viability was significantly inhibitted dose-dependently by treatment with bee venom and melittin in comparison with control. 2. The morphologic study demonstrated that synovial cell showed apoptosis after treatment with bee venom and melittin for 6 hours using microscope. 3. In case of NO generation bee venom group and melittin group showed significant inhibition in comparison with control. 4. The Flow cytometry demonstrated that apoptosis of synovial cell treated with bee venom and melittin was related with stop of cell cycle in stage of $G_0/G_1$. 5. DNA fragmenation demonstrated that synovial cell treated with bee venom and melittin showed DNA ladder below l Kbp. 6. Immunocytochemistry assay demonstrated that COX-II and PLA2 were strongly down-regulated by treatment with bee venom and melittin whereas iNOS was almostly not expressed by bee venom treatment and slightly expressed by melittin treatment. 7. RT-PCR analysis demonstrated that iNOS were strongly down-regulated by treatment with bee venom and melittin whereas COX-II was almostly not expressed by bee venom treatment and slightly expressed by melittin treatment. 8. Western blot demonstrated that iNOS were strongly down-regulated by treatment with $15{\mu}g/ml$ bee venom whereas COX-II was strongly down-regulated from $5{\mu}g/ml$ bee venom. Conclusions : These results suggest that bee venom and melittin have significant effect on cell death in synovial cell line and further study is needed in vivo.

The MTT assay demonstrated that synovial cell viability was significantly inhibitted dose-dependently by treatment with BV and PBV in comparison with control. And the inhibitory effect of BV and PBV was almost same. 2. The morphologic study demonstrated that synovial cell showed apoptotic body resulted from apoptosis after treatment with BV and PBV for 6 hours using microscope. 3. The Flow cytometry demonstrated that apoptosis of synovial cell treated with BV and PBV was related with stop of cell cycle in stage of G0/G1. 4. Immunocytochemistry assay demonstrated that COX-II and iNOS were slightly expressed by treatment with BV and PBV in comparison with control group. 5. RT-PCR analysis demonstrated that COX-II were almost down-regulated by high dose treatment with BV and PBV in comparison with control group. iNOS were well down-regulated by treatment with $5{\mu}g/ml$ BV and PBV whereas it was well expressed in control group. Conclusion : These results suggest that bee venom and purified bee venom have significant effect on cell death in synovial cell line and further study is needed in vivo.
Purpose : This study was aimed to investigate the inhibitory effect of Corydalis Tuber on the proliferation of human uterine leiomyoma cell and the expression of gene related the mechanism of cell apoptosis. Methods : We counted the number of suvival cells treated with indicated concentration of Corydalis Tuber and investigated cell viability by MTS assay. Furthermore, flow cytometric analyis were used to dissect between necrosis and apoptosis related with cell cycle and then we observed the differential gene expression by western blot analysis. Results : 1) The inhibitory effect on the proliferation of uterine leiomyoma cell treated with Corydalis Tuber was increased in a concentration and time proportional. 2) The result of flow cytometry analysis, subG1 phase arrest related cell apoptosis was not investigated in uterine leiomyoma cell treated Corydalis Tuber but showed G2/M phase prolongation. 3) The gene expression of p27, p21 related cell cycle was increased according to increasing concentration, but p53 was not exchanged. 4) The dephosphorylation of pRb gene were increased dependent on treatment concentration and pro-caspase 3, CDK4 were not exchanged. Conclusion : This study showed that Corydalis Tuber have the inhibitory effect on the proliferation of human uterine leiomyoma cell but the effect was thoughted no relationship with apoptosis. The inhibitory effect was suggested that dephosphorylation of pRb gene induced with increasing p21, p27 prolonged cell division in G2/M phase.
Performance and flow characteristics of a small forward swept propeller fan for home refrigerators are studied experimentally. An unusual discontinuity is observed in the performance curve of the fan. Mean flow fields measured with as-hole Pitot probe reveal that the flow is axial at the high flow rate and radial at the low flow rate. The flow structure changes abruptly across the discontinuity. Unsteady flow measurements with a set of hot-wire probes indicate that near the discontinuity a single-cell stall rotates at 40% speed of the fan speed, while away from the discontinuity the flow shows periodic variation corresponding to the blade passage frequency. Phase-lock averaged flow fields measured with a triple-sensor hot-wire probe show that there appears radially inward flow over the pressure side of the blade and the outward passage flow over the tip.
본 연구에서는 Moving Cell theory에 기반한 DNL(Dynamic Network Loading) 모형을 개발하고 이를 이용해 혼잡이 존재하는 교통망에서 교통류가 갖는 동적 특성을 분석하였다. 제시된 모형에서는 동일 시간대에 링크에 진입하는 교통량을 하나의 Cell로 형성하고 Cell following rule에 따라 링크에서 진행시킨다. 기존의 DNL 모형들은 링크에서 발생하는 물리적인 패기행렬을 묘사하기 위해 연속성을 갖는 단일 링크를 주행구간과 대기행렬 구간으로 분리하여 링크에서 발생하는 동적 상태(state)를 주행과 대기로 간단히 묘사하는 방법을 사용하고 있다. 하지만, 이러한 기법은 교통류의 다양한 동적 특성을 묘사하는데 한계점을 가지고 있었다. 이러한 문제점을 극복하기 위해 본 연구에서는 오염물질 확산분석 등에 주로 이용되었던 Lagrangian method과 차량 추종모형을 결합한 Moving Cell theory를 개발하였다. A boron doping process using a boron tri-bromide ($BBr_3$) as a boron source was applied to form a $p^+$ emitter layer on an n-type mono-crystalline CZ substrate. Nitrogen ($N_2$) gas as an additive of the diffusion process was varied in order to study the variations in sheet resistance and the uniformity of doped layer. The flow rate of $N_2$ gas flow was changed in the range 3 slm~10 slm. The sheet resistance uniformity however was found to be variable with the variation of the $N_2$ flow rate. The optimal flow rate for $N_2$ gas was found to be 4 slm, resulting in a sheet resistance value of $50{\Omega}/sq$ and having a uniformity of less than 10%. The process temperature was also varied in order to study its influence on the sheet resistance and minority carrier lifetimes. A higher lifetime value of $1727.72{\mu}s$ was achieved for the emitter having $51.74{\Omega}/sq$ sheet resistances. The thickness of the boron rich layer (BRL) was found to increase with the increase in the process temperature and a decrease in the sheet resistance was observed with the increase in the process temperature. Furthermore, a passivated emitter solar cell (PESC) type solar cell structure comprised of a boron doped emitter and phosphorus doped back surface field (BSF) having Ni/Cu contacts yielding 15.32% efficiency is fabricated.
Finite element analysis of fluid flow with moving free surface has been carried out in two and tree dimensions. The new VOF-based numerical algorithm that has been proposed by the present authors was applied to several 2-D and 3-D free surface flow problems. The proposed free surface tracking scheme is based on two numerical tools that have been newly introduced by the present authots; the orientation vector to represent the free surface orientation in each cell and the baby-cell to determine the fluid volume flux at each cell boundary. The proposed numerical algorithm has been applied to 2-D and 3-D cavity filling and sloshing problems, which demonstrated versatility and effectiveness of the new free surface tracking scheme as well as the overall solution procedure. The proposed numerical algorithm resolved successfully the interacting free surface with each other. The simulated results demonstrated the applicability of proposed numerical algorithm to the practical problems of large free surface motion. Also, it has been demonstrated that the proposed free surface tracking scheme can be easily implemented in any irregular non-uniform grid systems and can be extended to the 3-D free surface flow problem without additional efforts.
본 연구에서는 수계 레독스 흐름전지에서 사용하는 멤브레인 특성분석방법을 개선하여 비수계 레독스 흐름 전지를 위한 멤브레인 특성분석방법을 확립하였다. 비수계 레독스 흐름 전지에 적합한 멤브레인 특성을 확인하기 위해 상용 멤브레인의 이온교환능력, 이동수, 이온 전도도, 활물질 투과도, 전지효율 실험 등 특성분석들을 수행하였다. 상용 음이온 교환 멤브레인의 특성분석 실험을 통해 충 방전 효율 및 에너지효율과 이온 선택성의 상관관계를 조사하였다. Neosepta AHA 음이온 교환 멤브레인은 이동수 측정에서 0.81의 값으로 비수계 전해질에서 비교적 낮은 이온 선택성을 보였지만, 충방전 전지효율 평가에서는 92%의 충 방전효율과 86%의 에너지효율을 각각 나타내었다. 또한 이온의 선택성이 없는 다공성 멤브레인은 높은 전류밀도의 비수계 레독스 흐름 전지에 적절함을 알 수 있었다.
We have proposed a novel planar lightwave circuit (PLC) optical sensor to monitor the contamination in a flow-cell where water is continuously supplied through a water quality measurement system. We designed a PLC chip with a V-shape waveguide and the simulated its function as a sensor for monitoring contamination in a flow-cell using a numerical the FDTD (finite-difference time-domain) analysis. A novel cross type of waveguide was introduced to make the PLC chip of the V-shaped waveguide. The fabricated PLC was cut into the cross waveguide. A change in the optical propagation loss of the PLC sensor was observed after immersing the PLC sensor into city water. It was determined that the propagation loss of the PLC sensor was 3 dB at a wavelength of $1.55{\mu}m$ in the city water for 15 days.
