Use of Human Adipose Tissue as a Source of Endothelial Cells (혈관내피세포 채취의 원천으로 인간 지방조직의 활용)

  • Purpose: Adipose tissue is located beneath the skin, around internal organs, and in the bone marrow in humans. Its main role is to store energy in the form of fat, although it also cushions and insulates the body. Adipose tissue also has the ability to dynamically expand and shrink throughout the life of an adult. Recently, it has been shown that adipose tissue contains a population of adult multipotent mesenchymal stem cells and endothelial progenitor cells that, in cell culture conditions, have extensive proliferative capacity and are able to differentiate into several lineages, including, osteogenic, chondrogenic, endothelial cells, and myogenic lineages. Materials and Methods: This study focused on endothelial cell culture from the adipose tissue. Adipose tissues were harvested from buccal fat pad during bilateral sagittal split ramus osteotomy for surgical correction of mandibular prognathism. The tissues were treated with 0.075% type I collagenase. The samples were neutralized with DMEM/and centrifuged for 10 min at 2,400 rpm. The pellet was treated with 3 volume of RBC lysis buffer and filtered through a 100 ${\mu}m$ nylon cell strainer. The filtered cells were centrifuged for 10 min at 2,400 rpm. The cells were further cultured in the endothelial cell culture medium (EGM-2, Cambrex, Walkersville, Md., USA) supplemented with 10% fetal bovine serum, human EGF, human VEGF, human insulin-like growth factor-1, human FGF-$\beta$, heparin, ascorbic acid and hydrocortisone at a density of $1{\times}10^5$ cells/well in a 24-well plate. Low positivity of endothelial cell markers, such as CD31 and CD146, was observed during early passage of cells. Results: Increase of CD146 positivity was observed in passage 5 to 7 adipose tissue-derived cells. However, CD44, representative mesenchymal stem cell marker, was also strongly expressed. CD146 sorted adipose tissue-derived cells was cultured using immuno-magnetic beads. Magnetic labeling with 100 ${\mu}l$ microbeads per 108 cells was performed for 30 minutes at $4^{\circ}C$ a using CD146 direct cell isolation kit. Magnetic separation was carried out and a separator under a biological hood. Aliquous of CD146+ sorted cells were evaluated for purity by flow cytometry. Sorted cells were 96.04% positivity for CD146. And then tube formation was examined. These CD146 sorted adipose tissue-derived cells formed tube-like structures on Matrigel. Conclusion: These results suggest that adipose tissue-derived cells are endothelial cells. With the fabrication of the vascularized scaffold construct, novel approaches could be developed to enhance the engineered scaffold by the addition of adipose tissue-derived endothelial cells and periosteal-derived osteoblastic cells to promote bone growth.

Changes in CGRP-immunoreactive Nerve Fibers during Expansion of Midpalatal Suture of the Rat (백서 정중구개봉합 확대후의 CGRP 면역반응 신경섬유의 변화)

  • Midpalatal suture expansion is often used for patients haying narrow maxillary arch, cleft palate, respiratory handicap with narrow nasal cavity. CGRP has been known as a modulator of pain transmission in central nervous system and a local effector to peripheral tissue causing vasodilation, increase of blood flow, modulation of immune system, regulation of macrophagic function and stimulation of bone formation. To investigate changes of CGRP-immunoreactive nerve fibers in midpalatal suture during the expansion, immunohistochemical study was performed by using rats. Experimental rats (10 weeks, 250 gm) were divided into five groups (control, 1, 4, 7, 14 days group (each n=4) and applied orthodontic force (approximately 200gm) to upper anterior incisors. Frozen sections of midpalatal suture area were immunostained by using rabbit antisera. The results were as follows. ${\cdot}$ The CGRP-immunoreactive nerve fibers were hardly observed in control group. ${\cdot}$ In 1 day group, the CGRP-immunoreactive nerve fibers were more increased around the vessels than control group. ${\cdot}$ In 4 days group, the CGRP-immunoreactive nerve fibers were more increased than control group, but not more increased than 1 day group. Vascular diameter was more enlarged. ${\cdot}$ In 7 days group, especially, hematoxilin affinity of cells was remarkable and cells were arranged along the bone margin. The CGRP-immunoreactive nerve fibers were more reduced than 4 days group and vascular diameter was also reduced. ${\cdot}$ In 14 days group, the CGRP-immunoreactive nerve fibers were similar to those of 7 days group and the irregularity of bone margin was almost recoverd. In Conclusion, the CGRP-immunoreactive nerve fibers nay be related to initial neurogenic inflammatory reaction in expanding mid-palatal suture.

The Use of Interviews in Documentary Films (다큐멘터리에서의 인터뷰 활용 방식 연구)

  • In some documentary films, there are interviews, but in some other ones, there aren't any. As for the documentaries with interviews, the director uses talking heads to suggest their own arguments and to have audience agree. It is evident that interviews play a key role since they lead the story of a human-related documentary. Some directors prefer interviews where directors' questions and interviewees' answers are both heard. On the other hand, there a re directors who carry out the story just by using the answers from the interviewees. Then, it becomes crucial that we understand both of these styles and have new perspectives when watching documentary films. Ethics has always been considered one of the most important factors in documentaries and since they are believed to be "true," the influence documentaries have on the society is enormous. However, possibilities of exaggerating or manipulating the "truth" always exist. Therefore, it is important for the audience to identify the intentions of the director. As a matter of fact, there have been countless researches being done, but it is difficult to find studies that discuss the ways interviews are used in documentary films. The two different styles mentioned above do have huge differences in terms of directing methods. Thus, taking a look at differences from diverse angles will help us better understand the essentials of documentary films. This study will take examples from the interviews in Michael Moore's (2002), (2007) and observe how the flow of interviews can change when the voice of the director gets involved. There will also be a close examination of interviews in Kim Ilran and Hong Ji Yoo's (2011), and (2005) directed by Jim Butterworth, Aeron Lubarsky, and Lisa Sleeth, as well as Sebastian Junger and Tim Hetherington's (2010). By a careful review, use of dissolve, cross-cutting, and flashback will be analyzed to show how different editing techniques have an impact on subjective views of the director.

Stand Water Balance and Stream Water Quality in Small Forested Watershed Yangpyong Gyeonggido (경기도(京畿道) 양평지역(陽平地域) 산림(山林) 소류역(小流域)의 수수지(水收支)와 계류수(溪流水)의 수질특성(水質特性))

  • This study was carried out to investigate the characteristics of water quality variations by stand water balance in YangPyong-Gun Gejung-Lee small forest watershed. Water quantity. pH, $Cl^-$, $NO_3{^-}$, $SO_4{^{2-}}$, $Na^+$, $NH_4{^+}$, $K^+$, $Mg^{2+}$, $Ca^{2+}$ were monitored in open rainfall for one unit storm and long-term stream water in small forest watershed from January. 1998 to December. 1999. The results were summarized as follows: The runoff rate was 46.4% in 1998 and 52.2% in 1999. The average pH values of rainfall were 4.8 to 6.2 and those of stream water were 6.4 to 7.1 in small forest watershed. Total amount of input anion and cation values (kg/ha) in rainfall were $SO_4{^{2-}}>NO_3{^-}>Ca^{2+}>NH_4{^+}>Cl^->Na^+>K^+>Mg^{2+}$ and in stream water were $NO_3{^-}>Ca^{2+}>SO_4{^{2-}}>Na^+>Cl^->K^+>Mg^{2+}>NH_4{^+}$ in the order, respectively. The dissolved $NH_4{^+}$ was stored 5.29kg/ha and output of the other contents were more flow than input in small forest watershed.

Effects of Water Temperature and Body Weight on Oxygen Consumption Rate of Black Rockfish, Sebastes schlegeli (조피볼락, Sebastes schlegeli의 산소 소비율에 미치는 수온과 체중의 영향)

  • The effect of water temperature (T) and body weight (W) on the oxygen consumption of the fasted black rockfish, Sebastes schlegeli was investigated to provide empirical data for the culture management and bioenergetic growth model of this species. The mean wet body weights of two fish groups used for the present experiment were $12.9{\pm}2.7g$ ($mean{\pm}SD$) and $351.1{\pm}9.2g$. The oxygen consumption rate (OCR) was measured under three water temperature regimes (15, 20 and $25^{\circ}C$) at an interval of 5 minutes for 24 hours using a continuous flow-through respirometer. In each treatment three replicates were set up and 45 fish in small size groups and 6 fish in large size groups were used. The OCRs increased with increasing water temperature in both size groups (p<0.001). Mean OCRs at 15, 20 and $25^{\circ}C$ were 414.2, 691.5 and $843.8mg\;O_2\;kg^{-1}h^{-1}$ in small size groups, and 182.0, 250.7 and $328.2mg\;O_2\;kg^{-1}h^{-1}$ in large size groups, respectively. The OCRs decreased with increasing body weights in three water temperature groups (p<0.001). The mass effect on metabolic rate can be expressed by the power of 0.69~0.75. The data are best described by the relationship: OCR=89.12+28.79T-1.17W. $Q_{10}$ values ranged 1.90~2.79 between 15 and $20^{\circ}C$, 1.49~1.71 between 20 and $25^{\circ}C$, and 1.80~2.03 over the full temperature range, respectively. The energy loss by metabolic cost increased with increasing water temperature and decreasing body weight (p<0.001). Mean energy loss rates by oxygen consumption at 15, 20 and $25^{\circ}C$ were 282.9, 472.3 and $576.3kJ\;kg^{-1}d^{-1}$ in small size groups and 124.3, 171.3 and $224.1kJ\;kg^{-1}d^{-1}$ in large size groups, respectively.

Shallow subsurface structure of the Vulcano-Lipari volcanic complex, Italy, constrained by helicopter-borne aeromagnetic surveys (고해상도 항공자력탐사를 이용한 Italia Vulcano-Lipari 화산 복합체의 천부 지하 구조)

  • Helicopter-borne aeromagnetic surveys at two different times separated by three years were conducted to better understand the shallow subsurface structure of the Vulcano and Lipari volcanic complex, Aeolian Islands, southern Italy, and also to monitor the volcanic activity of the area. As there was no meaningful difference between the two magnetic datasets to imply an apparent change of the volcanic activity, the datasets were merged to produce an aeromagnetic map with wider coverage than was given by a single dataset. Apparent magnetisation intensity mapping was applied to terrain-corrected magnetic anomalies, and showed local magnetisation highs in and around Fossa Cone, suggesting heterogeneity of the cone. Magnetic modelling was conducted for three of those magnetisation highs. Each model implied the presence of concealed volcanic products overlain by pyroclastic rocks from the Fossa crater. The model for the Fossa crater area suggests a buried trachytic lava flow on the southern edge of the present crater. The magnetic model at Forgia Vecchia suggests that phreatic cones can be interpreted as resulting from a concealed eruptive centre, with thick latitic lavas that fill up Fossa Caldera. However, the distribution of lavas seems to be limited to a smaller area than was expected from drilling results. This can be explained partly by alteration of the lavas by intense hydrothermal activity, as seen at geothermal areas close to Porto Levante. The magnetic model at the north-eastern Fossa Cone implies that thick lavas accumulated as another eruption centre in the early stage of the activity of Fossa. Recent geoelectric surveys showed high-resistivity zones in the areas of the last two magnetic models.

Geoscientific land management planning in salt-affected areas* (염기화된 지역에서의 지구과학적 토지 관리 계획)

  • Over the last twenty years, farmers in Western Australia have begun to change land management practices to minimise the effects of salinity to agricultural land. A farm plan is often used as a guide to implement changes. Most plans are based on minimal data and an understanding of only surface water flow. Thus farm plans do not effectively address the processes that lead to land salinisation. A project at Broomehill in the south-west of Western Australia applied an approach using a large suite of geospatial data that measured surface and subsurface characteristics of the regolith. In addition, other data were acquired, such as information about the climate and the agricultural history. Fundamental to the approach was the collection of airborne geophysical data over the study area. This included radiometric data reflecting soils, magnetic data reflecting bedrock geology, and SALTMAP electromagnetic data reflecting regolith thickness and conductivity. When interpreted, these datasets added paddock-scale information of geology and hydrogeology to the other datasets, in order to make on-farm and in-paddock decisions relating directly to the mechanisms driving the salinising process. The location and design of surface-water management structures such as grade banks and seepage interceptor banks was significantly influenced by the information derived from the airborne geophysical data. To evaluate the effectiveness ofthis planning., one whole-farm plan has been monitored by the Department of Agriculture and the farmer since 1996. The implemented plan shows a positive cost-benefit ratio, and the farm is now in the top 5% of farms in its regional productivity benchmarking group. The main influence of the airborne geophysical data on the farm plan was on the location of earthworks and revegetation proposals. There had to be a hydrological or hydrogeological justification, based on the site-specific data, for any infrastructure proposal. This approach reduced the spatial density of proposed works compared to other farm plans not guided by site-specific hydrogeological information.

Risk Factors Analysis and Results of the Arterial Switch Operation for Transposition of the Great Arteries with Intact Ventricular Septum (심실중격결손을 동반하지 않은 대혈관전위증 환자에서 동맥 전환술의 결과 및 위험인자 분석에 관한 연구)

  • Background: To evaluate the risk factor and long-term result of arterial switch operation , a retrospective study was done. Material and Method: A retrospective analysis was done to evaluate the early and long-term results on 58 patients who underwent an arterial switch operation(ASO) for transposition of the great arteries(TGA) with intact ventricular septum, between January 1988 and December 1996. Beforesurgery, 36 patients(62.1%) underwent balloon atrial septostomy, 32 patients(51.7%) received PGE1 infusion, and preparatory banding of pulmonary artery was performed on 6 patients(mean LV/RV pressure ratio 0.53$\pm$0.11). Result: The age at operation ranged from 1 to 137 days(mean 24$\pm$26 days) and the weights ranged from 1.8 to 6.8 kg (mean 3.5$\pm$0.8 kg). There were 14 early deaths(24.1%), but of the last 24 patients operated on since 1994, there were only 2 early deaths(8.3%). In the risk factor analysis, the date of operation was the only risk factor for early death(p-value < 0.01). Eight of the 14 early deaths were due to acute myocardial failure(mainly inadequate coronary blood flow). The length of follow-up ranged from 2 months to 8 years, average of 36$\pm$27 months. The follow-up included sequential noninvasive evaluations and 21 catheterizations and angiographic studies performed 5 to 32 months postoperatively with particular attention to the great vessel and coronary anastomosis, ventricular function, valvular competence, and cardiac rhythm. There were 5 late deaths(11.4%), one of thesedeaths was related to the late coronary problems, two to aspiration, one to uncontrolled chronic mediastinitis, and one to progressive aortic insufficiency and heart failure. The most frequent postoperative hemodynamic abnormality was supravalvular stenosis and the degree of pulmonary or aortic obstruction had slowly progressed in some cases, however there were no children who had to undergo a reoperation for supravalvular pulmonary or aortic stenosis. Aortic regurgitation was identified in 9 patients, which was mild in 7 and moderate in 2 and had progressed in some cases. Two patients who had an unremarkable perioperative course were identified as having coronary artery obstructions. The other late survivors were in good condition, were in sinus rhythm, and had normal LV functions. Actuarial survival rate at 8 years was 68.8%. Conclusion: We concluded that anatomic correction will be established as the optimal approach to the TGA with intact ventricular septum, though further long-term evaluations are needed.

A study on the operation realities of the teenager internet shopping malls and the entrepreneurship education (청소년들의 인터넷 쇼핑몰 운영 실태 및 창업교육에 관한 연구)

  • The object of this study is to compare the cognitive differences between teenagers who are operating the internet shopping mall and those who are not operating to analyze the effect of the enterprise experience on the career maturity of teenager. And we are to present the operation methods of teenager entrepreneurship education at school by identifying the difficulties of teenager internet shopping malls and investigating demands of teenagers for the entrepreneurship education. The results of this study are as following. First, the internet shopping malls established by teenagers were mostly operated with small scale and capital dealt with clothing and fashion accessories. It is difficult for most of teenagers to inform the shopping malls and understand the flow of fashion and demands of consumers. They acquired the informations on enterprise using internet or acquaintances. This is because there are no professional teenager entrepreneurship education. And they chose the confidence and spirit of challenge which are mostly needed for success of the internet shopping malls. Therefore, they can acquire the confidence and spirit of challenge by effective entrepreneurship education on resource management, team management, business plan and marketing. Second, teenagers who are operating the internet shopping malls got higher scores on career maturity and degree of need in the entrepreneurship education than those who are not. Thus the expansion of the systemed and diversified entrepreneurship education at school is needed to increase business practice and entrepreneurship. Third, most teenagers wanted the entrepreneurship education at school. They preferred external lectures who can teach them with professional experiences and practical knowledges using discretional activity classes or club activity classes. Dividend classes of creative experience activities including career, voluntary and club activities are increased in 2009 revised education curriculum. Using these classes, it requires to operate the entrepreneurship education which make students decide their career themselves through concrete education and experience. Consequently, the expansion of the systemed and diversified teenager entrepreneurship education at school is needed using development of practical entrepreneurship education program, professional teacher training and revitalization of entrepreneurship club activities.

Study of Rainfall-Runoff Variation by Grid Size and Critical Area (격자크기와 임계면적에 따른 홍수유출특성 변화)

  • This study utilized the 1/25,000 topographic map of the upper area from the Geum-ho watermark located at the middle of Geum-ho river from the National Geographic Information Institute. For the analysis, first, the influence of the size of critical area to the hydro topographic factors was examined changing grid size to $10m{\times}10m,\;30m{\times}30m\;and\;50m{\times}50m$, and the critical area for the formation of a river to $0.01km^2{\sim}0.50km^2$. It is known from the examination result of watershed morphology according to the grid size that the smaller grid size, the better resolution and accuracy. And it is found, from the analysis result of the degree of the river according to the minimum critical area for each grid size, that the grid size does not affect on the degree of the river, and the number of rivers with 2nd and higher degree does not show remarkable difference while there is big difference in the number of 1st degree rivers. From the results above, it is thought that the critical area of $0.15km^2{\sim}0.20km^2$ is appropriate for formation of a river being irrelevant to the grid size in extraction of hydro topographic parameters that are used in the runoff analysis model using topographic maps. Therefore, the GIUH model applied analysis results by use of the river level difference law proposed in this study for the explanation on the outflow response-changing characters according to the decision of a critical value of a minimum level difference river, showed that, since an ogival occurrence time and an ogival flow volume are very significant in a flood occurrence in case of not undertow facilities, the researcher could obtain a good result for the forecast of river outflow when considering a convenient application of the model and an easy acquisition of data, so it's judged that this model is proper as an algorism for the decision of a critical value of a river basin.
