Variation of Decursin Contents of Root Ages in Floral Inhibition Cultured Angelica gigas Nakai (화성억제재배한 참당귀의 년근별 약효성분함량)

  • In order to clarify the contents of active principles in floral inhibition cultured Angelica gigas roots, the amounts of crude extract and the contents of decursin and decursinol angelate in the yearly roots were analyzed and compared with respect to the root age, root part and growth stage. The woody cell, weight and width of pith and cortex were also investigated at different growth stage to observe the developmental characteristics of lignification in the roots as bolting and flowering in normal cultured Angelica gigas. The amount of crude extracts did not differ with plant ages, whereas the contents of decursin and decursinol angelate were differed and the highest in 3 year old roots. The contents of decursin in 1, 2 and 3 year old roots were 3.71, 4.76 and 8.20% and those of decursinol angel ate were 2.84, 3.40 and 5.01%, respectively. The amount of crude extracts, and the contents of decursin and decursinol angelate were the highest in fine roots, followed by the lateral roots and the lowest in the primary roots. On the other hand, the amount of the constituents in the cortex were much higher than those in the pith of the root. The amounts of crude extract, and the contents of decursin and decursinol angelate showed the highest value at the vagetative stage and decreased with development to bolting and blooming stage. Woody cells were accumulated in the pith of the root as advancing growth stage, so that the weight and radius of the pith increased, whereas the relative weight and width of the cortex decreased slightly.

High-density genetic mapping using GBS in Chrysanthemum

  • Chrysanthemum is one of the most important floral crop in Korea produced about 7 billion dollars (1 billion for pot and 6 billion for cutting) in 2013. However, it is difficult to breed and to do genetic study because 1) it is highly self-incompatible, 2) it is outcrossing crop having heterozygotes, and 3) commercial cultvars are hexaploid (2n = 6x = 54). Although low-density genetic map and QTL study were reported, it is not enough to apply for the marker assisted selection and other genetic studies. Therefore, we are trying to make high-density genetic mapping using GBS with about 100 $F_1s$ of C. boreale that is oHohhfd diploid (2n = 2x = 18, about 2.8Gb) instead of commercial culitvars. Since Chrysanthemum is outcrossing, two-way pseudo-testcross model would be used to construct genetic map. Also, genotype-by-sequencing (GBS) would be utilized to generate sufficient number of markers and to maximize genomic representation in a cost effective manner. Those completed sequences would be analyzed with TASSEL-GBS pipeline. In order to reduce sequence error, only first 64 sequences, which have almost zero percent error, would be incorporated in the pipeline for the analysis. In addition, to reduce errors that is common in heterozygotes crops caused by low coverage, two rare cutters (NsiI and MseI) were used to increase sequence depth. Maskov algorithm would also used to deal with missing data. Further, sparsely placed markers on the physical map would be used as anchors to overcome problems caused by low coverage. For this purpose, were generated from transcriptome of Chrysanthemum using MISA program. Among those, 10 simple sequence repeat (SSR) markers, which are evenly distributed along each chromosome and polymorphic between two parents, would be selected.

Seed Transmission Rates of Bean pod mottle virus and Soybean mosaic virus in Soybean May Be Affected by Mixed Infection or Expression of the Kunitz Trypsin Inhibitor

  • To facilitate their spread, plant viruses have developed several methods for dispersal including insect and seed transmission. While insect transmission requires virus stability against insect digestion, seed-transmitted viruses have to overcome barriers to entry into embryos. Bean pod mottle virus (BPMV) is transmitted through seed at levels typically below 0.1%, but co-infection with Soybean mosaic virus (SMV) enhanced the seed transmission rate of BPMV in one experiment. In contrast, the rate of SMV seed transmission was not affected by BPMV co-infection. In a second preliminary study, the rate of SMV transmission was lower in an isoline of Williams 82 that contained a null mutation for the Kunitz trypsin inhibitor gene than in Williams 82. In this preliminary study, we observed that factors such as protease inhibitor expression and dual infection may affect the frequency of seed transmission of BPMV and SMV.

Occurrence of Off-type Plants in japonica/indica Hybrid Rice Cultivars

  • Frequent occurrence of off-type plants in a given cultivar has been a serious problem in both breeder's and farmer's fields. An experiment was designed to examine the differences in rate of occurrence of off-type plants among Tongil-type cultivars (high yielding cultivars derived from indica/japonica hybridization) from which the possible cause of higher occurrence of off-type plant in a specific cultivar was deduced. Among five Tongil-type cultivars examined for morphological variant in the field, only one cultivar, Dasanbyeo, had off-type plants. When analyzed with SSR markers, off-type plants showed different band patterns from original cultivar, having several extra bands in addition to cultivar-specific band, suggesting that off-type plants were originated from Dasanbyeo, rather than originated from mixing or mishandling of seed materials with other cultivars. The possible cause of off-type occurrence seems to be natural pollination with other cuItivars adjacent to the original cultivar during seed multiplication. This was supported from the observation that self-crossed progeny of the off type plants showed a wide range of variation of agronomic traits which could not be observed when there was a smaller introduction of genes to the fixed germplasm as happened in the case of cultivar mutation. Another evidence supported this idea that Dasanbyeo showed much of difference in floral organ and behavior to other cultivar to be subjected to higher out-crossing than other cultivars examined.

Morphometric Study of Achyranthes bidentata Complex Using Numerical Taxonomy (수리분류를 이용한 쇠무릎 분류군의 외부형태 연구)

  • 'Usul' is a traditional medicinal herb, which has anti-inflammatory activities is distributed in India, Nepal, China, Korea, Japan. Korea pharmacopeia listed 'Usul' as either a species of Achyranthes japonica (Miq.) Nakai or A. bidentata Blume. Recent taxonomic studies in China and Japan delimited these taxa as two varieties, A. bidentata Blume var. bidentata and var. japonica Miq. A multivariate morphometric study of Achyranthes bidentata complex was undertaken to assess the entities of taxa that usefully could be recognized. Five quantitative characters were reviewed and analyzed with 293 specimens from Korea. The univariate analysis of inflorescence length, interval between florets, angle between floret and floral axis indicated that ranges among all taxa were continuous. However, quantitative characters of membrane size and the number of hairs within 4 were useful to identify two varieties. In PCA, the first three principal components accounted for 89.4% of the total variance. PCA revealed that var. bidentata showed distinctions in morphological attributes from var. japonica entity. Therefore, continued recognition at the infraspecifc level for these taxa is supported.

AltMV TGB1 Nucleolar Localization Requires Homologous Interaction and Correlates with Cell Wall Localization Associated with Cell-to-Cell Movement

  • The Potexvirus Alternanthera mosaic virus (AltMV) has multifunctional triple gene block (TGB) proteins, among which our studies have focused on the properties of the TGB1 protein. The TGB1 of AltMV has functions including RNA binding, RNA silencing suppression, and cell-to-cell movement, and is known to form homologous interactions. The helicase domains of AltMV TGB1 were separately mutated to identify which regions are involved in homologous TGB1 interactions. The yeast two hybrid system and Bimolecular Fluorescence Complementation (BiFC) in planta were utilized to examine homologous interactions of the mutants. Helicase motif I of AltMV TGB1 was found to be critical to maintain homologous interactions. Mutations in the remaining helicase motifs did not inhibit TGB1 homologous interactions. In the absence of homologous interaction of TGB1, subcellular localization of helicase domain I mutants showed distinctively different patterns from that of WT TGB1. These results provide important information to study viral movement and replication of AltMV.

Identification of the Regulatory Region Responsible for Vascular Tissue-Specific Expression in the Rice Hd3a Promoter

  • Flowering time is determined by florigens. These genes include, Heading date 3a (Hd3a) and Rice FT 1 (RFT1) in rice, which are specifically expressed in the vascular tissues of leaves at the floral transition stage. To study the cis-regulatory elements present in the promoter region of Hd3a, we generated transgenic plants carrying the 1.75-kb promoter fragment of Hd3a that was fused to the ${\beta}$-glucuronidase (GUS) reporter gene. Plants expressing this construct conferred a vascular cell-specific expression pattern for the reporter gene. However, GUS was expressed in leaves at all developmental stages, including the early seedling stage when Hd3a was not detected. Furthermore, the reporter was expressed in roots at all stages. This suggests that the 1.75-kb region lackings cis-elements that regulate leaf-specific expression at the appropriate developmental stages. Deletion analyses of the promoter region indicated that regulatory elements determining vascular cell-specific expression are present in the 200-bp region between -245 bp and -45 bp from the transcription initiation site. By transforming the Hd3a-GUS construct to rice cultivar 'Taichung 65' which is defective in Ehd1, we observed that Ehd1 is the major regulatory element that controls Hd3a promoter activity.

Flowering Responses and Floral Characteristics of Ipomoea batatas Varieties (고구마 품종별 개화성 및 화기특성)

  • Korean sweetpotato, Ipomoea batatas, varieties were examined for their flowering response as a fundamental research to improve breeding efficiency by hybridization of this crop. Under 10.0~10.5 hours short day condition, self-rooted plants of three varieties such as Shinmi, Hongmi and Hwangmi flowered sparsely or moderately, and those of fourteen varieties including Eunmi flowered very sparsely. Under 10.6~12.0 hours day length condition, self-rooted plants of three varieties such as Shinmi, Hongmi and Hwangmi flowered very sparsely. Under the day length longer than 12 hours, none of the tested plants flowered. In all the sweetpotato plants grafted on morning glory, flowering occurred with varying degrees by varieties and day lengths. Under 10.0~10.5 hours day length, thirty two varieties including Hongmi flowered moderately or profusely, and one variety such as Mokgye 1 flowered sparsely. Under 10.6~12 hours day length, twenty three varieties including Sinchunmi flowered moderately or profusely, nine varieties including Jeungmi flowered sparsely, and Mokgye 1 didn't flower at all.

Foliar Application of Growth Bioregulators Influences Floral Traits, Cormassociated Traits and Chemical Constituents in Gladiolus grandiflorus L.

  • Gladiolus is one of the most popular cut flowers in the world floral market and is highly attractive to consumers. The production of poor-quality gladiolus spikes is one of the most important hurdles to obtaining high prices for this crop. The main objectives of this study were to improve the quantitative characteristics of gladiolus spikes, to increase propagule production and to enhance chemical constituent levels in leaves to help improve plant growth. We performed foliar application of bioregulators, including polyamines (putrescine and spermine) and vitamins (ascorbic acid and thiamine), at different concentrations (0, 0.1, 0.4, 0.7 and 1 mM) to gladiolus plants at the three leaf, five leaf and slipping stages. Application of 1 mM putrescine was the most effective treatment, with prominent effects on economically important spike traits including spike length (55.53 cm), spike diameter (7.53 mm), florets per spike (13.99), first floret diameter (9.66 cm) and first floret weight (4.90 g), followed by spermine treatment. The 1 mM putrescine treatment also had positive effects on corm-associated traits including corm diameter and corm weight, with values of 4.57 cm and 26.16 g compared to 3.47 cm and 17.16 g in control plants, respectively. The contents of chemical constituents in leaves, including total chlorophylls ($8.06mg{\cdot}g^{-1}$), total carotenoids ($1.66mg{\cdot}g^{-1}$), total soluble sugars ($4.75 mg{\cdot}g^{-1}$) and phenolics ($0.89mg{\cdot}g^{-1}$) increased in response to foliar application of 1 mM putrescine compared to the control. Ascorbic acid and thiamine also had positive effects on various plant morphological traits and chemical constituents in leaves compared to control plants, but the growth-promoting effects of polyamines were more pronounced. Hence, applying polyamines to gladiolus leaves will help improve spike production and increasing propagule (cormel) yields.

Response of Alisma plantago Varieties Cultivated After Early Maturing Rice Cropping to Fertilizer Levels

  • This study selects Sunwol local group, Gusang local group and Yongjun local group to examine the appropriate amount of applied fertilizer in cultivating the double crop seed of Alisma plantago by transplantation in the southern area, improve and complements the double cropping techniques and contribute to stable production. While the plant height is 45,47 and 49cm and the number of leaves is 11.7, 12.5 and 14.4 at non-fertilizing plot, the plant height is 67, 72 and 75cm and the number of leaves is 15.8, 17.2 and 19.3 at all fertilizing plot and their growth is active and in the character of plant height and the number of leaves, especially in the groove of N-P$_{2}$O$_{5}$-K$_{2}$O= 30-15-45kg/10a, the mean plant height of Sunwol is 75cm and mean number of its leaves is 19.3, the mean plant height and number of leaves in Gusang are 72cm and 17.2 respectively and those of Yongjun are 67cm and 15.8 respectively. While the number of floral axis is 1,2 in non-fertilizing plot, that of the whole varieties in all fertilizing plot is 3, 4, 5 and the mean floral axis of Sunwol is N-P$_{2}$O$_{5}$-K$_{2}$O = 10-5-15, 20-10-30, 30-15-45 kg/10a and 18-0-18 of complex fertilizer and 21-17-17kg/10a and it is small in all fertilizing plot. The yield of dry root per 10a is high in all fertilizing grooves and especially in the groove of N-P$_{2}$O$_{5}$-K$_{2}$O = 30-15-45 kg/10a, it is 372.6kg, in the groove of 18-0-18kg/10a using complex fertilizer, it is 389.1kg and in that of 21-17-17kg/10a, it is 376.7kg.6.7kg.
