  • The essential oil composition by means of gas liquid chromatography, and the occurrence and distribution of flavonoid glycosides in leaves, peels and barks of citrus plants native to Je Ju island were investigated. Results indicate that the occurrence of p-cymene, d,l-limonene, linalool, geraniol and linalyl acetate in the essential oils, and of hesperidin in leaves, peels and barks are fairly common to these species.

  • The effects of Citrus flavonoids, hesperetin, hesderidin, naringenin, and naringin, on nonenzymatic lipid peroxidation were studied in three different in vitro experimental models. Hesperetin showed the most antioxidant effect in this experimental condition by measuring the malondialdehyde production using the thiobarbiturate and thiocyanate methods, the lipid peroxidation of microsomal membranes, and DPPH (${\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl$) method. The antioxidative activity of the flavonoid aglycone forms, hesperetin, Citrus aglycone flavonoid, suggest the most antioxidant effect in this experimental condition, and this effect indicate more potent in the aglycones than their corresponding glycosides.

  • Activity-guided separation for the aerial parts of Rodgersia podophylla A. Gray gave a new acylated flavonoid, quercetin 3-O-${\alpha}$-L-(5$^2$-acetyl)-arabinofuranoside (1), together with six known flavonoids (2-7). Their hepatoprotective activities were determined by using the primary cultures of rat hepatocytes injured by H$_2$O$_2$. Quercetin 3-O-${\alpha}$-L-(3$^2$-acetyl)-arabinofuranoside (3), kaempferol 3-O-${\alpha}$-L-rhamnopyranoside (5) and quercetin 3-O-${\alpha}$-L-rhamnopyranoside (6) exhibited hepatoprotective activities comparable to silybin at the concentration of 50 mM (45.7, 50.8 and 57.3 %, respectively), and the new flavonoid 1 showed hepatoprotective activity at the concentration of 100mM (50.1%).

Phytochemical Screening and Biological Studies of Boerhavia Diffusa Linn

  • Hexane, ethyl acetate and methanol extracts of whole plant of Boerhavia diffusa were screened for phytochemical and biological activities. Qualitative phytochemical screening via colorimetric method and the quantitative estimation of phenolic and flavonoid content were performed. Antioxidant assay using DPPH scavenging method was studied. Antimicrobial screening of plant extracts was done by cup diffusion technique. Cytotoxic activity of B. diffusa was studied by brine shrimp bioassay and anthelminthic activity was evaluated in vitro in Pheretima posthuma. This study revealed B. diffusa as a source of various phyto-constituents such as alkaloids, glycosides, saponins, tannins, carbohydrates, cardiac glycosides, flavonoids and terpenoids. Quantitative estimation of total phenol was found to be maximum in BEE i.e. $29.73{\pm}0.88$, BME $19.8{\pm}2.02$ and in BHE $9.15{\pm}0.304mgGAE/g$. Similarly, the total flavonoid content was found to be $17.44{\pm}0.75$ in BEE, $14.43{\pm}0.23$ in BHE and 3.678 mg QE/g in BME. Ethyl acetate extract showed its antibacterial activity against all tested pathogens except Escherichia coli whereas Staphylococcus aureus and Salmonella Typhi were resistant to methanol and hexane extract. The zone of inhibition (ZOI) of ethyl acetate extract against S. Typhi and B. cereus was found to be 18 mm and 14 mm respectively. The MIC value of BEE in S. Typhi was $3.125{\mu}g/ml$ and in B. cereus was $12.5{\mu}g/ml$. The preliminary screening of anticancer property of B. diffusa i.e. BSLT in methanol was found to be $165.19{\mu}g/ml$. B. diffusa was also found to contain anthelmintic property. The study helped in further exploration of medicinal properties of B. diffusa by phytochemical screening and biological activities paving the path for study and investigation in this plant.

  • Juglans mandshurica barks were collected, extracted with acetone-$H_2O$ (7:3, v/v), fractionated with n -hexane, $CH_2Cl_2$, and EtOAc, and freeze dried to give some dark brown powder. The EtOA cand $H_2O$ soluble fractions were chromatographe d on a Sephadex LH-20 column using $H_2O$-MeOH and EtOH-hexane mixture as eluents. Spectrometric analysis such as NMR and MS, including TLC,were performed to characterize the structures of the isolated compounds. From the EtOAc and $H_2O$ soluble fractions, three flavanols (1~3), three flavonols (4~6) and five flavonol glycosides (7~11) were isolated and elucidated.

Active Compounds from The Aerial Parts of Polygonum multiflorum Thunberg (하수오 지상부의 활성 성분)

  • Polygoni Radix (Polygonaceae) has been used as renal disoder hematopoietic, menstrual irregularity and antiaging in Korean folk medicine. Specially aerial parts has been used for insomnia and sedative agent. In order to investigate the efficacy of antioxidant activity on these aerial parts, the bio-activity guided fraction and isolation of physiologically active substance were performed. H$_2$O, 30%, 60%, 100% MeOH were examined antioxidant activity by DPPH method. It was revealed that 30%, 60% MeOH fractions have significantly antioxidant activity. From 60% MeOH fraction, six known flavonoid glycosides (quercetin, quercetin 3-O-${\beta}$-D-galactopyranosyl-4'-O-glucopyrano side-3'-methyl ether, quercetin 3-O-${\alpha}$-L-rhannopyranoside, quercetin 3-O- ${\alpha}$-L-rhamnopyranoside-4'-methylethor, quorcetin 3-O-${\beta}$-D-galactopyranoside, quercetin 3-O-${\beta}$-D-galactopyranoside) and 3,4-Oihydrocinnarnic acid were isolated. To investigate the antioxidant activities of each com-pounds, we measured radical scavening activity with DPPH method and anti-lipid peroxidative efficacy on low density lipo-protein (LDL) with TBARS assay and anti-acetylcholinestrase activity with Ellman's Test. Four compounds (ll, IV Vl, Vll) of quercetin glycosides showed significant activity.

Flvonoids and Their Glycosides from the Bark of Salix rorida (분버들(Salix rorida) 수피의 후라보노이드 및 배당체 화합물)

  • The air-dried bark of Salix rorida was extracted with acetone-water(7:3, v/v) and its extractives were concentrated with a vacuum evaporator. The extractives were fractionated with a series of n-hexane, chloroform, ethylacetate(EtOAc) and water on a separatory funnel. Each fraction was freeze-dried to give some dark brown powder. The EtOAc and water soluble fractions were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. The isolated compounds were tested with a cellulose TLC developed with TBA and 6% acetic acid and then visualized on UV lamp or sprayed with vanillin-HCl-EtOH. The purified compounds were flavonoids and their glycosides as follows:(+)-catechin, naringenin, salipurposide, aromadendrin, isosalipurposide, aromadendrin-7-O-𝛽-D-glucopy- ranoside and taxifolin-7-O-𝛽-D-glucopyranoside. The structures of each compounds were confirmed by 1H-NMR, 13C-NMR and mass spectra.

Preparation and Quality Properties of Extruded Snack using Onion Pomace and Onion (양파착즙박과 양파를 이용한 압출스낵의 제조 및 품질특성)

  • To use onion pomace produced from concentrated onion juice processing, dried onion pomace or dried onions were mixed with com grits at levels of 10, 20, and 30% and were extruded in a twin-screw extruder. Quality properties of extruded onion snack were investigated. Low-lignin dietary fiber of dried onion pomace was approximately 2 times higher than that of dried onion. Browning increased with increasing onion content. Reducing sugar and free sugar in onion snack showed high content by increasing onion concentration and the majority of free sugar consisted of glucose and fructose. Flavonoid in control snack was not detected and the flavonoid content containing quercetin and quercetin glycosides was about 50% less in onion pomace snack than in onion snack. Sensory evaluation of onion snack containing 10% concentration of dried onion pomace or dried onion indicated the products were acceptable in overall eating-quality.

Phenolic Compounds from Antioxidant Plant Materials and their Protective Effect on PC12 cells (항산화 식물의 페놀성화합물에 의한 PC12 세포보호 효과연구)

  • This study was designed to evaluate the antioxidant activities and protective effects on PC12 cells of the extract of Epimedium koreanum and its main constituents icariin and icariside I. After screening the seven identified flavonoid glycosides from E. koreanum through DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay, E. koreanum, Icariin and Icariside I exhibited significant effect on radical scavenging activity. E. koreanum, icariin and icariside I were examined using DPPH, ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) and FRAP (ferric reducing ability power) assay. In all antioxidant assays, E. koreanum, icariin and icariside I showed high radical scavenging activities in a dose-dependent manner. Protective effects against $H_2O_2-induced$ PC12 cells were assessed with MTT assay. The results indicated that cell viability and protection on PC12 cells of icariside I and icariin increased dose dependently. These study results suggest that E. koreanum, icariin and icariside showed high antioxidant capacities and cell protective effects. Icariside I, one of the metabolites of icariin, may be a new and effective flavonoid compound as a functional component.

Screening for Antioxidant Activity of Plants and Marine Algae and Its Active Principles from Prunus davidiana

  • The antioxidant activity of methanol extracts of plants and marine algae was tested by using 1,1-diphenyl-2-picrylhydrazyl(DPPH). Five plant extracts(Prunus davidiana, Eriobotrya japonica, Artemisia iwayomogi, Stirodella tolyrrhiza and Ulmus davidiana) and two algae (Ecklonia stolonifera and Symphycoladia latiuscula) were found to be the most effective in DPPH radical scavenging activity. The methanol extract obtained from the stems of Prunus davidiana was fractionated with several solvents. The ethylacetate soluble fraction exhibiting the strongest antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Antioxidant flavonoids and flavonoid glycosides were isolated and the most active ones was identified as (+)-catechin by MS, $^1H-NMR$ and $^{13}C-NMR$. Its antioxidant activity was higher than that oil vitamin C.
