Fungi are wood-decaying organisms, and this is an important trait that should be considered in wood utilization. When fungi attack wood, it decreases the quality of the wood. The use of metal screws has become an important part of woodworking. The ability of fungi to decay wood and damage metal screws that are embedded into wood is varied. In this study, eight fungal species were evaluated with respect to their ability to decay Parashorea smythiesii and P. tomentella wood. In addition, the effect of fungi on corroding metal screws was determined using the Kolle flask method. The evaluation showed that the fungal species Schizophyllum commune, Pycnoporus sanguineus, and Polyporus arcularius were highly capable of decaying Parashorea spp. woods. The greatest wood weight loss occurred with the heartwood of P. tomentella exposed to S. commune. Based on the classification of wood resistance against fungal attack, the two Parashorea spp. were classified as moderately resistant woods (class III). Schizophyllum commune was classified as highly capable of decaying wood that was embedded with metal screws and was highly capable of corroding metal screws placed in fungi-culture media. The greatest weight of rust powder formed because of screw corrosion was obtained from screw-embedded wood exposed to S. commune. Additionally, the most severe corrosion of metal screws that were embedded into woods was caused by the activities of P. arcularius. Moreover, the average weight loss of screw-embedded wood was greater than that of unscrewed wood.
Isoprene has the potential to replace some petroleum-based chemicals and can be produced through biological systems using renewable carbon sources. Ralstonia eutropha can produce value-added compounds, including intracellular polyhydroxyalkanoate (PHA) through fatty acid and lipid metabolism. In the present study, we engineered strains of R. eutropha H16 and examined the strains for isoprene production. We optimized codons of all the genes involved in isoprene synthesis by the mevalonate pathway and manipulated the promoter regions using pLac and pJ5 elements. Our results showed that isoprene productivity was higher using the J5 promoter ($1.9{\pm}0.24{\mu}g/l$) than when using the lac promoter ($1.5{\pm}0.2{\mu}g/l$). Additionally, the use of three J5 promoters was more efficient ($3.8{\pm}0.18{\mu}g/l$) for isoprene production than a one-promoter system, and could be scaled up to a 5-L batch-cultivation from a T-flask culture. Although the isoprene yield obtained in our study was insufficient to meet industrial demands, our study, for the first time, shows that R. eutropha can be modified for efficient isoprene production and lays the foundation for further optimization of the fermentation process.
The rising demand for carotenoids can be met by microbial biosynthesis as a promising alternative to chemical synthesis and plant extraction. Several species of lactic acid bacteria (LAB) specifically produce C30 carotenoids and offer the added probiotic benefit of improved gut health and protection against chronic conditions. In this study, the recently characterized Lactiplantibacillus plantarum subsp. plantarum KCCP11226T produced the rare C30 carotenoid, 4,4'-diaponeurosporene, and its yield was optimized for industrial production. The one-factor-at-a-time (OFAT) method was used to screen carbon and nitrogen sources, while the abiotic stresses of temperature, pH, and salinity, were evaluated for their effects on 4,4'-diaponeurosporene production. Lactose and beef extract were ideal for optimal carotenoid production at 25℃ incubation in pH 7.0 medium with no salt. The main factors influencing 4,4'-diaponeurosporene yields, namely lactose level, beef extract concentration and initial pH, were enhanced using the Box-Behnken design under response surface methodology (RSM). Compared to commercial MRS medium, there was a 3.3-fold increase in carotenoid production in the optimized conditions of 15% lactose, 8.3% beef extract and initial pH of 6.9, producing a 4,4'-diaponeurosporene concentration of 0.033 A470/ml. To substantiate upscaling for industrial application, the optimal aeration rate in a 5 L fermentor was 0.3 vvm. This resulted in a further 3.8-fold increase in 4,4'-diaponeurosporene production, with a concentration of 0.042 A470/ml, compared to the flask-scale cultivation in commercial MRS medium. The present work confirms the optimization and scale-up feasibility of enhanced 4,4'-diaponeurosporene production by L. plantarum subsp. plantarum KCCP11226T.
In order to investigate the effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot, embryogenic callus were cultured in the medium supplemented with various concentrations of sucroseor glucose, and with combination of 2% sucrose and various concentrations of mannitol or sorbitol. The maximum number of somatic embryos formed per flask (1,555.70) was obtained in the medium supplemented with 2% sucrose rather than glucose alone or a combination of mannitol or sorbitol and 2% sucrose, and the number of somatic embryos was decreased with the increasing of mannitol or sorbitol concentration. The frequencies of somatic embryos with two cotyledons were 35.14% for sucrose, 19.88% for glucose, 32.55% for mannitol + 2% sucrose, and 38.59% for sorbitol + 2% sucrose, respectively, and the frequencies of abnormal somatic embryos having 3 or more cotyledons were 64.86% for sucrose, 80.12% for glucose, 67.44% for mannitol + 2% sucrose, and 61.41% for sorbitol + 2% sucrose, respectively. Particularly, the frequency of somatic embryos with two cotyledons (59.16%) was the highest in the 2% sucrose medium compared to the frequency of abnormal somatic embryogenesis with three or more cotyledons, and the frequency gradually decreased with increasing concentration of glucose, mannitol or sorbitol. According to these results, it was found that the ratio of abnormal somatic embryo was higher than the normal somatic embryo in carrot, and was shown that somatic embryogenesis and the cotyledon number was affected by the concentrations of sucrose, glucose as carbon source, and mannitol and sorbitol as osmotic agents in culture medium.
본 연구에서는 프로바이오틱 생균인 비스루트균의 최적 배지 탐색을 위해 탄소원, 질소원 그리고 아포형성의 극대화를 위한 인산염의 최적 농도를 검토하였다. 500 mL baffle flask 에서 배양을 실시한 결과, 탄소원 영향 실험에서는 포도당 2%(w/v)와 전분 2%(w/v)가 첨가되었을 때 최대 총균수$(4.7{\times}10^9\;CFU/mL)$와 활성아포수$(4.3{\times}10^9\;CFU/mL)$를 보였으며 아포형성율은 91%이었다. 질소원 영향 실험에서는 대두분이 5%(w/v) 첨가되었을 때 최대 총균수$(5.9{\times}10^9\;CFU/mL)$와 활성아포수$(5.7{\times}10^9\;CFU/mL)$를 나타내었다. 그리고, 인산염 실험에서는 최대 총균수$(6.3{\times}10^9\;CFU/mL)$와 활성아포수$(6.0{\times}10^9\;CFU/mL)$를 $KH_2PO_4$를 1%(w/v) 첨가되었을 때 얻을 수 있었으며 아포형성율은 95%를 보였다. 5L 발효조에서 최적 배지인 KH5 배지를 이용하여 $32^{\circ}C$의 온도에서 통기량 1 vvm, 교반속도 450 rpm, pH $7.0{\pm}0.1$로 맞추어 회분식 배양을 실시한 결과 최대 총균수는 $3.3{\times}10^{10}\;CFU/mL$을 보였으며 최대 활성아포수는 $3.0{\times}10^{10}\;CFU/mL$을 나타내었고, 아포형성율은 94%를 나타내었다. 이때 아포 생산성은 $5{\times}10^8\;spores/mL/h$이었다.
본 연구는 다양한 조건에서의 실험을 위한 C. minutissima의 500 mL flask의 종 배양을 통해 고농도의 균주 획득 후, 가장 효율적인 배양 방식을 보인 fed-batch를 통한 autotrophic, heterotrophic, mixotrophic 조건하에서 바이오디젤용 지질 생산의 최적 배양 조건을 찾아야했다. 먼저, fed-batch의 Heterotrophic 배양 시 유기 탄소 원으로 glucose의 가장 효율적인 안정한 초기농도가 10 g/L임을 확인했다. 이 후, 20 L 배양기를 이용한 batch, fed-batch의 heterotrophic 배양에서 fed-batch배양의 경우 지질 생성양이 1.62배 정도 더 높았으며, 최대 균체량 역시 4 (g-dry wt./L)정도 높으며, 또한 디젤용 지질로 적합한 $C_{18}$의 조성 역시 49%이상 차지하여, 옥외 대량 배양 시 유기탄소 원으로 적절한 glucose의 이용을 통한 효율적인 배양방법임을 알 수 있다. 이 결과를 바탕으로, glucose를 포함한 배지를 0.5 L/day로 공급하여 유가 식 배양을 각 배양 조건에 따라 비교 실시했다. 그 결과 mixotrophic의 경우 다른 배양 조건에 비해 각각 24 (g-dry wt./L)의 최대 군체 농도와 함께 43 (%, w/w)의 높은 지질 생성량을 보였다. 또한 생성되는 지질의 조성이 균체 당 바이오 디젤용 양질의 지질 생성이 가능하였다. 또한 fed-batch 배양 이후 0.047 (% lipid/g-dry wt./day)의 비 생산 속도를 유지하며, 일정 균체 농도를 유지하는 경우 지속적인 지질 생성이 가능함을 보여주었다. 이러한 세 가지 배양 조건하에서 지질 추출을 통한 지방산 조성을 살펴본 결과 mixotrophic 배양 하에 디젤용으로 잘 알려진 $C_{17}$, $C_{18}$의 지방산 조성은 각각 18%와 49%의 생성을 보여주었다. 이 결과들을 바탕으로 이 균주를 이용해 옥외 배양이나 배양 조에서 대량 배양이 가능함을 확인했으며, 특히, 옥외배양을 위한 혼합영양 배양조건하에 배양 공정의 scale-up에 대한 추가적인 연구를 통해 C. minutissima 미세조류로부터 바이오 디젤의 보다 경제적 생산이 가능하도록 하고자 한다.
Botrytis cinerea에 의한 들깨 잿빛곰팡이병의 생물학적 방제를 위해 선발한 Bacillus lirheniformis Nl 글주를 제제화하여 미생물농약을 제조하고 이의 방제 효과를 검정하였다. Nl균주의 길항력에 미치는 탄소원과 질소원의 영향을 실험한 결과 탄소원으로는 glucose, 질소원으로는 tryptone이 가장 높은 효과를 보였다. 또한 Nl 균주를 이들 glucose와 tryptone을 첨가한 NB배지에 대량배양하고 그 배양액에 콩가루, 쌀가루, glucose, FeSo$_4$~7$H_2O$ 및 MnCl$_2$. 4$H_2O$ 을 첨가하여 수화형 미생물농약 Soy제제로제조하고,이 병원균에 대한 방제효과를 하우스내에서 폿트 검정한 결과 방제가가 93.1%로서 유의성은 없으나 베노밀 수화제에 의한 86.1%보다 높았다.
We use two different methods for laboratory propagation from seed of lady's slipper orchid(Cypripedium macranthos Sw.); immature seed which also called green capsule or fully mature seed about 120~130 days from pollination. In green capsule culture, the seed pods should be collected within precisely right time. The right time of seed collection could be diverse under the wether conditions or nutritional factors of the plants. In fully matured seed culture, the more complicated procedures are needed to break the dormancy of the seed; thermal or chemical treatment. The seedlings in this study were easily germinated from immature seeds in Harvais medium; 53 days after pollination(DAP) in Cypripedium pubescens, DAP 65 in C. parviflorum and C. macranthos. The germinated seedlings were transplanted to hormone free media immediately to avoid abnormal growth of seedlings. When the seedlings have roots with a minimum length of around 2-3cm and have visible dormant buds, the seedlings were removed from the flask and stored in refrigerator for vernalization. To examine the correlation of seedlings and maternal plants, the 125 seedlings of C. macranthos were transplanted in the soil bed at a distance of 20-100 cm from mother plants on April 20. The survival rate of seedlings were 92% in 20 cm distance from the ripe plants, and 56 % in 100 cm distance. The seedlings which were transplanted near mother plants showed vigorous growth in plant height, leaf width, and especially dormant buds. Considering the existence of mycorrhiza which is a symbiotic association between a fungus and the roots of a orchid vascular, the various fungus from mother plants could affect the growth of the seedlings. These results indicate the possibility of high and stable production and practical industrialization of endangered lady's slipper orchids.
참깨의 뿌리를 바이오촉매로 이용한 뿌리 배양을 통하여 HMF를 생산하기 위하여 뿌리의 생장과 HMF의 합성에 적합한 뿌리배양 조건을 탐색하였으며, 이를 위하여 NAA(naphthalene acetic acid), silver nitrate의 적정 농도조건 및 적정 배양온도 조건에 대한 실험이 수행되었다. 뿌리의 생장이 가장 높은 처리구는 $30^{\circ}C$에서 NAA의 농도가 1.0 mg/l로 첨가된 처리구에서 측정되었으며, 평균적으로 볼 때 두 배양 온도($25^{\circ}C$와 $30^{\circ}C$)간에는 뿌리의 생장차이는 측정되지 않았다. NAA가 첨가되지 않은 처리에서는 두 배양온도 모두에서 참깨의 뿌리생장이 전혀 생기지 않은 반면에, NAA가 첨가된 배양에서는 두 배양온도에서 뿌리의 생장이 확인되었다. 그리고 0.5 mg/l의 질산은이 첨가된 처리구에서 참깨의 뿌리생장이 가장 높게 측정되었으며, $25^{\circ}C$에서 배양된 처리구 보다는 $30^{\circ}C$에서 배양된 뿌리의 생장이 약간 높게 나타났고, 질산은의 농도가 0.5 mg/l 이상의 높은 농도에서는 뿌리의 생장이 오히려 감소되는 경향이 있었다. 그리고 HMF의 합성의 경우 가장 높게 측정된 처리구는 배양온도 $25^{\circ}C$에서 NAA 0.5 mg/l이 첨가된 처리구에서 측정된 반면에 NAA 무처리구에서는 HMF 합성이 측정되지 않았다. 질산은이 첨가된 처리구에서는 HMF의 합성이 오히려 억제되는 경향을 보여준 반면에 질산은의 무처리구에서 HMF의 합성이 가장 높은 결과가 나타났다. 질산은의 농도가 0.5, 1.0 및 1,5 mg/l로 처리된 구에서는 질산은 무처리구에 비해서 약 8-10% 정도의 HMF 합성이 감소되는 경향이 나타났으며, 두 배양 온도($25^{\circ}C$와 $30^{\circ}C$ 배양) 간에도 HMF의 합성에는 차이가 보이지 않았다.
초임계수 처리를 통하여 얻어진 목질계 바이오매스 가수분해 물을 이용한 바이오 에탄올 생산에 대하여 연구하였다. 초임계수 처리 가수분해물은 바이오에탄올 생산을 위한 배지의 탄소원으로 사용되었다. 농축된 초임계수 처리 가수분해물 (SCW3)을 사용하여 효모를 배양하였을 때, 다른 두 가지 초임계수 처리 가수분해물 (SCW1, SCW2) 을 사용한 경우에 비하여 효모의 성장속도가 늦었다. 그리고 모든 경우에 0.51에서 0.56 (%, w/v)의 바이오 에탄올이 생산되었다. 그래서 농축된 초임계수 처리 가수분해물 (SCW3)을 활성탄과 수산화 칼슘으로 전처리하여 페놀류 독성물질을 제거하였다. 활성탄 전처리가 보다 효과적으로 94.6%의 페놀류 화합물을 제거하였고, 바이오 에탄올도 0.96 (%, w/v) 생산 할 수 있게 하였고, 환원당을 기준으로 한 바이오에탄올 수율도 0.5에 이르렀다.
