Investigation on Inhibitory Effect of ErmSF N-Terminal End Region Peptide on ErmSF Methyltansferase Activity In Vivo Through Development of Co-Expression System of Two Different Proteins in One Cell (서로 다른 두 단백질의 세포 내 동시 발현 체계의 개발을 통한 ErmSF에서 특이적으로 발견되는 N-Terminal End Region (NTER)을 포함하는 펩타이드의 생체내에서의 ErmSF 활성 억제 효과 검색)

  • Most problematic antibiotic resistance mechanism for MLS (macrolide-lincosamide-streptogramn B) antibiotics encountered in clinical practice is mono- or dimethylation of specific adenine residue at 2058 (E. coli coordinate) of 23S rRNA which is performed by Erm (erythromycin ribosome resistance) protein through which bacterial ribosomes reduce the affinity to the antibiotics and become resistant to them. ErmSF is one of the four gene products produced by Streptomyces fradiae to be resistant to its own antibiotic, tylosin. Unlike other Erm proteins, ErmSF harbors idiosyncratic long N-terminal end region (NTER) 25% of which is comprised of arginine well known to interact with RNA. Furthermore, NTER was found to be important because when it was truncated, most of the enzyme activity was lost. Based on these facts, capability of NTER peptide to inhibit the enzymatic activity of ErmSF was sought. For this, expression system for two different proteins to be expressed in one cell was developed. In this system, two plasmids, pET23b and pACYC184 have unique replication origins to be compatible with each other in a cell. And expression system harboring promoter, ribosome binding site and transcription termination signal is identical but disparate amount of protein could be expressed according to the copy number of each vector, 15 for pACYC and 40 for pET23b. Expression of NTER peptide in pET23b together with ErmSF in pACYC 184 in E. coli successfully gave more amounts of NTER than ErmSF but no inhibitory effects were observed suggesting that there should be dynamicity in interaction between ErmSF and rRNA rather than simple and fixed binding to each other in methylation of 23S rRNA by ErmSF.

  • The productionof the highly relible softwae systems and theirs performance evaluation hae become important interests in the software industry. The software evaluation has been mainly carried out in ternns of both reliability and performance of software system. Software reliability is the probability that no software error occurs for a fixed time interval during software testing phase. These theoretical software reliability models are sometimes unsuitable for the practical testing phase in which a software error at a certain testing stage occurs by causes of the imperfect debugging, abnornal software correction, and so on. Such a certatin software testing stage needs to be considered as an outlying stage. And we can assume that the software reliability does not improve by means of muisance factor in this outlying testing stage. In this paper, we discuss Bavesian software reliability growth modeling and estimation procedure in the presence of an imidentitied outlying software testing stage by the modification of Jehnski Moranda. Also we derive the Bayes estimaters of the software reliability panmeters by the assumption of prior information under the squared error los function. In addition, we evaluate the proposed software reliability growth model with an unidentified outlying stage in an exchangeable model according to the values of nuisance paramether using the accuracy, bias, trend, noise metries as the quantilative evaluation criteria through the compater simulation.

Considerations of Environmental Factors Affecting the Detection of Underwater Acoustic Signals in the Continental Regions of the East Coast Sea of Korea

  • This study considers the environmental factors affecting propagation loss and sonar performance in the continental regions of the East Coast Sea of Korea. Water mass distributions appear to change dramatically in a few weeks. Simple calculation with the case when the NKCW (North Korean Cold Water) develops shows that the difference in propagation loss may reach in the worst up to 10dB over range 5km. Another factor, an eddy, has typical dimensions of 100-200km in diameter and 150-200m in thickness. Employing a typical eddy and assuming frequency to be 100Hz, its effects on propagation loss appear to make lower the normal formation of convergence zones with which sonars are possible to detect long-range targets. The change of convergence zones may result in 10dB difference in received signals in a given depth. Thermal fronts also appear to be critical restrictions to operating sonars in shallow waters. Assuming frequency to be 200Hz, thermal fronts can make 10dB difference in propagation loss between with and without them over range 20km. An observation made in one site in the East Coast Sea of Korea reveals that internal waves may appear in near-inertial period and their spectra may exist in periods 2-17min. A simulation employing simple internal wave packets gives that they break convergence zones on the bottom, causing the performance degradation of FOM as much as 4dB in frequency 1kHz. An acoustic experiment, using fixed source and receiver at the same site, shows that the received signals fluctuate tremendously with time reaching up to 6.5dB in frequencies 1kHz or less. Ambient noises give negative effects directly on sonar performance. Measurements at some sites in the East Coast Sea of Korea suggest that the noise levels greatly fluctuate with time, for example noon and early morning, mainly due to ship traffics. The average difference in a day may reach 10dB in frequency 200Hz. Another experiment using an array of hydrophones gives that the spectrum levels of ambient noises are highly directional, their difference being as large as 10dB with vertical or horizontal angles. This fact strongly implies that we should obtain in-situ information of noise levels to estimate reasonable sonar performance. As one of non-stationary noise sources, an eel may give serious problems to sonar operation on or under the sea bottoms. Observed eel noises in a pier of water depth 14m appear to have duration time of about 0.4 seconds and frequency ranges of 0.2-2.8kHz. The 'song'of an eel increases ambient noise levels to average 2.16dB in the frequencies concerned, being large enough to degrade detection performance of the sonars on or below sediments. An experiment using hydrophones in water and sediment gives that sensitivity drops of 3-4dB are expected for the hydrophones laid in sediment at frequencies of 0.5-1.5kHz. The SNR difference between in water and in sediment, however, shows large fluctuations rather than stable patterns with the source-receiver ranges.

Korea's Optimal Basket Exchange Rate : Thoughts on the Proper Operation of the Market Average Rate Regime (우리나라의 적정(適正)바스켓환율(換率) : 시장평균환율제도(市場平均換率制度)의 운용기준(運用基準) 모색(模索))

  • For the last several years, considerable criticism has been leveled against Korea's exchange rate management. While Korea was designated a currency manipulator by the U.S., domestically it is often complained that the won/dollar rate did not adequately reflect changes in Korea's export competitiveness and fluctuations in the exchange rates of major currencies. In view of this situation, Korea changed its exchange regime at the beginning of March this year from the dual currency basket system to a more flexible one, called a "market average rate regime". Under this new regime, the won rate is determined in the exchange market based upon the supply of and demand for foreign exchange and is allowed to freely fluctuate each day within a + 0.4 % range. This paper, first, seeks to evaluate Korea's exchange rate management under the dual basket regime of the 1980s, and then to construct an optimal currency basket for the won which could provide a proper indicator for exchange market intervention under the new market average rate regime. The analysis of fluctuations in the real effective exchange rate (REER) of the won indicates that the won rates in the 1980s failed not only to offset changes in relative prices between home and trading partner countries, but also to properly respond to variations in major exchange rates as further evidenced by sizable fluctuations in the nominal effective rates of the won. In other words, the currency basket regime which was adopted in 1980 for the stabilization of the REER of the won has not been operated properly, mainly because authorities often resorted to policy considerations in determining the won's rate. In the second part of the paper, an optimal currency basket for Korea is constructed, designed to minimize the fluctuations in the REER of the won without including policy considerations as a factor. It is recognized, however, that both domestic and foreign price data are not available immediately for the calculation of the REER. For this problem, the approach suggested by Lipschitz (1980) is followed, in which optimal weights for currencies in the basket are determined based upon the past correlation between price and exchange rates. When the optimal basket is applied to Korea since the mid-80s, it is found that the REER of the won could have been much more stable than it actually was. We also argue for the use of variable weights rather than fixed ones, which would be determined by the changing relationship between exchange rates and relative prices. The optimal basket, and the optimal basket exchange rate based on that basket, could provide an important medium- or long-term reference for proper exchange market intervention under the market average rate regime, together with other factors, such as developments in the current account balance and changes in productivity.

  • Recently, interest in radiation protection is increasing because of the occurrence of accidents related to exposure dose. So, the nuclear safety act provides to install the shields to avoid exceeding the dose limit. In particular, when the worker conducts the non-destructive testing (NDT) without the fixed shielding structure, we should monitor the access to the workplace based on a constant dose rate. However, when we apply for permits for NDT work in these work environments, the consideration factors to the estimation of the distance and exposure dose are not legally specified. Therefore, we developed the excel model that automatically calculates the distance, exposure dose, and cost if we input the factors. We applied the assumption data to this model. As a result of the application, the distance change rate was low when the thickness of the lead blanket and collimator is above 25 mm, 21.5 mm, respectively. However, we didn't consider the scattering and build-up factor. And, we assumed the shape of the lead blanket and collimator. Therefore, if we make up for these limitations and use the actual data, we expect that we can build a database on the distance and exposure dose.

Semi-daily Variations in Populations of the Dinoflagellates Dinophysis acuminata and Oxyphysis oxytoxoides and a Mixotrophic Ciliate Prey Mesodinium rubrum in Masan Bay (마산만에서 와편모류 Dinophysis acuminata 및 Oxyphysis oxytoxoides와 먹이생물 섬모류인 Mesodinium rubrum의 단주기적 개체군 변동)

  • Recent laboratory studies have documented that mixotrophic dinoflagellates Dinophysis spp. and heterotrophic dinoflagellate Oxyphysis oxytoxoides share a common prey, i.e. the mixotrophic ciliate Mesodinium rubrum. Nonetheless, very little is known about the population dynamics and species interactions among these protists in natural environments. To investigate the interactions between the dinoflagellate predators and their ciliate prey in the field, we took the samples twice a day from 26 July to 28 August, 2011 at a fixed station in Masan Bay and analyzed their abundances. During this study, salinity was highly variable, ranging from 5 to 28, due to the periodic input of rainfalls to the sampling station. Water temperature was on average $26.5^{\circ}C$ until 20 August and thereafter was about $21^{\circ}C$ by the end of the sampling period. The ciliate M. rubrum occurred persistently throughout the sampling period, ranging from 13 to $492\;cells\;mL^{-1}$. Cell densities of D. acuminata and O. oxytoxoides ranged from undetectable level to $19,833\;cells\;L^{-1}$ and from undetectable level to $100,333\;cells\;L^{-1}$, respectively. The high abundance of D. acuminata mostly followed the blooming of the ciliate M. rubrum, but it often did not peak even during heavy blooms of the prey, probably due to sensitivity to large salinity fluctuation and also presumably overlapped grazing by other mixotrophic dinoflagellates. The abundance of O. oxytoxoides was detected only when water temperature was lower than $24^{\circ}C$, indicating that water temperature is an important environmental factor to control the population dynamics of the dinoflagellate species.

The Effects of Sight Alignment Method on Pistol Shooting Records of Security Guard (경호요원 권총사격시 조준선정렬방식과 기록변화와의 관계)

  • Some of individuals working at various fields of security guard are given the legal right to use a gun to cope with expected risks and crimes. Not only in Korea but also throughout the world, a pistol is being used as a device for the workers of security guard such as the army, the police and intelligence agencies to keep the law and social order. A pistol, which is regarded as the last safety measure of security guard, is easy to carry but, because of its short barrel for high portability, its hit ratio and long-range shooting performance are much lower than ordinary guns. Despite such weak points, a pistol can be a great tool because of its high portability and if the users make steady efforts to improve their skills and, for this reason, it has been being used widely until now. National agency workers' pistol shooting skills depend on several important factors, but for the fast and accurate shooting, the mot important factor is how to aim at the target. Because it takes a relatively longer time to align the sight on the target, the change of the sight alignment method is expected to improve pistol shooting skills a lot. In Korea, however, most of national agency training centers teach the basic aiming method that crosses the foresight and the backsight and aligns the crossing to the target. That is, they teach the sight alignment method for fixed targets. Some agencies in Korea and foreign countries that have used guns more frequently and longer than us are teaching different sight alignment methods as well. Representative ones of them are aiming only with the foresight and pointing shooting. These aiming methods are expected to produce good results against moving targets. While the basic sight alignment method has to cross the foresight and the backsight and then to align the crossing to the target, so takes a longer time than other aiming methods, the other two aiming methods are expected to be effective in fields where both swiftness and accuracy are require. Because domestic agencies are teaching mainly the basic sight alignment method, the present study purposed to examine the effects of different aiming methods on shooting records and ultimately to contribute to national agency workers' operation. For this purpose, we selected 20 policemen from local police commandos under the National Police Agency who were accustomed to pistol shooting and measured their pistol shooting. According to the results, the pointed shooting method showed the best record in 5m short.range shooting. As to hit ratio in 15m shooting, which is relatively long.range for pistol shooting, the basic aiming method showed the best record, but the results of statistical analysis showed that pointed shooting is most superior. Of course, the results were obtained from a group who were used to and skillful in pistol shooting, but they are considered meaningful in that all the police and national agency workers should have such high shooting skills.

The influence of Collar design on peri-implant marginal bone tissue (Collar design이 임플랜트 주위 변연골 흡수에 미치는 영향)

  • Statement of problem: Peri-implant marginal bone loss is an important factor that affects the success of implants in esthetics and function. Various efforts have been made to reduce this bone loss by improving implant design and surface texture. Previous studies have shown that early marginal bone loss is affected by implant neck designs. Purpose: The purpose of this study was to examine the influence of laser microtexturing of implant collar on peri-implant marginal bone loss. Materials and methods: Radiographical marginal bone loss was examined in patients treated with implant-supported fixed partial dentures. Marginal bone level was examined with 101 implant fixtures installed in 53 patients at three periods(at the time of implantation, prosthetic treatment and 6-month after loading). Four types of implants were examined. The differences of bone loss between implants(ITI standard) with enough biologic width and implants(ITI esthetic plus, Silhouette IC, Silhouette IC Laser-$Lok^{TM}$) with insufficient biologic width have been compared. Resorption angles were examined at the time of prosthetic delivery and 6-month after loading. Results and Conclusion: Within the limitation of this study, the following results were drawn. 1. The marginal bone loss of ITI standard and Silhouette IC Laser-$Lok^{TM}$ was less than that of ITI esthetic plus and Silhouette IC(P<0.05). The marginal bone loss between ITI standard and Silhouette IC Laser-$Lok^{TM}$ had no significant statistical difference(P>0.05). There was no significant statistical difference between marginal bone loss of ITI esthetic plus and Silhouette IC(P>0.05). 2. There was no significant difference in marginal bone loss between maxilla and mandible(P>0.05). 3. There was no significant difference in resorption angle among four types of implants(P>0.05). The marginal bone of implants with supracrestal collar design of less than that of biologic width had resorbed more than those with sufficient collar length. The roughness and laser microtexturing of implant neck seem to affect these results. If an implant with collar length of biologic width, exposure of fixture is a possible complication especially in the anterior regions of dentition that demand high esthetics. Short smooth neck implant are often recommended in these areas which may lack the distance between microgap and the marginal bone level. In these cases, the preservation of marginal bone must be put into consideration. From the result of this study, it may be concluded that laser microtexturing of implant neck is helpful in the preservation of marginal bone.

FEA estimates of margin design in all ceramic crowns (완전 도재관을 위한 지대치 형성시 변연 형태에 따른 응력 분포의 유한요소법적 비교)

  • Statement of problem: Over the past decade, increased demand for esthetically pleasing restorations has led to the development of all-ceramic systems. Recent reports suggest that the all-ceramic crowns have excellent physical properties, wear resistance, and color stability. In addition, numerous ceramics have excellent biocompatibility, a natural appearance, and improved physical bonding with resin composite luting agents. However, the brittle nature of ceramics has been a major factor in their restriction for universal usage. Functional occlusal loading can generate stress in the luting agent, and the stress distribution may be affected by the marginal geometry at the finish line. Tooth preparation for fixed prosthodontics requires a decision regarding the marginal configuration. The design dictates the shape and bulk of the all ceramic crowns and influences the fit at the margin. Purpose: The purpose of this study was to evaluate the stress distribution within marginal configurations of all- ceramic crowns (90-degree shoulder, 110-degree shoulder, 135-degree shoulder). Material and methods: The force is applied from a direction of 45 degrees to the vertical tooth axis. Three-dimensional finite element analysis was selected to determine stress levels and distributions. Results and conclusion: The result of stress level for the shoulder marginal configuration was more effective on stress distribution at 135-degree shoulder margin. But the stresses concentrated around at 135-degree shoulder margin. The stress decreased apically at the surface between cements and alumina core, and increased apically at the surface between alumina core and veneering porcelain.

The Comparison of the Solar Radiation and the Mean Radiant Temperature (MRT) under the Shade of Landscaping Trees in Summertime (하절기 조경용 녹음수 수관 하부의 일사와 평균복사온도 비교)

  • The purpose of this study was to compare the Solar Radiation(SR) and the Mean Radiant Temperature(MRT) under the shades of the three landscaping trees in clear summer daytimes. The trees were Lagerstroemia indica, Quercus palustris and Ulmus parvifolia. The solar radiation, the globe temperature and the air temperature were recorded every minute from the $1^{st}$ of April to the $30^{th}$ of September 2013 at a height of 1.1m above on the four monitoring stations, with four same measuring system consisting of a solar radiation sensor, two resistance temperature detectors(Pt-100), a black brass globe (${\phi}50mm$) and data acquisition systems. At the same time, the sky view photos were taken automatically hourly by three scouting cameras(lens angle: $60^{\circ}$) fixed at each monitoring station. Based on the 258 daily sky view photos and 6,640 records of middays(10 A.M.~2 P.M.) from the $1^{st}$ of June to the $30^{th}$ of August, the time serial differences of SR and MRT under the trees were analysed and compared with those of open sky, The major findings were as follows; 1. The average ratio of sky views screened by the canopies of Quercus palustris, Lagerstroemia indica and Ulmus parvifolia were 99%, 98% and 97%, and the SR were $106W/m^2$, $163W/m^2$ and $202W/m^2$ respectively, while the SR of open sky was $823W/m^2$. Which shows the canopies blocked at least 70% of natural SR. 2. The average MRT under the canopies of Quercus palustris, Lagerstroemia indica and Ulmus parvifolia were $30.34^{\circ}C$, $33.34^{\circ}C$ and $34.77^{\circ}C$ respectively, while that of open sky was $46.0^{\circ}C$. Therefore, it can be said that the tree canopies can reduce the MRT around $10{\sim}16^{\circ}C$. 3. The regression test showed significant linear relationship between the SR and MRT. In summary, the performances of the landscaping shade trees were very good at screening the SR and reducing the MRT at the outdoor of summer middays. Therefore, it can be apparently said that the more shade trees or forest at the outdoor, the more effective in conditioning the outdoor space reducing the MRT and the useless SR for human activities in summertime.
