Induced Epidermal Cell Turnover in the Seawater-Adopted Guppy, Poecilia reticulata

  • The turnover of epidermal cells after seawater adaptation of the freshwater fishwas studied in the guppy (Poecilia reticulata) by means of Proliferating cell nucleus antigen (PCNA) immunocytochemistry and transmission electron microscopy. The number of PCNA-immunoreactive cells in the epidermis of the seawater-adapted guppies, which becomes thinner than that in the fresh-water, generally increases four times as much. Degeneration of filament-containing cells by necrosis or apoptosis occurs mainly in epidermal cells. Apoptotic filament-containing cells seem to be shed into the water in the environment instead of phagocytosis by adjacent macrophages. The apoptotic chloride cell has a highly condensed cytoplasm and the lumen of tubular system is distended. The apoptotic mucous cell, which has an electron-dense cytoplasm, is characterized by the presence of a large multivesicular body of different electron densities. Macrophages contain many electron-dense lysosomal bodies and large vesicles filled with cellular debris. It is concluded that mitosis and apoptosis of epidermal cells are greatly stimulated when fish are adapting to seawater. This result reflects an increase in epidermal cell turnover by change of environmental salinity.

  • Bacterial disease caused by Aeromonas are among the most common and troublesome diseases of fish raised in aquaculture systems. In this study, some strains identified as belonging to the Aeromonas were isolated from neon tetra (Hyphessobrycon herbertaxelrodi) skin and fin, as well as from water samples. VITEK system and API ZYM examination of the isolated strains were undertaken, and it seemed to correlate with the Aeromonas, proved to be Aeromonas veronii. The antibiotic susceptibility test of isolated strains to different groups of antibiotics was evaluated using the disc diffusion method. Cefixime was the most sensitive antibiotic.

  • We reported an outbreak of Aeromonas (A.) veronii responsible for ulcerative dermatitis in Israeli carp (Cyprinus carpio). The major clinical signs were darkening body, abdominal extension, exophthalmos and severe ulcerative necrosis in the skin. The necropsy showed yellowish ascites, necrosis in liver and enlargement of kidney and spleen in the morbid fish. In blood agar for culturing bacteria, three different colonies were identified as A. veronii, Plesimonas shigelloides and Shewanella putrefaciens by phylogenetic identification using 16S rRNA or gyrB gene sequences. A. veronii was the most dominant species among them and was resistant to ampicillin, nalidixic acid and oxytetracycline.

  • Reproduction and organ structure of the pale chub, Zacco platypus in Isa stream were investigated by means of histological methods. The results of the study confirmed reproductive abnormality and histopathological features in the pale chub. The gonadosomatic index (GSI) of the fish showed two peak in April and August. In summer season, GSI of the male was about two times of the female's GSI. Monthly variation of the gonadal development was very irregular. From the histological analysis of the organ structure, epidermal atrophy, necrosis and hyperplasia of pigment cell were observed in the skin. Epithelial layer lifting and clubbing of the lamella and bifurcation of the filament were observed in the gill. Also histological changes as congestion, cytoplasmic degeneration of hepatic cell, degeneration of bile duct, glomerular dilatation, degeneration of renal tubule and pycnosis of interstitial cell were identified in the liver and kidney, respectively.

동물성 수산식품 중의 Sterol 조성과 Cholesterol ${\Delta}^7$-dehydrogenase의 존재에 관한 연구 (Studies on the Composition of Sterol and the Presence of Cholesterol ${\Delta}^7$-dehydrogenase in Marine Animal Products)

  • Levels of sterols including ${\Delta}^7$-dehydrogenase isolated from the tissues of marine animal products (20 species) were determined on 1.5% OV-17 columm of gas-liquid chromatography. The composition showed that the mussels and clams contained various sterols in their tissues : cholesterol, brassicasterol. 24-methylenecholesterol with some minor components such as 22-trans-norcholesta-5,22-dien-3${\beta}$-ol, 22-cis-dehydrocholesterol, 22-trans-dehydrocholesterol, desmosterol, 7-dehydrocholesterol, campesterol, stigmasterol, ${\beta}$-sitosterol, isofucosterol, and 7-dehydrocholesterol which could be converted into vitamin $D_3$ in the skin tissue of animal was present in the muscle of oyster, Crassostrea gigas. On the other hand, the others including gastropoda were predominantly composed of cholesterol. The minor sterols such as 24-methylenecholesterol, stigmasterol and ${\beta}$-sitosterol in the fish intestines are supposed to be derived from dietary plankton. Cholesterol ${\Delta}^7$-dehydrogenase which could convert cholesterol into ${\Delta}^7$-dehydrogenase was present in the pickles of Tricurus haumela intestine.

  • The Aeromonas strains were isolated from diseased cultured catfsh (Silurus asotus) in the ponds near by Gochang of Chunbuk. The present isolates were identified as A. hydrophila based on their biological and biochemica1 characteristics. The isolates of A. hydrophila were named GC-1, GC-2, GC-3, GC-4 and GC-5, Five strains were grew optimally at temperatures $35^{\circ}C,$ pH 7.5 and in 0.5 to l% NaCl. This bacterium(GC-1) was injected into health catfish in order to prove the causative agent of ascites and haemorrhagic ulcers. The symptoms in cat fish infected by this challenge method were observed to be very similar to the symptoms of a natural infection, but controls did not show any abnormal symptoms during the experimental period. At 24 post-injection, the red spot developed around the injection site and the haemorrhagic ulcers was extended near the gill and ventral fin. Five strains were tested for drug sensitivity by plate method. All strains were sensitivity to gentamicin and resistant to penicillin, streptomycin and tetracycline.

황다랑어 복부 껍질을 이용한 콜라겐의 제조 및 물리화학적 특성 해석 (Processing and Physicochemical Properties of Collagen from Yellowfin Tuna (Thunnus albacares) Abdominal Skin)

  • Processing of collagen from yellowfin tuna (Thunnus albacares) abdominal skins was optimized by response surface methodology and central composite design. The values of independent variables at optimal conditions were NaOH concentration: 0.5 N, NaOH treatment time: 36.2 hr, pepsin concentration: 1:4.9 ratio (0.245%, w/v), and digestion time: 48.1 hr, respectively. The collagen content estimated under optimal conditions was 33.1%, and the actual experimental collagen content was 32.3%. Physicochemical properties of collagen from yellowfin tuna abdominal skin were investigated by amino acids analysis, SDS-PAGE, FT-IR, viscosity and denaturation temperature. Amino acids content of the collagen was 21.0%. SDS-PAGE pattern of the collagen showed two different $\alpha$-chain (${\alpha}_1$- and ${\alpha}_2$- chain), $\beta$-component and $\gamma$-component. The spectrum of FT-IR of the collagen showed wavenumber at 3,434, 1,650, 1,542 and $1,235\;cm^{-1}$ representing the regions of amide A, I, II and III, respectively. Relative viscosity of the collagen decreased continuously on heating up to $32^{\circ}C$, and the rate of decrease was retarded in the temperature range of $35-50^{\circ}C$. Denaturation temperature (Td) of the collagen solution (0.06%, w/v) was $31^{\circ}C$ and was lower than calf skin collagen ($35^{\circ}C$).

가자미피 젤라틴 가수분해물로부터 항산화성 펩티드의 분리${\cdot}$정제 및 특성 (Isolation and Characterization of Antioxidative Peptides from Enzymatic Hydrolysates of Yellowfin Sole Skin Gelatin)

  • 연속식 3단계 막반응기로부터 각 단계별로 분리한 가자미피 젤라틴 가수분해물의 항산화성을 측정한 결과, 2단계 가수분해물의 항산화력이 가장 뛰어날 뿐만 아니라, 천연항산화제인 $\alpha-tocopherol$보다 $10\%$ 정도의 높은 항산화력을 나타낸 반면, 1단계 및 3단계 가수분해물은 $\alpha-tocopherol$보다 오히려 $10\~15\%$ 정도 낮은 항산화력을 보였다. 가수분해물의 첨가한 농도에 따른 항산화성은 유지중량에 대해 $1.0\%(w/w)$로 첨가한 농도에서 최대 항산화력을 나타내었다. 한편, 천연항산화제인 $\alpha-tocopherol$과 합성항산화제인 BHT와의 상승효과를 검토한 결과, 각 단계별 가수분해물은 $\alpha-tocopherol$과 우수한 상승효과를 관찰할 수 있었으며, 그 중 2단계 가수분해물의 상승효과가 가장 강하였다. 또한 항산화성이 가장 뛰어난 2단계 가수분해물로부터 gel column, ion exchange column 및 ODS column을 사용하여 항산화력이 특히 우수한 부분만을 분리한 단일 펩티드를 간세포에 첨가하여 세포생존율에 미치는 효과를 관찰한 결과, TBHP의 독성에 대해 펩티드 첨가구가 무첨가구에 비해 세포생존을 연장시켰으며, lipid peroxidation측정에서도 세포의 산화를 억제함으로써 세포의 생존율을 높였다.

숙시닐화에 의한 각시가자미껍질 젤라틴의 탄닌산과의 반응성 개선 (Improvement on the Reactivity Properties with Tannic Acid of Yellowfin Sole Skin Gelatin by Succinylation)

  • 어류껍질 젤라틴의 탄닌산과의 반응성을 개선하여 청정제 및 chewing gum base 등으로 유효 이용할 목적으로 각시가자미껍질 젤라틴의 숙시닐화를 시도하여 탄닌산과의 반응특성에 대하여 검토하였다. 각시가자미껍질 젤라틴의 숙시닐화는 succinic anhydride의 농도가 10%까지는 직선적으로 증가하였고, 그 이상의 농도에서는 거의 변화가 없었다. 본 실험에서는 젤라틴에 대하여 15%에 해당하는 succinic anhydride로 숙시닐화시켜 탄닌산과의 반응성을 살펴 보았고, 이 젤라틴의 숙시닐화 정도는 약 80%이었다. 등전점은 숙시닐화 어류껍질 젤라틴의 경우 4.08로 대조 어류껍질 젤라틴의 5.54에 비하여 감소하였고, 일반성분 및 아미노산 조성은 두 젤라틴이 유사하였다. 용액의 pH가 숙시닐화 어류점질 젤라틴의 경우 4.0부근에서, 대조 어류껍질 젤라틴의 경우 4.8 부근에서 젤라틴 및 탄닌산의 침전율이 최대이었다. 젤라틴 농도는 젤라틴 침전율의 경우 숙시닐화에 관계없이 농도가 증가할수록 감소하였고, 탄닌산의 침전율의 경우 탄닌산에 대하여 숙시닐화 젤라틴은 $2{\sim}4$배, 대조 젤라틴은 $2{\sim}3$배로 첨가한 것이 최대이었다. 젤라틴 및 탄닌산의 침전율은 에탄올의 존재에 의해서는 영향을 받았으나, sucrose의 존재에 의해서는 거의 영향을 받지 않았다.

저수온 양식 넙치 Paralichthys olivaceus의 Cryptocaryoniasis (Cryptocaryoniasis of cultured flounder, Paralichthys olivaceus in low temperatures)

  • 1995년 겨울 경북 구룡포에 있는 축제식 양식장에서 사육중이던 넙치(평균 체장 : 31cm, 평균체중 : 300g)에 백점병이 발생하여 대량 폐사를 일으켰다. 감염 넙치로부터 병리조직학적, 원인충의 형태학적 및 생물학적 특성을 조사한 결과, 아가미 및 지느러미의 병리조직학적 검사에서 많은 섬모충들이 상피 하층으로 뚫고 들어감으로써 기생 부위의 상피 세포와 점액 세포가 증식을 일으킨 것을 볼 수 있었고 병어의 지느러미, 체표 및 아가미에서 작은 백점을 나타낸 충체는 기생성 섬모충 Cryptocaryon irritans의 특징을 갖추고 있었다. 그러나 이 충의 증식 방법은 다분열증식법 이외에도 출아와 다분열을 혼합한 또하나의 증식법(혼합 증식법)을 가진 것으로 조사되었으며 수온 $16^{\circ}C$에서는 위와 같은 두가지의 증식 방법이 약 3대 2의 비율로 나타났고 다분열 증식형의 충체는 8~14일만에, 혼합 증식형의 충체는 13~15일 만에 각각 자충을 방출하였다. 수온 $12\sim24^{\circ}C$의 조건에서 염분 농도 범위 $0\sim35\%_{\circ}$$5\%_{\circ}$ 간격으로 조절하여 충체의 생존능력을 살펴본 결과 다분열 증식형의 총체는 $12^{\circ}C$에서 $30\%_{\circ}$이하, $16^{\circ}C$에서 $20\%_{\circ}$ 이하, $20^{\circ}C$에서 $15\%_{\circ}$이하 그리고 $24^{\circ}C$에서 $25\%_{\circ}$ 이하에서 각각 폐사된 반면 혼합 증식형의 충체는 $12^{\circ}C$에서 $20\%_{\circ}$ 이하, $16\sim20^{\circ}C$에서 $15\%_{\circ}$ 이하 그리고 $24^{\circ}C$에서 $25\%_{\circ}$ 이하에서 각각 폐사를 나타내어 저수온($12\sim16^{\circ}C$)에서 혼합 증식형의 충체가 다분열 증식형의 충체보다 다소 낮은 염분 농도에서 생존이 가능함을 보여주었다. 수온 $12\sim24^{\circ}C$의 조건에서 탈낭 소요 시간 및 탈낭률은 다분열 증식형의 총체가 $12^{\circ}C$에서 8일 및 30%, $16^{\circ}C$에서 6.5일 및 50%, $20^{\circ}C$에서 5.5일 및 75% 그리고 $24^{\circ}C$에서 7일 및 10%를 나타낸 반면 혼합 증식형의 충체는 $12^{\circ}C$에서 15.5일, $16^{\circ}C$에서 14일 및 76.6%, $20^{\circ}C$에서 12일 및 72.2% 그리고 $24^{\circ}C$에서 10일 및 31.6%를 나타내어 모든 수온 조건에서 혼합 증식형의 충체가 다분열 증식형의 충체 보다 탈낭 소요 시간이 길며 수온 $16^{\circ}C$에서 최고의 탈낭율을 나타내어 저수온에 다소 안정적인 것을 보여주었다.
