Morphological characterization of Vibrio alginolyticus specific bacteriophage isolated from fish farms on west coast of Korea (서해안 양식장에서 분리한 Vibrio alginolyticus의 특이 bacteriophage에 대한 구조적 특성)

  • Vibrio alginolyticus (V. alginolyticus), which is one of bacterial pathogens evoking severe infection in fish and shellfish as well as in human has been found at high frequency around all coast areas in Korea. Both V. alginolyticus and V. alginolyticus specific bacteriophage were isolated from sea water and various fishes from fish farms on west coast in Korea. In a morphological study based on electron microscope, the purified phage appeared to be composed of hexagon head of 60 nm and short tail of 20 nm. In the denatured SDS-PAGE analysis, the structural proteins of the phage were found to be 7 different protein fractions ranging from 37.8 to 198 kda. The kind of nucleic acid of the phage was ascertained to a double stranded DNA.

A Survey of Ecosystem Structure in the Watershed of the Seom River and Hoengseong Reservoir, Kangwon Province, Korea (강원도 섬강 (횡성호) 유역의 하천생태계 조사)

  • This study was conducted to elucidate the structure of river ecosystem in the watershed of the Seom River and Hoengseong Reservoir located in Hoengseong of Kangwon Province from February to October 2007. Topics of the survey were mainly rainfall, discharge, water quality in abiotic factors and attached algae, benthic macroinvertebrates, fish, birds and mammals of flora and fauna in a biotic factors, respectively. Specifically, the Seom River could be seen as a typical flow rate of the stream is controlled to the effect of the dam. Basic water qualities were great to seasonal effects, it was relatively clean. Diatom Achnanthes, Cymbella, Gomphonema, and Navicula were distributed predominantly in the periphytic algae. Benthic macroinvertebrates were mostly aquatic insects and freshwater shellfish, the aquatic insects were abundant Ephemeroptera and Trichoptera. Freshwater fish was the dominant Zacco platypus, fish species varied toward the downstream. Birds were mainly observed in four species, and species Egretta garzetta, which was distributed in a wide area of the Seom River. In mammals, Lutra lutra of Mustelidae was identified that the number of inhabit widely. In aspects of the ecological trophic level, the Seom River was maintained at a relatively stable state in the producer and the consumer relationship. The results of this study will expected to be utilized as a useful data for understanding the structure and function of the lotic and lentic ecosystems.

Effect of Preservative, Ever-Fresh, on Fish and Shrimp Freshness during Ice Storage (선도유지제(鮮度維持劑)(Ever-Fresh) 처리(處理)가 붕장어 학꽁치 및 새우의 선도(鮮度) 유지(維持)에 미치는 효과(效果))

  • In this paper, the effect of Ever-Fresh, a kind of preservatives for fish and shellfish, on fish and shrimp during ice storage were investigated. The results are as follows: 1. K value representing the ratio of optical densities of inosine and hypoxanthine fraction and total nucleotide fraction at 250nm could be useful as an index to measure the freshness of fish and shrimp during ice storage. 2. Volatile basic nitrogen could be also useful as an index to measure the freshness of shrimp during ice storage. 3. Ever-Fresh treatment of conger eel, half-beak, and shrimp appeared effective on keeping freshness during the early stage of ice storage. 4. Ever-Fresh treated shrimp showed lower thiobarbituric acid value as compared with the control during the early stage of ice storage.

Antibacterial effects of natural products on Vibrio vulnificus growth in seawater and shellfish (바닷물과 조개에서 패혈증비브리오균 증식에 대한 천연물의 항균 효과)

  • Vibrio vulnificus, an opportunistic pathogen, causes septicaemia when raw shellfish and fish are eaten by patients with hepatic diseases or reduced immunity. In this study, we evaluated inhibitory effects of some natural products on V. vulnificus growth using 96-well microplate assay. We found that Phyllanthus emblica L., Rosa chinensis Jacq., Rosa rugose Thub., and Chukrasia tabularis A. Juss. significantly inhibited V. vulnificus growth in Luria-Bertani (LB) broth. Among these four extracts, the inhibition diameter of Chukrasia tabularis was 16.00 ± 0.58 mm in disc diffusion assay on V. vulnificus growth. In addition, these four natural products protected HeLa cells from V. vulnificus-induced cytotoxicity. A cocktail containing these four products showed an inhibitory effect on V. vulnificus growth in seawater and shellfish by reducing its growth by 75.7% and 97%, respectively. These results suggest that these four natural products are safe and effective natural antimicrobial candidates to prevent V. vulnificus infection.

Analysis of Current Status of Marine Products and Characteristics of Processed Products Seafood in Joseon - via the Veritable Records of the Joseon Dynasty based data - (『조선왕조실록(朝鮮王朝實錄)』 속 수산물 현황과 가공식품 특성 분석)

  • This study used the big data method to analyze the chronological frequency of seafood appearance and variety mentioned by the veritable records of the Joseon dynasty. The findings will be used as a basis for Joseon Period's food cultural research. The web-crawling method was used to digitally scrap from the veritable records of the Joseon dynasty of Joseon's first to the twenty-seventh king. A total of 9,536 cases indicated the appearance of seafood out of the 384,582 articles. Seafood were termed "seafood" as a collective noun 107 times (1.12%), 27 types of fish 8,372 times (87.79%), 3 types of mollusca (1.28%), 18 types of shellfish 213 times (2.23%), 6 types of crustacean 188 times (1.97%), 9 types of seaweed 534 times (5.60%). Fish appeared most frequently out of all the recorded seafood. Sea fish appeared more frequently than the freshwater fish. Kings that showed the most Strong Interest Inventory (SII) were: Sungjong from the 15thcentury, Sehjo from the 15th, Youngjo from the 18th, Sehjong from the 15th, and Jungjo from the 18th respectively. Kings of Chosen were most interested in seafood in the 15th and 18th centuries.

A Bibliographical Study on the Gooumguk in Korea (고음국의 문헌적 분석 고찰)

  • The Guk was divided Malgunjangguk, Tojangguk, and Gooumguk as to cooking method. Gooumguk was boiled with several materials of beef, bone, organs and water for the long time, and a good source of protein, calcium, and phosphorus. In the thesis, according to the kinds of Gooumguk, the reference frequency to them, the adding foods in them, and the variety cooking method in the Gooumguk were analyzed by the cooking books published from 1670 to 1988 in Korea. 1. There were 41 kinds of Gooumguk and Kaejang, Joktang and Byultang were firstly published to Omshikdimibang in 1670. 2. Many kinds of Gooumguk were generally used on royal parties in Yi dynasty and were used by several substances. 3. The main substances of Gooumguk were meat, poultry, fish, and shellfish. The seasoning substances were green onion, garlic, ginger and black pepper etc, and were used for the removal of the meat's or fish's off-flavor. 4. Yangtang, Kalbitang, Joktang, Yeongebacksuk, and Chooatang etc expressed the taste of the one main substance and Japtang, Sulnongtang, Yukgejang, Yongbongtang etc were combinated the taste of the several materials. 5. Cooking methods of Yukgaejang and Koritang before 19 century were different in present.: Yukgaejang was not used Gochoojang or powdered red pepper and Koritang was used Gochoojang.

Recent Insights into Aeromonas salmonicida and Its Bacteriophages in Aquaculture: A Comprehensive Review

  • The emergence and spread of antimicrobial resistance in pathogenic bacteria of fish and shellfish have caused serious concerns in the aquaculture industry, owing to the potential health risks to humans and animals. Among these bacteria, Aeromonas salmonicida, which is one of the most important primary pathogens in salmonids, is responsible for significant economic losses in the global aquaculture industry, especially in salmonid farming because of its severe infectivity and acquisition of antimicrobial resistance. Therefore, interest in the use of alternative approaches to prevent and control A. salmonicida infections has increased in recent years, and several applications of bacteriophages (phages) have provided promising results. For several decades, A. salmonicida and phages infecting this fish pathogen have been thoroughly investigated in various research areas including aquaculture. The general overview of phage usage to control bacterial diseases in aquaculture, including the general advantages of this strategy, has been clearly described in previous reviews. Therefore, this review specifically focuses on providing insights into the phages infecting A. salmonicida, from basic research to biotechnological application in aquaculture, as well as recent advances in the study of A. salmonicida.

Comparison of Perception of the Neutropenic Diet between Nurses and Patients (호중구감소증 환자의 식품별 섭취허용에 대한 간호사와 환자의 인식 비교)

  • Purpose: This descriptive study aimed to compare the perception of the acceptable foods for the neutropenic diet between nurses and patients by food type. Methods: The participants were 225 nurses working at chemotherapy wards and 71 patients in chemotherapy treatment. Data were collected with a self-administered questionnaire from January 2 to February 24, 2012, and analyzed with SPSS 12.0 program using descriptive statistics and the ${\chi}^2$-test. Results: Eighty-eight point eight percent of nurses and 76.1% of cancer patients thought the patients needed the neutropenic diets. The most important decisional criteria to determine dietary restriction was neutrophil count for nurses and food type for patients. The two groups showed significantly different allowance to raw fruits and raw vegetables, sterilized canned juice, fried vegetables, yogurt, uncooked tofu, salted fish, cooked fish, cooked shellfish, uncooked grain powder, jellified food, home-made bread/cookies, nuts including peanuts, instant coffee or tea and tea brewed. In general, patients were more permissive about the neutropenic diet than nurses. Conclusion: It is recommended to consider patients' preference as well as nurses' professional knowledge and publish standardized clinical diet guidelines for neutropenic patients with collaboration between nurses and patient representatives.

A Study on 'Tang', Traditional Soup Originated in Kyoung-Nam Province (경남지방의 향토요리에 관한 연구 -탕류를 중심으로-)

  • Currentry, the originality of traditional soups in Kyoung-Nam province is vanishing by the inflex of processed and instant food, food industrial development. Especially, such factors as the influence of alien cooked, the develoment of transportation, the movement of population have made it hard to preserve the traditionnal food preparations. The purpose of this thesis was to seek out the various kinds of Kyoung-Nam province soups preparation. To do this, about 60 kinds of soups recipe were reviewed and charactered into several categories. The findings of the study are as follows. 1) Soup is made by fish or shellfish, freshwater fish. Meat and fowls is rarely used in Kyoung-Nam province (coastal region). 2) Soups of inland area is made by most vegetables and these soup were always harmonized with soybean powder or perilla powder. 3) Various soup season with the mag-jang, mul-jang sauce, hab-ja-jang sauce. 4) These soups were in general to make much red pepper and much salt.

Inhibitory Effects of Candidate Probiotic Bacteria on the Growth of Fish Pathogenic Bacteria, Streptococcus sp. (Candidate Probiotic Bacteria의 어류병원성 Streptococcus sp. 성장에 대한 억제 효과)

  • For the treatments and protection of bacterial fish disease, many requirements are needed for aquatic probiotics so that they are effective in aquaculture animals but are also harmless to humans. In the present study, among 17 candidate probiotic bacteria (CPB) obtained from the edible part of the shellfish, Bacillus sp. CPB-St (CPB-St) were selected and in vitro evaluated for the possibility as a probiotic strain for the control of fish streptococcosis which frequently occurs in the olive flounder farms. CPB-St showed inhibitory effects on the growth of various fish pathogenic bacteria, Streptococcus sp., S. parauberis, S. iniae, Lactococcus garvieae and L. piscium by the double layer test ranging about 18~26 mm of clear zone. Inhibitory activity of CPB-St to Streptococcus sp. was observed 6 hours after and the growth of Streptococcus sp. was decreased to 8~55 folds in the co-culture of CPB-St with Streptococcus sp.. The safety of CPB-St to fish and survival of CPB-St in the intestine were assessed in the olive flounder, Paralichthys olivaceus. Fish mortality was not observed in artificial infection with CPB-St for 2 weeks. CPB-St was entirely excreted from the stomach and intestine 24 hours after oral injection. This results indicate that CPB-St has potential applications as a probiotic for the control of fish streptococcosis in aquaculture.
