In separating mycelia from antibiotic fermentation broths, high permeate flux was obtained by cross-flow filtration using modified regenerated cellulose membrane. The flux was increased most effectively by increasing the flow rate. There existed a critical mycelium density (about 20% PMV) at which the highest flux was observed. In a batchwise concentration of the fermentation broth, the system suffered from a severe fouling problem, which was relieved drastically by applying diafiltration technique, although it increased the permeate volume. A combined concentration/diafiltration process was ideal in keeping relatively high flux together with a high product recovery yield. The best result was obtained by starting diafiltration after concentrating the broth to 20% PMV. By doing so, a 98% product recovery yield was achieved in the shortest time while keeping the permeate volume at a minimum level.
There are many design parameters affecting filtration efficiency such as filteration rate, media packing depth, size distribution, and so on. The sphericity, the ratio of the surface area of an equal volume sphere to the real surface area of the particles, is one of major physical characters of media. The effect of sphericity on the performance of anthracite filter has been investigated. Media from eight water treatment plants have been collected. The sphericity of each media has been calculated by using well known headloss equations such as Kozeny equation, Dahmarajah equation etc.. Columns packed with anthracite media having different sphericity have been used to compare headloss development, floc accumulation in the bed, particles in bed water, filtrate turbidities after backwash and so on. The repeated experiments have indicated that the sphericity of anthracite media may not have remarkable influence on the filter performance as it has been suspected. It also has been prospected in the experiment that the media of higher sphericity would store more particles in the bed and give better filtrate quality, if provided that the effective size and the size distribution of media would be the same.
The Pseudomonas fluorescens KCTC 1767, a selected and identified as potential candidate for stereo-specific resolution of rac-ketoprofen ethyl ester, was systematically investigated in order to induce the high level expression and detailed characterization of the expressing enzyme esterase. We cloned the esterase gene from chromosomal DNA of Pseudomonas fluorescens KCTC 1767 by PCR with two synthetic primers that desinged for simple purification. The recombinant esterase from Pseudomonas fluorescens KCTC 1767 exibited a high conversion rate and enantioselectivity to the (S)-ketoprofen ethyl ester as expected. The enzyme was easily purified to homogeniety by using a metal chelating affinity chromatography as a protein with poly histidine taq, and thus obtained 0.6 mg of protein from a 100 mL culture broth in a single step. The purified enzyme was steadily stable at the pH range from 7.0 to 10. The activity was also retained to be about 70% after the preincubation at $40^{\circ}C$ but over $50^{\circ}C$ lost the activity completely. The molecular mass of the esterase was estimated to be about 43 kDa on SDS-PAGE, and an identical result was also shown in gel filteration chromatography. The specific activity was calculated 27 mM/mg-protein/min by using the rac-ketoprofen ethly ester as a substrate.

As the integrated circuit device shrinks to smaller dimensions, chemical mechanical polishing (CMP) process was requirdfo the global planarization of inter-metal dielectric (IMD) layer with free-defect. However, as the IMD layer gest thinner, micro-scratches are becoming as major defects. However, as the IMD layer gets thinner, micro-scratches are becoming as major defects. Micro-scratches are generated by agglomerated slurry, solidified and attached slurry in pipe line of slurry supply system. To prevent agglomerated slurry particle from inflow, we installed 0.5${\mu}{\textrm}{m}$ point of use (POU) filter, which is depth-type filter and has 80% filtering efficiency for the 1.0${\mu}{\textrm}{m}$ size particle. In this paper, we studied the relationship between defect generation and polished wafer counts to understand the exact efficiency fo the slurry filteration, and to find out the appropriate pad usage. Our experimental results showed that it sis impossible to prevent defect-causing particles perfectly through the depth-type filter. Thus, we suggest that it is necessary to optimize the slurry flow rate, and to install the high spray bar of de-ionized water (DIW) with high pressure, to overcome the weak-point of depth type filter.
The studies carried out to investigate the detection method of Fasciola eggs from positive cattle feces in the intradermal reaction method with laborsaving composition of sieves and Kim's sedimental tube method. The results obtained are summarized as follows: 1. The detection method of Fasciola eggs was improved by 3 meshes (100mesh, 150mesh, 250mesh) for filteration of fecal fluid contained Fasciola eggs. 2. Third type of tube in the 3 kinds of sedimental tube had the highest recovers rate of Fasciola eggs. Thus, this type of tube was selected and called as Kim's sedimental tube. 3. A-II, A-III, B-III and C-III type by Kim's sedimental tube method had the recovery rates of eggs in the 1st drop as ranged 99 to 100% and, A-I, B-II, C-II as 71 to 91%. 4. These Type of tubes were useful to detect Fasciola eggs and to shorten the time of the diagnostic procedures. 5. The Kim's sedimental tube method showed high recovery rates and convenient procedures as compared with other detection methods reported. In addition, the EPG value can also be obtained by this method. Therefore, it is desirable that the Kim's sedimental tube method could be recommended to clinicians.
Seasonal variations of dominant algae species and the effects of these algae on coagulation and filteration of water treatment were investigated at Chilseo water treatment plant in downstream of Nakdong river from January in 1995 to Desember of 1998. The water quality of Nakdong river was found to be a hyper eutrophic state during the investigation periods. In the measurement, Chlorophyll-a contents ranged $20.7{\sim}180.9{\mu}g/l$ and total nitrogen contents(T-N) and total phosphorus contents(T-P) exceeded more than 3.4mg/l and 0.1mg/l, respectively. The changes in dominant algae species was in the order of Stepanodiscus sp., Asterionella sp., Melosira sp., Microcystis sp. and Synedra sp. from spring to winter. Microcystis sp. especially, was blooming during summer and Synedra sp. and Stepanodiscus sp. during winter. Although most diatomous algae appeared in the water treatment process caused filter clogging and reduced efficiency of coagulation and sedimentation, Synedra sp. and Stepanodiscus sp were revealed as the main trouble algae. Malfunction of water treatment process caused by Synedra sp. and Stepanodiscus sp. started at the algae concentrations of 800cells/ml and 1,820cells/ml, respectively. When chlorophyll-a content was $18.9{\mu}g/l$, the optimum amounts of coagulant were found to be 40mg/l of Alum and 16mg/l of PACS. Under condition of chlorophyll-a content of $154.1{\mu}g/l$, addition of Alum at the level of 75mg/l and PACS at the level of 35mg/l showed the lowest turibidity. The result indicates that increased amounts of the coagulants should be added for a better water treatment as chlorophyll-a contents increased. Addition of Alum at the amount of 60mg/l and 30mg/l of PACS removed Stepanodiscus sp. algae at the rate of 85% and 83%, respectively. In case of Synedra sp., 50mg/l of Alum and 25mg/l of PACS showed removal rates of 79% and 81%, respectively. Synedra sp. algae at the standing crops of 1,500cells/ml started filter clogging and a filtering process was completely inhibited after 8 hours. At this situation the filter clogging by Synedra sp. algae occurred at the depth of 5cm from the top anthracite layer. On the other, other algae did filter clogging at the depth of 10cm.
We designed high dose rate Ir-192 source which was prepared for substitute the Co-60 source in Ralstron unit (Simatsu, Japan) which is supplied for cervical cancer treatment. The source dimension is 1.5 mm in a diameter and 1.5mm thickness of cylinder and encapsulated with 3 mm diameter of stainless steel(SUS316L) to substituted for the Co-60 source size. The Ir-192 source was prepared the dose model for tissue dose computation through the experimental determination of apparent activity and applied the empirical tissue correction factors extended to 20cm distance. The tissue dose model was applied the 4.69 R/cm-mCi-hr gamma constant and the ratio of energy absorption coefficient of water to that of air showed 1.112 include filteration of the self-absorptions. In this experiments, we prepared the dose computation software to clinical usefulness.
This research aims to remove the heavy metals, nonbiodegradable COD(NBDCOD), and color using the coal waste. The experimental by heat treatment was performed to advance the adsorption capacity. The results are as follows ; i) The coal waste had the adsorption capacity of heavy metals and the rates were in the range of 20 to 30 percents. ii) The heat treatment was the optimum condition that the reaction time was 6 hours at $500^{\circ}C$, , iii) In the column experimen, non-treated coal waste removed the COD and color in the range of 20 to 60 percents. iv) Heat-treated coal waste showed higher removal rate of the color in biological effluent, and heavy metal and COD removal rates were changed by the filteration rates.
To study the effects of the age and the dietary protein content on Ca metabolism male rats of 1 month 6 month 12 month of age were fed experimental diets containing 5%, 15% or 50% casein for 4 weeks. Food and ca intake were higher in old rats and in high protein groups. The weight ash and Ca contents of femur and tibia were higher in old rats. The higher dietary protein level resulted in higher skeletal weigh ash and Ca contents. But high protein diet(50% casein) lead to reduced bone mineral density(ash/dry bone weight) and Ca density(Ca/dry bone weight) in 1 month old rats. Low protein diet(5% casein) on the other hand reduced the bone growth even though the bone density was higher in this group. The ill effect of low protein diet was not evident in 12 month old rats. Glomerular filteration rate(GFR) and urinary Ca excretionincreased with age and with dietary protein level especially in 12 month old rats. Serum immunoreactive parathyroid hormone(iPTH) level tended to be higher in aged rats but was not affected by dietary protein level except 1 month old rats where 50% protein group showed significantly higher value. This study showed that the dietary protein level seemed to have different effect on Ca metabo-lism in rats of different age., The low bone density in the high protein group of growing rats may be due to the higher iPTH level and increased urinary Ca. The dietary protein level however had no effects on the bone composition in aged rats even though the higher urinary Ca excretion. In conclusion this study suggests that high protein intake from young may lead to less peak bone mass and to increase the bone loss in later years, which would increase the risk for osteporosis.
The purpose of this study is to manufacture lightweight aggregates for recycling water treatment sludge, to identify the physical properties of the aggregates, and present a method of utilizing the manufactured lightweight aggregates. The chemical composition and thermal properties were examined via a raw materials analysis. The aggregate examined here was fired by the rapid sintering method and the single-particle density and water absorption rate were measured. Water treatment sludge has high ignition loss and high fire resistance. When 30wt% of purified sludge was added, the single-particle density of the aggregates was in the range of 0.8~1.2g/cm3 at a temperature of 1,150~1,200℃. At temperatures of 1200℃ or higher, ultra-light aggregates having a single-particle density of 0.8 or less could be produced. When applied to concrete by replacing the general aggregate in the concrete, a specimen having strength values of 200 to 450 kgf/cm2 on 28 days was obtained, and when applied as a filter material, the performance was equal to or higher than that of ordinary sand.
