The Yellow needle crystal was isolated from Atractylis Rhizoma, having mp $124{\sim}126^{\circ}C$(decomp.), the chemical composition $C_{16}H_{21}N_{3}O_{6}$, and its m.w. 251. The pharmacological actions of this alkaloid were studied by various psycopharmacological experiments. 1) In order to see the effect of this Atractylis(=At.) alkaloid on gross general behaviors in mice, a behavioral analysis experiment was adapted. The occurrence number of sleep and lying in At. alkaloidal animals with the doses 10mg/kg or 20mg/kg was increased but the number of jumping, exploration, rearing and defecation was significantly decreased than those of placebo. 2) The effect of the At. alkaloid on unlearned emotional behaviors of mice was studied with an open-field method. The At. alkaloidal groups with the doses 20mg/kg or 30mg/kg showed less often the frequency of locomotion than that of placebo. 3) To know the effect of the At. alkaloid on the learning, a standard water maze experiment and conditioned avoidance response were conducted. As compared to placebo control, the aquisition rate of the maze learning in the alkaloidal mice with the dose of 10mg/kg or 20mg/kg was significantly impaired and the speed of swimming was also signficantly delayed. In the conditioned avoidance response, the extinction performances of the alkaloidal rats with doses of 20mg/kg or 30mg/kg did not differ significantly than that of placebo.
Age variation and secondary sexual dimorphism in four external and 27 cranial characters of 102 specimens of striped firld mice, Apodemus agrarius coreae Thomas, from Cheongju, Korea, were analyzed statistically. Specimens were classified as subadults, young adults, middle-aged adults, and old adults, and variation of morphometric characters among the four age classes was evident. Significant secondary sexual dimorphism among sexes of the same age class was not revealed. Teh specimens of the same age class have to be used in further analyses to clarify geographic variation of morphometric characters in Apodemus agrarius from Korea.
Thirty nine samples of striped field mice (Apodemus agrarius coreae Thomas) from eight localities in the Korean peninsula were used for the analyses of mitochondria1 DNA (mtDNA) fragment patterns resulted from the digestion with eight restriction enzymes. A total of 31 fragments were recognized and seven mtDNA clones were revealed: one clone consisted of 32 among 39 samples from eight localities (1 of 1 from Sogcho, 4 of 5 from Mt. Chiak, 3 of 3 from Mt. Weolak, 2 of 2 from Mt. Sogri, 2 of 2 from Mt. Deokyoo, 3 of 4 from Mt. Jiri, 2 of 4 from Haenam, and 15 of 18 from Cheongju). The nucleotide-sequence divergences (p) among seven mtDNA clones ranged from 0.2% to 2.3% and distinct subgroups were not resulted from the grouping of these clones. It is confirmed that striped field mcie from the Korean peninsula is a single subspecies of Apodemus agrarius (A. agrarius coreae) because they were not divided into separate subgroups in their mtDNA genotypes.

Atopic dermatitis (AD), a chronic inflammatory skin disease, is characterized by itchy and age-dependent lesions. Previous studies have shown that pulsed electromagnetic field (PEMF) significantly improved chronic ulcers and ununited fractures, providing an evidence for the application of PEMF in resolving inflammation caused by AD. This study investigated the anti-inflammatory effect of PEMF on DNCB-induced AD in animal models. Five male hairless mice (6 weeks old) per group were assigned to a normal group, a sham group, and two PEMF groups (15Hz, 75Hz). Mice were treated with 2,4-Dinitrochlorobenzene (DNCB) to induce uniform AD among all groups excluding a normal group. To examine the inflammatory progress and the improvement of AD after the PEMF stimulation, images are taken with various cameras for non-invasive evaluation and the results are expressed using principal component analysis (PCA) for visualization. The results of this study demonstrated that PEMF effectively improved skin lesions without the use of drugs.

Stauntonia hexaphylla (SH) and Vaccinium bracteatum (VB) are herbal extracts widely used in food and traditional herbal medicine, and have the ability to perform a wide range of biological activities. We aimed to investigate the effects of the SH and VB combination (SHVB) on mice models of chronic restraint stress (CRS) and pentobarbital-induced sleeping behaviors to elucidate its possible mechanisms of action. CRS-exposed mice treated with SHVB showed significantly decreased immobility time, increased swimming and climbing times in the forced swim test (FST), and increased locomotor activity in the open field test (OFT). SHVB decreased serum CORT levels, but enhanced brain monoamine neurotransmitters. SHVB significantly decreased the sleep latency and increased total sleep duration in pentobarbital-induced sleeping behavior in mice. SHVB showed inhibitory effect on 5-HT2A receptor-mediated ERK1/2 phosphorylation. These results suggest that SHVB has antidepressant and hypnotic effects by regulating the 5-HT2A receptor.
The motor neuron degeneration 2 (mnd2) mice carry a point mutation of A to T nucleotide transversion at the serine 276 residue of high temperature requirement A2 (HtrA2), resulting in losses of an AluI restriction enzyme site (5'AGCT3') and the HtrA2 serine protease activity. Moreover, dysfunctions of HtrA2 are known to be intimately associated with the pathogenesis of neurodegenerative diseases, including Parkinson's disease. Thus, this mnd2 mouse is an invaluable model for understanding the physiological role of HtrA2 and its pathological role in neurodegenerative diseases. Nevertheless, many molecular and cellular biologists in this field have limited experience in working with mutant mouse models due to the necessity of acquired years of the special techniques and knowledges. Herein, using the mnd2 mouse model as an example, we describe easy-to-use standard protocols for web-based analyses of target genes, such as HtrA2, and a novel approach for simple and accurate PCR-AluI-RFLP-based genotype analysis of mnd2 mice. In addition, band resolution of AluI-RFLP fragments was improved in 12% polyacrylamide gel running in 1X Tris-Glycine SDS buffer. Our study indicates that this PCR-AluI-RFLP genotype analysis method can be easily applied by the molecular and cellular biologist to conduct biomedical science studies using the other mutant mouse models.
Objectives : We aimed to identify the antidepressant effect of liver tonification acupuncture treatment (ACU (LT); KI10, LR8, LU8, LR4) and four gate acupuncture treatment (ACU (FG); LI4, LR3) and its brain neural activity in the normal and chronic restraint stress (CRS)-induced mouse model. Methods : Firstly, normal mice were given ACU (LT) or ACU (FG) and the c-Fos expressions in each brain region were analyzed to examine brain neural activity. Secondly, CRS was administered to mice for 4 weeks, then ACU (LT) or ACU (FG) was performed for 2 weeks. The depression-like behavior was evaluated using open field test (OFT) before and after acupuncture treatment. Then, the c-Fos expressions in each brain region were analyzed to examine brain neural activity. Results : In normal mice, ACU (FG) regulated brain neural activities in the hypothalamus, hippocampus, and periaqueductal gray. ACU (LT) changed more brain regions in the prefrontal cortex, insular cortex, striatum, and hippocampus, including those altered by ACU (FG). In CRS-induced model, ACU (LT) alleviated depression-like behavior more than ACU (FG). Also, brain neural activities in the motor cortex area 2 (M2), agranular ventral part and piriform of insular cortex (AIV and Pir), and cornu ammonis (CA) 1 and CA3 of hippocampus were changed by ACU (LT), and those of AIV and CA3 were also changed by ACU (FG). As in normal mice, ACU (LT) resulted in changes in more brain regions, including those altered by ACU (FG) in CRS model. M2, Pir, and CA1 were only changed by ACU (LT) in depression model, suggesting that these brain regions reflect the specific effect of ACU (LT). Conclusions : ACU (LT) relieved depression-like behavior more than ACU (FG), and this acupuncture effect was associated with modulation of brain neural activities in the motor cortex, insular cortex, and hippocampus.
Depression is a major mood disorder. Abnormal expression of glial glutamate transporter-1 (GLT-1) is associated with depression. Schisantherin B (STB) is one bioactive of lignans isolated from Schisandra chinensis (Turcz.) Baill which has been commonly used as a traditional herbal medicine for thousands of years. This paper was designed to investigate the effects of STB on depressive mice induced by forced swimming test (FST). Additionally, we also assessed the impairment of FST on cognitive function in mice with different ages. FST and open field test (OFT) were used for assessing depressive symptoms, and Y-maze was used for evaluating cognition processes. Our study showed that STB acting as an antidepressant, which increased GLT-1 levels by promoting PI3K/AKT/mTOR pathway. Although the damage is reversible, short-term learning and memory impairment caused by FST test is more serious in the aged mice, and STB also exerts cognition improvement ability in the meanwhile. Our findings suggested that STB might be a promising therapeutic agent of depression by regulating the GLT-1 restoration as well as activating PI3K/AKT/mTOR pathway.
Background: A chronic social defeat stress (CSDS) model has been proposed as relevant to stress-induced behavioral change in humans. In this study, we examined the effect of Korean Red Ginseng (KRG) on CSDS-induced mood disorders and protein expression in an animal model. Methods: To evaluate the effect of KRG on social defeat stress, test mice were exposed in the resident aggressor's home cage compartment for 14 days beginning 1 h after KRG treatment (10, 20, and 40 mg/kg, per oral (p.o.)). After the exposure, behavioral tests to measure anxiety, social interaction, and depression-like behavior were performed. To investigate the underlying mechanism, N-methyl-D-aspartate receptor expression levels in CSDS-induced mice were evaluated using Western blot analysis. Results: CSDS induced anxiety-like behaviors by decreasing central activity in the open-field test and open-arm approach in the elevated plus maze test and led to social avoidance behavior in the social interaction test. CSDS mice showed upregulated NR1, NR2A, and NR2B expression in the hippocampus. KRG 20 and 40 mg/kg ameliorated anxiety-like activities and KRG 20 mg/kg alleviated social avoidance by decreasing time in the corner zone. KRG treatment recovered CSDS-induced NR1, NR2A, and NR2B protein levels in the hippocampus. Conclusion: These results indicate that KRG has a therapeutic effect on CSDS-induced mood disorder by alleviating N-methyl-D-aspartate receptor overexpression in the hippocampus.

Purpose: Traumatic brain injury (TBI) is a significant cause of morbidity and mortality worldwide. Severity of the initial insult is one of the most significant factors affecting outcome following TBI. In order to investigate the mechanisms of cellular injury and develop novel therapeutic strategies for TBI, we designed a standardized animal TBI model and evaluated histological and functional outcomes according to the degree of impact severity. Methods: Male adult C57Bl/6 mice underwent controlled cortical impact (CCI) at varying depths of deflection (1.0-2.0 mm). We performed hematoxylin and eosin staining at 7 days after recovery from TBI. Neurobehavioral characterization after TBI was analyzed by the Barnes maze test, passive avoidance test, open field test, rotarod test, tail suspension test, and light/dark test. Results: We observed a graded injury response according to the degree of deflection depths tested (diameter, 3 mm; velocity, 3 m/s; and duration, 500 ms) compared to sham controls. In the Barnes maze test, the severe TBI (2 mm depth) group showed reduced spatial memory as compared with the sham and mild TBI (1 mm depth) groups at 7 days after TBI. There was a significant difference in the results of the open field test and light/dark test among the three groups. Conclusion: Our findings demonstrate that the graded injury responses following TBI resulted in differential histopathological and behavioral outcomes in a mouse experimental CCI model. Thus, a model of CCI with histologic/behavioral outcome analysis may offer a reliable and convenient design for preclinical TBI research involving mice.
