Characterization of Fibrinolytic Proteases from Gloydius blomhoffii siniticus Venom

  • Objectives : This study was undertaken to identify fibrinolytic proteases from Gloydius blomhoffii siniticus venom and to characterize a major fibrinolytic protease purified from the venom. Methods : The venom was subjected to chromatography using columns of Q-Sepharose and Sephadex G-75. The molecular weights of fibrinolytic proteases showing fibrinolytic zone in fibrin plate assay were determined in SDS-PAGE (Sodium dodecyl sulfate-polyacrylamide gel electrophoresis) The effects of inhibitors and metal ions on fibrinolytic protease and the proteolysis patterns of fibrinogen, gelatin, and bovine serum albumin were investigated. Results : 1) The fibrinolytic fractions of the three peaks isolated from Gloydius blomhoffii siniticus venom contained two polypeptides of 46 and 59 kDa and three polypeptides of 32, 18, and 15 kDa and a major polypeptide of 54 kDa, respectively. 2) The fibrinolytic activity of the purified protease of 54 kDA was inhibited by metal chelators, such as EDTA, EGTA, and 1,10-phenanthroline, and disulfhydryl-reducing compounds, such as dithiothreitol and cysteine. 3) Calcium chloride promoted the fibrinolytic activity of the protease, but mercuric chloride and cobalt(II) chloride inhibited it. 4) The fibrinolytic protease cleaved preferentially A${\alpha}$-chain and slowly B${\beta}$-chain of fibrinogen. It also hydrolyzed gelatin but not bovine serum albumin. Conclusions : The Gloydius blomhoffii siniticus venom contained more than three fibrinolytic proteases. The major fibrinolytic protease was a metalloprotease which hydrolyzed both fibrinogen and gelatin, but not bovine serum albumin.

  • Antithrombotic and fibrinolytic activity of natto was evaluated on platelet aggregation in vitro and in vivo. Natto showed inhibitory effects on platelet aggregation induced by adenosine 5’diphosphate (ADP) and collagen. Orally administered natto also showed fibrinolytic activity in hypercholesterolemia rats. Normal levels of natto, when administered for four weeks, shortened euglobulin clot lysis time (ECLT) and prolonged partial thromboplastin time (PATT) significantly compared to non-treated group. In addition, the natto treatment decreased total cholesterol in serum. These results showed that intake of normal levels of natto can elicit antithrombotic and fibrinolytic effects, suggesting its consumption may improve blood circulation.

보리 입국과 두류 첨가가 전통주의 품질과 혈전용해활성에 미치는 영향 (Effects of Barley Koji and Legumes on the Quality and Fibrinolytic Activity of Korean Traditional Rice Wine)

  • 혈전용해 효과를 가진 전통주를 개발하기 위하여 우선 혈전용해 활성이 있다고 확인된 몇 가지 곡류를 이용하여 입국들을 만들어 이들의 알콜 발효특성을 조사하였고 선정된 입국과 각종 두류 분말 및 기호성 음료 재료들을 술덧에 첨가하여 전통주를 제조한 후 품질 특성과 혈전용해 활성 등을 조사하였다. 보리 입국을 이용하여 전통주를 제조하였을 때 에탄올 생성량은 16.8%로 시판 입국으로 제조한 전통주(15.6%)보다 높았고 혈전용해 활성도 20.0 U로 제일 높았다. 또한 보리입국을 10% 첨가한 술덧에 녹두가루를 50% 첨가하고 대추를 3% 첨가하여 3$0^{\circ}C$에서 10일간 발효시켰을 때 혈전용해 활성이 26.0 U로 증가하였고 맛과 향도 우수하였다.

Effects of Water-soluble and Water-insoluble Fractions of Kimchi on the Alteration of Plasma Lipids and Fibrinolytic Activity in Middle-aged Healthy Subjects

  • Freeze-dried kimchi fractions, water-soluble (WSK) versus water-insoluble (WISK), on their effects on plasma lipids and for their fibrinolytic activities were studied. Nineteen middle-aged healthy subjects were assigned to WSK, WISK, or placebo during 6 weeks of kimchi supplementation. Subjects in the WSK and WISK groups were supplemented with 3 grams of kimchi pills (equivalent to 60 grams of fresh kimchi), while the placebo group was on their normal diets. LDL/HDL and atherogenic index decreased in the subjects who received kimchi pills, but not in those in the placebo group. The levels of plasma triglyceride were significantly decreased in the WSK group compared with those in the WISK group (p<0.05). However, there was no difference in the levels of total cholesterol and HDL between the two groups. Fibrinolytic activity of WISK was significantly higher than that of WSK.

Effects of Insect Crude Drugs on Blood Coagulation and Fibrinolysis System

  • The in vitro anticoagulant and fibrinolytic activities of crude extracts from insects were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. We prepared three types of extracts (water, methanol and ethylacetate) from 28 insects for use as raw materials for the activity assays. The fibrinolytic activity was tested using the fibrin plate method and the activated partial thromboplastin time and thrombin time were measured for blood clotting activity. With regards to the fibrinolytic system, water extracts of six kinds of insects displayed a remarkable level of activity with a plasmin-like action. The water extracts of [Catharsius molossus, Eupolyphaga sinensis, Huechys sanguinea, Mantidis $o\ddot{o}theca$, Mimela splendens, and Polistes mandarinus (Vespae Nidus)] exhibited the activity. On the other hand, the methanol extracts did not display any fibrinolytic activity. In terms of the coagulation system, an aqueous extract of silkworm Tongchunghacho (Paecilomyces japonica), Oxya japonica japonica and Buthus martensi (Scorpion) increased the clotting time significantly longer (181 times) than the control. These results suggest that crude drugs from insects are useful sources for the development of new drugs for use in treatments involving blood coagulation and fibrinolysis.

Purification and Biochemical Characterization of a Novel Fibrinolytic Enzyme from Streptomyces sp. P3

  • A novel proteolytic enzyme with fibrinolytic activity, FSP3, was purified from the recently isolated Streptomyces sp. P3, which is a novel bacterial strain isolated from soil. FSP3 was purified to electrophoretic homogeneity by ammonium sulfate precipitation, anion exchange, and gel filtration. FSP3 is considered to be a single peptide chain with a molecular mass of 44 kDa. The maximum activity of the enzyme was observed at 50℃ and pH 6.5, and the enzyme was stable between pH 6 and 8 and below 40℃. In a fibrin plate assay, FSP3 showed more potent fibrinolytic activity than urokinase, which is a clinical thrombolytic agent acting as a plasminogen activitor. The activity was strongly inhibited by the serine protease inhibitor PMSF, indicating that it is a serine protease. Additionally, metal ions showed different effects on the activity. It was significantly suppressed by Mg2+ and Ca2+ and completely inhibited by Cu2+, but slightly enhanced by Fe2+. According to LC-MS/MS results, its partial amino acid sequences are significantly dissimilar from those of previously reported fibrinolytic enzymes. The sequence of a DNA fragment encoding FSP3 contained an open reading frame of 1287 base pairs encoding 428 amino acids. FSP3 is a bifunctional enzyme in nature. It hydrolyzes the fibrin directly and activates plasminogen, which may reduce the occurrence of side effects. These results suggest that FSP3 is a novel serine protease with potential applications in thrombolytic therapy.

율무, 홍화, 아욱종자의 혈전용해 효소활성 및 감마선 조사의 영향 (Fibrinolytic Activities and Effects of Gamma-Irradiated on Seeds from Coix lacryma-jobi L. Carthamus tinctorius L. and Malva verticillata L.)

  • 미생물 및 동물에 비해 식물에서는 혈전용해효소에 대한 연구가 부족한 실정이며, 기존의 혈전용해효소가 가지는 혈전에 대한 비특이적, 부작용, 고가 등의 단점을 해결할 수 있는 새로운 혈전용해효소의 개발을 위하여 율무, 홍화, 아욱의 종자로부터 추출된 수용성 단백질의 혈전용해 활성을 조사하였다. 각각의 식물들로부터 추출된 조효소 용액은 기존 혈전 용해효소인 plasmin과 양성 대조군으로 하여 비교하여 fibrin 평판법으로 확인한 결과 피브린 응집을 효과적으로 분해하였다. 그 중 율무종자의 수용성 추출물의 혈전용해 활성은 양성 대조군인 plasmin과 비교하여 1.3배의 높은 활성을 나타내었다. 전체 수용성 단백질은 50-75% 에탄올을 이용하여 농축하였으며 율무의 혈전용해효소는 fibrin zymography를 수행하여 확인하고 직접 추출하였다. SDS-PAGE에 의하여 추출효소의 분자량을 측정한 결과 7.8 kDa으로 단일 polypeptide임을 확인하였으며, 효소 활성에 미치는 온도의 효과는 $50^{\circ}C$ 이상에서는 비교적 안정하였으나 더 낮은 온도에서는 급격히 효소활성이 감소하였다. 또한, 각종 단백질분해효소 저해제에 의한 영향을 조사한 결과 APMSF, PMSF, pepstatin A 그리고 TPCK에 강력하게 저해되는 것으로 보아 추출효소는 chymotrypsin과 유사한 serine protease의 하나로 생각되었다. 그러나 EGTA와 EDTA 처리에 의해서는 효소활성의 저해가 두드러지게 나타나지 않았다. 더욱이, 종자저장 중에 미생물에 의한 부패, 활력저하, 생리활성물질의 감소와 장기저장에 따른 에너지소비 증가 등이 문제가 되고 있어 저선량의 감마선 조사를 통해 율무, 홍화, 아욱의 종자로부터 혈전용해 효소활성에 미치는 효과 및 선량에 따른 차이를 조사하였는데 비조사 종자인 대조구와 비교하여 1 Gy, 4 Gy, 16 Gy, 32 Gy선량에서는 낮은 활성을 보였으면 반면에 8 Gy와 64 Gy의 선량에서는 더 높은 활성을 나타내었다. 이러한 결과는 Y선 조사가 종자의 혈전용해 활성을 향상시킬 가능성이 있을 것으로 생각된다. 이상의 모든 결과로 볼 때 율무의 추출 효소는 chymotrypsin-like serine protease에 속하는 혈전용해효소임을 확인할 수 있었다.

플라스크 배양에서 Bacillus subtilis BK-17의 혈전용해효소 생산에 대한 환경 및 영양 조건의 영향 (Effects of Environmental and Nutritional Conditions on Fibrinolytic enzyme Production from Bacillus subtilis BK-17 in Flask Culture)

  • The production of fibrinolytic enzyme from Bacillus subtilis BK-17 was studied in the shake flask cultures. The important medium components studied include nitrogen source, carbon source and inorganic salts. The environmental conditions include initial pH, temperature, shaking speed and working volume. Among various N-sources, C-sources and inorganic salts tested, soybean flour, D-glucose and Na2HPO4 gave the best results, and their optimal concentrations were 1.5%, 0.5% and 0.05%, respectively. The optimal pH and temperature were 9.0 and 37$^{\circ}C$. With decreasing working volume in the range of 25∼100ml in the 250ml flask or increasing shaking speed in the range of 100∼300rpm, the enzyme production was greatly enhanced. The enzyme activity under the optimal conditions was about 1400I.U./ml with urokinase as a standard.

Numerical modeling of thrombolysis - Effects of nozzle types and ejection velocities

  • Direct injection of a fibrinolytic agent to the intra-arterial thrombosis may increase the effectiveness of thrombolysis by enhancing the permeation of thrombolytic agents into the blood clot. Permeation of fibrinolytic agents into a clot is influenced by the surface pressure, which is determined by the injection velocity of fibrinolytic agents. Computational fluid dynamic methods were used in order to predict clot lysis for different jet velocities and nozzle arrangements. Firstly, thrombolysis of a clot was mathematically modeled based on the pressure and lysis front velocity relationship. Direct injection of a thrombolytic agent increased the speed of thrombolysis significantly and the effectiveness was increased as the ejecting velocity increased. The nine nozzles model showed about 20% increase of the lysed volume, and the one and seventeen nozzles models did not show significant differences. Secondly, thrombolysis was modeled based on the enzyme transport and the fluid flow equations, and quasi steady numerical analysis was performed. Clot lysis efficiency was also increased as injection velocity increased.

Physiological Effects of Hot Water Extract and Solvent Fractions of Carthamus tinctorius L.

  • In this study, we investigated the biological activities of Carthamus tinctorius including antioxidative, fibrinolytic, thrombin inhibitory, ${\alpha}$-glucosidase inhibitory and collagenase inhibitory activities. Carthamus tinctorius, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The hot water extract showed high antioxidative activity and thrombin inhibitory activity at 90.17% and 97.10% respectively. In the fraction activity tests, chloroform fraction showed the highest antioxidative activity at 81.85%. The fibrinolytic activity was strong only in the butanol fraction at 0.70 plasmin units/ml. The thrombin inhibitory activities of hexane, ethyl acetate and butanol fractions were 97.35%, 86.74% and 93.18% respectively. In collagenase inhibitory activity test, hexane fraction showed the highest activity at 87.78%. In conclusion, the hot water extract and solvent fractions of Carthamus tinctorius L can be used as a material for the development of biofunctional tea and foods respectively.
