통계학적인 방법과 왕겨를 기질로 사용하여 해양에서 분리한 Bacillus licheniformis LBH-52 를 사용한 carboxymethylcellualse의 생산조건 최적화 (Statistical Optimization for Production of Carboxymethylcellulase from Rice Hulls by a Newly Isolated Marine Microorganism Bacillus licheniformis LBH-52 Using Response Surface Method)

  • 왕겨를 기질로 사용하여 carboxymethylcellualse (CMCase)를 생산하는 미생물을 해수에서 분리하였으며 16S rDNA의 염기서열을 분석하여 동정한 결과, Bacillus lichemiformis로 확인되었다. CMCase를 생산하기 위한 최적의 탄소원과 질소원은 왕겨와 암모니움 나이트레이트이었다. 통계학적인 방법인 response surface method (RSM)을 사용하여 CMCase를 생산하기 위한 조건을 최적화하였다. 통계학적인 분석 결과, 왕겨가 균체의 생육에 미치는 영향이 가장 높았으며, 왕겨와 배지의 초기 pH가 CMCase 생산에 미치는 영향이 높았다. Design Expert Software를 사용하여 결과를 분석한 결과, 균체의 생장에 최적인 조건은 48.7 g/l 왕겨, 1.8 g/l 암모니움 나이트레이트, 배지의 초기 pH 6.8 및 배양온도 35.7$^{\circ}C$이었으나, CMCase의 생산에 최적인 조건은 43.2 g/l 왕겨, 1.1 g/l 암모니움 나이트레이트, 배지의 초기 pH 6.8 및 배양온도 35.7$^{\circ}C$이었다. 최적화된 조건에서 왕겨를 기질로 사용하여 B. lincheniformis LBH-52가 생산하는 CMCase는 79.6 U/ml이었다. 본 연구를 통하여 왕겨와 암모니움 나이트레이트를 CMCase를 생산하는 기질로 개발하였으며, 해수에서 분리한 미생물을 사용하여 생산기간을 3일로 단축하였다.

Saccharification of Foodwastes Using Cellulolytic and Amylolytic Enzymes from Trichoderma harzianum FJ1 and Its Kinetics

  • The study was targeted to saccharify foodwastes with the cellulolytic and amylolytic enzymes obtained from culture supernatant of Trichoderma harzianum FJ1 and analyze the kinetics of the saccharification in order to enlarge the utilization in industrial application. T. harzianum FJ1 highly produced various cellulolytic (filter paperase 0.9, carboxymethyl cellulase 22.0, ${\beta}$-glucosidase 1.2, Avicelase 0.4, xylanase 30.8, as U/mL-supernatant) and amylolytic (${alpha}$-amylase 5.6, ${\beta}$-amylase 3.1, glucoamylase 2.6, as U/mL-supernatant) enzymes. The $23{\sim}98\;g/L$ of reducing sugars were obtained under various experimental conditions by changing FPase to between $0.2{\sim}0.6\;U/mL$ and foodwastes between $5{\sim}20\%$ (w/v), with fixed conditions at $50^{\circ}C$, pH 5.0, and 100 rpm for 24 h. As the enzymatic hydrolysis of foodwastes were performed in a heterogeneous solid-liquid reaction system, it was significantly influenced by enzyme and substrate concentrations used, where the pH and temperature were fixed at their experimental optima of 5.0 and $50^{\circ}C$, respectively. An empirical model was employed to simplify the kinetics of the saccharification reaction. The reducing sugars concentration (X, g/L) in the saccharification reaction was expressed by a power curve ($X=K{\cdot}t^n$) for the reaction time (t), where the coefficient, K and n. were related to functions of the enzymes concentrations (E) and foodwastes concentrations (S), as follow: $K=10.894{\cdot}Ln(E{\cdot}S^2)-56.768,\;n=0.0608{\cdot}(E/S)^{-0.2130}$. The kinetic developed to analyze the effective saccharification of foodwastes composed of complex organic compounds could adequately explain the cases under various saccharification conditions. The kinetics results would be available for reducing sugars production processes, with the reducing sugars obtained at a lower cost can be used as carbon and energy sources in various fermentation industries.

Characteristics of Wet and Dried Distillers Grains on In vitro Ruminal Fermentation and Effects of Dietary Wet Distillers Grains on Performance of Hanwoo Steers

  • Two experiments were conducted to evaluate the nutrient composition, in vitro dry matter disappearance (IVDMD) and organic matter disappearance (IVOMD) of three kinds of distillers grains (DG); i) wet distillers grains (WDG, KRW 25/kg), ii) dried distillers grains (DDG, KRW 280/kg), iii) dried distillers grains with solubles (DDGS, KRW 270/kg) produced from tapioca 70% and rice 30%, and to evaluate dietary effects of WDG on the performance of Hanwoo steers. In Exp. 1, twelve-WDG, four-DDG and one-DDGS were collected from seven ethanol plants. Average crude protein, crude fiber, neutral detergent fiber, and acid detergent fiber of WDG, DDG, and DDGS were: 32.6%, 17.8%, 57.5%, and 30.2% for WDG, 36.7%, 13.9%, 51.4%, and 30.5% for DDG, and 31.0%, 11.9%, 40.3%, and 21.2% for DDGS (DM basis), respectively. The DDGS had a higher quantity of water-soluble fraction than WDG and DDG and showed the highest IVDMD (p<0.05) in comparison to others during the whole experimental time. The IVDMD at 0 to 12 h incubation were higher (p<0.05) in DDG than WDG, but did not show significant differences from 24 to 72 h. The same tendency was observed in IVOMD, showing that DG made from tapioca and rice (7:3) can be used as a feed ingredient for ruminants. Considering the price, WDG is a more useful feed ingredient than DDG and DDGS. In Exp. 2, 36 Hanwoo steers of 21 months ($495.1{\pm}91kg$) were randomly assigned to one of three dietary treatments for 85 days; i) Control (total mixed ration, TMR), ii) WDG 10% (TMR containing 10% of WDG, as fed basis), and iii) WDG 20% (TMR containing 20% of WDG, as fed basis). With respect to body weight and average daily gain, there were no differences between control and WDG treatments during the whole experimental period. Dry matter intake of control (9.34 kg), WDG 10% (9.21 kg) and 20% (8.86 kg) and feed conversion ratio of control (13.0), WDG 10% (13.2) and 20% (12.1) did not show differences between control and WDG treatments. Thus, the use of WDG up to 20% in TMR did not show any negative effect on the performance of Hanwoo steers.

Effect of Additive on the Chemical Composition of Tra Catfish (Pangasius hypophthalmus) By-product Silages and Their Nutritive Value for Pigs

  • Two experiments were conducted to determine i) the fermentation characteristics of catfish by-product (C) ensiled with rice bran (RB) or sugarcane molasses (M) in different ratios, and ii) the digestibility of the silages in growing pigs. In the ensiling experiment, there were three ratios of C, ensiled with RB or M, of 8:2, 7:3 and 6:4 (wet basis for C and air-dry basis for RB and M). The six treatments were CRB8:2, CRB7:3, CRB6:4, CM8:2, CM7:3 and CM6:4, with 3 replications per treatment and seven sampling times. The pH of CRB7:3 and CRB6:4 decreased (p<0.05) from the first week and stayed stable until 8 weeks of ensiling, but the pH did not decrease in CRB8:2. The pH of CM8:2, CM7:3 and CM6:4 decreased rapidly from the first week until week 8. Dry matter (DM) and crude protein (CP) contents were slightly lower when catfish by-product was ensiled with RB than with M. Ammonia content in all treatments increased (p<0.05) during ensiling. Lactic acid content in silages with molasses increased from the first week, with the highest value at week 4. However, the lactic acid content in CRB8:2 did not increase with time and had the lowest value of all treatments. Acetic acid proportions of total volatile fatty acids were low at day 0, with the highest value at day 7, decreasing slowly until 8 weeks. The butyric acid proportion was highest at day 0 and decreased up to week 8. The proportion of propionic acid increased during ensiling. The digestibility experiment had a 4${\times}$4 Latin-square design and included 4 castrated crossbreed (Yorkshire${\times}$Landrace) male pigs fed four diets. The basal diet (BD) included rice bran, broken rice and maize meal, and the other three diets included fish meal (FMD) or catfish by-product ensiled with rice bran (CRBD) or sugarcane molasses (CMD). The silages chosen were those which gave the best results in the ensiling experiment for each additive. The coefficient of total tract apparent digestibility (CTTAD) of DM and organic matter (OM) was not significantly different among treatments. The CTTAD of CP was not different among the silage diets, but was lower in BD (p<0.01). The CTTAD of ether extract (EE) was higher in the catfish by-product silage diets than in FMD and BD. There was no significant difference in the digestibility of DM, OM, CP and EE between the fish meal and the by-product silages. In conclusion, catfish by-product can be successfully preserved by ensiling. Moreover, the total tract apparent digestibility of OM, CP and EE in ensiled catfish by-product was comparable with that of fish meal.

Effects of Methylcellulose on Fibrolytic Bacterial Detachment and In vitro Degradation of Rice Straw

  • Two in vitro experiments were conducted to evaluate the effect of methylcellulose (MC) on i) bacterial detachment from rice straw as well as ii) inhibition of bacterial attachment and fiber digestibility. To evaluate the effect of MC on fibrolytic bacterial detachment (Exp 1), in vitro bacterial cultures with 0.1% (w/v) MC solution were compared with cultures without MC after 8 h incubation. The effect of MC on inhibition of bacterial attachment was determined by comparing with real-time PCR the populations of F. succinogenes, R. flavefaciens and R. albus established on rice straw pre-treated with 0.1% MC with those on untreated straw after incubation for 0, 6 and 12 h (Exp 2). The major fibrolytic bacterial attachment on rice straw showed significantly lower populations with either the addition of MC to the culture or pre-treated rice straw compared to controls (p<0.05). Also, the digestibility of rice straw with MC was significantly lower compared with control (p<0.05). The F. succinogenes population did not show detachment from rice straw, but showed an inhibition of attachment and proliferation on rice straw in accordance with a decrease of fiber digestion. The detachments of Ruminococcus species co-existed preventing the proliferations with subsequent reduction of fiber degradation by MC during the incubation. Their detachments were induced from stable colonization as well as the initial adhesion on rice straw by MC in in vitro ruminal fermentation. Furthermore, the detachment of R. albus was more sensitive to MC than was R. flavefaciens. These results showed the certain evidence that attachment of major fibrolytic bacteria had an effect on fiber digestion in the rumen, and each of fibrolytic bacteria, F. succinogenes, R. flavefaciens and R. albus had a specific mechanism of attachment and detachment to fiber.

주류의 풍미 및 품질 향상을 위한 야생 효모의 분리 및 특성분석 (Isolation and Characterization of Wild Yeasts for Improving Liquor Flavor and Quality)

  • non-Saccharomyces 효모는 야생효모로서, 다양한 효소를 세포 밖으로 분비하여 와인의 향기 성분 증가에 중요한 영향을 준다고 알려져 있다. 본 연구에서는 한국의 발효식품에서 분리된 효모 1,007주의 세포외 효소 활성을 조사하였다. 그 결과, β-glucosidase 566, glucanase 45, protease 401로 나타났으며, AA decarboxylase 활성 균주는 각 아미노산 별로 His 69, Tyr 306, Phe 171, Trp 23, Lys 197, Leu 198 균주로 나타났다. Cerulenin 과 TFL 내성 균주는 각각 563, 610주로, 15% 에탄올 내성 균주는 307주로 나타났다. 황화수소 생성균은 500주로 조사되었다. AA decarboxylation와 황화수소 저생성 균주 중 유용 효모 12 균주를 선발하여 26S rDNA 염기서열을 분석한 결과, C. tropicalis, H. opuntiae, H. uvarum, P. kudriavzevii, S. cerevisiae, W. anomalus로 각각 동정되었다. 12주의 효모 중 당 내성 우수 균주는 9주이며, 알코올 내성 우수 균주는 5 균주로 나타났다. 알코올 발효능은 Saccharomyces 효모는 8% 이상으로 나타났으며, non-Saccharomyces 효모는 3% 내외로 나타났다.

생선 폐기물의 혐기성 소화 처리(II) : Biochemical Methane Potential을 이용한 생분해도 평가 (Anaerobic Digestion of Fish Offal(II) : Evaluation of Biodegradability Using Biochemical Methane Potential)

  • 본 연구에서는 생선폐기물의 혐기성 소화처리를 위한 생선폐기물의 혐기성 생분해 특성을 평가하기 위해 serum bottle을 이용한 BMP(Biochemical Methane Potential) test를 수행하였다. 어판장에서 배출되는 생선폐기물 중 고등어 손질 시 발생되는 폐기물 중 내장을 제외한 부분과 갈치손질 시 발생되는 폐기물 중 내장을 제외한 부분, 생선손질 시 발생되는 내장류 3개 군을 실험대상 기질로 선정하여 사용하였다. 각각의 생선폐기물들을 분쇄 후 serum bottle에 11 50 ml, 100 ml, 150 ml씩 혐기적으로 넣은 다음 BMP test를 실시하였으며 실험의 정확도를 위하여 동일 시료에 대하여 3개씩 중복으로 조제하여 실험하였다. 배양시간에 따른 누적 메탄가스 생성량과 메탄생성속도를 평가하였다. 이들 값들은 기질의 특성에 따라 다르기는 하나 통상 휘발성 고형물 (Volatiles Solids: VS) 1 kg당 420-470 ml의 메탄가스를 생성하는 것으로 나타났다. 기질 종류별로 볼 경우 갈치와 고등어군에 비하여 내장 군이 고형물 단위 중량당 메탄발생량과 분해속도 측면에서 모두 높은 것으로 나타났다. 대상 시료에 대해 원소분석을 실시한 결과, C/N비는 5.2로 나타났다. 원소분석 자료를 근거로 계산한 이론적인 최종 메탄생성량은 $522ml{\cdot}CH_4/g{\cdot}VS$로 나타났다. 이를 본 실험에서 구한 실제 최종 메탄생성량과 비교해 볼 때 생분해도는 0.847로 평가되었다.

미세조류로부터 바이오 수소 생산을 위한 산(acid) 전처리의 최적화 (Optimization of bio-$H_{2}$ production from acid pretreated microalgal biomass)

  • 본 연구에서는 제 3세대 바이오매스 미세조류를 이용한 혐기성 암발효 수소 생산 과정에서 산 전처리의 최적화를 통계학적 실험방법인 반응표면법을 적용하여 도출 하였다. 1~3% (v/w)의 산 농도와 10~60 min 전처리 시간을 최적화 실험 범위로 설정하였으며 기질농도 76 g dcw/L와 초기 pH는 7.4로 고정하였고 수소발효 운전 중에 pH는 조절하지 않았다. 최적화 결과 HCl 1.2%와 반응시간 48 min에서 가장 높은 수소전환율인 36.8 mL $H_{2}/g$ dcw을 얻었으며 이때 가용화율은 18.9%로 나타났다. 정확도는 $R^{2}$=0.95로 매우 정확한 상관계수를 보였고 ANOVA test를 통해 예측된 수소전환율에 관련한 경험식은 a quadratic polynomial equation 으로 나타났으며 반응시간보다 산주입농도가 수소 생산에 큰 영향을 미치는 것으로 나타났다.

저염 건식절임법으로 제조한 오이지의 저온저장 중 이화학적$\cdot$미생물학적 품질특성 (Physicochemical and Microbial Characteristics of Oiji Prepared with Dry Salting Methods during Low Temperature Storage)

  • The physicochemical and microbial characteristics of Oiji prepared with dry salting method, which has been used industrially for industry, were investigated. Low salting and low storage temperature were employed:extremely low salting extremely low temperature; ESET $(5\%,\;0^{\circ}C)$, very low salting extremely low temperature;VSET $(10\%,\;0^{\circ}C)$, extremely low salting very low temperature; ESVT$(5\%,\;5^{\circ}C)$, low salting very low temperature; VSVT$(10\%,\;5^{\circ}C)$ and high salting low temperature;HSLT$(30\%,\;10^{\circ}C)$ for control. Acidity was lower, and pH was higher in VSET, in of which the fermentation pattern was similar with that of HSLT The time required to reach the optimum acidity ($0.3\%$ lactic acid) was longer delayed for VSET (168 days), than for compared to ESVT (51 days). During storage of Oiji, greenness (-a) as measured with of the Hunter color system wasshowed the highest in VSET, and the lowest while in ESVT, the lowest. Total microbial and lactic acid bacteria counts number were the lowest in HSLT and VSET and were the lowest than in other groups, while the highest in ESVT. Yeast was not detected in HSLT, but was the highest while in VSVT. E coli coliform and listeria were detected in the $5\%$ salting groups, although Salmonella was not detected in any of the all groups. Texture profile analysis demonstrated exhibited that fracturability and hardness were highest in HSLT and VSET, compared to the other groups. Scores of over-all preference for ESVT and HSLT were higher atwith 6.3 and 6.2, respectively, compared to the other products. Based on these results, lower saltiness less than $10\%$ and lower storage temperature (less than $5^{\circ}C$) condition was optimum for maximizing the better for good quality of industrial Oiji preparation in industry.

산분해 대두 단백질로부터 분리된 Esterase 생성균의 생육 및 효소생성 특성 (Esterase Production and Culture Characteristics of Bacteria Isolated from Acid Hydrolysed Soybean Protein)

  • 산분해 대두 단백질로 부터 식품 보존료인 p-hydroxybenzoic acid butyl ester를 분해하고, esterase 활성을 갖는 균주들을 분리, 동정하여 그들의 생육 및 효소 생성 특징을 조사하였다. 분리된 균주들은 전부 Baillus sp. 로 동정되었으며, specific growth rate는 균주에 따라 $0.844{\sim}1.213\;h^{-1}$였으며, esterase activity 는 Baillus sp. KB8 균주가 222 mU/ml로 대비균주인 B. subtilis ATCC6633의 21 mU/ml보다 10배 높은 활성을 보였으나 p-hydroxybenzoic acid butyl ester 분해 활성도는 $5.4{\sim}8.1\;mU/ml$로 균주에 따른 큰 차이는 볼 수 없었다. Baillus sp. KB8 균주의 배양시 NaCl 첨가는 생육을 저해시키며, esterase의 세포외 축적과 세포내 합성을 억제시키는 것으로 나타났으며, 특히 NaCl 첨가 농도의 증가는 세포외 축적을 더 억제시키는 것으로 나타났다. 플라스크 배양실험 결과 Baillus sp. KB8 균주의 효소 생성은 생육이 stationary phase 이후 점차 증가하여 배양 64시간에 420 mU/ml를 생성하였다. 온도에 대한 esterase의 활성은 $50^{\circ}C$에서 30분간 실활없이 유지되었으나 $70^{\circ}C$에서는 전부 실활되었다. $60^{\circ}C$$65^{\circ}C$ 에서의 효소 활성은 노출 시간에 따라 지수적인 실활 경향을 보여 3시간 후 $60^{\circ}C$에서 76%, $65^{\circ}C$에서는 3%의 잔존 효소활성을 보였다.
