Response of Saccharomyces cerevisiae to Ethanol Stress Involves Actions of Protein Asr1p

  • During the fermentation process of Saccharomyces cerevisiae, yeast cells must rapidly respond to a wide variety of external stresses in order to survive the constantly changing environment, including ethanol stress. The accumulation of ethanol can severely inhibit cell growth activity and productivity. Thus, the response to changing ethanol concentrations is one of the most important stress reactions in S. cerevisiae and worthy of thorough investigation. Therefore, this study examined the relationship between ethanol tolerance in S. cerevisiae and a unique protein called alcohol sensitive RING/PHD finger 1 protein (Asr1p). A real-time PCR showed that upon exposure to 8% ethanol, the expression of Asr1 was continuously enhanced, reaching a peak 2 h after stimulation. This result was confirmed by monitoring the fluorescence levels using a strain with a green fluorescent protein tagged to the C-terminal of Asr1p. The fluorescent microscopy also revealed a change in the subcellular localization before and after stimulation. Furthermore, the disruption of the Asr1 gene resulted in hypersensitivity on the medium containing ethanol, when compared with the wild-type strain. Thus, when taken together, the present results suggest that Asr1 is involved in the response to ethanol stress in the yeast S. cerevisiae.

  • 목질계 바이오 매스 전처리에 사용되는 ionic liquid는 전처리 후 100% 회수되지 않아 잔존하는 ionic liquid의 독성이 직접적으로 미생물 균주의 생육에 나쁜 영향을 미쳐 에탄올 발효의 수율 및 생산성을 저해하는 문제를 가지고 있다. 본 연구에서는 ionic liquid에 저해를 받지 않으며 높은 ethanol 생산 효율을 가진 균주를 얻고자 유도적 돌연변이 유발 실험을 진행하였다. 선별된 돌연변이 균주 D452-B2와 D452-S3는 3% [EMIM][Ac]가 포함된 배지에서 glucose 소비속도는 $4.5g{\cdot}l^{-1}{\cdot}h^{-1}$$4.4g{\cdot}l^{-1}{\cdot}h^{-1}$로 모균주인 S. cerevisiae D452-2 균주에 비해 6배 가량 증가하였으며, ethanol 생산성은 각각 $1.99g{\cdot}l^{-1}{\cdot}h^{-1}$$2.0g{\cdot}l^{-1}{\cdot}h^{-1}$로 27배 가량 증가하였다.

Inhibitory Effect of Lactococcus lactis HY 449 on Cariogenic Biofilm

  • Dental caries is caused by cariogenic biofilm, an oral biofilm including Streptococcus mutans. Recently, the prevention of dental caries using various probiotics has been attempted. Lactococcus lactis HY 449 is a probiotic bacterium. The aim of this study was to investigate the effect of L. lactis HY 449 on cariogenic biofilm and to analyze its inhibitory mechanisms. Cariogenic biofilm was formed in the presence or absence of L. lactis HY 449 and L. lactis ATCC 19435, and analyzed with a confocal laser microscope. The formation of cariogenic biofilm was reduced in cultures spiked with both L. lactis strains, and L. lactis HY 449 exhibited more inhibitory effects than L. lactis ATCC 19435. In order to analyze and to compare the inhibitory mechanisms, the antibacterial activity of the spent culture medium from both L. lactis strains against S. mutans was investigated, and the expression of glucosyltransferases (gtfs) of S. mutans was then analyzed by real-time RT-PCR. In addition, the sucrose fermentation ability of both L. lactis strains was examined. Both L. lactis strains showed antibacterial activity and inhibited the expression of gtfs, a nd t he d ifference b etween both strains did not show. In the case of sucrose-fermenting ability, L. lactis HY 449 fermented sucrose but L. lactis ATCC 19435 did not. L. lactis HY 449 inhibited the uptake of sucrose and the gtfs expression of S. mutans, whereby the development of cariogenic biofilm may be inhibited. In conclusion, L. lactis HY 449 may be a useful probiotic for the prevention of dental caries.

Polyphasic Microbial Analysis of Traditional Korean Jeung-Pyun Sourdough Fermented with Makgeolli

  • Jeung-pyun, a fermented rice cake, is prepared by fermenting rice sourdough using makgeolli, a traditional Korean rice wine, in the presence of yeast and lactic acid bacteria (LAB). The goal of this study was to conduct biochemical and microbial analyses of five different rice sourdoughs, each fermented with a different commercial makgeolli, using culture-dependent and culture-independent approaches. All sourdough samples fermented with different makgeolli for 6.5 h showed different profiles in pH, total titratable acidity, organic acid concentration, and microbial growth. LAB belonging to different genera were identified based on colony morphology on modified MRS and sourdough bacteria agar medium. PCR-denaturing gradient gel electrophoresis analyses of the five sourdoughs showed different bands corresponding to LAB and yeast. 16S/26S rRNA gene sequence analyses of the samples confirmed that the predominant LAB in the five fermented rice doughs was Lactobacillus plantarum, Lb. pentosus, and Lb. brevis. Various other Lactobacillus spp. and Saccharomyces cerevisiae were common in all five fermented samples. This study provides comprehensive and comparative information on the microflora involved in fermentation of rice sourdough and signifies the need to develop effective starters to enrich the quality of jeung-pyun.

  • Bacillus thuringiensis var. thuringiensis H1균을 4종류의 배지에서 배지 조성 및 pH 조건에 따라서 성장 양상과 아포와 내독소 결정체 생산을 조사했다. 1. 4개의 M-배지중 pH 9인 M-3배지에서 BTT의 증식량과 내독소 결정체 및 아포 생산수가 가장 높았다. 150ml배양에서 BTT의 생산량은 3.218g이었고, 생존 아포수는 ml당 $3.3{\times}10^{10}$개였고, 내독소 결정체의 회수된 무게비는 20.05%였다. 2. M-1배지에서 BTT의 세대시간은 평균 47.6분이었고, M-2에서는 평균 49.6분, M-3에서는 평균 132.9분, M-4에서는 평균 110.2분이었다. 3. M-배지의 초기 pH는 72시간 배양 후에 pH $6.5{\sim}7.5$로 변화하였으며, 최적 배양 pH는 $6.5{\sim}7.0$이었다.

재조합 Aspergillus niger에 의한 글루콘산나트륨의 산업적 생산 (Overproduction of Sodium Gluconate Using the Recombinant Aspergillus niger)

  • Polymerase chain reaction(PCR) was conducted to obtain the gene encoding glucose oxidase(GOD) from Aspergillus niger(ATCC 2110) and the DNA sequence determined was coincided with published GOD sequence from A. niger. Recombinant transforming vector containing GOD and hygromycin B(hyg.B) resistant gene(hph) was constructed and used for further transformation of A. niger ATCC 2110. Selectivity of hyg.B against A. niger differed depending on which media were used i.e., nutrient-rich media such as potato dextrose agar(PDA) and complete medium(CM) showed only 50% growth inhibition at 400 $\mu$m ml$^-1$ of hyg.B while the minimal media inhibited mycelial growth completely at 200 $\mu$m ml$^-1$ of hyg.B. Twenty to sixty putative transformants were isolated from the hyg.B-containing minimal top agar, transferred successively onto alternating selective and nonselective media for a mitotic stability of hyg.B resistance and, then, single-spored. Among the stable transformants, the transformant(GOD1-6) grown by flask culture showed the considerable increase of extracellular GOD activity, which was estimated to the degree of 50% - 100% comparing to that of wild type. Transformation of tGOD1-6 was resulted from integration of the vectors into heterologous as well as homologous regions of the A. niger genome. Southern blot analysis revealed that there were two independent integrations of vector into fungal genome and one into the GOD gene due to homologous recombination. In addition, GOD activity and sodium gluconate production when tGOD1-6 was fed-batch fermented were enhanced 11 fold and 2.25 fold, respectively, compared to that of the wild type.

Bacillus polymyxa KS-1에 의한 다당류 KS-1 생산의 발효 조건 (Cultural Conditions of Exopolysaccharide KS-1 Produced by Bacillus polymyxa KS-1)

  • 토양으로부터 분리 동정된 균주인 Bacillus polymyxa KS-1을 이용하여 이들이 생산하는 미생물 생 다당듀 KS-1의 생산을 위한 배지 및 배양조건을 조사하였다. 다당류 KS-1의 생산을 위한 애지의 조성은 증류수 1 liter당 glucose 30g, yeast extract $2.5g, KH_2PO_4 2.5g, NaCl 0.5g, MgSO_4.7H_20 0.3g, CaC0_3 0.05g, FeSO_4.7H_2O 0.05g, 그리고 MnS0_4 . 4H_20 0.05g$ 이 첨가된 배지에서 높은 정성 벚 다탕류 생산 을 얻을 수 있었으며, 이때 배양조건은 초기 pH와 배양온도는 각각 7.0과 $30^{\circ}C$, C에서 다당류 생산이 가 장 좋은 갯으로 조사되였다. 또한 이상의 최적조건 에서 발효조를 이용한 경우 통기량 빛 교반속도를 각각 2vvm과 400rpm으로 조절하여 배양한 경우 생산량은 liter당 약 10.3g의 다당류 KS-1이 얻어졌다.

  • S. marcescens QM 81466 균주는 chitin 분해 효소(1mg/Lmedium)를 선택적으로 높게 생성시킬 수 있는 균주로서, chitin을 N-acetyl-$\beta$-D-glucosa­m mine(NAG)으로 효소적 가수분해를 할 때 chitinase와 chitobiase의 두 가지 가수분해 효소계를 구 성시킨다. 본 연구에서는 이 균주의 chitinase/chitobiase 생성을 위한 chitin 입자크기에 대한 최적화와, 회분 발효계에서 이 균주의 세포 밀도 배양에 따른 두 효소 생성의 변화를 조사하여 NAG 생산성의 증대를 시도하였다. 아울러. chitin과 CM­ chitin이 chitinase/chito biase 생성비 와 NAG 생성 에 미치는 영향을 검토하였는데, CM-chitin을 colloidal 및 결정성 chitin 대신에 사용했을 때, chitinase 활성을 약 7~10U/mL 증가시켰다. 이 경우에 있어서, chitinase/chitobiase의 비는 9:1로 서 NAG의 생성량이 3.0g/L로서 높게 나타났다.

Molecular Cloning and Expression of Grass Carp MyoD in Yeast Pichia pastoris

  • MyoD, expressed in skeletal muscle lineages of vertebrate embryo, is one of muscle-specific basic helix-loop-helix (bHLH) transcription factors, which plays a key role in the determination and differentiation of all skeletal muscle lineages. In this study, a cDNA of grass carp MyoD was cloned and characterized from total RNA of grass carp embryos by RT-PCR. The full-length cDNA of grass carp MyoD is 1597 bp. The cDNA sequence analysis reveals an open reading frame of 825 bp coding for a protein of 275 amino acids, which includes a bHLH domain composed of basic domain (1-84th amino acids) and HLH domain (98-142th amino acids), without signal peptide. Then the MyoD cDNA of grass carp was cloned to yeast expression vector pPICZ$\alpha$A and transformed into P. pastoris GS115 strain, the recombinant MyoD protein with a molecular weight of about 31KD was obtained after inducing for 2d with 0.5% methanol in pH 8.0 BMGY medium, and the maximum yield was about 250 mg/L in shaking-flask fermentation. The results were expected to benefit for further studies on the crystal structure and physiological function of fish MyoD.

Mycoflora of Soybeans Used for Meju Fermentation

  • Diverse fungi are present in Korean traditional meju and they are known to play an important role in fermented soybean products. To determine the origin of the fungi in meju, we examined the mycoflora of soybeans from 10 traditional meju factories. The samples were untreated or treated with sodium hypochlorite, and placed on malt extract agar (MEA), dichloran 18% glycerol agar (DG18), and dichloran rose bengal chloramphenicol agar (DRBC) medium. A total of 794 fungal strains were isolated and they were identified as 41 genera and 86 species. From sodium hypochlorite untreated soybeans, the genera, Cladosporium (55%), Eurotium (51%), Fusarium (33%), Penicillium (22%), and Aspergillus (exclusion of Eurotium) (20%), were mainly isolated, and Eurotium herbariorum (22%), Eurotium repens (18%), Cladosporium tenuissimum (18%), F. fujikuroi (18%), Aspergillus oryzae/flavus (7%), and Penicillium steckii (6%) were the predominant species. In case of sodium hypochloritetreated soybeans, Eurotium (31%) and Cladosporium (5%) were frequently isolated, but Aspergillus (excluding Eurotium), Penicillium and Fusarium which were frequently isolated from untreated soybeans, were rarely isolated. Eurotium herbariorum (21%), Eurotium repens (8%), and Cladosporium tenuissimum (3%) were the predominant species. Of the 41 genera and 86 species isolated from soybeans, 13 genera and 33 species were also found in meju. These results suggest that the fungi on soybeans may influence the mycoflora of meju.
