Fabrication and Characterisation of a Novel Pellicular Adsorbent Customised for the Effectvie Fluidised Bed Adsorption of Protein Products

  • A dense pellicular solid matrix has been fabricated by coating 4% agarose gel on to dense zironia-silica(ZS) spheres by watr-in-oil emulsification . The agarose evenly laminated the ZS bead to a depth of 30㎛, and the resultin gpellicular assembly was characterised by densities up to 2.39g/mL and a mean particle dimeter of 136 ㎛. In comparative fluidisation tests, the pellicular solid phase exhibited a two-fold greater flow velocity than commercial benchmark ad-sorbents necessary to achieve common values of bed expansion. Furthermore, the perlicular parti-cles were characterised by improved qualities of chromatographic behaviour, particularly with re-spect to a three-fold increase in the apparent effective diffusivity of lysozyme within a pellicular assembly modified with Cibacron Blue 3GA. The properties of rapid protein adsorption/desorp-tion were attributed to the physical design and pellicular deployment of the reactive surface in the solid phase. When combined with enhanced feedstock throughput, such practical advantages recommend the pellicular assembly as a base matrix for the selective recovery of protein products from complex, particulate feedstocks(whole fermentation broths, cell disruptates and biological extracts).

팽이버섯 부산물 발효에 따른 한우 거세우 반추위 성상 및 소화율에 미치는 영향 (Effects of Applying Microbial Additive Inoculants to Spent Mushroom Substrate (Flammulina velutipes) on Rumen Fermentation and Total-tract Nutrient Digestibility in Hanwoo Steers)

  • We inoculated a spent mushroom substrate from Flammulina velutipes (SMSF) with a microbial additive and assessed the effects on chemical composition, ruminal fermentation parameters, and total-tract nutrient digestibility. In Exp. 1, three cannulated Hanwoo steers were used in an in situ trial to determine the degradation kinetics of dry matter (DM) and crude protein (CP). In Exp. 2, three Hanwoo steers were randomly assigned to experimental diets according to a $3{\times}3$ Latin square for a 3-week period (2 weeks for adaptation and 1 week for sample collection). Experimental diets included the control diet (3.75 kg/d formulated concentrate mixture + 1.25 kg/d rice straw), SMSF diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d SMSF), and inoculated SMSF (ISMSF) diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d ISMSF). The chemical composition of ISMSF did not differ from that of SMSF. Microbial additive inoculation decreased pH (P<0.05) and improved preservation for SMSF. The percentages of DM, neutral detergent fiber (NDF), and acid detergent fiber (ADF) in ISMSF were slightly lesser than those in SMSF. Ruminal fermentation characteristics and total-tract nutrient digestibility were not affected by diet. Overall, microbial additive inoculation improved preservation for SMSF and may allow improved digestion in the rumen; however, the total digestible nutrients (TDN) of SMSF and ISMSF diets were slightly lesser than the control diet. The ISMSF can be used as an alternative feedstuff to partially replace formulated concentrate feed.

  • Streptococcus lactis IFO 12007의 nisin 생산을 위한 발효조건을 검토하였다. Nisin 생성을 위한 포도당의 농도는 60g/ι이며 pH와 온도는 각각 6.5와 3$0^{\circ}C$이었다. 이 조건에서 최대 2,000IU/$m\ell$의 생산량을 보이며 이 때 specific glucose uptake rate: 0.59g/g/h, specific nisin productivity는 34924iu/g/h, growth yield는 0.24, 7시간 후 균체 생산량은 4.81g/ι이었다. 비성장속도는 온도와 pH에 의하여 영향을 많이 받으며 증식활성화 에너지는 1.35kca1/ mole이었다. 유가배양에 의하여 1420IU/$m\ell$의 nisin을 생산하며 연속배양은 0.38h$^{-1}$까지 가능하고 이때 nisin 농도는 740IU/m1, specific nisin productivity는 45000IU/g/h, true growth yield 는 0.144, maintenance energy는 207mg glucose/g-cell/h 이었다.

Comparison of in vitro digestibility and chemical composition among four crop straws treated by Pleurotus ostreatus

  • Objective: The effects of Pleurotus ostreatus on the feed utilization of broad bean stalks (BBS), rape straw (RS), paddy straw (PS), and corn stalk (CS) was examined. Methods: The four roughages were co-cultured with Pleurotus ostreatus. The chemical composition; enzyme activities of laccase, carboxymethylcellulase (CMCase) and xylanase; carbohydrate and protein fractions (based on The Cornell Net Carbohydrate and Protein System [CNCPS]) were assessed at different days after inoculation (7, 14, 21, 28 d) and un-inoculated roughages (control, 0 d). The digestibility of nutrient components and the gas production of roughage with various incubation times were monitored at 0, 2, 4, 6, 9, 12, 24, 36, 48, 60, and 72 h using an in vitro ruminal fermentation method. Results: A higher CMCase activity (0.1039 U/mL) and earlier time to peak (14 d) were detected in Pleurotus ostreatus cultured with CS (p<0.05). Significantly, the incubation length-dependent responses of cumulative gas production were observed from 24 to 72 hours post fermentation (p<0.05), and these incubation length-dependent effects on cumulative gas production of PS and CS appeared earlier (24 h) for PS and CS than those (48 h) for BBS and RS (p<0.05). The fast-degradable carbohydrate (CA) content for all four roughages significantly increased over time (p<0.05). Nonetheless, increased degradation efficiency for CA treated with Pleurotus ostreatus was detected at both 21 and 28 days of incubation (p<0.05). With the exception of PS (p<0.05), there were no significant difference among the roughages (p>0.05) in slowly-degradable carbohydrate (CB2) at different incubation times (p<0.05). Conclusion: Assessment of the alterations in chemical composition, CNCPS system fractions, and the fermentation kinetics after biological pretreatment may yield a valuable database for evaluating the biological pretreatment of Pleurotus ostreatus in ruminant feed.

Effect of Sodium Hydroxide plus Hydrogen Peroxide Treated Mustard (Brassica campestris) Straw Based Diets on Rumen Degradation Kinetics (In sacco), Fermentation Pattern and Nutrient Utilization in Sheep

  • Two experiments were conducted to determine the effect of alkaline hydrogen peroxide (AHP) treatment (1% NaOH+1.5% $H_2O_2$; 1 AHPMS, 2% NaOH+1.5% $H_2O_2$; 2AHPMS) on rate and extent of degradation of mustard straw (MS) in sacco in sheep, and its in vivo digestion and ruminal fermentation characteristics when fed to sheep with concentrate (200 g per sheep daily). The treatment of straw with 1 and 2% AHP increased its sodium content by 148 and 296% to that of untreated straw (UMS). There was significant decrease in NDF and hemicellulose contents of AHP treated straw and increase in cellulose and lignin contents. Phenolic acids like ferrulic, $\rho$-coumaric and o-coumaric significantly (p<0.001) reduced by AHP treatment of mustard straw. In first experiment the in sacco degradation of DM, OM and NDF was significantly (p<0.01) greater for 2 AHPMS than for UMS at all incubation periods. The disappearance of nutrient from 1 AHPMS and 2 AHPMS treated straws continue to increase up to 96 h whereas in UMS the peak disappearance was found at 48 h. By using the equation {(y=a+b) ($1-e^{-ct}$)} the degradation rates (c) for DM, OM, and NDF were significantly higher for UMS than AHP treated straws. Level of alkali (1 and 2%) had significant effect on degradation characteristics (a, b, c and $P_{0.05}$) of DM and NDF fraction of MS. However, the effect was not pronounced on OM fraction of MS. In feeding experiment, the intake of nutrients for DM, OM, cell wall constituents and energy was higher on 2 AHPMS, whereas no effect on the digestibility of these nutrients was observed. The apparent nitrogen retention was higher (p<0.05) both in 1 and 2 AHPMS groups. Water intake by animals was significantly increased due to AHP treated mustard straw feeding. Rumen liquor pH was higher in 2 AHPMS fed animals. The $NH_3-N$ of rumen liquor was not affected by feeding of AHP treated MS based diets. Total VFA concentration was significantly (p<0.01) higher in UMS fed group. The fractional out flow rate of DM was higher (p<0.05) in animals fed on 2 AHPMS diets compared to UMS and lAHPMS fed groups. The population of large holotrichs was higher (p<0.05) on AHP treated MS fed diets compared to UMS. The study indicated that treatment of mustard straw with AHP changed its chemical composition towards a better feed. The nutritive value of 2% AHP treated mustard straw was better in terms of dry matter intake and apparent nitrogen retention. The higher in sacco DM, OM and NDF disappearance however, was not confirmed by in vivo data in this study.

Arachidonic Acid 생산을 위한 Mortierella alpina 곰팡이의 심부 및 고체 발효 특성 연구 (Characteristics of Submerged and Solid-State Fermentations for Production of Arachidonic Acid Mortierella alpina)

  • 본 연구의 목적은 최근 DHA와 더불어 분유첨가제 등 기능성 신소재로서 수요가 증가되고 있는 arachidonic acid (AA)를 곰팡이 Mortierella alpina ATCC 32222를 이용하여 실질적으로 값싸게 생산할 수 있는 고체발효공정을 개발하기 위한 것이다. 본 연구에서는 회분식 심부 및 고체발효연구를 통하여 균체성장과 AA 생성에 대한 패턴을 조사하였는데, 심부발효에서는 포도당과 효모추출물을 그리고 고체발효에서는 농산 부산물인 저렴한 쌀겨를 원료로 하여 실험하였다. 심부발효 및 고체발효의 속도론적 관점에서 두 공정 모두 균체성장과 AA 생성간에 전형적인 mixed-growth associated pattern이 얻어졌다. 고체발효 배지의 최적화를 위하여 carbon to nitrogen (C/N) ratio를 6.5-20 범위로 조절하여 실험해 본 결과 쌀겨에 탄소원인 포도당을 첨가해 주지 않은 경우 즉, 6.5에서 가장 좋은 결과가 얻어졌다. 또한 sodium glutamate를 쌀겨에 소량 $(0-4\%,\;w/w)$ 첨가했을 때 보고된 문헌과는 달리 균체성장은 오히려 감소되는 경향을 보였으며, 고온에서 볶은 참깨로부터 얻어진 sesame oil $(7.5\%,\;w/w)$을 첨가했을 경우 AA 생산성은 약 1.2배가 향상되었다.

In Vitro 반추위 발효를 통한 볏짚 분해와 섬유소 박테리아 군집 변화의 관계 연구 (Study on the Relationships between Rice Straw Degradation and Changes of Fibrolytic Bacteria Population by in Vitro Rumen Fermentation)

  • 본 연구는 반추동물이 볏짚을 소화하는 과정 중에 주요 섬유소 분해 박테리아의 분포 변화를 연구하여 반추위내 볏짚이용효율을 증진시키기 위한 진보적 연구 자료를 제공하고자 실시하였다. 본 연구를 수행하기 위하여 분쇄 볏짚을 기질로 사용한 In vitro 반추위 발효를 실시하였으며, 배양 0. 4, 8, 12, 48시간에 볏짚 건물 소화율 및 섬유소 분해 박테리아 균수 변화를 조사하였다. 섬유소 분해 박테리아 균수는 F. succinogenes. R. albus와 R. flavefaciens를 대상으로 Quantitative real-time PCR을 이용하여 16S rDNA의 copy No의 log값(Log copy No)을 측정하였다. 섬유소 분해 박테리아의 총 균수는 F. succinogenes, R. albus와 R. flavefaciens의 볏짚 표면 부착 및 배양액내 부유 박테리아의 총합으로 배양 시간에 따라 증식을 하여 24시간에서 최고 군집(29.0 Log copy No)을 형성하고, 이후 감소하였다. 이에 볏짚 소화율도 24시간까지 점점 큰 폭으로 증가하였고, 이후에는 점점 둔화되는 경향을 나타냈다. F. succinogenes, R. flavefaciens 및 R. albus는 배양 시작 즉시 볏짚 표면에 부착이 발생됨을 발견하였으며, 이때 이들의 부착 군집의 비율은 각각 34.5%, 84.4% 및 67.9%로 차지하였다. 그리고 이들 섬유소 분해 박테리아의 볏짚 부착 비율은 모든 균수가 공히 배양 4시간째부터 94.7% 이상 최고 수준에 이르렀고, 부착 균수는 균종에 따라 각각 배양 12시간 또는 24시간째에 최고 수준을 나타냈다. F. succinogenes, R. flavefaciens 및 R. albus의 최고 수준의 볏짚 표면 부착 속도는 배양 12시간에 시작되어 배양 24시간에 각각 10.33, 9.28 및 8.30 Log copy No/h/g DM에 도달하였다. 그리고 섬유소 분해 속도는 표면 부착 속도보다 좀 늦은 배양 24시간에 0.95% DM/h로 최고 정점에 도달하였다. 따라서 본 연구결과는 반추위 발효 진행 과정에서 섬유소 분해 박테리아 총균의 증식에 따라 조사료의 소화가 증가되며, 상당히 많은 섬유소 분해 박테리아들이 조사료 유입과 동시에 표면 부착이 발생하고, 이들 부착 섬유소 분해 박테리아의 활성화가 먼저 진행된 후에 섬유소 분해 활성화가 이루어져야 조사료 소화의 극대화를 가져올 수 있다는 사실을 제시하여 주었다.

영지의 액체배양에 의한 세포외 다당 생산의 동력학적 특성 (Batch Kinetics of Exo-polysaccharide Production by Submerged Cultivation of Ganoderma lucidum)

  • 영지버섯의 세포외 다당발효중 동력학적 특성을 기질(포도당, 전분), 기질농도$(1{\sim}7%)$ 및 계대배양(3회)의 함수로서 조사하였다. 영지버섯 균사체 증식은 logistic 모델이 Monod 모델 및 two- thirds power 모델과 비교하여 실험값에 잘 일치하였고, 기질 및 생성물은 Luedeking-Pirt 식에 의하여 잘 설명되었다. 또 다당 생성의 발효 기작은 증식연동형과 비증식 연동형이 함께 존재하는 혼합형이었으나 기질에 상관없이 비증식연동형 기작이 더 중요하였다. Glucose는 농도 증가에 따라 다당생성 및 기질소비의 기작이 증식연동형이 감소하고 비증식연동형이 증가하는 경향을 보였다. 그러나 starch를 사용하였을 경우는 glucose와는 달리, 기질소비의 증식연동형과 비증식연동형 기작이 모두 증가하여 높은 기질 이용성을 보였다. 아울러 starch배지에서는 glucose배지에서 보다 비증식속도의 증가와 계대배양시의 안정성을 보였다. 따라서 영지버섯의 배양시 starch배지는 비증식연동형에 의한 균체 생육 및 다당생성의 생합성 촉진에 의해 이들 생산성 및 계대배양시의 안정성을 증가시키는 것으로 생각되었다 최대의 균사체 생육 및 다당 생산은 각각 9.463 및 10.410 g/l로, 7% starch을 함유한 배지에서 $30^{\circ}C$로 7일간 진탕배양하였을 때 얻어졌다.

Use of In vitro Gas Production Technique to Investigate Interactions between Rice Straw, Wheat Straw, Maize Stover and Alfalfa or Clover

  • Measurement of gas produced during in vitro fermentation was used to investigate the fermentation characteristics and interactions of rice straw, wheat straw or maize stover mixed with alfalfa or clover at proportions of 100:0, 75:25, 50:50, 25:75 and 0:100, respectively. Cumulative gas production was recorded at 2, 4, 8, 12, 16, 24 and 48 h of incubation, and the Gompertz function was used to describe the kinetics of gas production. In vitro dry matter and organic matter disappearances (IVDMD and IVOMD) were determined after 48 h incubation. The rate of gas production of clover was higher (p<0.05) than that of rice straw, wheat straw, maize stover and alfalfa when straws and hays were incubated separately. Increasing the proportion of alfalfa in the straw-alfalfa mixtures increased (p<0.05) the rates, but not the maximum volume of gas production. However, both rate and the maximum volume of gas production were increased (p<0.01) as the proportions of clover increased in the straw-clover mixtures. The cumulative gas production at 48 h, IVDMD and IVOMD showed no consistent interaction effects between different mixtures of cereal straws and hays. The extent of interactive effects was affected by the types of cereal straw, legume hay and their proportions in the mixture. The appropriate combination for the mixture of rice straw or maize stover with leguminous hays was 75:25 and 25:75, respectively. The better combination occurred at a proportion of 50:50 for the mixture of wheat straw and alfalfa. We conclude that the suitable proportion of low-quality straw and high quality legume hay combination should be considered in the ration formulation system of ruminants according to the extent of positive interactive effects.

어장유의 속성발효와 동력학적 고찰 (Rapid Fermentation of Fish Sauce and Its Kinetics)

  • A study on the rapid fermentation of fish sauce has been carried out for effective utilization of sardine. The frozen sardine was thawed at room temperature, chopped, homogenized with equal amount of water and then hydrolyzed by addition of commercial proteolytic enzymes such as bromelain, papaya protease, ficin and a enzyme mixture under different conditions of hydrolysis. The effect of wheat gluten for masking fishy odor and color development during thermal treatment were also tested. The reaction mixture was heated for 30 minutes at $100^{\circ}C$ for enzyme inactivation, pasteurization and color development and then centrifuged for 20 minutes at 4,000 rpm. Finally, table salt and benzoic acid were added for bacteriostatic effect. The results were summarized as follows ; 1. The hydrolyzing temperature, time, pH and the concentration of enzymes based on the weight of whole sardine for optimal hydrolysis were as follows: autolysis, $52.5^{\circ}C$, 4 hours, pH 8.0: with $0.25\%$ bromelain, $52.5^{\circ}C$, 4 hours, pH 6.6 :with $0.25\%$ ficin, $52.5^{\circ}C$, 4 hours, pH 6.8: with $0.3\%$ papaya protease, $52.5^{\circ}C$, 4 hours, pH 6.6: with $6\%$ enzyme mixture, $52.5^{\circ}C$, 4 hours, pH 6.9, respectively. But pH control was not much beneficial in increasing yield. 2. The hydrolytic reaction of chopped sardine with proteolytic enzymes could be interpreted as a first order reaction that devided into 2 periods with different reaction rate constsnts. $Q_{10}$ values of the first period prior to 4 hours were 1.23 to 1.31, and those of post 4 hours were 1.25 to 1.55. The corresponding activation energies were $1.81{\times}10^4\;to\;2.34{\times}10^4\;kJ/kmol$ and $1.92{\times}10^4\;to\;3.77{\times}10^4\;kJ/kmol$, respectively. 3. The reasonable amount of $75\%$ vital wheat gluten for addition was $9\%$ of chopped sardine. 4. The dark brown color was mainly developed during the thermal treatment for 30 minutes at $100^{\circ}C$ and not changed during storage.
