The objective of this study was to evaluate the accuracy and efficiency of different methods used for the fermentable sugar assay in the production of Makgeolli sul-dut. In the initial stage of fermentation, Ipguk treatment produced a higher alcohol content compared to the Nuruk treatment. However, the alcohol content was not significantly different between the two starters at the final stage of fermentation. Acidity in the Ipguk treatment was higher than that of Nuruk throughout the fermentation period. After analyzing the fermentable sugars using dinitrosalicylic acid (DNS), Fehling's method, refractometer, glucose kit, and high performance liquid chromatography (HPLC), it was confirmed that the HPLC method was the most accurate for fermentable sugar quantification. In both types of starters, DNS and Fehling's methods showed results comparable to HPLC in terms of fermentable sugar content, while the glucose kit and refractometer analyses showed relatively large discrepancies, indicating that the Fehling's method could also be effective for the analysis of fermentable sugars in the manufacture of Makgeolli.
In order to study the prediction of shelf-life and the relation between the initial reducing sugar content (So) and the fermentation period (T) to produce 0.75% acid in kimchi, kimchis were prepared with Chinese cabbage from which fermentable sugars were removed by 0%, 30%, 50%, 74. In kimchis with 2.3%, 0.97%, 0.60%: initial reducing sugar content, fermentation periods to produce 0.75% acid took 2, 7, 12 days, respectively. This relation can be expressed as the following equation; T= -16.82 LogSo+7.66. Kimchi with cabbage removed by about 80% fermentable sugar showed out about 0.8% total acidity during 30 days's storage at $25^{\circ}C$.
For obtain fermentable sugar, we conducted acid hydrolysis with lignocellulosic biomass without enzyme. The lignocellulosic biomass used pinus rigida and Palm residues (EFB; empty fruit bunches). In the acid hydrolysis, we consider the hydrolysis condition to reduce a denatured sugar. So this study was conducted 2-step acid hydrolysis. First-step hydrolysis used high concentration (72 wt%) sulfuric acid at $80^{\circ}C$. At the condition, we obtained 11.49 wt%, 32 wt% glucose conversion for pinus rigida and EFB. After the step, the liquor was dilute until 9~15 wt% acid concentration and conducted second hydrolysis at $50{\sim}120^{\circ}C$. In the second hydrolysis, we obtained maximum glucose conversion (pinus rigida 86.8 wt% (39 g/L) and EFB 95.3 wt% (32.4 g/L)) at 9 wt% acid concentration and $120^{\circ}C$ for 80 min. All samples through the process are analyzed on the basis of mass balance.
The microalgae have cellulose as a main structural component of their cell wall and the lignin content in microalgae is much lower than other lignocellulosic biomass. Therefore, fermentable sugar production from microalgae (Tetraselmis KCTC 12236BP) can be carried out under pretreatment without high temperature and high pressure. It was investigated that the effect of hot-water pretreatment using sulfuric acid for lipid extracted algae which is expected to be a next generation biomass. The effects of three major variables including extraction temperature, acid concentration and time on the enzymatic hydrolysis were investigated. Among the tested variables, temperature and acid concentration showed significant effects and optimum pretreatment conditions for the economic operation criteria were obtained as follows: reaction temperature of $120^{\circ}C$, sulfuric acid concentration of 2 mol and pretreatment time of 40 min. Under the optimum conditions of acidic hot water pretreatment, experimentally obtained hydrolysis yield were 95.9% which showed about 2.1 fold higher compared with enzymatic hydrolysis process. Therefore, acid pretreatment under mild condition was proven to be an effective method for fermentable sugar production from lipid extracted microalgae.
A cell-recycled continuous fermentation process was studied to produce ethanol from molasses using Sacchoromyces uvarum ATCC26602 at 35 $^{\circ}C$. The fermentation system was divided into two stages in order to reduce the inhibitions of ethanol and substrate for cell growth and fermentation rate. The first reactor was aerated at the rate of 0.12 vvm whilst the second was kept anaerobic. In medium composition studies, it was revealed that inorganic nutrient supplement to the diluted molasses with 14% fermentable sugar was not needed for the fermentation, however, phosphate limitation was observed when cell propagation was contemplated. By using the cell-recycled continuous fermentation system, 14.5 hour was required to produce 8.4-9.0% (v/v) of ethanol from diluted molasses containing 14% of fermentable sugar. The ethanol productivity was 6.2g/$\ell$hr with the yield of 88.1-94.4% to the theoretical value.
This study reviewed on the research trend of sources and utilization of the byproducts(Lignin) from bioethanol production process with lignocellulosic biomass such as wood, agri-processing by-products(corn fiber, sugarcane bagasse etc.) and energy crops(switch grass, poplar, Miscanthus etc.). During biochemical conversion process, only Cellulose and hemicellulosic fractions are converted into fermentable sugar, but lignin which represents the third largest fraction of lignocellulosic biomass is not convertible into fermentable sugars. It is therefore extremely important to recover and convert biomass-derived Lignin into high-value products to maintain economic competitiveness of cellulosic ethanol processes. It was introduced that lignin types and characteristics were different from various isolation methods and biomass sources. Also utilization and potentiality for market of those were discussed.
Green alga, Ulva pertusa kjelmann has been known to be one of the largest pollutants in Korea. Therefore, the efficient pretreatment processes have been required to improve the yields of fermentable sugar. The optimal pretreatment conditions were determined to be $195^{\circ}C$ for 15 min. The sugar yield of glucose and xylose were estimated as 20.5%, and 5.0% respectively, based on theoretical yields. However solid residues were estimated enzymatic digestibility of 90-95% with cellulase loading of 15 FPU/g glucan. This process was proved to generate the low concentration of Hydroxy-Methyl-Furfural (51 ppm), which resulted in ethanol production with 95% of the maximum conversion yield from glucose in the culture of Saccharomyces cerevisiae (ATCC, 24858). This study showed that Ulva pertusa kjellmann can be used as a bioetahnol resource using the high temperature liquefaction process.
Brettanomyces custersii CBS 5512 which has reported as a thermotolerant glucose-cellobiose co-fermentable yeast strain was mutated with UV and NTG to improve ethanol yield at higher than 4$0^{\circ}C$ B. custersii H1-23, H1-39, H1-55 and H1062 were finally selected for hyper-fermentable strains at higher than 4$0^{\circ}C$ from thermotolerant 7510 colonies through 5th selection. Among the selected strains, H1-39 mutant had better fermentability at 4$0^{\circ}C$ and 43$^{\circ}C$ from different concentrations of glucose. H1-39 and H1-23 mutants yielded more than 70% of the theoretical ethanol yield in 4 and 8% mixed sugars at above 4$0^{\circ}C$, which was 5-11% higher than those by original strain. Especially, H1-39 mutant had better fermentability in 4% mixed sugar. It showed 78.5% of the theoretical yield at 4$0^{\circ}C$ and 72.2% of the theoretical yield at 43$^{\circ}C$. On the other hand, theoretical yield of ethanol by H1-39 mutant in 8% mixed sugar at 4$0^{\circ}C$ and 43$^{\circ}C$ were 75.2% and 70.2%, respectively. Theses values increased up to 7-11% as compared to those by orginal strain. By the simultaneous saccharification and fermentation, ethanol production by H1-39 mutant increased up to more than 23% as compared to that by original strain.
A novel brown rot fungus Phaeolus schweinitzii IUM 5048 was firstly used for ethanol production. It was found that this fungus produced ethanol with various sugars, such as glucose, mannose, galactose and cellobiose at 0.28, 0.22, 0.06, and 0.22 g of ethanol per g of sugar consumed, respectively. This fungus showed relatively good ethanol production from xylose at 0.23 g of ethanol per g of sugar consumed. However, the ethanol conversion rate of arabinose was relatively low (at 0.08 g of ethanol per g sugar). P. schweinitzii was capable of producing ethanol directly from rice straw and corn stalks at 0.11 g and 0.13 g of ethanol per g of substrates, respectively, when the fungus was cultured in a basal medium supplemented with 20 g/L rice straw or corn stalks. These results suggest that P. schweinitzii can hydrolyze cellulose or hemicellulose to fermentable sugars and convert them to ethanol simultaneously under oxygen limited condition.
Sugar beet pulp is an abundant industrial waste material that holds a great potential for bioethanol production owing to its high content of cellulose, hemicelluloses, and pectin. Its structural and chemical robustness limits the yield of fermentable sugars obtained by hydrolyzation and represents the main bottleneck for bioethanol production. Physical (ultrasound and thermal) pretreatment methods were tested and combined with enzymatic hydrolysis by cellulase and pectinase to evaluate the most efficient strategy. The optimized hydrolysis process was combined with a fermentation step using a Saccharomyces cerevisiae strain for ethanol production in a single-tank bioreactor. Optimal sugar beet pulp conversion was achieved at a concentration of 60 g/l (39% of dry weight) and a bioreactor stirrer speed of 960 rpm. The maximum ethanol yield was 0.1 g ethanol/g of dry weight (0.25 g ethanol/g total sugar content), the efficiency of ethanol production was 49%, and the productivity of the bioprocess was 0.29 $g/l{\cdot}h$, respectively.
