Feeding Value of Urea Treated Wheat Straw Ensiled with or without Acidified Molasses in Nili-Ravi Buffaloes

  • Thirty early lactating Nili-Ravi buffaloes, six animals in each group, were used in a completely randomized design to examine the feeding value of 4% urea treated wheat straw (UTWS) ensiled with 6% or without acidified molasses. Five experimental diets were formulated. The control ration was balanced to contain 30% DM from UTWS ensiled without acidified molasses. The other four diets were formulated to have 30, 40, 50 and 60% DM from UTWS ensiled with 6% acidified molasses, respectively. Dry matter and neutral detergent fiber (NDF) intakes were higher in buffaloes fed diets containing UTWS ensiled with acidified molasses compared with those fed a diet containing UTWS ensiled without acidified molasses. Intake of DM was not significantly different in buffaloes fed diets containing varying levels of UTWS ensiled with acidified molasses. A similar trend was observed for crude protein (CP) intake. Apparent DM and NDF digestibilities were significantly higher in buffaloes fed diets containing UTWS ensiled with acidified molasses compared with those fed UTWS ensiled without acidified molasses. However, differences in DM and NDF digestibilities were non-significant across buffaloes fed diets containing varying levels of UTWS ensiled with acidified molasses. Milk yield (4% fat corrected) was significantly higher in buffaloes fed diets containing UTWS ensiled with acidified molasses than those fed a diet containing UTWS ensiled without acidified molasses. Milk yield was similar in buffaloes fed varying level of UTWS ensiled with acidified molasses. Milk CP, true protein, solid-not-fat and total solids were similar in buffaloes fed UTWS ensiled with or without acidified molasses. The UTWS ensiled with 6% acidified molasses can be included at up to 60% DM of lactating buffalo rations without any ill effect on productivity.

Influence of Fiber Content and Concentrate Level on Chewing Activity, Ruminal Digestion, Digesta Passage Rate and Nutrient Digestibility in Dairy Cows in Late Lactation

  • The influence of fiber content of hay (low-fiber 47% NDF and high-fiber 62% NDF of DM) and concentrate level (high 50% and low 20% of ration DM) on chewing activity, passage rate and nutrient digestibility were tested on four restrict-fed (11.1 to 13.7 kg DM/d) Holstein cows in late lactation. Aspects of ruminal fermentation and digesta particle size distribution were also investigated on two ruminally cannulated (100 mm i.d.) cows of the same group of animals. All digestion parameters studied were more affected by the fiber content of the hay and its ratio to non structural carbohydrates than by the concentrate level. Giving a diet of high-fiber (62% NDF) hay and low concentrate level (20%) increased chewing activity but decreased solid passage rate and total digestibility of nutrients due to a limited availability of fermentable OM in the late cut fiber rich hay. A supplementation of high-fiber hay with 50% concentrate in the diet seems to improve the ruminal digestion of cell contents, whilst a depression of the ruminal fiber digestibility was not completely avoided. Giving a diet of low-fiber (47% NDF) hay and high concentrate level (50%) reduced markedly the chewing and rumination activity, affected negatively the rumen conditions and, consequently, the ruminal digestion of fiber. A reduction of the concentrate level from 50 to 20% in the diet of low-fiber hay improved the rumen conditions as reflected by an increase of the ruminal solid passage rate and of fiber digestibility and in a decrease of the concentration of large particles and of the mean particle size of the rumen digesta and of the faeces. Generally, it can be summarised that, (i) concentrate supplementation is not a strategy to overcome limitations of low quality (fiber-rich) hay, and (ii) increase of the roughage quality is an effective strategy in ruminant nutrition, especially when concentrate availability for ruminants is limited.

Effect of Additives on the Fermentation Quality and Residual Mono- and Disaccharides Compositions of Forage Oats (Avena sativa L.) and Italian Ryegrass (Lolium multiflorum Lam.) Silages

  • This study aimed to evaluate the effects of silage additives on the fermentation qualities and residual mono- and disaccharides composition of silages. Forage Oats (Avena sativa L.) and Italian Ryegrass (Lolium multiflorum Lam.) were ensiled with glucose, sorbic acid and pre-fermented juice of epiphytic lactic acid bacteria (FJLB) treatments for 30 days. In both species grass silages, although the respective controls had higher contents of butyric acid (20.86, 33.45g $kg^{-1}$ DM) and ammonia-N/total nitrogen (100.07, 114.91 g $kg^{-1}$) as compared with other treated silages in forage oats and Italian ryegrass, the fermentation was clearly dominated by lactic acid bacteria. This was well indicated by the low pH value (4.27, 4.38), and high lactic acid/acetic acid (6.53, 5.58) and lactic acid content (61.67, 46.85 g $kg^{-1}$ DM). Glucose addition increased significantly (p<0.05) lactic acid/acetic acid, and significantly (p<0.05) decreased the values of pH and ammonia-N/total nitrogen, and the contents of butyric acid and volatile fatty acids as compared with control, however, there was a slightly but significantly (p<0.05) higher butyric acid and lower residual mono- and di-saccharides as compared with sorbic acid and FJLB additions. Sorbic acid addition showed the lowest ethanol, acetic acid and ammonia-N/total nitrogen, and highest contents of residual fructose, total mono- and di-saccharides and dry matter as well as high lactic acid/acetic acid and lactic acid content. FJLB addition had the lowest pH value and the highest lactic acid content, the most intensive lactic acid fermentation occurring in FJLB treated silages. This resulted in the faster accumulation of lactic acid and faster pH reduction. Sorbic acid and FJLB additions depressed clostridia or other undesirable bacterial fermentation, thus this decreased the water-soluble carbohydrates loss and saved the fermentable substrate for lactic acid fermentation.

Urea Treated Corncobs Ensiled with or without Additives for Buffaloes: Ruminal Characteristics, Digestibility and Nitrogen Metabolism

  • Influences of urea treated corncobs (UTC) ensiled with or without different additives on ruminal characteristics, in situ digestion kinetics, nutrient digestibility and nitrogen metabolism were examined in a $5{\times}5$ Latin square design using five ruminally cannulated buffalo bulls. Five iso-caloric and iso-nitrogenous diets were formulated to contain 30% dry matter (DM) from concentrate and 70% DM from 5% UTC ensiled without any additive (U) or with 5% enzose (EN), 5% acidified molasses (AM), 5% non-acidified molasses (NM) and 5% acidified water (AW), respectively. These diets were fed to buffalo bulls at 1.5% of their body weight daily. Ruminal $NH_3$-N concentration at 3 hours (h) post feeding was significantly higher in bulls fed U, NM and AW diets, however, at 6, 9 and 12 h post feeding it was significantly higher in bulls fed EN and AM diets. Ruminal total volatile fatty acids (VFA) and acetate concentrations were significantly higher with EM and AM diets compared with other diets at 3, 6, 9 and 12 h post feeding. Ruminal pH at 6 and 9 h post feeding was higher with EN and AM diets; however; it was notably lower with these diets at 3 h post feeding. Total ruminal bacterial and cellulolytic bacterial counts were higher in bulls fed EN and AM diets than in those fed the other diets. In situ ruminal DM and NDF degradabilities and total tract digestibilities were significantly higher with UTC ensiled with enzose and acidified molasses than those ensiled without any additive or other additives. Nitrogen balance was significantly higher in bulls fed EN and AM diets than those fed U, AW and NM diets. The UTC ensiled with enzose or acidified molasses resulted in better digestibility and N utilization than those ensiled without any additive, with non-acidified molasses and acidified water in buffaloes.

효소당화를 위한 목질계 바이오매스의 유기용매 침출 전처리 공정 (Enhancement of Enzymatic Hydrolysis of Lignocellulosic Biomass by Organosolv Pretreatment with Dilute Acid Solution)

  • 오르가노솔브를 이용한 전처리는 목질계 바이오매스의 주요성분을 분별하고 전처리 바이오매스의 효소당화를 효과적으로 유도할 수 있는 공정이다. 이에 사용되는 오르가노솔브는 증류와 재순환공정으로 쉽게 재사용이 가능하고 고형성분에서 리그닌을 화학적으로 분별시킬 수 있다는 장점이 있다. 경제적인 이유로서 낮은 분자량의 알코올(에탄올, 메탄올 등)이 사용되어 왔으며, 무기산(염산, 황산, 인산 등)이 촉매로 첨가되었을 경우 탈리그닌 효과 및 자일로스 성분의 수율을 증대시킬 수 있다. 본 연구에서는 오르가노솔브로는 에탄올을 사용하였고 소량의 황산 촉매를 첨가해 전처리를 수행하였다. 바이오매스는 침출식반응기에서 40~50 wt%의 에탄올을 이용해 20~60분 동안 $170{\sim}180^{\circ}C$에서 전처리가 되었다. 전처리를 마친 바이오매스에 대해서는 전처리 효과를 알아보기 위하여 72시간 동안 효소당화를 수행하였다. 그 결과 $180^{\circ}C$에서 50 wt% 에탄올을 이용한 리기다의 2단 전처리에서 당화율은 40.6%로 나타났지만 같은 조건에서의 1단 전처리에서는 55.4%로 가장 높게 나타나는 것을 보았을 때, 용매와 촉매를 균일하게 섞는 것이 전처리에 효과적이라는 것을 알 수 있었다.

순환식 암모니아 반응기(Ammonia Circulation Reactor (ACR))를 이용한 옥수수대의 전처리 및 효소 당화율 향상 (Pretreatment of Corn Stover for Improved Enzymatic Saccharification using Ammonia Circulation Reactor (ACR))

  • 목질계 바이오매스인 옥수수대 전처리를 위하여 고안된 순환식 암모니아 전처리 반응기(Ammonia Circulation Reactor (ACR))를 이용하여 연구하였다. 이 전처리 방법은 적은 양의 액체를 사용하도록 고안되었으며 이 연구에선 기존의 전처리 공정보다 낮은 전처리 온도($60{\sim}80^{\circ}C$), 반응시간(4~12 hour) 그리고 고체:액체 비율(1:3~1:8) 등의 공정 조건을 실험 하여 효과를 비교하였다. 즉 여러 공정 조건에서 전처리 후 고형물의 잔류 고체량, 당, Lignin 함량, 그리고 효소당 화율 등을 측정하였다. 여러 실험 조건에서 공통적으로 관찰된 것은 전처리 조건이 더 가혹해 지면 Lignin의 제거율이 가장 큰 영향을 받았으며, 47.6~70.6% 범위로 나타났다. 반면 다른 당(Glucan, Xylan)은 손실이 비교적 작게 나타났다. 모든 실험 조건에서, 전처리된 고형물의 Glucan 손실율은 4.7~15.2% 범위로 변화가 크지 않았으며 Xylan 손실율은 여러 조건의 변화에 따라 7.4~25.8% 정도 범위로 나타났다. 암모니아 순환 전처리로 8~12 hour 동안 처리된 옥수수대는 90.1~94.5%의 높은 72-h Glucan 당화율을 (15 FPU-GC220+30 CBU)/g-glucan의 효소 투입으로 나타냈으며 순수 Cellulose인 Avicel의 당화율(92.7%)과 비슷하거나 높았다. 또한 8~12 hour 처리된 옥수수대의 초기 24-h Glucan 당화속도는 73.0~79.4%로 Avicel의 같은 시간 당화율인 69.5% 보다 높게 나타났다. 반응시간을 증가는 보다 많은 Lignin을 제거하였으며 따라서 효소 당화율 증가에 기인한 것으로 보인다.
