Antiviral Effects of Fermented Lonicerae Flos on A Type Influenza Virus (발효 금은화의 인플루엔자 바이러스 A형에 대한 저해효과)

  • Objective : Lonicerae Flos has detoxifying properties and been used as antipyretic, antibacterial and antitumor. Fermentation of herbal medicine is known to increase the absorption, enhance effectiveness, decrease herbal toxicity and reduce side-effects. This study was performed to measure the effects of fermented Lonicerae Flos on influenza A/WSN (H1N1) virus replication. Material and Methods : Lonicerae Flos was fermented by Lactobacillus casei PM1. Fermented Lonicerae Flos was treated for 12 hours to MDCK (Mardin Darby canine kidney) cells, then cell-virulence was observed by MTT assay for 12 hours, 24 hours, and 36 hours after treatment. Following cases were conducted for 0, 10, 100, and $1000{\mu}g/ml$ concentrations of fermented Lonicerae Flos under the same time-frame; the fermented Lonicerae Flos was treated to MDCK cells before and after contamination by A-type influenza virus. The fermented Lonicerae Flos and the virus were mixed directly. The influence was observed by MTT assay and plaque assay. Results : These findings suggest that the fermented Lonicerae Flos inhibited the virulence of influenza A virus in MDCK cells and suppressed the plaque forming colonies induced by influenza A virus. Furthermore, pretreatment with fermented Lonicerae Flos was more effective than post-treatment. The titer of influenza virus was reduced for all before and after influenza A virus inoculation.

Nodakenin and Decursin Contents of Fermented Angelicae Gigantis Radix by 4 Species Strain (4종의 발효균주가 당귀의 nodakenin과 decursin의 함량에 미치는 변화)

  • Objectives : The purpose of this study was to investigate the nodakenin and decursin contents in each fermented Angelicae gigantis Radix by 4 species of ferment strains. Methods : The strains of fermented Angelicae gigantis Radix were Lactobacillus plantarum(SE1), L. acidophilus(AC), Bacillus subtilis(B2) and B. licheniformis(BL2). The fermentative changes of nodakenin and decursin were analyzed using HPLC. Results : All of 4 species strains reduced nodakenin and decursin concentration in Angelicae gigantis Radix by fermentation. But fermentability were difference between strains. The nodakenin concentration of AC was lower than B2. The decursin concentrations of SE1 and AC were lower than B2 and BL2. Conclusions : 4 species of fermental strain were difference decomposable rate with nodakenin and decursin in fermented Angelicae gigantis Radix.

Characterization of degradation of fish wastes using mixed microorganisms (복합미생물을 이용한 수산폐기물의 분해특성)

  • Fifteen species of microorganisms were isolate form the intestines of fishes, fish feed, and ferment. Eleven microorganisms except HY4, HY8, HY12, and HY13 were Gram-positive, and HY1, HY2, HY3, HY5, HY6, and HY7 produced lactic acid. The species of HY1, HY2, HY3, HY4, HY5, HY6, HY13, and HY14 showed some growth in the medium containing 1% of NaCl. Except HY6, HY7, HY8, HY12 and HY5, 10 isolates had proteolytic activity, whereas only HY13 and HY14 had lipase activity. From all the results four isolates (HY3, HY4, HY13 and HY14) were chosen for the degradation of fish wastes. There was no mutual inhibition among the microorganisms, and the optimum temperature and pH for the growth of the mixed culture were found to be 3 2$\^{C}$ and 7, respectively. Under the optimum growth conditions the maximum optical density and the maximum specific growth rate were estimated to be 2.35 and {TEX}$0.46h^{-1}${/TEX}, respectively. Major microorganisms in the mixed culture at the log-phase were HY3 and HY4, which occupied 70%. The degrading efficiency of fish waste by the mixed microorganisms was 2.3 times higher, compared to control. The total amount of free amino acids in the degraded products from fish wastes was 39g/100g protein and little odor was produced by the mixed microorganisms after 48 hours.

  • A promising application of Lactococcus lactis is its use as live vehicles for production and delivery of heterologous proteins of vaccines and therapeutic substances. Because L. lactis has GRAS ('generally regarded as safe') status, we tested whether L. lactis could function as the carrier of the Ll protein of human papillomavirus (HPV) type 16. The RNA level expression of Ll gene was detected in L. Lactis. The Ll protein was expressed in L. lactis with Ll gene. The growth of strains L. lactis with an empty plasmid (pAMJ328) and L. lactis with Ll-encoding plasmid (pAMJ328-Ll) was slightly decreased in comparison with the growth of strains L. lactis (wild type). However, all the three strains of L. lactis maintained the ability to ferment sugars primarily into lactic acid, indicating that Ll protein did not affect the biochemical property of L. lactis. These results suggest that L. lactis, capable of carrying Ll protein, might be further developed as a biocompatible oral protein delivery system.

Characterization of airag collected in Ulaanbaatar, Mongolia with emphasis on isolated lactic acid bacteria

  • Background: Airag, alcoholic sour-tasting beverage, has been traditionally prepared by Mongolian nomads who naturally ferment fresh mares' milk. Biochemical and microbiological compositions of airag samples collected in Ulaanbaatar, Mongolia and physiological characteristics of isolated lactic acid bacteria were investigated. Methods: Protein composition and biochemical composition were determined using sodium dodecyl sulfate-gel electrophoresis and high performance liquid chromatography, respectively. Lactic acid bacteria were identified based on nucleotide sequence of 16S rRNA gene. Carbohydrate fermentation, acid survival, bile resistance and acid production in skim milk culture were determined. Results: Equine whey proteins were present in airag samples more than caseins. The airag samples contained 0.10-3.36 % lactose, 1.44-2.33 % ethyl alcohol, 1.08-1.62 % lactic acid and 0.12-0.22 % acetic acid. Lactobacillus (L.) helveticus were major lactic acid bacteria consisting of 9 isolates among total 18 isolates of lactic acid bacteria. L. helveticus survived strongly in PBS, pH 3.0 but did not grow in MRS broth containing 0.1 % oxgall. A couple of L. helveticus isolates lowered pH of skim milk culture to less than 4.0 and produced acid up to more than 1.0 %. Conclusion: Highly variable biochemical compositions of the airag samples indicated inconsistent quality due to natural fermentation. Airag with low lactose content should be favorable for nutrition, considering that mares' milk with high lactose content has strong laxative effect. The isolates of L. helveticus which produced acid actively in skim milk culture might have a major role in production of airag.

Production of Ethanol from D-Xylose by Fusarium sp. (Fusarium sp.에 의한 D-Xylose로부터 Ethanol 생산)

  • Microorganisms capable of utilizing D-xylose as a sole carbon and energy source were isolated to ferment D-xylose directly to ethanol. Among them, the strain, which showed the best ability to pro-duce ethanol, was selected and was identified as Fusarium sp. The optimal conditions for the pro-duction of ethanol were 8.0 of initial pH, 33$^{\circ}C$ of temperature, and 2% of substrate concentration. Under this optimal condition, the following results were obtained : maximum ethanol concentration, 7.0g/$\ell$; ethanol yield, 0.35g of ethanol per g of D-xylose (68.6% of theoretical); biomass yield, 0.27g of dry biomass per g of D-xylose.

  • The purpose of this study was to investigate the method of making whole Chinese cabbage Kimchi, the family's taste in Kimchi and the reason for disliking the taste of Kimchi in the Taegu area. The results were as follows; 1) Most housewives learned the method of making the Kimchi from their mothers. Method was as follows; First, scatter salt on the whole Chinese cabbage and soak that for 3∼6 hours. After, wash the whole Chinese cabbage 3 times. Then, add red pepper powder, garlic, ginger and salted fermented anchovy as basic seasoning. Be sure to, mix the red pepper powder and salted fermented anchovy until they taste hot and salty. Store the whole Chinese cabbage until they ferment. Afterwards store the whole Chinese cabbage in the refrigerator. 2) It was the whole Chinese cabbage Kimchi that most family members preferred. 3) Among the family members, the old age group preferred newly made Kimchi because of the cool and fresh taste. The adult group preferred fresh taste, also smell and peculiar taste of the seasoning were important. The adolescent group preferred fresh taste and sour taste. 4) It shows that most of the family likes Kimchi, and among the family members, about 25% of the sons and daughters dislike Kimchi. And it shows that all family members dislike salty taste. 5) The older housewives’ Kimchi was preferred over the younger housewives’ Kimchi.

Biochemical Characteristics of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus and Their Growth on Chinese-Style Beaker Sausage

  • This study was conducted to investigate protein and carbohydrate utilization of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus. Sensitivity to pH, sodium chloride, potassium sorbate and sodium nitrite of these strains was also determined. In Chinese-style beaker sausage manufacturing, the growth rate of these strains during the curing period ($20^{\circ}C$ and $30^{\circ}C$) was evaluated. The results indicated that no strains could hydrolyze azo-casein and sarcoplasmic protein and only S. xylosus could hydrolyze gelatin at $30^{\circ}C$. All of these strains could oxidize and ferment fructose and mannitol. S. carnosus and S. xylosus could slightly oxidize lactose and utilize citrate. Arabinose was oxidized by S. xylosus and sorbitol was oxidized by S. carnosus. Growth of M. varians was restricted at pH 5.0 and S. carnosus and S. xylosus were restricted at pH 4.5. S. xylosus and S. carnosus were able to grow with 0.1~0.5% potassium sorbate, 50~200 ppm sodium nitrite or 1~15% sodium chloride. S. xylosus had a higher growth rate than the other strains. Staphylococcus species grew well during curing period of Chinese-style beaker sausage then followed by Micrococcaceae.

Fermentation of Pentose and Hexose Derived from Cellulosic Food Wastes by Mixed Yeast (공기 주입 방법에 따른 셀룰로오스계 음식물류 폐기물 유래의 오탄당과 육탄당의 동시발효)

  • It is indispensable to increase the conversion rate of a reducing sugars such as pentose and hexose derived from cellulosic wastes for a highly efficient bioethanol fermentation from food wastes. The saccharification liquid from cellulosic substrates such as vegetable food wastes contained lots of hexose like glucose and pentose like xylose. Since Saccharomyces-based yeasts could not convert xylose to bioethanol, Pichia stipitis which could directly ferment xylose to ethanol was chosen. After selecting Saccharomyces coreanus and P. stipitis, fermentation characteristics by mixture of two yeasts were investigated. As a result, it was verified the production of ethanol was enhanced by the co-fermentation, although there were somewhat differences between the fermentation characteristics by the aeration methods. Moreover, the consumption of pentose, hexose and disaccharide was obviously observed, and aeration in the process of fermentation seemed to stimulate the activity of P. stipitis.

A Case of Flavobacterium indologenes Meningitis in Neonate (신생아의 Flavobacterium indologenes 뇌막염 1례)

  • Flavobacterium species are nonfastidious, oxidase-positive gram-negative rods that do not ferment glucose. These organism are widely distributed in nature and in hospital environments. In past, one of flavobacteria, Flavobacterium indologenes, was treated with non-pathogenic organism. Recently, several investigators have demonstrated the infection of this organism in human. Nowadays, the growth of F. indologenes in specimen should be considered a potential pathogen in infectious patients including neonate, especially in the setting of malignancy and with use of invasive procedures. The resistance of this organism to multiple antibiotics and the high incidence of polymicrobial bacteremia make it difficult to determine optimal therapeutic options. We experienced a case of neonatal bacterial meningitis causing by F. indologenes. So we report this case to evoke more concerns about the infections of this organism in human.
